Description
Fremgangsmåte ved fremstilling av terapeutisk aktive
pyrrylaminoethanoler.
Foreliggende oppfinnelse angår en fremgangsmåte ved fremstil-
ling av N-substituerte pyrrylaminoethanoler.
Ved foreliggende fremgangsmåte fremstilles terapeutisk aktive substituerte pyrrylaminoethanoler med den generelle formel:
hvor R er et lavere alkyl-, fenyl- eller benzylradikal som kan være substituert med et eller flere klor- eller bromatomer, lavere alkyl-,
alkoxy- eller tr if luorme.thylgrupper,
R-^er hydrogen, et lavere alkyl- eller fenyl-lavere alkyl-radikal, R2er hydrogen, et lavere alkyl- eller fenyl-lavere alkyl-radikal, eller kan sammen med R^ og det nitrogenatom til hvilket de er bundet, danne, en pyrrolidino-, piperidino- eller morfolinogruppe, og R^er hydrogen eller methyl, og salter av disse forbindelser med organiske eller uorganiske syrer og alkylhalogenider, og fremgangsmåten er særpreget ved at en forbindelse med den generelle formel:
hvor R, , R2og R^ har samme betydning som angitt ovenfor, hydrogeneres med lithiumaluminiumhydrid, natriumborhydrid eller aluminiumisopropoxyd og at eventuelt en erholdt forbindelse med R2

Hydrogeneringen med lithiumaluminiumhydrid, natriumborhydrid eller aluminiumisopropoxyd utfores fortrinnsvis i nærvær av et inert opplosningsmiddel ved en temperatur av 0 - 90°C i lbpet av 2 - 60 timer. Dersom R2er en benzylgruppe, kan denne spaltes av med hydrogen i nærvær av f.eks. 5 % palladium-på-carbon ved atmosfære-trykk, en temperatur av 15-60°C og i nærvær av et inert opplosningsmiddel inntil den teoretiske hydrogenmengde er blitt tatt opp.
Syreaddisjonssaltene kan fremstilles ifolge foreliggende oppfinnelse ved å omsette pyrrolderivatene med formelen I med enten den kjemisk ekvivalente mengde av organisk eller uorganisk syre i et med vann blandbart opplosningsmiddel, som f.eks. aceton eller ethanol, og med separering av saltet ved konsentrering og avkjoling, eller med et overskudd av syren'i et med vann blandbart opplosningsmiddel, som f.eks. diethylether eller kloroform, hvorved det onskede salt utskilles direkte.
Eksempler på slike organiske salter er salter med malein-, fumar-, benzoe-, ascorbin-, succin-, methansulfon-, benzensulfon- eller hydroxybenzoesyrer. De foretrukne salter er salter med aromatiske hydroxycarboxylsyrer, som f.eks. p-hydroxy-,2,5-dihydroxy-, 3, h,5-trihydroxy-, 3)^-dihydroxy- eller a-resorcylsyre. Som eksempler på slike uorganiske salter kan nevnes salter med hydrogen-klorid-,, hydrogenbromid- og svovelsyrer.
De kvaternæreammoniumsalter kan også fremstilles ved å omsette et nytt pyrrolderivat med den generelle formel I med et lavere alkyl-halogenid, som f.eks. methylbromid eller methyljodid.

II fremstilles ved fremgangsmåten ifolge norsk patent nr. 121.95CL
Fra de norske patenter nr. 112Li-ll+ og nr. 116119 er det kjent å fremstille terapeutisk aktive amino-hydroxyalkyl-isoxazoler ved hydrogenering av aminoalkanoyl-isoxazolforbindelser med et alkali-aluminiumhydrid eller et alkaliborhydrid i et inert opplosningsmiddel.
De ved foreliggende fremgangsmåte fremstilte N-monosubstituerte pyrrylaminoethanoler viser fordelaktige farmakologiske virkninger på det sentrale nervesystem, det periferiske autonome system eller cardiovasculare system.
Det sentrale nervesystem
De N-monosubstituerte derivater fremstilt ifolge oppfinnelsen, hvor er et lavere alkylradikal, som f.eks. methyl og ethyl,
viser merkbare virkninger på det sentrale nervesystem. Disse virkninger er sympatetisk-eksitatoriske virkninger som avhengig av den administrerte dose består av hypermotilitet, tremorer og konvulsjoner. . De nye forbindelser forbedrer den codein-analgetiske aktivitet og har direkte analgetiske egenskaper.
Det autonome nervesystem
De N-monosubstituerte derivater fremstilt ifolge oppfinnelsen samvirker med sympatetisk virkning og viser både sympathomimetiske og sympatholytiske virkninger. På den annen side er den parasympa-/tetiske del av mindre interesse. De foretrukne forbindelser med hensyn til den sympatholytiske aktivitet er forbindelser med den generelle formel I, hvor R2= H og R-^er et lavere alkylradikal med mer enn 2 carbonatomer, og deres syreaddisjonssalter.
l-[a-(N-benzyl)-pyrryl]-2-isopropylaminoethanol virker anta-gonistisk på det inhibitoriske sympathomimetiske system ved selektiv blokkering av p-adrenergiske receptorer. Studiumet av disse egenskaper er blitt utfort-ved å undersoke: blodtrykkets reaksjon overfor N-ethyl-noradrenalin hvis hypotensive virkning inverteres av l-[a-(N-benzyl)-pyrryl]_ 2-isopropylaminoethanol, og den tachycardia og blodtrykks-'forhoyelse som fåes i dyr in vivo med N-isopropyl-noradrenalin._ Den effektive dose for tilveiebringelse av de ovennevnte virkninger var 3-5 mg/kg intravenost.
Det cardiovasculare system
Dersom R-^er et alkylradikal med 1-2 carbonatomer, viser de N-monosubstituerte forbindelser fremstilt ifolge foreliggende oppfinnelse hypertensiv aktivitet og letter hjertefrekvensen mens dersom er et lavere alkylradikal med mer enn 2 carbonatomer, viser disse forbindelser hypotensive og bradycardialske virkninger.
Toksisitet
I mus og rotter er LD-verdiene for de N-monosubstituerte forbindelser fremstilt ifolge foreliggende oppfinnelse hoyere enn 20
mg pr. kg ved intravenos undersøkelse. Doser av ca. 100 mg pr. kg gitt oralt kan godt tolereres. Disse derivater kan administreres oralt, subcutant eller intravenost i en hvilken som helst farma-søytisk form som i alminnelighet anvendes for disse administreringsmetoder.
De N,N-disubstituerte forbindelser fremstilt ifolge foreliggende oppfinnelse har betydelige egenskaper med hensyn til påvirk-ning av det sentrale eller perifere nervesystem. De sedative og analgetiske virkninger på det sentrale nervesystem er spesielt merkbare. Den analgetiske aktivitet er blitt undersokt i mus.ved hjelp av varmplatetesten og ved strekking ved injisering av eddiksyre.
Med hensyn til den sistnevnte undersøkelse er ED^Q-verdiene i mus 0,7 mg pr. 100 g gitt subcutant for l-a-(N-o-klorbenzyl)-pyrryl-2-disek.-butylaminoethanol og 1,5 mg pr. 100 g gitt oralt. Ved oral administrering er l-a-(N-o-klorbenzyl)-pyrryl-2-disek.-butylamino-ethanol tre ganger sterkere enn codein, ni ganger sterkere enn aminopyrin og tolv ganger sterkere enn acetylsalicylsyre. Den hoyes-te analgesia fremkommer etter 60 minutter, men virkningen er også markert h timer etter administrering av medisinen.
Den sedative aktivitet er blitt studert ved å undersoke:
a) oppforselen til dyr behandlet med de N,N-disubstituerte nye forbindelser. Det ble notert en nedsatt motilitet og forminsket reaktivitet. b) innflytelsen av virkningene av subhypnotiske og hypnotiske doser av barbiturater, c) den sedative virkning på hostesentret som på forhånd var blitt irritert med en ammoniakkalsk aerosol i rotter og ved elektrisk sti-mulering av hovedlaryngealnerven i decerebrerte katter. Ifolge den sistnevnte undersøkelse er ED-verdiene i rotter 8 - 20 mg pr. kg og i katt"er? 5 - 10 mg pr. kg gitt intraperitonealt.
De N,N-disubstituerte derivater fremstilt ifolge foreliggende oppfinnelse har vist en meget lav toksisitet: LD^Q-verdier hoyere enn 100 mg/kg ved intraperitoneal administrering i mus og hoyere enn ^00 mg/kg ved oral administrering i mus og rotter. En daglig administrering av 100 mg/kg tolereres lett av rotter. Undersøkelse av muskelvev etter avlivning ga intet tegn på pathologiske forandringer, og de hematologiske verdier ble funnet å være upåvirkede.
Disse forbindelser kan administreres oralt, subcutant eller intravenost i en hvilken som helst farmasøytisk form som er egnet for anvendelse ved disse administreringsmetoder.
Eksempel 1
7 g (0,022 mol) l-La-(N-benzyl)-pyrryl]-2-(N'-benzyl-N'-methyl)-aminoethanol, 1 g 5 % palladium på carbon og 25 ml absolutt ethanol fylles i en 100 ml flaske til et hydrogeneringsapparat med atmos-færetrykk. Blandingen oppvarmes til ca. 50°C og holdes ved denne temperatur inntil den teoretiske hydrogenmengde er blitt tatt opp. Katalysatoren fjernes fra reaksjonsblandingen ved filtrering, og oppløsningen konsentreres under redusert trykk. Den faste rest av l-|.a-(N-benzyl)-pyrryl]-2-methylaminoethanol veide 5,*+ g og ble renset ved krystallisering fra diisopropylether. Smeltepunktet var &V-86°C.
De følgende forbindelser er blitt fremstilt analogt:
Eksempel 2
6,85 g (0,03 mol) l-benzyl-2-methylaminoacetylpyrrol og 55 ml methylalkohol anbringes i en 100 ml firehalset kolbe utstyrt med en omrorer, en dråpetrakt, en tilbakelopskondehsator og et termometer. Oppløsningen omrores, og en blanding av 2,27 g (0,06 mol) natriumborhydrid i 6 ml vann tilsettes langsomt gjennom dråpetrakten med en slik hastighet at opplosningsmidlet tilbakelopsdestilleres forsiktig uten utvendig oppvarming. Når tilsetningen er fullstendig og den opprinnelige reaksjon opphorer, omrores blandingen og oppvarmes under forsiktig tilbakelopsdestillering i 6 timer.
Opplosningsmidlene destilleres av under redusert trykk. Re-siduet suspenderes i vann og rystes med diethylether, opplosningsmidlet fjernes ved fordampning under redusert trykk, og den faste rest av l-[a-(N-benzyl)rpyrryl]-2-methylaminoethanol som veier 6,2 g, renses ved krystallisering fra diisopropylether..

5A g (0,022 mol) l-benzyl-2-dimethylaminoacetyl-pyrrol og
<*>f0 ml methylalkohol anbringes i en 100 ml fire-halset kolbe utstyrt med en omrorer, en dråpetrakt, en tilbakelopskondensator og et termometer. Oppløsningen omrores, og en blanding-av -1,7 g (0,0^+ mol) natriumborhydrid i h ml vann tilsettes langsomt gjennom dråpetrakten med en slik hastighet at opplosningsmidlet tilbakelopsdestilleres forsiktig uten utvendig oppvarming. Når tilsetningen er ferdig og den opprinnelige reaksjon opphorer, omrores blandingen og oppvarmes ved forsiktig tilbakelopsdestillering i 6 timer. Opplosningsmidlene destilleres av under redusert trykk, og de faste rester suspenderes i" vann og rystes med diethylether. Den. annqn ekstrakt
;6rres over vannfritt magnesiumsulfat, opplosningsmidlet fjernes red fordampning under redusert trykk, og den oljeaktige rest frak-sjoneres. Utbyttet var ^,9 g, og produktet hadde et kokepunkt av
.20°C ved et trykk av 0,<*>+ mm Hg.
De folgende forbindelser er blitt fremstilt analogt:
Eksempel h
10 g (0,0278 mol) l-(o-klor)-benzyl-2-disek.-butylamino-acetylpyrrol og 300 ml vannfri diethylether fylles i en 500 ml fire-halset kolbe med en kvikksolvforseglet omrorer, et termometer, en dråpetrakt og en tilbakelopskondensator med et på toppen anbragt ror inneholdende vannfritt kalsiumklorid. Oppløsningen omrores,
og en blanding av 1 g (0,026<*>+ mol) lithiumaluminiumhydrid i 20 ml diethylether tilsettes langsomt gjennom dråpetrakten med en slik hastighet at opplosningsmidlet tilbakelopsdestillerer forsiktig uten utvendig oppvarming. Når tilsetningen er ferdig og den opprinnelige reaksjon opphorer, omrores blandingen og oppvarmes under forsiktig refluks i 2 timer.
Blandingen avkjoles, og overskuddet av lithiumaluminiumhydrid dekomponeres med isbiter. Vannlaget fraskilles og vaskes med diethylether. De samlede etherekstrakter torres over vannfritt magnesiumsulfat, og opplosningsmidlet fjernes ved destillering under redusert trykk.
Utbyttet var 8,8 g, og produktet hadde et kokepunkt av
160 - 165°C/0,1 mm Hg..
Claims (1)
- Fremgangsmåte ved fremstilling av terapeutisk aktive substituerte pyrrylaminoethanoler med den generelle formel:hvor R er et lavere alkyl-, fenyl- eller benzylradikal som kan være substituert med et eller flere klor- eller bromatomer, lavere alkyl-, alkoxy- eller trifluormethylgrupper, er hydrogen, et lavere alkyl-, eller fenyl-lavere alkyl-radikal, R9 er hydrogen, et lavere alkyl- eller fenyl-lavere alkyl-radikal, eller kan sammen med og det nitrogenatom til hvilket de er bundet, danne en pyrrolidino-, piperidino- eller morfolinogruppe, og R^ er hydrogen eller methyl, og salter av disse forbindelser med organiske eller uorganiske syrer og alkylhalogenider, karakterisert ved at en forbindelse med den generelle formel:hvor R, R-p R£ og R^ har samme betydning som angitt ovenfor, hydrogeneres med lithiumaluminiumhydrid, natriumborhydrid elleral uminiumisopropoxyd og at eventuelt en erholdt forbindelse med R2 i betydning av en benzylgruppe derefter hydrogeneres med hydrogen i nærvær av palladium-på-carbon, for å fraspalte benzylgruppen, og de således fremstilte forbindelser eventuelt omdannes til deres salter ved anvendelse av organiske eller uorganiske syrer eller alkylhalogenider.
