Abstract
본 발명은 색소체 2-옥소글루타레이트/말레이트 트랜스로케이터를 코딩하는 DNA 분자에 관한 것으로서, 상기 DNA 분자는, 박테리아, 진균류, 유전전환성 식물 세포 및 상기 DNA 분자를 함유하는 유전전환성 식물 뿐 아니라, 특히 스피나시아 올레라세아로부터 얻어서, 색소체 2-옥소글루타레이트/말레이트 트랜스로케이터를 코딩하는 것을 특징으로 한다.

도 1은 효모에서 발현되는 재조합 2-옥소글루타레이트/말레이트 트랜스로케이터의 수송활성의 재구성을 나타낸다. 상기 도면은 에스, 폼베(S.pombe) 세포막을 함유하는 재구성 리포좀에서 〔14C〕말레이트의 흡수를 나타낸다,
도 2는 플라스미드 pEVP11-211(도 2A)및 플라스미드 pBinAR-211(도 2B)의 클로닝을 도식적으로 나타낸다.
도 3은 2-옥소글루타레이트/말레이트 트랜스로케니터 전구체 단백질을 엽록체로의 표적 및 내부 엔버로프막내에서 성숙한 단백질의 에너지-의존 삽입을 나타내는 것이다.
Claims (18)
- (a) 서열번호 1 로 나타난 아미노산 서열을 갖는 단백질을 코딩하는 DNA 분자; (b) 서열번호 1로 나타난 코딩 영역을 나타내는 DNA 분자; (c)(a) 또는 (b)에서 언급된 DNA 분자에 하이브리드화되는 DNA 분자; (d) (a),(b) 또는 (c)에서 언급된 DNA 분자의 서열과 비교되는 유전 코딩의 결과 퇴화된 DNA 분자 서열; 및 (e)(a),(b),(c) 또는 (d)로 언급된 분자에 상보적인 DNA 분자로 구성된 그룹에서 선택되는 것을 특징으로 하는, 색소체 2-옥소글루타레이트/말레이트 트랜스로케이터를 코딩하는 DNA 분자.
- 제 1항에 따른 DNA 분자를 포함하고 있는 것을 특징으로 하는 재조합 DNA 분자.
- 제 2 항에 있어서, 제 1 항에 따른 DNA 서열이 진핵- 또는 원핵 세포에서의 전사를 확실하게 하는 DNA서열과 연결되어 있는 것을 특징으로 하는 재조합 DNA 분자.
- 제 3 항에 잇어서, 2-옥소글루타레이트/말레이트 트랜스로케이터의 활성을 갖는 단백질을 코딩하는 전사가능한 mRNA의 발현을 확실하게 하는 것을 특징으로 하는 재조합 DNA 분자.
- 제 3 항에 있어서, 안티-센스 효과 또는 리보자임 활성에 의하여, 하나 이상의 2-옥소글루타레이트/말레이트 트랜스로케이터의 합성을 방해하는 비-전사성 RNA를 발현시키는 것을 특징으로 하는 재조합 DNA 분자.
- 제 3 항에 있어서, 변형된 특이성을 갖는 색소체 디카르복실레이트 트랜스로케이터를 발현시키는 것을 특징으로 하는 재조합 DNA 분자.
- E. coli 균주 TG1 pBSC-211 에서 DSM 9238로 기탁된 플라스미드 pBSC-211.
- E. coli 균주 TG1 pEVP11-211 에서 DSM 9237로 기탁된 플라스미드 pEVP11-211.
- E. coli 균주 TG1 pBinAR-211 에서 DSM 9239로 기탁된 플라스미드 pBinAr-211.
- 제 1 항에 따른 DNA 분자, 제 2 항 내지 제 6 항 중 어느 한 항에 따른 재조합 DNA 분자 똔느 제 7 항 내지 제 9 항 중 어느 한 항에 따른 플라스미드를 함유하는 것을 특징으로 하는 박테리아.
- 제 1 항에 따른 DNA 분자, 제 2 항 내지 제 6 항 중 어느 한 항에 따른 재조합 DNA 분자 또는 제 8 항에 따른 플라스미드를 포함하는 것을 특징으로 하는 진균류.
- 제 2 항 내지 제 6 항 중 어느 한 항에 따른 재조합 DNA 분자, 또는 제 9 항에 따른 플라스미드를 포함하는 것을 특징으로 하는 유전전환성 식물 세포.
- 제 12 항에 따른 식물세포로부터 재생가능하거나 제 12 항에 따른 식물 세포를 포함하는 것을 특징으로 하는 유전전환성 식물.
- 제 1 항에 따른 DNA 분자에 의하여 코딩되는 것을 특징으로 하는, 색소체 2-옥소글루타레이트/말레이트의 활성을 갖는 단백질.
- 디카르복실레이트 트랜스로케이터, 특히 2-옥소글루타레이트/말레이트 트랜스로케이터 또는 글루타메이트/말레이트 트랜스로케이터의 생물학적인 활성을 갖는 폴리펩티드를 코딩하는 DNA 분자를 분리할때 제 1 항에 따른 DNA 분자를 사용하는 것을 특징으로 하는 DNA 분자의 용도.
- 원핵성 또는 진핵성 단백질을 상기 서열을 사용하여 색소체 엔버로프 막내로, 플라스미드 스트로마내로 또는 티라코이드내로 이동시키기 위해서 표적 서열로서 사용하는 것을 특징으로 하는, 제 1 항에 따른 DNA 분자 또는 그의 일부의 용도
- 일부가 2-옥소글루타레이트/말레이트 운반자의 생물학적 활성을 갖는 성숙한 단백질을 코딩하는 코딩 영역을 함유하여, 다른 세포의 부분 또는 세포막 시스템을 위한 표적 서열과 결합함으로써 성숙한 단백질을 다른 부분 또는 막 시스템내로 이동시키는데 사용하는 것을 특징으로 하는, 제 1항에 따른 DNA 분자 또는 그 분자의 일부의 용도.
- 색소체의 내부 엔버로프막을 가로질러 디카르복실레이트 또는 글루타메이트의 수송시 억제 효과를 갖는 물질을 동정하는데 사용하는 것을 특징으로 하는, 제 1 항에 따른 DNA 분자의 용도.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
