Abstract

Las células endoteliales de la córnea humana y/o las células precursoras endoteliales de la córnea humana se conservan con una alta tasa de supervivencia de estas células manteniéndose, y se suprime la tasa de aparición de células contaminadas en el cultivo posterior a la conservación. Un método de almacenamiento de células endoteliales de la córnea humana y/o células precursoras endoteliales de la córnea humana se caracteriza porque las células endoteliales de la córnea humana y/o las células precursoras endoteliales de la córnea humana que han sido cultivadas utilizando un medio de cultivo que contiene un inhibidor de ROCK, y en el que el contenido de factor de crecimiento epidérmico (EGF) es menor que una concentración que causará una transformación se recolectan en un tiempo en que se cumple cualquiera de o una pluralidad de las condiciones (a) ~ (d) que se indican a continuación, y se colocan en un estado de suspensión y luego de conservado. (a) Durante un período inmediatamente después de que una morfología de las células endoteliales de la córnea humana y/o de las células precursoras endoteliales de la córnea humana haya cambiado de una forma fusiforme a una forma poligonal o una forma elíptica cuya proporción eje mayor-eje menor es cercana a 1, hasta inmediatamente antes de que los límites entre las células se vuelvan indistintos. (b) Durante un período desde que un nivel de expresión de CD44 se vuelve igual o inferior a la mitad del valor máximo observado después del subcultivo más reciente hasta que este nivel de expresión alcanza una meseta. (c) Cuando la densidad celular de las células endoteliales de la córnea humana y/o de las células precursoras endoteliales de la córnea humana no sea inferior a 900 células/mm2 ni superior a 2500 células/mm2. (d) Cuando el número de días de cultivo desde el subcultivo más reciente no sea inferior a 4 días ni superior a 14 días.
