제1도는 본 발명의 실시예 1에서의 단백질 1차분획에 대한 AG 1-X4 칼럼의 크로마토그램이고, 제2도는 본 발명의 실시예 1에서의 단백질 2차분획에 대한 Bio-Rex70 컬럼의 크로마토그램이며, 제3도는 본 발명의 실시예에 따라 얻어진 트리코산틴(Trichosanthin)에 대한 SDS-PAGE의 Semi-leg plotting(분자량 측정).
Claims (7)
- 식물체의 근경으로부터 유효생체활성물질을 분리, 정체함에 있어서, 신선한 천화분을 완충용액에 대해 0.5∼0.7g/㎖의 비율로 완충용액에 넣은 다음, 여기에다 아세톤을 부피비로 40∼60% 또는 황산암모늄을 포화농도비로 20∼80%로 가하여 단백질 분획을 침전물로 얻은다음, 이를 이온교환 컬럼 크로마토그래피를 이용하여 트리코산틴(Trichosanthin)을 정제하는 것을 특징으로 하는 식물체의 근경으로부터 유효생체 활성물질을 제조하는 방법.
- 제1항에 있어서, 상기 이온교환컬럼크로마토그래피를 할때에 완충용액의 pH는 6.5∼7.0이고, 흡착된 단백질을 분리하기 위한 완충용액중의 염류의 농도는 70∼100mM인 것을 특징으로 하는 유효생체 활성물질의 제조방법.
- 식물체의 근경으로부터 유효생체활성물질을 분리, 정제함에 있어서, 건조된 천화분을 완충용액에 대해 0.25∼0.35g/㎖의 비율로 완충용액에 넣은 다음, 여기에다 아세톤을 부피비로 40∼60% 또는 황산암모늄을 포화농도비로 20∼80%로 가하여 단백질 분획을 침전물로 얻거나, 아세톤 40부피%의 분획을 제거하지 말고 아세톤 60 부피%의 총분획을 취하여 이를 이온교환 컬럼크로마토그래피를 이용하여 크리코산틴(Trichosanthin)을 정제하는 것을 특징으로 하는 식물체의 근경으로부터 유효생체 활성물질을 제조하는 방법.
- 제3항에 있어서, 상기 이온교환컬럼크로마토그래피를 할 때에 완충용액의 pH는 6.5∼7.0이고, 흡착된 단백질을 분리하기 위한 완충용액중의 염류의 농도는 7∼100mM인 것을 특징으로 하는 유효생체 활성물질의 제조방법.
- 트리코산틴(Trichosanthin)을 유효성분으로 함유하는 백혈종양치료제.
- 트리코산틴(Trichosanthin)을 유효성분으로 함유하는 낙태제.
