Abstract

본 발명은 연골세포에서는 발견되나 연골세포로부터 탄분화된 세포에서는 발견되지 않는 콘카나발린 A-결합 단백질에 관한 것이다. 이 단백질은 바람직하게는 엔도글리코시다제로 처리한 후 SDS-PAGE 분석에 따른 분자량이 약 76kD 또는 약 67kD이며, 하나 이상의 서열 확인 번호 1 내지 5에 나타난 아미노산 서열을 나타낸다.
본 발명의 단백질은 연골세포의 분화 및 또한 연골세포에 의한 작용 조절 및 연골성 골형성 조절에 관련된 물질이며, 연골대사 질환 및 대사성 골 질환의 치료, 예방 및 진단을 위한 신규한 유형으로서 사용될 수 있다.

제1도는 콘카나발린 A-결합 당단백질의 전기 영동 양상을 나타낸다. 제2도는 정제된 CMP의 엔도글리코시다제 분해를 나타낸다.

Claims (13)

  1. 연골세포에는 존재하나 연골세포로부터 탄분화된 세포에는 존재하지 않는 콘카나발린 A결합 단백질(CMP).
  2. 제1항에 있어서, 엔도글리코시다제로 처리한 후 SDS-PAGE로 분석한 분자량이 약 76kD 또는 약67kD인 단백질.
  3. 제1항 또는 제2항에 있어서, 서열 확인 번호 1 내지 5에 나타낸 아미노산 서열중 하나 이상을 포함하는 단백질.
  4. (a)연골세포를 분리시키고, (b) 단계(a)의 연골세포로부터 막 단백질을 분리시킨 후, (c) 콘카나발린 A-친화성 컬럼을 통해 콘카나발린 A-결합 당단백질을 분리시키며, (d) CMP를 분리시켜 수득할 수 있는 CMP.
  5. 제4항에 있어서, 단계(c)를 수행하기 전에 콘카나발린 A-결합 당단백질 이외의 막 단백질을 분리 시킨 CMP.
  6. (a)연골세포를 분리시키고, (b) 단계(a)의 연골세포로부터 막 단백질을 분리시킨 후, (c) 콘카나발린 A-친화성 컬럼을 통해 콘카나발린 A-결합 당단백질을 분리시키며, (d) CMP를 분리시키는 단계를 포함하는, CMP 분리 방법.
  7. 제6항에 있어서, 단계(c)를 수행하기 전에 콘카나발린 A-결합 당단백질 이외의 막 단백질을 분리 시키는 방법.
  8. CMP를 함유하는 약제.
  9. CMP에 대한 항체를 함유하는 약제.
  10. CMP를 함유하는 진단 보조제.
  11. CMP에 대한 항체를 함유하는 진단 보조제.
  12. 연골 대사 질환 및/또는 대사성 골 질환의 치료, 예방 및 진단을 위한 CMP의 용도.
  13. 연골 대사 질환 및/또는 대사성 골 질환의 치료, 예방 및 진단을 위한 CMP에 대한 항체의 용도.
