Aminocyclopentanonamiden, werkwijze ter bereiding daarvan en farmaceutische preparaten, die deze verbindingen bevatten.
De endoperoxyden prostaglandinen Gg en Hg en 5 thromboxan Ag zijn in de natuur voorkomende, reactieve meta- holieten van arachidonzuur in menselijke bloedplaatjes. Zij zijn niet alleen sterke aggregerende middelen, maar zij vertonen eveneens een samentrekkende werking op de vasculaire en bronchiale gladde spier, en daarom zijn stoffen, die de ef-*10 fecten daarvan antagoniseren, van groot belang in de mense lijke geneeskunde.
Gevonden werd nu een nieuwe groep van verbindingen die een endoperoxyde- en thromboxan-antagonistische activiteit vertonen, en die daarom van belang zijn bij de behande-15 ling van asthma en cardiovasculaire ziekten. De verbindingen van deze groep vertonen eveneens een goede chemische bestendigheid in vergelijking met de verwante carbonzuren.
De uitvinding voorziet aldus in verbindingen met de algemene formule I, waarin R^ voorstelt (i) recht of ver-20 takt C.j ,. alkyl, gesubstitueerd door (a) fenyl [eventueel ge substitueerd door g alkyl, ^ cycloalkyl, fenylalkyl met een ^ alkylgedeelte, thienyl of fenyl (eventueel gesubstitueerd door C.j_k alkyl, alkoxy of fenyl)], (b) thienyl [eventueel gesubstitueerd door alkyl, g alkoxy, ^ 25 cycloalkyl of fenyl (eventueel gesubstitueerd door ^ alkyl, C. _ alkoxy of halogeen, of (c) naftyl (eventueel gesubsti- 2 tueerd door alkyl of ^ alkoxy), of (ii) cinnamyl, R voorstelt een waterstofatoom, een methylgroep, -(CH„) 0R^ 3 ^ji11 (waarin m 2-5 is en R voor stelt C , alkyl) of -CHR (CH0)

leaten. Ook kunnen wanneer de groep R voorstelt -CHR (CH_) d m C00H, zouten worden gevormd met basen. Voorbeelden van dergelijke zouten zijn alkalimetaal (bijvoorbeeld natrium)·!·, 15 aardalkalimetaal (bijvoorbeeld calcium)- en amine (bijvoor beeld }piperazine)-zouten.

Voorbeelden van groepen R van de types -(CH_)

0RJ en -CHR (CH2)mC00H zijn -(CH2)20CH3, -(CH^OCHg en -CHgCOOH. R^ is bij voorkeur een waterstofatoom.

De heterocyclische aminogroep Y kan bijvoorbeeld een 5?» 6- of 7-ring omvatten, bijvoorbeeld pyrrolidino, pi-peridino, morfolino, piperazino» thiamorfolino, 1 ,.T:-dioxo-25 thiamorfolino, homomorfblino en hexamethyleenimino. Voorbeelden van eventuele substituenten, die aanwezig kunnen zijn aan een tweede stikstofatoom in de ring, zijn methyl, ethyl en benzyl. De koolstofatomen van de heterocyclische ringen kunnen bijvoorbeeld gesubstitueerd zijn door methyl of ethyl. 30 Verbindingen, waarin Y een morfolino- of piperi- dino-groep voorstelt, genieten de voorkeur.
Wanneer R1 een gesubstitueerde alkylgroep is kan het alkyleen-gedeelte bijvoorbeeld 1-3 koolstofatomen bevatten en is deze bij voorkeur een methyleengroep.
— 35 R is bij voorkeur een fenylalkylgroep waarin het 8103 1 9 9 i i - 3 ~ alkylgedeelte 1-3 koolstofatomen bevat en de fenylgroep is gesubstitueerd door thienyl of fenyl (welke fenylgroep op zichzelf eventueel gesubstitueerd kan zijn door alkyl of alkoxy), of is een f enylthienylalkylgroep, waarin het alkyl-5 gedeelte 1-3 koolstofatomen bevat.
Groepen R^, die in het bijzonder de voorkeur ge—' nieten, zijn benzylgroepen, waarin de fenylgroep (bij-voorkeur op de para-plaats) een fenyl-, tolyl- of methoxyfenyl-sübsti-tuent draagt.
10 Een groep van verbindingen, die in het bijzonder de voorkeur geniet, bezit aldus de formule I, waarin W voorstelt -CHgCHgCHg-, X voorstelt cis -CH-CH-, Y voorstelt mor-folino of piperidino, R* voorstelt benzyl, waarbij de fenylgroep is gesubstitueerd door fenyl, tolyl of methoxyfenyl, o 15 en R voorstelt een waterstofatoom, en de fysiologische aan vaardbare zouten en solvaten daarvan.
De verbindingen met de formule I remmen de bloed-plaatjes-aggregatie en de broncho-vemauwing. Teneinde de remming van de bloedplaatj es-aggregatie te bepalen wordt aan 20 guinese biggetjes, die men heeft laten vasten, oraal de te onderzoeken verbinding in een geschikt vehicle toegediend. Bloedplaatjes-rijk plasma wordt bereid uit elk dier en de aggregatie voor een traject van collageen-concentraties wordt gemeten volgens de methode van Born, Nature 19^% 92T-929, 25 (1962). De collageen-concentraties-effect-curven voor elk mon ster van plasma worden berekend en de resultaten worden :uit-gedrukt via de verschuiving van de curven na de behandeling met de verbinding.
Het vermogen van de verbindingen volgens de uit-30 vinding de broncho-vernauwing te remmen wordt bepaald bij ver doofde guinese biggetjes door meting van het effect daarvan op de dosis-responsie-curve van de broncho-vemauwènde stof [lR-[l<x, ha, 5B(Z),6a(lE, 3S*)]]-7-[β-(3-hydroxy-1 -octenyl)- 2-oxabicyclo 2,2,1 hept-5-ylJ-5-heptienzuur (U-H6619).
35 De verbindingen zijn dus van belang bij de behan- 8103 199 i 4 -Indeling van asthma en als remmers van de bloedplaatj es-aggrega-tie en thrombose ten gebruike bij de nier-dialyse en de behandeling en voorkoming van vasculaire verstoppingsziekten, zoals arteriosclerosis, atherosclerosis, periphere vasculaire 5 ziekten, cerebrale vasculaire ziekten, waaronder voorbijgaan de ischaemische aanvallen, beroerten, longemboly, diabetische retinopatie, na-operatieve thrombose, angina en myocardinale infarcten. Zij kunnen op een gebruikelijke wijze worden gerecepteerd voor het gebruik met een of meer farmaceutische dragers. 10 Voor de orale toediening kan het farmaceutische preparaat in de vorm worden gebracht van bijvoorbeeld tabletten, capsules, poeders, oplossingen, stropen of suspensies, die bereid worden onder toepassing van gebruikelijke middelen met aanvaardbare excipiëntia.
15 De verbindingen kunnen voor de parenterale toe diening worden gerecepteerd door bolus-injecties of continue infusie. Preparaten voor injecties kunnen worden bereid in eenheidsdoseringsvorm in ampullen of in multi-dosis-houders met een toegevoegd conserveermiddel. De preparaten kunnen in 20 de vorm zijn van bijvoorbeeld suspensies, oplossingen of emulsies in olieachtige of waterige vehikels en kunnen récepteer-middelen bevatten, zoals suspendeer-, stabiliseer- en/of dis-pergeermiddelen. Anderzijds kan het actieve bestanddeel in poedervorm zijn voor de reconstitutie daarvan, alvorens te 25 worden gebruikt, met een geschikt vehikel, bijvoorbeeld steriel pyrogeen-vrij water.
De verbindingen worden bij voorkeur oraal toegediend, bij-voorbeeld in hoeveelheden van 0,05-10 mg/kg lichaamsgewicht, 1-1+ maal dagelijks. Ten gebruike bij de behandeling 30 van asthma kunnen de verbindingen worden gebruikt in combinatie met andere anti-asthmatische middelen. De toegediende exacte dosis zal natuurlijk afhangen van de leeftijd en de conditie van de patiënt.
Geschikte methodes voor de bereiding van de ver-____ 35 bindingen met formule I zijn hieronder beschreven, waarbij de 810 3 19 9 * * - 5 - 1 2 de groepen R , R , W, X en Y de boven aangegeven betekenissen bezitten, tenzij anders is aangegeven.
(a) De verbindingen met de formule I kunnen worden bereid door oxydatie van een overeenkomstige hydroxy-ver-5 binding, bijvoorbeeld een verbinding met de formule II.
Geschikte oxydati e-methodes omvatten de toepassing van een Cr^ oxydatiereagens in een geschikt oplosmiddel, bijvoorbeeld ehroomzuur in aceton (bijvoorbeeld Jones reagens, bij voorkeur gebruikt in tegenwoordigheid van een diatomeën-1° achtig siliciumoxyde, zoals celiet) of CrO^ in pyridine. Deze reagentia worden bijvoorbeeld gebruikt bij temperaturen van -20°C tot kamertemperatuur.
Andere belangrijke methodes omvatten de toepassing van een geaktiveerd zwavel-reagens, bijvoorbeeld (i) N-15 chloorsuccinimide-dimethylsulfide-complex in een geschikt oplosmiddel (bijvoorbeeld tolueen of dichloormetaan) bij temperaturen van bijvoorbeeld -25 tot +25°C , bij voorkeur bij 0-5°C, (ii) een dialkylsulfoxyde (bijvoorbeeld dimethylsul-foxyde), geaktiveerd door een geschikt electrofiel reagens 20 (zoals oxalylchloride, acetylbromide of thionylchloride) in een geschikt oplosmiddel (bijvoorbeeld tolueen of dichloormetaan), bijvoorbeeld bij -70 tot -20°C; dicyclohexylcarbodiimide kan eveneens worden gebruikt als het electrofiele reagens (bij voorkeur in tegenwoordigheid van CF^COOH of het pyridini-25 umzout daarvan) bij bijvoorbeeld -10°C tot kamertemperatuur

De uitgangsmaterialen met de formule II kunnen worden bereid uit de moeder-carbonzuren met de formule III — 35 (of een g alkylester daarvan) onder toepassing van gebrui- 8103 199 * % - 6 - kelijke methodes voor de omzetting van zuren in amiden.
Bijvoorbeeld kan een reactief derivaat van een zuur met de formule III worden behandeld met ammonia of een . 2 . ...
amine H^NR m een geschikt oplosmiddel, bijvoorbeeld aceton 5 of acetonitril. Het.reactieve derivaat is op geschikte wijze een gemengd anhydride van het zuur (lil), bijvoorbeeld verkregen door behandeling van het zuur (lil) met een chloorfor-miaat in tegenwoordigheid van een geschikte base, bijvoorbeeld triethylamine of pyridine.
10 Het chloorformiaat kan bijvoorbeeld zijn een g alkyl (bijvoorbeeld methyl, ethyl, isobutyl, sec.-butyl, t-butyl, isopropyl of isoamyl)-, aryl (bijvoorbeeld fenyl)-of aralkyl (bijvoorbeeld benzyl)-chloorformiaat.
Anderzijds kan het reactieve derivaat een imida-15 zolide, bijvoorbeeld verkregen door behandeling van het zuur (III) met 1,1'-carbonyldiimidazol, zijn. De ring-hydroxygroep moet worden beschermd gedurende deze laatste reactie, bijvoorbeeld in de vorm van een tetrahydro-2H-pyran-2-yloxy- of t-butyldimethylsilyloxygroep. Deze beschermingsgroepen kunnen 20 daarna worden verwijderd door zuurhydrolise.
Volgens een anderè methode kan een ester van een zuur met de formule III bijvoorbeeld worden verhit (bijvoor- o 2 beeld op 100-130 C) met ammonia of een amine H^NR , eventueel onder gebruikmaking van een oplosmiddel, zoals een alkohol.
25 De bereiding van vele van de zuren met de formule III en esters daarvan, waarin Y aanwezig is op de 3-plaats en de ring-hydroxygroep op de α-plaats, is beschreven in het Britse octrooischrift 2.028.805A. Dezelfde algemene methodes kunnen worden toegepast voor de bereiding van al deze verbin-30 dingen onder gebruikmaking van uitgangsmaterialen, die de vereiste groep R1 bevatten,
De zuren met de formule III en esters daarvan, waarin Y en de ring-hydroxygroep'beide aanwezig zijn in de α-configuratie, kunnen eveneens worden bereid volgens de in __ 35 het Britse óctrooischrift 2.028.805A beschreven methodes onder 8103199 * φ - 7 - gebruikmaking van een norbomanon met de formule IV als uitgangsmateriaal.
De norbornanonen met de formule IV kunnen vorden bereid volgens reactieschema A, vaarin Rh een hydroxyl-bescher-5 mingsgroep voorstelt. Aldus vordt het norbomanon (V) eerst gereduceerd (bijvoorbeeld met NaBH^) tot de alkohol (VI), vaarin de groep R^ vervolgens vordt ingevoerd (bijvoorbeeld door reactie met R^L, vaarin L een verdvijnende groep is, bijvoorbeeld halogeen of tosylaat) onder vorming van de verbinding h.
10 (Vil). De beschermingsgroep R vordt vervolgens vervijderd en de hydroxygroep geoxydeerd (bijvoorbeeld als beschreven voor proces (a)) onder vorming van het norbomanon (IV).
De norbornanonen (V) kunnen vorden bereid volgens de algemene methode, die beschreven is in het Britse octrooi-15 schrift 2.028.805A.
De verbindingen met de formule (lil) en de esters daarvan, vaarin Y aanvezig is in de α-configuratie en de ring-hydroxygroep in de 6-configuratie, kunnen vorden bereid door epimerisering van de overeenkomstige verbinding, vaarin de 20 ring-hydroxygroep aanvezig is op de a-plaats. Dit kan bijvoor beeld vorden beverkstelligd met trifenylfosfine in tegenvoor-digheid van een zuur (bijvoorbeeld mierezuur o'f benzoeenzuur) en (C2Hj_00C.N)2 bij kamertemperatuur, gevolgd door behandeling met een base (bijvoorbeeld KgCO^) in een oplosmiddel, zoals 25 methanol.
De zuren met de formule (lil) en esters daarvan, vaarin zovel Y als de ring-hydroxygroep aanvezig zijn op de β-plaats, kunnen vorden bereid uit de overeenkomstige verbinding met de formule (lil) vaarin de hydroxygroep aanvezig is 30 op de o-plaats, door oxydatie van de hydroxygroep (bijvoorbeeld volgens de methode van proces (a)), gevolgd door reductie (bijvoorbeeld met lithium-tri-sec.-butylboorhydride).
(b) De verbindingen met de formule (i) kunnen eveneens vorden bereid door selectieve reductie van een over-.. 35 eenkomstige verbinding met de formule I, vaarin X een ethyn- 8103199

Geschikte reductiemethodes omvatten de toepassing van waterstof in tegenwoordigheid van een katalysator, bijvoorbeeld palladium op' een drager (bijvoorbeeld CaCO^ of BaSO^) en vergiftigd door bijvoorbeeld lood of pyridine. Geschikte 5 oplosmiddelen zijn onder andere ethylacetaat of methanol.
De voor deze reactie als uitgangsmaterialen benodigde ethynen kunnen worden bereid uit een amide met de formule VIII, waarin X voorstelt -CH=CH-.
Het hydrochloride van een amide (VIII) wordt eerst 10 gebromeerd onder vorming van de overeenkomstige verbinding, waarin X voorstelt -CHBr-CHBr-, bijvoorbeeld onder gebruikmaking van broom in CH^Cl^ bij 0°C.
De dibroomverbinding wordt vervolgens gedehydro-bromeerd ónder vorming van de ethyngroep, bijvoorbeeld in 15 twee trappen, onder gebruikmaking van kalium-t-butoxyde, eerst bij 0°C en vervolgens bij kamertemperatuur. Oxydatie, bijvoorbeeld zoals beschreven voor proces”(a), geeft vervolgens het benodigde uitgangsmateriaal.
(c) De verbindingen met de formule I kunnen even-20 eens worden bereid door amidering van een reactief derivaat van hèt overeenkomstige'carbonzuur. Deze reactie kan worden uitgevoerd op de hierboven voor de bereiding van amiden met de formule II uit zuren met formule III beschreven wijze.
Aldus kan bijvoorbeeld een .reactief derivaat van een zuur 25 (IX) (bijvoorbeeld een anhydride of imidazolide als hierboven beschreven) in reactie worden gebracht met ammonia of een 2 amine HgNR .
De zuren met de formule IX kunnen worden bereid door,oxydatie van het overeenkomstige zuur met de formule III 30 of een beschermd derivaat daarvan, bijvoorbeeld een tr&alkyl-silylester, bijvoorbeeld als beschreven voor proces (a).
(d) De verbindingen met de formule (i), waarin X voorstelt -CHgCHg-, kunnen worden bereid door katalytische hydrogenering van een overeenkomstige verbinding, waarin X 35 voorstelt -CÏÏ=CH-, onder gebruikmaking van een katalysator, 8103199 # .. - - ......... y « - 9 - zoals palladiumoxyde.
(e) De zouten van de verbindingen met de formule I kunnen worden bereid door behandeling van een amide met 2 de formule I met een zuur of, wanneer de groep R voorstelt 5 -Cïïr\cH_) COOH, een base, in een geschikt oplosmiddel, zoals dm ether.
De onderstaande voorbeelden illustreren de uitvinding. De temperaturen zijn aangegeven in °C. De droging werd bewerkstelligd met watervrij MgSO^. "Hyflo" is een fil-10 tratie-hulpmiddel.

Het mengsel werd gedurende 1 uur bij kamertemperatuur geroerd 15 en vervolgens af gekoeld in ijs. Waterig NaOH (IM, 2,1 ml) werd toegevoegd en na roeren (15 min) werd een overmaat watervrij Na2S0^ toegevoegd. Het mengsel werd afgefiltreerd en het fil-traat ingedampt tot een vaste stof. Kristallisatie uit cyclo-hexaan-methanol gaf de titelverbinding (1,0U g); smeltpunt = 20 128-13-1 °C.
Intermediair IVb U-Broommethvl-U*-methyl-(1,1 *-bifenyl)
Aan een koude (0°C) oplossing van intermediair IVa (0,917 g) in droge CHgCl^ (1^ ml) werd PBr^ (0,29 m l) 25 toegevoegd. Na roeren -gedurende 1 uur bij 0°C werd een 8%
NaHCO^-oplossing (30 ml) toegevoegd en werden de lagen gescheiden. De waterige laag werd geëxtraheerd met CH^Clg (2 x 30 ml), gedroogd en ingedampt, waarbij een vaste stof werd verkregen (0,99 g). Kristallisatie uit PE (kookpunt 60-80°c) gaf de ti-30 telverbinding (0,9-1 g); smeltpunt = 100-102°C.

De titelverhinding werd gekristalliseerd uit ether-PE in de vorm van naalden (0,14 g); smeltpunt = 75-77°C.
15 Intermediair IV
Cla(Z), 2g,3a.5cü-(-)-7-l3-Hydroxy-5-ED* f-methyl(1,1’-hifenyl)-U-yllmethoxy3-2-(U-morfonolinyl)cyclopentylJ-5-hepteenzuur Een suspensie van intermediair IVc (0,UU g) in methanol (2 ml) en water (U ml), bevattende Κ0Η (0,35 g)s 20 werd gedurende 7 uur hij kamertemperatuur geroerd. De methanol werd verwijderd en het residu verder verdund met water (75 ml), gewassen met ether (75 ml) en vervolgens voorzichtig aangezuurd tot een pH van 6 met IIN chloorwaterstofzuur. Extractie met ether (¼ x 50 ml), gevolgd door droging en indam-25 ping, gaf de titelverhinding (0,35 g) als een schuim. IR
(CHBr3) 3500, 3200 (hr.), 1730, 1700 cm"1. TLC (D) Rf Ö,25. Intermediair Va (endo,anti)-(-)-6- Γ0* *-Methoxy (1,1'-hifenyl)-U-yllmethoxyJ- 8-(U-morf olinyl)-2-oxabi cyclo[3,2, lloctan-3-on 30 U-Broomanisool (7,U8 g) in ether (30 ml) werd toegevoegd aan Mg-spaanders (1,06 g) in THP (Uo ml), bevattende een klein jodiumkristal. Na 0,5 uur trad een exotherme reactie op, die gematigd werd door afkoeling in een waterbad.
Het resulterende mengsel werd gedurende nog 0,5 uur hij 20°C
35 geroerd en vervolgens in een stikstof atmosfeer toegevoegd aan ________ _____ _ _ _____ ________________ * 8103199 % - 12 - een geroerde afgekoelde (5°C) oplossing van watervrij ZnBr^ (9,0 g) in THF (Uo ml) en.gedurende 1 uur geroerd.
Dibal (IM in hexaan; 5,72 ml) werd druppelsgewijs toegevoegd aan een suspensie van bis(trifenylfosfine)palladium-5 chloride (2,0 g) in THF (20 ml). Na roeren gedurende 5 minuten werd een oplossing van intermediair VlIId (2,38 g) in THF (35 ml) toegevoegd, gevolgd, na nog 5 minuten, door de hierboven beschreven oplossing van 1-methoxy-fenylzinkbromide.
Het resulterende mengsel werd gedurende 18 uren in een stik- 30. stofatmosfeer bij 20°C geroerd. Het oplosmiddel werd vervol gens in vacuo verwijderd, NH^Cl-oplossing (1.50 ml) werd toegevoegd en het mengsel werd geëxtraheerd met EA (3 x 100 ml). Indamping van de gedroogde extracten gaf een olie, die werd ge-chromatografeerd (EA), waarbij de titelverbinding als een vaste 15 stof werd verkregen. Kristallisatie uit EA-PE (kookpunt 60-80°C) gaf een materiaal (1,58 g) met een smeltpunt van 123-125°C. Intermediair Vb (tot,'26, 3a. 5a)-(-)-7- 3-Hydroxy-5-LD1 -methoxy( 1,1'-bifenyl)-^-yljmethoxyj -2- (U-morfolinyl) cyclopentaanacét aldehyde.
20 Een oplossing van intermediair Va (1,6 g) in CH«- o ^
Cl2* (25 ml) bij -70 C in een droge stikstofatmosfeer werd geroerd gedurende de toevoeging van Dibal (IM in hexaan, 8,7 ml). Na 1,5 uren bij -70°C werd zorgvuldig methanol (25 ml) toegevoegd en men liet de temperatuur van het mengsel vervolgens 25 stijgen tot omgevingstemperatuur, waarna het roeren gedurende 18 uren werd vooi^ezet. Het mengsel werd afgefiltreerd door "ïïiflo" en het filtraat ingedampt, waarbij de titelverbinding werd verkregen als een schuim (1,68 g). IR (CHBr^) 3580/3560 (br), 1715, 121*0, 10^0 cm”1.
30 Intermediair V
De (Z) ,26,3a,5cJ-(-)-7- D-Hydroxy-5-LD» * -methoxy(1,1f-bi fenyl)-l*-ylimethoxy3-2-(^-morfolinyl)cyclopentyl]-5-hepteenzuur
Aan een oplossing van kalium-t-butoxyde (2,28 g) in THF (15 ml) in een droge stikstofatmosfeer werd (l*-carboxy-... 35 butyl)trifenylfosfoniumbromide (1*,5 g) toegevoegd. Na 15 min.
8103 199 - 13 - bij 20°C werd een oplossing van intermediair Vb (1,6 g) in THF toegevoegd en het roeren werd gedurende nóg 1 uur voortgezet. Water (2 ml) werd toegevoegd en het THF in vacuo verwijderd. Het residu werd opgenomen in water (100 ml), basis 5 gemaakt (pH 10) met NaOH-oplossing en gewassen met ether (2 x 75 ml)· De waterige laag werd op een pH van 6 ingesteld met IXN chloorwaterstofzuur, geëxtraheerd met ether (5 x 75 ml), gedroogd, ingedampt en weer opgelost in EA (25 ml) en ether (U0 ml). Aan de oplossing werd een overmaat etherische 10 chloorwaterstof-oplossing toegevoegd, waarna werd afgekoeld totdat een kristallisatie optrad. De filtratie en zuivering uit EA-methanol gaf de titelverbinding (1,2 g) in de vorm van het hydrochloride daarvan; smeltpunt = l6k-l66°C. Intermediair Vla 15 fr-(1,3-Dioxolan-2-yl)-2-fenylthiofeen
Een oplossing van 5-broom-3-thiofeenearboxaldehyde (32,5 g) in benzeen (500 ml) werd behandeld met FTSA (0,323 g) en ethyleenglycol (21,1 g) en het mengsel werd in een Dean-Stark-apparaat onder terugvloeikoeling verhit totdat het the-20 oretische volume water was verwijderd. Na de afkoeling werd het mengsel gewassen met water (2 maal) en vervolgens met zoutoplossing, gedroogd, afgefiltreerd en geconcentreerd en het residu werd vervolgens gedestilleerd (kookpunt 96-100°C hij 0,¼ mm), waarbij de titelverbinding werd verkregen als een 25 olie (24 g).
Analyse: gevonden: C 35,8; H 3,0$

Intermediair VIb 5-Fenyl-3-thiofeenc arboxaldehyde
30 Een oplossing van fenylmagnesiumchloride in THF
(82,9¼ ml; 2,39 M) werd toegevoegd aan een geroerde oplossing van ZnBr2 (¼¼^ g) in droge THF (350 ml) in een stikstofatmos-feer. Het mengsel werd gedurende 15 minuten bij kamertemperatuur geroerd.
35 Dibal (9,91 ml, I M) in hexaan-oplossing werd 8103199 - 1l* - druppelsgewijs toegevoegd aan een geroerd mengsel van tri-fenylfosfiné (10,39 g) en nikelacetonaat (2,55 g) in droge THP (160 ml) in een stikstofatmosfeer. Een oplossing intermediair Vla (23,3 g) in droge THP (150 ml) werd toegevoegd 5 na 10. minuten. De-oplossing, bevattende het organozinkreagens, werd vervolgens druppelsgewijs toegevoegd en het mengsel werd gedurende 1 uur geroerd.
IIN chloorwaterstofzuur (1)-00 ml) werd toegevoegd bij 0°C en het mengsel werd gedurende 0,5 uur bij hamertem-10 peratuur geroerd. De twee lagen werden gescheiden en de wateri
ge laag werd geëxtraheerd met ether (2 x 1*00 ml), gewassen met NaHCO^-oplossing en keukenzoutoplossing en vervolgens gedroogd. De verwijdering van het oplosmiddel in vacuo gaf een vaste stof (32,8 g), die werd gechromatografeerd (Q), waarbij de 15 titelverbinding werd verkregen (13,35 g)j smeltpunt = 61*-65°C
(uit PE(kookpunt 6Ο-8Ο0)).
Intermediair VIc 5-Penyl-3-thiofeenmethanol
Een geroerde oplossing van intermediair VIb (12 20 g) in methanol (120 ml) werd behandeld met NaBH^ (1,82 g) bij kamertemperatuur gedurende 15 minuten. Het mengsel werd afgekoeld tot 0°C en behandeld met Nïï^Cl-oplossing (200 ml), gevolgd door water (200 ml) en ether (1*00 ml). Het etherextract werd afgescheiden en de waterige fase verder geëxtraheerd met 25 ether (1*00 ml). De gecombineerde extracten werden gewassen met keukenzoutoplossing, gedroogd, afgefiltreerd en ingedampt, waarbij de titelverbinding werd verkregen als een vaste stof (11,5 g); smeltpunt = 92-93°C).
Intermediair VId' 30 1*- (Broommet hyl) -2-fenylthi of een
Een oplossing van intermediair VIc (1* g) in CH^-Cl2 (TO ml) bij 0°C werd behandeld met fosfortribromide (1,32 ml) in CHgClg (5 ml). Na 10 minuten werd 8$ NaHCO^-oplossing (TO ml) toegevoegd en werden de lagen gescheiden. De waterige 35 fase werd geëxtraheerd met CHgClg (1*0 ml). De gecombineerde 8103199 - 15 - organische lagen werden gewassen met keukenzoutoplossing (50 ml), gedroogd en ingedampt, waarbij de titelverbinding werd verkregen (4,87 g); TLC (L) Ef 0,58.
Intermediair Vie 5 0 a(Z),2g,3a.5a]-(-)-Methyl-7- Os-hydroxy-2-(4-morfolinyl)-5- E( 2-fenylthien-4-yl)methoxylcyclopentyl3-5-heptenoaat
Natriumhydride (0,88 g, 50$1s dispersie in olie) werd toegevoegd aan een geroerde oplossing van intermediair VId (4,6 g) en |ja(Z),2$,3α,5α1-(-)-7-|5--hydroxy-2-(4-morfolinyl)-10 3- Btetrahydro-.2H-pyran-2-yl) oxyJcyclopentyl3-5-hepteenzuur- methylester (2,5 g) in droge DMF (15 ml) bij 0°C. Ha 1 uur bij omgevingstemperatuur werd ammoniumchloride-oplossing (100 ml) toegevoegd bij 0°C en de pH ingesteld op 6,5 onder gebruikmaking van KHgPO^. Het mengsel werd geëxtraheerd met ether (3 x 15 100 ml) en de gecombineerde extracten werden gewassen met water (2 x 200 ml), gedroogd en ingedampt. Het residu werd geroerd met 9:1 methanol-geconcentreerd zwavelzuur (20 ml) gedurende 3 uren, 8$’s NaHC0^-oplossing (100 ml) werd toegevoegd en het mengsel werd geëxtraheerd met EA (3 x 70 ml). De gecombineerde 20 extracten werden gewassen met keukenzoutoplossing (100 ml), gedroogd en ingedampt en het residu werd gezuiverd door ehro-matografie (B), waarbij de titelverbinding werd verkregen als een olie (0,98 g). IB (CHBr^) 3590, 1728 cm~1.

Aan een geroerde oplossing van kalium-t-butoxyde 10 (1,12 g) in droge THF (10 ml) werd in droge stikstofatmosfeer toegevoegd (ίι-carboxybutyl)trifenylfosfoniumbromide (2,21 g) en het mengsel werd gedurende 15 minuten bij 22°C geroerd, waarna een oplossing van het intermediair VIIc (0,9 g) in THF (5 ml) werd toegevoegd en het roeren gedurende nog 30 minuten 15 werd voortgezet. Water (50 ml) werd toegevoegd en het mengsel werd geëxtraheerd met EA (3 x 25 ml). De waterige fase werd met KHgPOj^-oplossing ingesteld op een pH van 6,5 en vervolgens geëxtraheerd met EA(3x30ml), gewassen met keukenzoutoplos-sing, gedroogd en behandeld met etherisch diazomethaan. Intermediair Villa 25 (endo, anti )-(-)-7-(^-Morfolinyl)-5-Ebetrahydro-2H-nyran-2- yl) oxylbi cylo Β?,2,ΐ] hept aan-2-on
Morfoline (76 ml) werd druppelsgewijs over een periode van 15 minuten toegevoegd aan een geroerde oplossing van (exo, endo)-(-)-2-broom-3- £( tetrahydro-2H-pyran-2-yl)oxyj-30 bicyclojj3,2,o|heptaan-6-on (100,8 g) in aceton (500 ml) bij 0°C. Na 2 uren bij 5°C werd het.mengsel gedurende 18 uren bij 20°C geroerd en vervolgens afgefiltreerd. Door indamping van filtraat verkreeg men een olie, die werd opgenomen in ether (350 ml), afgefiltreerd en gewassen (water, 2 x 100 ml). De 35 etherische oplossing werd gedroogd, afgefiltreerd en ingedampt, 8103 199 j. Η - 18 - waarbij de titelverbinding werd verkregen als een vaste stof. Door zuivering uit PE verkreeg men een materiaal (85,5 g) met een smeltpunt van 86-88°C.
Intermediair VlIIb 5 (endo, anti)-(-)-5-Hydroxy-T-(4-morfolinyl)bicyclo[g,2jl-hep- tan-2-on-hydrochloride
Aan een geroerde oplossing van intermediair Villa (96,4 g) in methanol (600 ml) werd een etherische oplossing van HC1 (2^0 ml) toegevoegd en het mengsel werd gedurende 2,5 10 uren bij 20°C (pH 1,5-2) geroerd. Door filtratie, gevolgd door indamping van het filtraat, verkreeg men een olie, die bij samenwrijven met EA (2 x 200 ml) vast werd. Gekleurde verontreinigingen werden verwijderd door extractie met kokende isopro-panol onder achterlating van de titelverbinding als een vaste 15 stof (70,6 g); smeltpunt - 181-182°C.
Intermediair VIIIc (endo, anti)—(—)—5—(4-Broomfenylmethoxv)-7-(4-morfolinyl)-bicycloL? «2 %1 heptan-2-on
Waterige ïïaOH-oplossing (10N; 200 ml) werd toege-20 voegd aan een oplossing van de vrije base van intermediair VlIIb (21,1 g), benzyltriethylammoniumchloride (4 g) en 4-broombenzylbromide (27,5 g) in CHgClg (400 ml) en het mengsel werd gedurende 4 uren heftig geroerd. Een verdere portie 4-broombenzylbromide (9 g) werd vervolgens toegevoegd en het roe-25 ren werd gedurende 68 uren voortgezet. Water (200 ml) werd toegevoegd en de lagen werden' gescheiden. De waterige laag werd geëxtraheerd met EA (2 x 75 ml), gewassen met water, gedroogd en ingedampt, waarbij een olie (48 g) werd verkregen, die bij staan vast werd. De overmaat alkyleermiddel werd verwijderd 30 door samen wrijven met PE (kookpunt 60-80°C) en kristallisatie uit EA-PE (kookpunt 60-80°C) gaf vervolgens de titelverbinding (34,1 g) als een vaste stof; smeltpunt = 130-131°C.

Intermediair VIIIc (13,3 g) in azijnzuur (110 ml) en vater (55 ml), bevattende CH^COONa.311^0 (23,7 g), verd afgekoeld (ongeveer 5-10°c) en geroerd gedurende de druppelsgewijze toevoeging van perazijnzuur (β,1Μ; 28,5 ml). De resulterende op-5 lossing verd gedurende U8 uren bij 20°C geroerd en vervolgens verd 10^’s NagSO^-oplossing (200 ml) toegevoegd, vaarbij de temperatuur van het mengsel· verd gehouden op 10-15°C. Na 1,5 uren verden de oplosmiddelen in vacuo bij 35°C verwijderd, verd het residu opgenomen in water (150 ml) en basisch gemaakt 10 tot een pH van 9 met Na^CO^-oplossing. Extractie met EA (3 x 200 ml), gevolgd door droging en indamping, gaf een vaste stof, die uit EA kristalliseerde, waarbij de titelverbinding (5,^9 g) verd verkregen; smeltpunt = 15^-156°C.
Intermediair YlIIe 15 (endo, anti)-(-)-&-(Thien-2-yl)fenylmethoxy]-8-(U-morfolinyl)- 2-oxabieyclo 13,2, ll oct an-3-on
Het Grignardreagens uit 2-broomthiofeen (U,1 g) en magnesium (0,7 g) verd bereid in droge ether (50 ml); de exotherme reactie verd gematigd door afkoeling in koud water.
20 Het reagens verd toegevöegd aan een geroerde afgekoelde (5°C) oplossing van vatervrij ZnBrg (5,6 g) in THE (30 ml) en gedurende 1 uur geroerd.
Dibal (IM in hexaan; 3,6 ml) verd druppelsgewijs toegevoegd aan een suspensie van bis(trifenylfosfine)palladium-25 chloride (1,25 g) in THE (20 ml). Door indaging van de gedroogde extracten verkreeg men een olie, die gezuiverd verd door chromatografie EA-PE (kookpunt 60-80°C) (achtereenvolgens 1:2, 1:1 en 2:1), waarbij de titelverbinding verd verkregen als een vaste stof; — 35 smeltpunt = 2l6-217°C.

Na 10 minuten bij 20°C werd een oplossing van intermediair VlIIf (0,55 g) in THF (30 ml) toegevoegd en het roeren werd gedurende 5 20 uren voortgezet. Het mengsel werd uit gegoten in ammoniumchlo-ride-oplossing en geëxtraheerd met ether. De gedroogde extracten werden ingedampt en het residu werd gezuiverd door chro-matografie (PE), gevolgd door (D), waarbij de titelverbinding (0,M g) werd verkregen. IR (zuiver) 3360, 1710 cm”"*.
25 Intermediair IXa (endo ,anti)-(-)-7-Azido-5-hydróxybieyclo «2, l]hept an-2-on Een oplossing van (exo, endo)-(-)-3-acetoxy-2-broombicyclo[3,2,ÖJheptan-6-on (50 g) en kalium-t-butoxyde (27,25 g) in THF (1,5 1) werd.gedurende 1 uur bij -75°C ge-30 roerd. Men liet de temperatuur van de oplossing oplopen tot 0° C en vervolgens werd een oplossing van natriumazide (16,1+5 g) in water (600 ml) toegevoegd, waarna het roeren gedurende 18 uren bij 20°C werd voortgezet.
De twee lagen werden gescheiden en ether werd toe-— 35 gevoegd aan de organische laag, werd gewassen met water (2 x 8103199 * * - 21 - 250 ml). De gecombineerde waterige lagen werden geëxtraheerd met ether (2 x 250 ml). De gecombineerde organische lagen werden gedroogd en ingedampt, waarbij een gom (28,1 g) werd verkregen. Een oplossing van de gom in methanol (225 ml) werd ge-5 roerd met K^CO^ (18,37 g) gedurende 3,5 uren bij kamertempe ratuur. Het mengsel werd afgefiltreerd en het filtraat in vacuo ingedampt, waarbij een vaste stof werd verkregen, die werd opgenomen in ether (150 ml) en gewassen met water (150 ml).
De waterige laag werd geëxtraheerd met ether (3 x 125 ml) en 10. de gecombineerde organische lagen werden gedroogd en ingedampt, waarbij een olie (2k,5 g) werd verkregen, die werd gechromato-grafeerd over silica. Elutie met 2:1 ether-PE gaf een olie (18,7 g), die werd samengewreven met ether, waarbij de titel-verbinding werd verkregen als een vaste stof (lt,6 g); smelt-15 punt = T2-jk°C.
Intermediair IX b (3act ,kct.5S.6aa)-(-) -t-Azi do-hexahydro-5-hydroxy-gH-cyclopenta-(b)-furan-2-on.
t0*'s Perazijnzuur (6^,35 ml) werd toegevoegd aan 20 een wafgekoelde (0°C) geroerde oplossing van intermediair IXa (12,9 g) en natriumacetaat (31,2 g) in azijnzuur (155 ml) en water (15,5 ml) en de resulterende oplossing werd vervolgens gedurende 2k uren bij omgevingstemperatuur geroerd. Een overmaat NagSO^-oplossing werd toegevoegd aan de afgekoelde oplos-25 sing en het roeren werd gedurende 1 uur voortgezet. Na de in-damping in vacuo werd het residu opgelost in VN.NaOH-oplos sing (^00 ml) onder afkoeling en de oplossing gedurende 0,5 uur geroerd. Geconcentreerd chloorvaterstofzuur (30 ml) werd onder koeling toegevoegd en de oplossing werd continu geëxtraheerd 30 met CHgClg (600 ml) gedurende 18 uren. De organische extracten werden gewassen met IIN.Na^O^-oplossing (100 ml) en keuken-zout-oplossing (100 ml), gedroogd en ingedampt, waarbij een vaste stof (3,5 g) werd verkregen. Een gedeelte (1 g) werd geherkristalliseerd uit ether-PE (kookpunt 60-80°c), waarbij 35 de titelverbinding (816 mg)werd verkregen; smeltpunt * 73-7t°C.

Een mengsel'van intermediair IXd (6 g), waterig NaHCO^ (5,2 g), NaJ (9,72 g) en bis- (2-chloorethyl)-sulfide (5,15 g) in acetónitril (250 ml) werd gedurende 18 uren onder 30 terugvloeikoeling verhit. Het oplosmiddel werd in vacuo verwijderd en'het'residu in water (200 ml) werd geëxtraheerd met EA (i x 200 mj); De gecombineerde éxtracten'werden gewassen met keukenzoutoplossing (200 ml),gedroogd en ingedampt, waarbij een olie (10,2 g) werd verkregen, die werd gezuiverd door chro-— 35 matografie óver silica. Dibal (1 ,UM in hexaan; 20 ml) werd druppelsgewijs 15 toegevoegd aan een geroerde oplossing van intermediair IXe (U,7 g) in droge CHgClg (100 ml) in een stikstofatmosfeer bij -70°C. Na 2 uren werd methanol (100 ml) voorzichtig toegevoegd en vervolgens liet men de temperatuur van; .het mengsel oplopen tot omgevingstemperatuur. Na 2,5 uren werd het mengsel afge-20 filtreerd en het filtraat ingedampt. Het residu werd opgelost in CH2C12 (150 ml), gedroogd en ingedampt, waarbij de titel-verbinding werd verkregen als een olie (^,25 g). TLC 9:1 benzeen-methanol Rf 0,25.

Aan een geroerde oplossing van kalium-t-butoxyde (3,36 g) in droge THF (30 ml) in een droge stikstofatmosfeer werd (H-carboxybutyl)tri fenylfos foniumbrcmide (6,65 g) toege-20 voegd en het mengsel werd gedurende 15 minuten bij omgevings- ’ temperatuur geroerd. Een oplossing van intermédiair X (1,58 g) in THF (10 ml) werd toegevóegd en het roeren werde gedurende nog 2 uren bij 0°C voortgezet. Het mengsel werd uitgegoten in water (50 ml) en het THF werd in vacuo verwijderd. De resi-25 duale oplossing werd ingesteld op een pH van 6 met fosfaatbuffer en geëxtraheerd met CH2C12 (3 x 100 ml). De gecombineerde extracten, werden gedroogd en behandeld met etherische diazo-* methaan. Door concentrering verkreeg men een olie, die werd opgelost in pyridine (29 ml) én'behandeld met azijnzuuranhydride 30 (1^,5 ml}) De oplossing werd gedurende 22 uren bij omgevings temperatuur geroerd. De oplosmiddelen werden afgedampt en het residu in 8$’s NaHCO^ (250 ml) werd geëxtraheerd met CHgClg (3 x 100 ml). De gecombineerde extracten werden gedroogd en ingedampt en het residu gezuiverd door chromatografie (L), waar-35 bij de titelverbinding werd verkregen als een olie (1,12 g).

Aan een geroerde oplossing van intermediair XI (2,7 g) en NaH (^6^'s dispersie in olie, 1,32 g) in droge DMF (20 ml) in een stikstofatmosfeer bij 0°C werd (broommethyl)-20 l,1f-bifenyl (6,25 g) toegevoegd. Na 3 uren bij 20°C werd het · o mengsel voorzichtig uitgegoten in fosfaatbuffer met een pH van 6 (100 ml) en geëxtraheerd met CH^Clg (k x 50 ml). De gecombineerde extracten werden gewassen met water (2 x 100 ml), gedroogd en ingedampt. Het residu in 9:1 methanol-zwavelzuur 25 (50 ml) werd gedurende 0,5 uur bij 20°C geroerd. Een geroerde suspensie van intermédiair IV (1,5 g) in aceton (15 ml) werd behandeld met triethylamine (1,6 ml).
De oplossing werd af gekoeld tot -10°C en isobutylchloorformi-15 aat (1,3 ml) werd toegevoegd. 8103199 i t· -f -3^- DMSO (0,1*9 uil) werd druppelsgewijs toegevoegd aan een afgekoelde (-78°C), geroerde oplossing van oxalylchloride (0,3 ml) in CHgCl^ (10. ml) in een stikstofatmosfeer. Na ongeveer 10 minuten werd een oplossing van intermediair XVII g 5 (0,67 g) in CHgClg (20 ml) over een periode van 0,5 uur toege- vaegd. De organische laag werd afgescheiden, gedroogd en ingedampt en het residu werd gezuiverd door chroma-tografie (F), waarbij de titelverhinding (0,53 g) werd verkregen; smp. - 9H-96°C.

Deze kunnen worden bereid door directe samenper-20 sing of natte granulering. De directe samenpersingsmethode geniet de voorkeur, maar is mogelijker wijze niet in alle gevallen geschikt, omdat dezè afhankelijk is van de dosis-hoeveelheid en de fysische eigenschappen van het actieve bestanddeel.

Het actieve bestanddeel wordt gezeefd door een -6 30 zeef van 250 m , gemengd met de excipiëntia en samengeperst onder gebruikmaking van ponzen van 10,0 mm. Tabletten met andere sterkten' kunnen worden bereid door verandering van het samenpersingsgewicht en door toepassing van geschikte ponzen.

Het actieve bestanddeel wordt gezeefd door een zeef van 250 en gemengd met.de lactose, het zetmeel en de voor-gegelatiniseerde zetmeel. De gemengde poeders worden bevochtigd met gezuiverd water, korrels worden vervaardigd, gedroogd, ge-10 zeefd en gemengd met het magnesiumstearaat. De gesmeerde korrels worden samengeperst tot tabletten als beschreven voor het di-rekte samenpersingsrecept.
De tabletten kunnen met een film worden bekleed onder gebruikmaking van geschikte filmvormende materialen, bij-15 voorbeeld methylcellulose of hydroxypropylmethyicellulose, onder toepassing van standaard-technieken. Anderzijds kunnen de tabletten met suiker worden bekleed.

25 Het actieve bestanddeel wordt gezeefd door een zeef van 250 m”^ en gemengd met de andere materialen. Het mengsel wordt afgevuld in nummer II harde gelatine-capsules onder toepassing van een geschikte vulmachine. Andere doseringen kunnen worden bereid door verandering van het vulgewicht en, indien 30 noodzakelijk, verandering van de capsule-grootte om een geschikte vulling mogelijk te maken.

Gefractioneerde kokosnootolie tot 5»00 ml.
Het aluminiummonosteraat wordt gedispergeerd in 5 ongeveer 90% van de gefractioneerde kokonootolie. De resulterende suspensie wordt onder roeren verhit op 115°C en vervolgens af gekoeld. De smaak- en kleurstoffen worden toegevoegd en het actieve bestanddeel en de sucrose worden op geschikte wijze gedispergeerd. De suspensie wordt op volume gebracht met de res-10 terende gefractioneerde kokosnootolie en gemengd.

Een steriele presentatie van het actieve bestand-15 deel in een ampul of flesje samen met een ampul, bevattende een geschikt vehikel. De eerste kan worden bereid door (a) afvullen van een steriel materiaal in flesjes onder aceptische omstandigheden en (b) vriesdrogen van een steriele oplossing van het actieve bestanddeel onder aceptische omstandigheden.

. of (b) water voor injectie B.P, bevattende (i) natriumchloride tér instelling van de toniciteit van de oplossing en/of (II) bufferzouten of verdund zuur of alkali teneinde het oplossen van het actieve bestanddeel te vergemakkelijken.
25 Het vehikel wordt bereid, geklaard en afgevuld in ampullen met een geschikte grootte, die afgesloten worden door smelting van het glas. Het vehikel wordt gesteriliseerd door verhitting in een autoclaaf onder toepassing van êên van de aanvaarde cycli.

Claims (10)
1. Verbindingen met de algemene formule I, waarin R voorstelt (i} recht of vertakt C^ ^ alkyl, gesubstitueerd door (a) fenyl ^eventueel gesubstitueerd door C^_g alkyl, C,__^ cycloalkyl, fenylalkyl met een C^ ^ alkyl-gedeelte, thienyl of 5 fenyl (eventueel gesubstitueerd door C^ ^ alkyl, C^^ alkoxy of fenyl)^J , (b) thienyl [eventueel gesubstitueerd door C^_g alkyl, C^_g alkoxy, C^_^ cycloalkyl of fenyl (eventueel gesubstitueerd door C.j 2 alkyl, C^ ^ alkoxy of halogeen)]] , of (c) naftyl (e-ventueel gesubstitueerd door C^ ^ alkyl of C^ ^ alkoxy), of 10 (ii) cinnamyl, R^ voorstelt een waterstofatoom, een methylgroep, O O -(CH^OR3 (waarin n 2-5 is en R3 voorstelt C^ ^ alkyl) of -CHR (CEL) C00H (waarin R een waterstofatoom of een methylgroep d m is en m 0-2 is), W voorstelt recht of vertakt alkyleen, X voorstelt cis of trans -CH=CH of -CH^CH^-, Y voorstelt een 15 verzadigde heterocyclische aminogroep (vastgehecht aan de cy- clopentaan-ring via het stikstofatoom), die 5-8 ring-leden be- 5 vat en (a) eventueel in de ring bevat -0-, -S-, -SCL-, of -NR C ^ (waarin R3 voorstelt een waterstofatoom, C^ ^ alkyl of araiyl met een C^ ^ alkyl-gedeelte), en/of (b) eventueel is gesubsti-20 tueerd door een of meer C^_^ alkylgroepen, en de fysiologische aanvaardbare zouten en solvaten daarvan.
2. Verbindingen volgens conclusie 1, met het kenmerk, dat Y voorstelt morfolino, piperidino, homomorfolino, thiamor-folino of 1,1-dioxothiamorfolino.
3. Verbindingen volgens conclusies .1 of 2 , met het kenmerk, dat X voorstelt cis-CH=CH. h. Verbindingen volgens êên of meer der voorgaande conclusies, met het kenmerk, dat W voorstelt -CH^CH^CHg-.
5. Verbindingen volgens een of meer der voorgaande 2 30 conclusies, met het kenmerk, dat R een waterstofatoom voorstelt.

Verbindingen volgens conclusie 5» met het kenmerk:, dat r"* voorstelt een gesubstitueerde of ongesubstitueerde fenyl (C.^) alkyl of naftylgroep als gedefinieerd in conclusie 8103 199 ' \ - 38 - “ 'Γ V I (anders dan een thienylfenylalkylgroep) en X voorstelt cis " of trans -CH=CH-. Verbindingen volgens conclusies 1-5, met het kenmerk, dat R voorstelt fenyl (C^_^) alkyl, waarin de . 5 fenylgroep -gesubstitueerd door thienyl' of fenyl, waarbij de ' - laatste fenylgroep eventueel gesubstitueerd is door alkyl of alkoxy, of voorstelt een fenylthienylalkylgroep, waarin het alkylgedeelte. 1-3 koolstofatomen bevat.
8- Verbindingen volgens conclusie 1, met het 10 kenmerk, dat W voorstelt -Cï^CI^CI^··, X voorstelt cis -CH=CH-, Y voorstelt morfolino of piperidino, voorstelt benzyl, waarin de fenylgroep is gesubstitueerd door fenyl, tolyl of 2 methoxyfenyl, en R voorstelt een waterstofatoom, en de fysiologische aanvaardbare zouten en solvaten daarvan.
9. Een farmaceutisch preparaat, bevattende een verbinding als gedefinieerd in conclusie 1 en één of meer farmaceutische dragermaterialen.

Werkwijze ter bereiding van een verbinding als gedefinieerd in conclusie 1, met het kenmerk, dat 20 men (a) een overeenkomstige hydroxyverbinding met de formule II oxydeert, (b) een overeenkomstige verbinding, waarin X een ethiengroep.is, reduceert, (c) een reaktief derivaat van een overeenkomstig carbonzuur met de formule IX amideert, (d) bij de bereiding van een verbinding, waarin X voorstelt -CI^C^··, 25 een overeenkomstige verbinding, waarin X voorstelt -CH=CH-, katalytisch hydrogeneert of (e) bij de bereiding van een zout een amide-met de formule I behandelt met een zuur of, indien geschikt, met een base. 11., Verbindingen, farmaceutische preparaten 30 en werkwijzen als beschreven in de beschrijving en/of voor-. beelden. f r 35_________________ ! ί 8103199 'V __ s OR1 J. j:h2xwconhr2
