Description
Analogifremgangsmåte for fremstilling av
terapeutisk virksomme purinforbindelser.
Foreliggende oppfinnelse angår analogifremgangsmåter for fremstilling av en terapeutisk virksom purinforbindelse med den generelle formel:
samt farmasøytisk akseptable salter derav. Disse forbindelser karw brukes for å undertrykke immunitetsreaksjoner og som antivirale midler.
Salter som er spesielt hensiktsmessige for terapeutisk an-vendelse er salter av farmasøytisk akseptable karboksylsyrer såsom melkesyre, eddiksyre eller maleinsyre, såvel som salter av farmasøyt-isk akseptable mineralsyrer såsom saltsyre eller svovelsyre. Aktivi-teten i eventuelle tilførte salter ligger i basen. I tillegg til det som er nevnt ovenfor kan toksiske salter fremstilles og omdannes enten til basen eller farmasøytisk akseptable salter ved standard fremgangsmåter, f.eks. ved en metatetisk reaksjon.
Forbindelsen med formel I er spesielt brukbar ved behandling av virusinfeksjoner som er et resultat av DNA-virus, såsom kopper og helvetesild. Den kan dessuten brukes for å undertrykke immunitetsreaksjoner hos dyr eller mennesker ved transplantasjoner av fremmede celler i legemet, og ved behandling av autoimmune lidelser hos pattedyr, såsom Lupus erythematosus, Haemolytic anaemia, Ulcerative colitis og nephrosis.
Slik det brukes her betyr en effektiv doseringsenhet en for-utbestemt antiviral mengde som er tilstrekkelig tii å være effektiv overfor organismen in vivo. Farmasøytisk akseptable bærere er forbindelser som er anbefalt for administrasjon av forskjellige typer medikamenter, såsom flytende, faste eller gassformede materialer, og som ellers er inerte eller medisinisk akseptable og forenlige med de aktive bestanddeler.
Farmasøytiske preparater inneholdende den fremstilte forbindelse kan administreres parenteralt, oralt, som et suppositorium, påført som en oppløsning eller påført topikalt som en salve, en krem, et pulver, etc, alt avhengig av om preparatet skal brukes, for en ytre eller indre virusinfeksjon, eller skal brukes som et undertrykkende middel for immunitets- eller autoimmunitets-reaksjoner.
For indre virusinfeksjoner blir preparatene vanligvis an-vendt oralt eller parenteralt i doser beregnet ut fra basen, varierende fra 5 til 250 mg/kg, fortrinnsvis fra 20 til 100 mg/kg kroppsvekt, og anvendes fortrinnsvis hos mennesker i enhetsdoseringsform tilført et par ganger daglig i mengder fra 10 til 250 mg/enhetsdose.
Som undertrykkende midler for immunitetsreaksjoner anvendes preparatene internt et par ganger i døgnet i dosér fra 3 til 10 mg/kg kroppsvekt.
Por oral administrasjon kan man anvende fine pulvere eller granulater inneholdende fortynnede, dispergerende og/eller overflate-aktive midler, og kan være tilstede i drikker, i vann eller i siruper, i kapsler eller i ikke-vandige suspensjoner, hvor suspenderende midler kan være tilsatt, i tabletter som kan omfatte bindemidler og.smøre-midler, eller i suspensjoner i vann eller i en sirup. Når det er ønskelig eller nødvendig.kan man anvende smakstilsetninger, konserver-ende, suspenderende og fortykkende midler. Tabletter og granulater er foretrukket og disse kan være belagt.
Por parenteral administrasjon kan forbindelsene være tilstede i vandige injeksjonsoppløsninger i konsentrasjoner fra 0.1 til 10 % t fortrinnsvis 1 og nevnte oppløsninger kan inneholde antioksyderende midler, buffere, etc.
Por virusinfeksjoner i øyet eller andre ytre vev blir preparatene fortrinnsvis påført .den infiserte del av legemet , som en to-pikal salve, f.eks. i doser fra ca. halvparten av det som anvendes for indre bruk, dvs. opp til 125 mg/kg eller 5 til 125 mg per enhets-dose.
Den fremstilte forbindelse 2,6-diamino-9-(3-D-arabinofurano-syDpurin har ekstremt høy antiviral aktivitet, f.eks. som middel mot helvetesild-virus. Man har også kunnet påvise en vesentlig og relativt uventet høy aktivitet overfor koppevirus, og man har funnet at forbindelsen er meget brukbar for å undertrykke immunitetsreaksjoner overfor transplanterte celler.
Forbindelsen med formel I fremstilles ifølge oppfinnelsen
ved å redusere en forbindelse med formel II
hvor Z er en blokkerende gruppe slik som en alkylgruppe, fortrinnsvis' en p-fenylbenzylgruppe, eller en a- eller 3-menaftylgruppe, mest foretrukket en benzylgruppe. Z-gruppen må være slik at den oppfyller to betingelser, nemlig: (a) at når den er knyttet til oksygenatomet i posisjon to i sukkerdelen, må den ikke innvirke på;reaksjoner ved C(l) i sukkeret, og (b) den må la seg fjerne når dette er ønskelige, under relativt milde -betingelser.
En eller to av de tre mulige Z-grupper, f.eks. i tredje og fjerde stilling, kan imidlertid være et hydrogenatom.
Reduksjonen kan utføres katalytisk, f.eks. ved å bruke hydrogen og en hydrogeneringskatalysator, slik som palladium, palla-disert trekull, platinasort, Raney-nikkel, etc. Noen av disse er svakere katalysatorer, og det kan være nødvendig å anvende trykk eller lange hydrogeneringsperioder for å oppnå full effekt. Anvendelsen av platinaoksyd eller platinasort er mest hensiktsmessig i så henseende. Alternativt kan forbindelser med formel II omsettes med et reduksjons-middel, spesielt av den type som frembringer hydrogenatomer in situ, slik som alkalimetaller, fortrinnsvis natrium, i flytende ammoniakk.
Forbindelser med formel II kan fremstilles ved en av de følgende fremgangsmåter:
A. En forbindelse med formel III:

hvor X og X hver er et halogenatom, fortrinnsvis klor eller brom, og Z er en blokkerende gruppe slik denne er definert ovenfor, fortrinnsvis en benzylgruppe, kan omsettes med et alkalimetallazid, fortrinnsvis natriumazid. Det resulterende 2,6-diazidderivat kan så katalytisk hydrogeneres til 2,6-diamidet med formel II med en ytter-ligere dannelse av en mindre mengde av den tilsvarende forbindelse med formel I.
<y>tterligere reduksjon av forbindelsen med formel II under de betingelser som er beskrevet ovenfor, dvs. en katalytisk hydr.o-genering, eller fortrinnsvis ved å anvende et middel som'natrium i flytende ammoniakk, gir så det ønskede sluttprodukt med formel I. I praksis kan det være fordelaktig å forené de to trinn til ett trinn,
ved å anvende en av de mer effektive sterke■katalysatorer slik som platinaoksyd eller platinasort, fortrinnsvis.under trykk eller med noe forlenget hydrogeneringstid, hvorved man får fremstilt mellomproduktet med formel II som så umiddelbart reduseres til en forbindelse med formel I.
B. En forbindelse med formel IV:

hvor Q og Q hver er en YCONH-gruppe, hvor Y er en alkylgruppe med fra 1 til 6 karbonatomer fortrinnsvis en metylgruppe, og Z er en blokkerende gruppe som definert ovenfor, fortrinnsvis en benzylgruppe, kan deacyleres ved hydrolyse under basiske betingelser. Dette kan f.eks. utføres ved å oppløse produktet i en lavere alkohol slik som metanol, og så tilsette, ét alkalimetallhydroksyd slik som natrium-hydroksyd.
For å få fremstilt en forbindelse med formel III, kan et 2,6-dihalogenpurin, fortrinnsvis 2,6-diklorpurin, kondenseres med et arabinosesukkerhalogenid som har en eller flere av sine hydroksyl-grupper blokkert med fjernbare blokkerende grupper Z slik disse er definert ovenfor, ved hjelp av den fremgangsmåte som er beskrevet av Keller et al i J. Org. Chem. (1967), 32, 1644. Det foretrukne sukker for dette formål er 2,3,5-tri-O-benzyl-ct-D-arabinofuranosylklorid. Acylering av 2,6-diaminopurin, fortrinnsvis med eddiksyreanhydrid, fulgt av en analog sukkerkondensasjqn, gir forbindelsen med formel IV.
"Forbindelser med formel IV slik disse er definert ovenfor er nye og verdifulle mellomprodukter for fremstilling av ovennevnte sluttprodukt.
Forbindelsen med formel I kan også fremstilles fra anomer-iske xylosider via lyxosider slik som beskrevet av Reist et al., Ibid og Biochemistry, 1964, 3> 15-18.
Ifølge oppfinnelsen blir således et (2,3-anhydro-3-D-lyxo-furanosyl)pyrinderivat med formelen:
omsatt med en base slik som natriumacetat i et inert oppløsningsmiddel slik som vandig dimetylformamid hvorved det skjer en ringåpning. Lyxo-furanosylderivåtet kan på sin side fremstilles ved en epoksyddannelse av det tilsvarende 9"(2-0-metansulfonyl-3-D-xylofuranosyl) med en egnet alkoholisk alkoksydoppløsning, såsom metanolisk natriummetoksyd. Ut-gangsforbindelsenj nemlig xylofuranosylforbindelsen kan fremstilles på den følgende måte.
Nevnte 9-(B-D-xylofuranosyl)purin, med•substituenten i 2-og 6-stillingene, omsettes med aceton inneholdende etansulfonsyre, noe som gir det tilsvarende 9-(3,5-0-isopropyliden-3-D-xylofuranosyl)-derivat.' Dette blir så omsatt med metansulfonylklorid til 2-metan-sulfonylforbindelsen som så hydrolyseres, f.eks. i vandig eddiksyre, for derved å få fjernet den.beskyttende isopropylidengruppe. Det fremstilte 9-(2-0-metansulfonyl-g-D-xylofuranosyl)-derivat kan så omsettes videre slik det er beskrevet ovenfor.
Førstnevnte xylofuranosylpurin kan fremstilles ved å konden-sere 1,2,3,5-tetra-O-acetyl-D-xylofuranose med et passende purin som har sin frie aminogruppe beskyttet ved acylering, fortrinnsvis ace-tylering.
Den ifølge oppfinnelsen fremstilte forbindelse anvendes hensiktsmessig for behandling,av infeksjoner som er forårsaket av DNA-virus i pattedyr (f.eks. mus, rotter, hunder, mennesker, etc.) ved'at man tilfører, en effektiv, ikke-toksisk, antiviral mengde av forbindelsen med formel I slik denne er definert ovenfor, til det infiserte dyr eller den infiserte pasient, eller til et dyr eller en pasient som har vært eksponert overfor en infeksjon.
Følgende eksempler illustrerer oppfinnelsen.
Eksempel I
a) 2, 6- diazido- 9-( 2, 3, 5- tri- 0- benzyl- 3- D- arabinofuranosyl) purin
En oppløsning av 2.1 g 2,6-diklor-9-(2,3,5-tri-0-benzy1-3-D-arabinofuranosyl)purin (III) i 8 ml metanol og 2 ml aceton ble oppvarmet sammen med 440 mg (2 molekvivalenter) natriumazid i 6 timer ved koking under tilbakeløp. Det utfelte natriumklorid ble frafilt-rertj og det gule filtrat som inneholdt'2,6-diazido-9~(2,3,5_tri-0-benzyl-g-D-arabinofuranosyl)purin viste absorbsjonsmaksima ved 245,
270 (skulder); 300 my i 95 % etanol. Filtratet ble brukt direkte for reduksjon -i neste trinn.
b) 2, 6- diamino- 9-( 2, 3»5~ tri- 0- benzy1- 3- D- arabinofuranosyl) purin( II)
Reaksjonsproduktet ble redusert med en 5 ? palladiumtrekull-katalysator og hydrogen ved 2 atm. trykk ved 25°C. Etter 4 timer ble katalysatoren fjernet, og filtratet fordampet til tørrhet under redusert trykk. Resten (1.88 g) ble omkrystallisert fra 95 % etanol,
og man fikk 1.0 g blekt gule krystaller, smeltepunkt 158°-159°C, som viste seg å være homogent ved tynnsjiktkromatografi. Analysen til-svarte 2,6-diamino-9~(2,3,5-tri-0-benzyl-6-D-arabinofuranosyl)purin.
c) 2, 6- diamino- 9-( B- D- arabinofuranosyl) purin ( I)
1 g av produktet (II) ble tilsatt 200 ml flytende ammoniakk.
Små mengder av natrium ble tilsatt inntil den blå farge vedvart i
flere minutter. Totalt anvendte man 350 mg natrium. Den blå fargen ble fjernet med et par krystaller ammoniumklorid. Ammoniakken ble fordampet under en strøm av nitrogen, og resten behandlet med 50 ml benzen. Den uoppløselige rest ble oppløst i et par ml vann og nøy-tralisert med eddiksyre. Produktet (500 mg) etter omkrystallisasjon fra vann, smeltet ved 257° - 259°C ved dekomponering. 2,6-diamino-9_

Eksempel II
a) 2, 6- diacetamidopurin
En godt omrørt suspensjon av 10 g vannfri 2,6-diaminopurin
i 150 ml frisk eddiksyreanhydrid ble oppvarmet til 70° - 80°C. Det faste stoff ble isolert ved filtrering under vakuum og vasket med eter. Det faste stoff var en blanding av di- og tri-acetyiert materiale, smeltepunkt 244° - 250°C, dekomponering.
■ Det ovennevnte materiale ble tilsatt 1 liter absolutt metanol inneholdende 100 ml konsentrert ammoniumhydroksyd. Kolben ble holdt ved romtemperatur i 15" minutter med tilfeldig omrøring. De uoppløse-lige faste stoffer ble fjernet ved filtrering under vakuum og tørket ved 100°C.
Utbytte 10.6 g (68 % totalt utbytte), smeltepunkt 296° - 300 C. Strukturen ble bekreftet ved NMR-spektroskopi. •
b) 2, 6- diamin0- 9-( 2, 3» 5~ tri- 0- benzy1- g- D- arabinofuranosy1) purin ( II)
En.suspensjon av 16.0 g 2,6-diacetamidopurin og molekyl-sikter i tørr metylenklorid inneholdende 20.6 g 2,3,5-tri-O-benzyl-B-D-arabinof uranosylklorid ble omrørt i to uker ved romtemperatur. Opp-løsningen ble filtrert gjennom et "Celite"-lag, og oppløsningsmidlet fjernet.i vakuum. Ekstraksjon av råoljen med cykloheksan ga 10.3 g (47 %) av et urent 2,6-diacetamido-9-(2,3,5-tri-O-benzy1-3-D-arabinofuranosyl)purin.
Denne forbindelse ble så deacetylert ved behandling med en etanolisk oppløsning inneholdende: 2 molekvivalenter av en 1 N vandig NaOH eller med et lite overskudd av base. Oppløsningen ble omrørt
ved 70° - 80°C i \ time. Produktet ble ekstrahert fra oppløsningen med kloroform, tørket over magnesiumsulfat, hvorpå kloroformen ble fjernet i vakuum, noe som ga (II) direkte eller ved krystallisasjon.
Når deacetyleringen hadde skjedd ble forbindelsen med formel II utfelt fra oppløsningen og isolert ved filtrering under vakuum.
Omkrystallisasjon fra metanol ga 2,6-diamino-9-(2,3,5-tri-0-benzyl-g<->D-arabinofuranosyl)purin, smeltepunkt l6l° - 162°C. c) Denne forbindelse ble deretter behandlet slik som angitt i eksempel I(c) for dannelse av det ønskede produkt, nemlig 2,6-diamino-9-(3-D-arabinofuranosyl)purin.
Eksempel III
2, 6- diamino- 9-( B- D- arabinofuranosy1) purin ( I)
2,6-diaminopurin ble kondensert med en ekvimolar mengde 1,2,3,5-tetra-0-acetyl-D-xylofuranose-for dannelse av 2,6-diamino-9-(B-D-xylofuranosyl)purin. Denne forbindelse ble deretter omsatt med aceton inneholdende etansulfonsyre slik at man oppnådde 2,6-diamino-9-(3,5-0-isopropyliden-3-D-xylofuranosyl)purin som deretter ble-hydro-lys ert i vandig eddiksyre til 2,6-diamino-9-(2-0-metansulfonyl-g-D-xylofuranosyl)purin. Behandling av denne forbindelsen med natriummetoksyd ga 2,6-diamino-9~(2,3-anhydro-g-D-lyxofuranosyl)purin, som deretter ble omdannet til 2,6-diamino-9~(B-D-arabinofuranosyl)purin ved oppvarming med natriumacetat i vandig dimetylformamid. Reaksjons-blandingen ble deretter inndampet. til tørrhet og resten krystallisert og omkrystallisert fra vann hvilket ga det ønskede produkt, smeltepunkt 256° - 258°C med dekomponering.

- Analogifremgangsmåte for fremstilling av en terapeutisk virksom purinforbindelse med den generelle formel:samt farmasøytisk akseptable salter derav, karakterisert v e d at a) en forbindelse med formelen:hvor Z er en blokkerende gruppe slik som en benzylgruppe, reduseres, eller b) et (2,3-anhydro-3-D-lyxofuranosyl)purinderivat med formelen:omsettes med en base, slik som natriumacetat, i et inert oppløsnings-middel, slik som dimetylformamid,hvoretter, om ønsket, de erholdte forbindelser omdannes til farmasøyt-isk akseptable salter derav.
