Abstract
Un metodo para la identificacion de compuestos que se enlazan a la polimerasa VHC que comprende las etapas de: poner en contacto la polimerasa VHC o un analogo de la misma con una Formula de prueba I: (ver formula) en donde A es O, S, N, NR1, o CR1, en donde R1 es como se define en la presente; ---representa ya sea un enlace doble o sencillo; R2 se selecciona de: H, halogeno, R21, OR21, SR21, COOR21, S02N (R22)2, N( R22)2, CON (R22)2, NR22C (O) R22 o NR22C(O)NR22 en donde R21 y cada una de R22 esta definida en la presente; B es NR3 o CR3 en donde R3 es como se define en la presente; con la condicion de que cuando A no es N, luego una de A o B es ya sea CR1 o CR3, K es N o CR4, en donde R4 esta definida en la presente; L es N o CR5, en donde R5 tiene la misma definicion como R4 como se ha definido anteriormente. M es N o CR7, en donde R7 tiene la misma definicion como R4 como se ha definido anteriormente; R5 es C(Y1)Z en donde Y1 es O o S; y Z es N(R6a)R6 o OR6, en donde R6a es H o alquilo o NR61R62 en donde R61 y R62 estan definidas en la presente; y R6 es H, alquilo, cicloalquilo, alquenilo, Het, alquil-arilo, alquil-Het o R6 es En donde R7, R8 y Q son como se han definido en la presente; Y2 es O o S; R9 es H, alquilo de (C1-6), cicloalquilo de (C3-7) o alquilo de (C1-6)-cicloalquilo de (C3-7), arilo, Het, alquilo de (C1-6)-arilo o alquilo de (C1-6)-Het los cuales estan opcionalmente sustituidos con R90; o R9 se enlaza covalentemente a ya sea R7 o R8 para formar un heterociclo de 5- o 6-miembros; o una sal de la misma; en donde la prueba comprende una etiqueta detectable que se une cualquier posicion adecuada, con lo cual la prueba se enlaza a una polimerasa VHC o a un analogo de la misma y que es capaz de ser desplazada por un inhibidor de la misma.
