Description
Titel: Functioneel serumeiwitproduct voor toepassing in kindervoeding en therapeutische samenstellingen, en werkwijzen ter bereiding daarvan.
De uitvinding heeft betrekking op een serumeiwitproduct, geschikt als ingrediënt voor voedingsmiddelen en therapeutische samenstellingen, in het bijzonder kinder- en babyvoedingen. De uitvinding voorziet tevens in een werkwijze voor de bereiding van het serumeiwitproduct, gebaseerd op 5 microfiltratie van melk van ruminanten, en de toepassing daarvan in voedingsmiddelen en therapeutische samenstellingen.
Als moedermelk in onvoldoende mate voorhanden is, of indien voeding met moedermelk om andere redenen niet mogelijk of niet gewenst is, wordt kindervoeding op basis van koemelk in het algemeen als een goed 10 alternatief beschouwd. Omdat koemelk en moedermelk significant verschillen in samenstelling, in het bijzonder eiwitsamenstelling, is er al veel onderzoek verricht om de samenstelling van kindervoeding zo goed mogelijk die van moedermelk te laten benaderen. Dit proces wordt ook wel het “humaniseren” van koemelk genoemd. Uitgangspunt is dan dat de 15 specifieke samenstelling van moedermelk de gewenste voedingskundige functionaliteiten voor het kind met zich meebrengt.
Voor wat betreft de voedingskundige functionaliteit van moedermelk is steeds meer kennis beschikbaar. Een recente ontwikkeling op dit gebied is het bekend worden van het belang van een juiste 20 beschikbaarheid van het aminozuur proline, via de voeding, voor de darmwandrijping na de geboorte, danwel ter preventie en behandeling van overmatige of ongewenste permeabiliteit van de darmwand, respectievelijk een goede sluiting van de tight junctions. Dit is onder meer beschreven in NL-1023239, NL-1025900 en NL-1027262. Proline dient als precursor voor 25 de vorming van polyaminen, die in het lichaam worden gesynthetiseerd met 1033698 2 ornithine als tussenproduct. Naast proline is ook glutamaat en arginine voor deze vorming nodig. Polyaminen hebben vervolgens de bovengenoemde positieve effecten op de darmwand. Deze effecten zijn niet alleen van belang voor kinderen, maar ook voor zieke volwassenen.
5 Daarnaast kan worden gewezen op WO 01/58283, waarin het belang van een goede voorziening van glutamaat en/of een precursor voor glutamaat aan de orde komt voor de behandeling of preventie van hyperpermeabiliteit of ongewenste permeabiliteit van de darmwand. WO 01/58283 geeft ook het belang aan van de beschikbaarheid van polyaminen 10 en/of precursors voor polyaminen, zoals proline. Verder beschrijft J. Nutr. Biochem. 15, 2004, 442-451 dat in prematuur geboren kinderen de synthese van intestinaal citrulline en arginine nog beperkt is, met als één van de mogelijke gevolgen een tekort aan polyaminen. Hieruit volgt dat een goede prolinevoorziening ook van belang is voor prematuren.
15 In de kindervoedingen van NL-1023239, NL-1025900 en NL- 1027262 wordt een voldoende hoog proline gehalte bereikt door het verrijken van een koemelkeiwitfractie die rijk is aan wei-eiwitten, met proline in vrije aminozuurvorm. Met name voor kindervoeding is het echter van belang dat de gewenste samenstelling zo veel mogelijk wordt bereikt op 20 basis van melkcomponenten, dus zonder de toevoeging van melkvreemde componenten. Daarnaast is het een belangrijk streven om niet alleen de aminozuursamenstelling, maar ook de eiwitsamenstelling van de kindervoeding zo veel mogelijk op die van moedermelk te laten lijken. Dit impliceert dat proline (en eventueel andere aminozuren) bij voorkeur niet 25 als vrij aminozuur, maar in de vorm van eiwit in een kindervoeding aanwezig dient te zijn.
De onderhavige uitvinding heeft als doel het verschaffen van een melkeiwitproduct dat geschikt is als eiwitbron in een voedingsmiddel of therapeutische samenstelling. In het bijzonder beoogt de uitvinding een 30 melkeiwitproduct te verschaffen waarmee een kinder- of babyvoeding kan 3 worden bereid die zo goed mogelijk de voedingskundige functionaliteit, zoals de aminozuur- en eiwitsamenstelling, van moedermelk benadert. Voorts beoogt de uitvinding een werkwijze te verschaffen die de bereiding van een dergelijk melkeiwitproduct op een industriële schaal aantrekkelijk maakt.
5 Verrassenderwijs is gevonden dat bovengenoemde doelstellingen bereikt kunnen worden door het toepassen van een werkwijze waarbij melk wordt gemicrofiltreerd met een membraan met een porie grootte tussen 0.3en 0.5 pm en bij een temperatuur tussen 8 en 20°C. Een melkeiwitproduct verkrijgbaar volgens een werkwijze van de onderhavige 10 uitvinding wordt gevormd door het microfiltratiepermeaat, dat qua eiwitsamenstelling bestaat uit serumeiwitten (>60%) en caseïne-eiwitten (<40%) en waarvan de caseïne-eiwitten uit minimaal 75% β-caseïne bestaan. Het betreft hier een prolinerijk serumeiwitproduct door de aanwezigheid van het prolinerijke β-caseïne. Het prolinegehalte van het product volgens 15 onderhavige uitvinding bedraagt 5 tot 10 g proline per 100 g eiwit en is daarmee vergelijkbaar aan het gehalte van moedermelk. Dit maakt toevoeging aan kindervoeding van proline in vrije aminozuurvorm en als melkvreemde component overbodig. Daarnaast wordt hiermee beter voldaan aan de wens om de eiwitsamenstelling van kindervoeding beter op die van 20 moedermelk te laten lijken.
De uitvinding verschaft derhalve een werkwijze voor de bereiding van een serumeiwitproduct, omvattende de bereiding van een permeaat door middel van het microfïltreren van koemelk met een membraan met een poriegrootte 25 tussen 0.3 en 0.5 pm, bij een temperatuur tussen 8 en 20°C. Het gebruik van microfiltratie om caseïne- en serumeiwitten in melk van elkaar te scheiden is op zich bekend, evenals de toepassing van het microfiltratiepermeaat na verdere opwerking in kindervoeding. EP-1133238 beschrijft bij voorbeeld het winnen van serumeiwitten uit melk door middel 4 van microfiltratie van melk in een conventionele cross-flow micro filtratie-installatie bij een temperatuur van 50-55°C.
Microfiltratie in een werkwijze van onderhavige uitvinding wordt daarentegen uitgevoerd bij een temperatuur tussen 8 en 20°C, bij voorbeeld 5 8 tot 15 °C, zoals bij 10 tot 12°C. Het is gebleken dat een dergelijk temperatuurbereik bijzonder geschikt is voor de isolatie van beta-caseïne in het permeaat. Bovendien vereist het geen bijzondere en/of kostbare maatregelen, zoals vergaande koeling of verhitting, om het proces op industriële schaal toe te passen. Microfiltratietemperaturen lager dan 8°C of 10 hoger dan 20°C hebben als nadeel dat dit op grote schaal lastig realiseerbaar is en relatief duur is.
Een werkwijze volgens de uitvinding is niet bekend uit de stand der techniek. US 5,169,666 openbaart microfiltratie van melk bij een lagere temperatuur en onder toepassing van een kleinere poriegrootte (0.1 of 0.2 15 pm). WO 96/08115 betreft het scheiden van serum- en wei-eiwitten uit ondermelk middels microfiltratie en ultrafiltratie. Net als in US 5,169,666 wordt de voorkeur gegeven aan membranen met een relatief kleine poriegrootte, namelijk 0.07-0.2 pm. De temperatuur ligt typisch tussen 5 en 60°C en bij voorkeur tussen 10 en 50°C. Alle voorbeelden WO 96/08115 20 hebben betrekking op microfiltratie bij 50 °C met een poriegrootte van 0.1 of 0.2 pm. WO 94/13148 beschrijft microfiltratie van rauwe melk onder toepassing van een keramische membraan met een poriegrootte van ongeveer 0.1 pm bij een temperatuur tussen 40 en 50°C, teneinde een serumalbuminegehalte van 10% of hoger te verkrijgen.

Als uitgangsmateriaal kan melk van ruminanten worden gebruikt, zoals koemelk, geitenmelk, kamelenmelk, ezelinnenmelk, buffelmelk, schapenmelk, paardenmelk of lamamelk. Veelal betreft het koemelk, bij voorkeur de vetarme fractie van de rauwe melk (magere melk of ondermelk 5 genoemd). Deze kan volgens standaard methode worden bereid, bij voorbeeld door het centrifugeren van rauwe melk gevolgd door het thermiseren om het initiële kiemgetal van de melk te verlagen.
Voor de microfiltratie kan elke gebruikelijke inrichting voor cross-5 flow microfiltratie worden toegepast. Zo kan bij voorbeeld gebruik worden gemaakt van een spiraalgewonden microfiltratiemembraan, bij voorbeeld zoals beschreven in EP-A- 1673975. Bij voorkeur wordt een processysteem met meerdere spiraalgewonden modules gebuikt. Het is gebleken dat het behulpzaam is dat er in het cross-flow microfiltratieproces maatregelen 10 worden genomen ter reductie van de transmembraandruk over het membraan, op zodanige wijze dat de transmembraandruk maximaal 2.5 bar bedraagt. Daarom wordt bij voorkeur de transmembraandruk tijdens de microfiltratie in een werkwijze volgende de uitvinding relatief laag wordt gehouden, dat wil zeggen maximaal 2.5 bar. Goede resultaten wat betreft de 15 eiwitsamenstelling van het permeaat zijn bij voorbeeld verkregen bij een maximale transmembraandruk van 2 bar. De gemiddelde transmembraandruk kan variëren, en is bij voorbeeld 1.5 of 1.3 bar.
In plaats van reductie van de transmembraandruk kan een andere oplossing de toepassing van microfiltratiemembranen met een 20 gradiënt in de porositeit of dikte van de membraanlaag zijn.
In een werkwijze volgens de uitvinding kunnen standaard microfiltratiemembranen met een poriegrootte tussen 0.3 en 0.5 pm worden toegepast. Zoals in het algemeen bekend is, heeft de poriegrootte invloed op de uiteindelijke eiwitsamenstelling van het permeaat en het retentaat. In 25 het licht van de onderhavige uitvinding blijkt de poriegrootte onder meer invloed te hebben op zowel de serumeiwit/caseïne-verhouding en op het aandeel beta-caseïne in de caseïne-fractie. In een uitvoeringsvorm wordt gebruik gemaakt van een membraan, bij voorbeeld een spiraalgewonden membraan, met een poriegrootte tussen 0.3 en 0.5 pm, zoals 0.45 pm.

In een specifieke uitvoeringsvorm verschaft de uitvinding een werkwijze voor de bereiding van een serumeiwitproduct, omvattende de bereiding van een permeaat door middel van het microfiltreren van magere (koe)melk met een membraan met een poriegrootte tussen 0.3 en 0.5 pm, bij 5 een temperatuur tussen 8 en 15°C, bij voorbeeld 10-14°C, waarbij de transmembraandruk tijdens microfiltratie maximaal 2.5 bar is, bij voorkeur maximaal 2 bar.
10 Na de uitvoering van de microfiltratiestap kan het microfiltratiepermeaat verder worden behandeld volgens één of meer gangbare processen, zoals ultrafiltratie, nanofiltratie, ionenwisseling, electrodialyse, omgekeerde osmose, ontzouting, indampen en sproeidrogen. Bij voorbeeld worden Na en K verwijderd.

Een verder aspect van de uitvinding betreft het verschaffen van een serumeiwitproduct verkrijgbaar volgens de werkwijze van de uitvinding. Afhankelijk van de microfiltratiecondities (bij voorbeeld poriegrootte, temperatuur, transmembraandruk) kan de verhouding serumeiwit/caseine 20 en/of het gehalte aan proline-rijk beta-caseïne variëren. De uitvinding voorziet bij voorbeeld in een serumeiwitproduct bevattende minimaal 60% serumeiwit en maximaal 40% caseïne, en waarbij de caseïnefractie voor ten minste 75% bestaat uit beta-caseïne. Het serumeiwitproduct volgens de onderhavige uitvinding vindt zijn toepassing in baby- en kindervoeding en 25 therapeutische samenstellingen. Doorgaans wordt een serumeiwit:caseïne verhouding van ongeveer 60:40 beoogd in kindervoeding om de eiwitsamenstelling zo goed mogelijk in overeenstemming met moedermelk te brengen. Bij toepassing van het serumeiwitproduct volgens de onderhavige uitvinding kan dit worden bereikt door naast het 30 serumeiwitproduct gebruik te maken van een caseïnebron zoals ondermelk, 7 caseïnaat, zuur caseïne of melkeiwitconcentraat bereid middels ultra- of microfiltratie. Een specifiek voordeel van het serumeiwitproduct volgens de uitvinding is dat de keuze van de caseïnebron vrij is voor de producent en daarmee bijvoorbeeld af kan hangen van de beschikbaarheid op de 5 betreffende plaats en tijd. Mede in verband hiermee bevat het serumeiwitproduct volgens onderhavige uitvinding bij voorkeur minimaal 65% serumeiwit en maximaal 35% caseïne, en bij nadere voorkeur minimaal 70% serumeiwit en maximaal 30% caseïne.
Onder toepassing van een werkwijze van de uitvinding kan binnen de 10 caseïne fractie een zeer hoog gehalte aan beta-caseïne worden bereikt. Bij voorbeeld verschaft het een serumeiwitconcentraat met een caseïnegehalte van 5,5% en een serumeiwitgehalte van 33,4% (resulterend in een eiwitsamenstelling met 86% serumeiwit en 14% caseïne), waarbij de caseïnefractie voor 95% uit beta-caseïne bestaat en daarnaast 5% alfa-15 caseïne. Een ander voorbeeld betreft een serumeiwitproduct met 68% serumeiwit en 32% caseïne, waarvan 79% beta-caseïne.
Zoals eerder genoemd heeft een serumeiwitproduct volgens de uitvinding een aantal belangrijke kenmerken wat betreft de functionele 20 voedingswaarde. In het bijzonder benadert het op verschillende punten de eiwitsamenstelling van moedermelk. Zo ligt het proline-gehalte van een serumeiwitproduct tussen 5 tot 15 gram proline per 100 g eiwit, bij voorkeur 6 tot 11 gram proline per 100 g eiwit. In tegenstelling tot bekende (ingrediënten voor) (kinder)voedingsmiddelen met dergelijke gehalten aan 25 proline, is het proline niet aanwezig als vrij aminozuur maar, net zoals in moedermelk, als onderdeel van een polypeptide uit melk. Supplementatie met proline in de vorm van vrije aminozuren is derhalve niet nodig. De uitvinding voorziet daarom in een serumeiwitproduct waarbij het proline in hoofdzaak aanwezig is als onderdeel van een polypeptide. Hiermee wordt 8 voldaan aan de wens om de eiwitsamenstelling van kindervoeding beter op die van moedermelk te laten lijken.
Een andere van belang geachte ontwikkeling op het gebied van het 5 "humaniseren" van koemelk betreft het aminozuur threonine. De aanwezigheid van een relatief hoog gehalte aan threonine in kindervoeding wordt meestal gekoppeld aan de aanwezigheid van glycomacropeptide (GMP), zoals onder andere blijkt uit J. Ped. Gastr. Nutr. 32, 2001, 127-130. GMP is een afsplitsingsproduct van κ-caseïne, dat onder invloed van het 10 enzym chymosine tijdens de kaasproductie wordt gevormd. Het komt hierdoor voor in zoete wei, dat veelal wordt toegepast als wei-eiwitbron voor kindervoeding. Het oligopeptide GMP is rijk aan threonine, dat bij overdosering hyperthreoninemia kan veroorzaken in prematuren.
Er zijn een aantal oplossingen bekend om een te hoge belasting met 15 threonine te voorkomen. Dit betreft het verlagen van threonine-gehalte van de koemelkproducten. Toepassing van zure wei in plaats van zoete wei is een bekende oplossing, zoals onder meer duidelijk is uit J. Ped. Gastr. Nutr. 32, 2001, 127-130. Zure wei bevat geen GMP omdat er geen enzymatische stremming van de melk plaatsvindt in de productie van zure wei. Een 20 andere bekende oplossing betreft het verwijderen van GMP uit zoete wei alvorens dit toe te passen in kindervoeding. Dit is bekend uit EP-1048226. Een derde bekende oplossing betreft, in plaats van zoete wei, de toepassing van een serumeiwitproduct op basis van microfiltratie-permeaat van melk. Dit is beschreven in EP-1133238. Ook hier is geen enzymatische stremming 25 van de melk uitgevoerd in de productie van het betreffende serumeiwitproduct, dat een product met een relatief laag gehalte aan threonine oplevert.
Een serumeiwitproduct volgens de onderhavige uitvinding heeft gewoonlijk een threonine-gehalte tussen 4,7 en 6 gram per 100 gram eiwit, 30 waarbij het threonine net als in moedermelk aanwezig is in intacte 9 eiwitmoleculen. Zonder aan enige theorie te willen worden gebonden, stellen de onderhavige uitvinders voor dat niet zozeer het gehalte aan threonine relevant is voor de threonine-belasting van kinderen, maar in welke vorm het threonine aanwezig is. Uit J. Dairy Sci. 75, 1992, 1380-1388 dat GMP al 5 in intacte vorm, dus zonder verdere hydrolyse, opgenomen kan worden door de dunne darm. Deze snelle opname van GMP heeft mogelijk negatieve gevolgen voor de synthese van mucine in de darm, waar threonine een belangrijke aminozuurbron voor is. Dit zou pleiten voor de beschikbaarheid van threonine in een minder snel te adsorberen vorm, zodat de 10 beschikbaarheid van threonine in de darm beter is. Ook in moedermelk komt threonine in intacte eiwitmoleculen voor. Het feit dat threonine in een serumeiwitproduct volgens de uitvinding in hoofdzaak aanwezigheid is in de vorm van intacte eiwitmoleculen, in plaats van in oligopeptiden of als vrije aminozuren, zou hier aan bij kunnen dragen. Het product volgens de 15 uitvinding bevat bij voorbeeld tussen 4,7 en 6 g threonine per 100 gram eiwit, zonder dat er sprake is van een te hoge threonine-belasting. Men kan derhalve spreken over een "slow-release-threonine" gehalte in het serumeiwitproduct volgens de onderhavige uitvinding, vergelijkbaar met dat van moedermelk.
20 Nog een ander gunstige nutritionele eigenschap van een serumeiwitproduct verkrijgbaar door microfiltratie volgens de uitvinding, is dat het een relatief hoog gehalte aan vrij beschikbaar (ionisch) calcium bevat (gewoonlijk circa 600-700 mg per 100 g eiwit). Hierdoor kan reeds bij geringe verhitting van het product een maximale denaturatie van met name 25 wei-eiwitten worden bereikt. Zie bij voorbeeld EP-311795. Het resultaat is een product, dat optimaal kan worden ingezet ter preventie van allergie voor eiwitten en/of een product dat tolerantieverhogend werkt, mede dankzij de bevordering van een optimale sluiting van de "tight junctions".
Een verder aspect van de uitvinding betreft een werkwijze voor de 30 bereiding van voedingsmiddel of therapeutische samenstelling, bij voorkeur 10 een baby- of kindervoeding, onder toepassing van ten minste het serumeiwitproduct volgens de onderhavige uitvinding. De werkwijze omvat gewoonlijk de standaard stappen van het mengen van ten minste één eiwit-, lipide- en koolhydraatbron, eventueel aangevuld met mineralen, oligo-5 componenten en andere ingrediënten. Ook wordt een voedingsmiddel of therapeutische samenstelling, bij voorkeur een baby- of kindervoeding, verschaft die verkrijgbaar is volgens een dergelijke werkwijze. In vergelijking met de tot dusver meest gangbare koemelkeiwitbronnen op basis van wei-eiwit en caseïne, zoals toegepast bij de bereiding van 10 moedermelkvervangers, zijn de melkeiwitten in een serumeiwitproduct volgens de uitvinding niet of nauwelijks geassocieerd. Dit heeft onder meer als voordeel dat op basis van deze eiwitbron in combinatie met de gebruikelijke vetten een zeer stabiele en fijne emulsie gevormd kan worden. Hiermee kan de vertering van het eindproduct worden verbeterd, en/of het 15 optreden van verteringsstoornissen worden voorkomen.
In de (baby- en kindervoeding en therapeutische samenstellingen volgens de onderhavige uitvinding is het ook mogelijk dat deze naast het serumeiwitproduct nog andere eiwitten bevat zoals wei-eiwitten, a-20 lactalbumine, lactoferrine en plantaardige eiwitten, zoals uit soja of tarwe. In een uitvoeringsvorm betreft de werkwijze de bereiding van een baby- of kindervoeding, bij voorkeur met een met serumeiwit:caseïne verhouding van ongeveer 60:40 op basis van gewicht. Als additionele eiwitbron kunnen ondermelk, caseïnaat, zuur caseïne of melkeiwitconcentraat wordt 25 toegepast. Bovendien kunnen zowel het serumeiwitproduct als andere eiwitten een hydrolysestap hebben ondergaan. Dit is bijvoorbeeld gebruikelijk voor het voorkomen van allergieën, waarbij eiwit wordt gehydrolyseerd onder invloed van pancreasenzymen.

Het is verder gebruikelijk dat aan baby- en kindervoeding en therapeutische samenstellingen koolhydraten, zoals lactose en oligosacchariden, lipiden en ingrediënten als vitaminen, aminozuren, mineralen, taurine, carnitine, nucleotiden en polyaminen, en antioxidanten als BHT, ascorbylpalmitaat, 5 vitamine E, a- en β-caroteen, luteine, zeaxanthine, lycopeen en lecithine. De lipiden zijn meest van plantaardige oorsprong. Daarnaast kan het voedingsmiddel of de therapeutische samenstelling worden verrijkt met meervoudig onverzadigde vetzuren, zoals gamma-linoleenzuur, dihomo-gamma-linoleenzuur, arachidonzuur, stearidonzuur, eicosapentaeenzuur, 10 docosahexaeenzuur en docosapentaeenzuur. Met het oog op een goede ontwikkeling van de darmflora kunnen probiotica worden toegevoegd, zoals lactobacillen en/of bifido-bacteriën, alsmede prebiotica. Een voorkeurscombinatie van probiotica betreft bijvoorbeeld Bifidobacterium lactis met L. casei, L. paracasei, L. salivarius of L. reuter. Voorbeelden van 15 prebiotica betreffen onder meer fuco-, fructo- en/of galacto-oligosacchariden, zowel met korte als met lange ketens, (fuco)sialyloligosacchariden, vertakte (oligo)sacchariden, sialinezuur-rijke melkproducten of derivaten daarvan, inuline, johannesbroodpitmeel, gommen, die al dan niet zijn gehydrolyseerd, vezels, eiwithydrolysaten, nucleotiden etc.

Een voedingsmiddel of therapeutische samenstelling volgens de uitvinding kan met voordeel worden toegepast ter bevordering van de rijping van de darmwand en/of het een goede sluiting van de “tight junctions”.
Ook is gebleken dat het de mucusvorming van de darmwand kan stimuleren 25 en/of de kolonisatieweerstand van de darmflora kan bevorderen. De uitvinding verschaft hiermee een "weerstandverhogend' eiwitconcentraat. Zonder aan enige theorie te willen worden gebonden, stellen de onderhavige uitvinders voor dat, waar threonine een belangrijke aminozuurbron is voor een optimale synthese van mucine, het met name de peptiden welke uit 30 enzymatische vertering van beta-caseïne resulteren, welke de mucus- 12 afscheiding bevorderen. Dit betekent dat een serumeiwitproduct verrijkt aan beta-caseïne volgens de uitvinding met een threonine-gehalte van 4.7-6 g/100 g, niet alleen geen nadelen kent (aangezien het als "slow-release-threonine" aanwezig is) maar dat het tevens kan bijdragen aan een optimale 5 mucusvorming op het niveau van de darmwand. Dit kan zowel de inductie als het behoud van mucusvorming betreffen. Een serumeiwitproduct volgens de uitvinding kan derhalve, door gestimuleerde mucusvorming, een gunstige invloed hebben op de opbouw van weestand tegen pathogenen. Daarenboven is gevonden, dat een serumeiwitproduct volgens de uitvinding 10 een specifiek gunstige werking heeft op de darmflora in vergelijking met traditioneel toegepaste wei-eiwitten uit zoete wei. Zie voor meer details Voorbeeld 4 hieronder.
De uitvinding wordt geïllustreerd aan de hand van de volgende 15 Voorbeelden.
Voorbeeld 1 (ver gelijkingsvoor beeld)
Er is ondermelk bereid door het centrifugeren van rauwe melk en 20 vervolgens de ondermelk te thermiseren gedurende 15 s bij 67°C. Deze ondermelk is gemicrofiltreerd in een proces systeem met 1 spiraalgewonden module (DSS, poriegrootte 0.15 μπι, membraanoppervlak 14 m2), bij een temperatuur van 10°C en een maximale transmembraandruk van 1.8 bar (gemiddeld 1.3 bar). De ondermelk is batch-gewijs gefiltreerd tot een 25 volumereductiefactor (VRF) van 3.3. Het permeaat is vervolgens geconcentreerd door middel van ultrafiltratie (UF) en gedroogd tot een poedervormig serumeiwitconcentraat. Het caseïne gehalte van het serumeiwitconcentraat was 5.5% en het serumeiwitgehalte 33.4%, resulterend in een eiwitsamenstelling met 86% serumeiwit en 14% caseïne. 30 De caseïnefractie bestond voor 95% uit β-caseïne en daarnaast 5% a-caseïne.

De serumeiwitfractie bestond voor 24% uit α-la en voor 75% uit β-lg. De aminozuursamenstelling is weergegeven in tabel 1.

Er is ondermelk bereid door het centrifugeren van rauwe melk en vervolgens de ondermelk te thermiseren gedurende 15 s bij 76°C. Deze ondermelk is gemicrofiltreerd in een proces systeem met vier 15 spiraalgewonden modules (DSS, poriegrootte 0.45 pm, membraanoppervlak 56 m2), bij een temperatuur van 10°C en een maximale transmembraandruk 14 van 2.5 bar (gemiddeld 1.5 bar). De ondermelk is gefiltreerd tot een VRF van 3.3 in een continue proceswijze. Het permeaat is vervolgens geconcentreerd door middel van UF en gedroogd, zodat een poedervormig serumeiwitconcentraat is verkregen. Het caseïnegehalte van het 5 serumeiwitconcentraat was 15.9% en het serumeiwitgehalte 33.4%, resulterend in een eiwitfractie met 68% serumeiwit en 32% caseïne. De caseïnefractie bestond voor 79% uit β-caseïne en daarnaast 18% a-caseïne en 3% K- en γ-caseïne. De serumeiwitfractie bestond voor 25% uit α-la en voor 73% uit β-lg en 1% uit BSA. De aminozuursamenstelling is 10 weergegeven in tabel 2.

Bereid werd een kindervoeding voor zuigelingen, welke werd samengesteld zoals aangegeven in onderstaande tabel 3, uitgaand van het serumeiwitconcentraat van voorbeeld 1. Het serumeiwitconcentraat bevatte 5.6% proline en 5.2% threonine en het Na-caseïnaat 10.5% proline en 4.9% 5 threonine, uitgedrukt als percentage van het totaal ruw eiwit.

Dit voorbeeld illustreert het gunstige effect op de darmflora van een serumeiwitproduct volgens de uitvinding (hierin SPC genoemd) Oin 15 vergelijking met DEMINAL90, een conventioneel wei-eiwitproduct op basis van zoete wei.

Een serumeiwitproduct verkrijgbaar volgens een werkwijze van de uitvinding onder toepassing van een 0.45 pm membraan en DEMINAL90 werden verwerkt in media die als voedingsbron hebben gediend in pH-gecontroleerde batch culturen (resp. BATCH 2 en BATCH 3). Daarnaast 5 werden beide producten in 2 aparte media verwerkt na te zijn behandeld met proteasen. Deze media zijn ook als voedingsbron gebruikt in pH gecontroleerde batch cultures (resp. BATCH 4 en BATCH 5). De proteasen waren dusdanig geselecteerd en dusdanig geincubeerd met de eiwitproducten dat ze zo goed mogelijk de condities in het gastro-intestinale 10 deel ven het humane lichaam nabootsen, dat wil zeggen pepsine behandeling bij pH 3.0 en een behandeling met een 'pancreas-extract' bij pH 6.5. Het medium bevatte gist extract, NaHC03, KH2PO4, K2HPO4, NaCl, cysteine.HC1, MgSCL, CaCl2, hemin, Resazurine, Tween-80 en vitamine K.
De steriele, pH-gecontroleerde batchculture met medium werd 15 anaëroob gemaakt en beënt met vers verkregen baby faeces van een gezond kind, waarbij de faeces was verwerkt in een slurry op basis van PBS. De verschillende eiwitbronnen werden aan de batchcultures toegevoegd. Tevens werd een referentieculture met een minimale hoeveelheid eiwit en zonder additionele te testen eiwitbronnen (BATCH 1) bereid die tevens met 20 faecesslurry werd beënt.
De batchcultures werden gedurende 6 uren bij een temperatuur van 37°C geincubeerd, waarna monsters werden genomen die werden geanalyseerd op aanwezigheid van microbiële organismen. De analyse werd uitgevoerd door middel van DNA amplificatie en een DNA-hybridisatie 25 assay. De assay was opgezet en gevalideerd voor detectie en quantificatie van zowel Bifidobacterie genera, Lactobacillus genera en andere bacteriële soorten, zoals Escherichia coli, Clostridium dificile, Salmonella, Bifidobacterium longum, Lactobacillus casei. De hybridisatie werd uitgevoerd met fluorescente probes specifiek voor de bacteriële soorten.
30 Specifiek gebonden probes werden gekwantificeerd met een daarvoor 17 ontwikkelde scanner. Het gemeten fluorescentiesignaal werd door middel van software gecorreleerd met de hoeveelheden bacteriën. De signalen werden gecorrigeerd voor het achtergrondsignaal verkregen uit BATCH 1.
5 De gecorrigeerde signalen van de geteste batches die corresponderen met Bifidobacteriën zijn getoond in Figuur 1. Hierbij zijn de volgende coderingen gebruikt: DD = gedigesteerde DEMINAL90 DS = gedigesteerd serumeiwitproduct van de uitvinding 10 UD = ongedigesteerde DEMINAL90 US = ongedigesteerd serumeiwitproduct van de uitvinding De y-as geeft de aantallen bacteriën weer in arbitraire eenheden die overeenkomen met de fluorescentie die is gemeten na hybridisatie en standaardisatie voor elke bacteriële soort.

De figuur toont het stimulerende effect van een gedigesteerd serumeiwitproduct volgens de uitvinding in vergelijking met een conventioneel wei-eiwitpreparaat. Naar verwachting is dit gunstige effect althans voor een deel toe te schrijven aan de aanwezigheid van beta-caseïne.

Claims (16)
- 2. Werkwijze volgens conclusie 1, waarin de temperatuur 8 tot 15 °C bedraagt, bij voorkeur 10 tot 12°C.
- 3. Werkwijze volgens conclusie 1 of 2, waarbij de transmembraandruk tijdens microfiltratie maximaal 2.5 bar is, bij voorkeur maximaal 2 bar.
- 4. Werkwijze volgens een der conclusie 1-3, waarin gebruik wordt 10 gemaakt van een spiraalgewonden microfiltratiemembraan.
- 5. Werkwijze volgens een der conclusies 1-4, waarbij de poriegrootte van het membraan ongeveer 0.45 pm is.
- 6. Werkwijze volgens een der conclusies 1-5, waarin het microfïltratiepermeaat verder wordt behandeld met één of meer van de 15 volgende processen: ultrafiltratie, nanofiltratie, ionenwisseling, electrodialyse, omgekeerde osmose, ontzouting, indampen en sproeidrogen.
- 7. Serumeiwitproduct verkrijgbaar volgens de werkwijze van een der conclusies 1-6.
- 8. Serumeiwitproduct volgens conclusie 7, bevattende minimaal 60% 20 serumeiwit en maximaal 40% caseïne, en waarbij de caseïnefractie voor meer dan 75% bestaat uit beta-caseïne,
- 9. Serumeiwitproduct volgens conclusie 8, bevattende minimaal 65% serumeiwit en maximaal 35% caseïne.
- 10. Serumeiwitproduct volgens een der conclusies 7-9, bevattende 6 tot 25 11 gram proline per 100 g eiwit. II. Serumeiwitproduct volgens een der conclusies 7- 10, waarbij het aanwezige proline in hoofdzaak aanwezig is in de vorm van intacte eiwitten. 1033698
- 12. Serumeiwitproduct volgens een der conclusies 7-11, bevattende meer dan 4,7 gram threonine per 100 g eiwit, bij voorkeur tussen 4,7 en 6 gram threonine per 100 g eiwit, in hoofdzaak in de vorm van intacte eiwitten.
- 13. Werkwijze voor de bereiding van voedingsmiddel of therapeutische samenstelling, bij voorkeur een baby- of kindervoeding, omvattende het mengen van ten minste één eiwit-, lipide- en koolhydraatbron, eventueel aangevuld met mineralen, oligo-componenten en andere ingrediënten, waarbij ten minste het serumeiwitproduct volgens een der conclusies 7-12 10 wordt toegepast.
- 14. Werkwijze volgens conclusie 13 voor de bereiding van een baby- of kindervoeding, bij voorkeur met een met serumeiwitrcaseïne verhouding van ongeveer 60:40 op basis van gewicht.
- 15. Werkwijze volgens conclusie 13 of 14, waarbij als additionele 15 eiwitbron ondermelk, caseïnaat, zuur caseïne of melkeiwitconcentraat wordt toegepast.
- 16. Voedingsmiddel of therapeutische samenstelling verkrijgbaar volgens de werkwijze van een der conclusies 13-15.
- 17. Toepassing van een voedingsmiddel of therapeutische 20 samenstelling volgens conclusie 16, ter bevordering van de rijping van de darmwand en/of een goede sluiting van de “tight junctions”.
