Description
본 발명은 성인 T세포 백혈병 관련 세포주와 그의 제조방법과 이 세포주를 동물에게 접종함으로써 성인 T세포 백혈병 관련 항혈청을 제조방법에 관한 것이다.
성인 T세포 백혈병은 1977년에 고즈끼(Kozuki)씨등에 의해 밝혀진 근래에 확인된 형의 백혈병이다. 임상적으로, 이 병은 T세포를 불규칙하고 분할된 핵을 갖게 하여 임파선 확장, 간종(肝腫) 또는 비종(脾腫)을 일으키는 것으로서 성인들이 걸린다. 많은 경우에 아급성으로 진행하여 종종 피부병적 장해를 형성하는 원인이 된다. 이외의 동물에 접종하는 것을 가능하게 함이 발견되었다.
본 발명을 좀더 설명하기 위해서 실시예들을 든다.
[실시예 1]
말초혈 10ml를 수토끼(일본 백색종)로부터 수집하여 Ficoll Hi-pac
로 비중원심 분리함으로써 백혈구를 분리 하였다. 다음에 그것을 1×106세포/ ml의 농도에서 2일동안 페트리접시에서 배양하였다. 배양용액은 인제대 혈청 10%, 소의 태아혈청 10%, 스트렙토마이신 100㎍/dl, 페니실린 100u/ml를 RPMI 1640에 가함으로써 제조되었다.
다음에 10,000R의 선량으로 조사된 1×106개의 MT-2세포를 가하고 37℃에서 CO2농도 7.5%에서 4주동안 배양하였다. 이 배양의 동안에 주 2회의 빈도로 새로운 매제를 가하였다.
Claims (6)
- 사람이나 원숭이 이외의 동물의 백혈구와 성인 T세포 백혈병 관련 항원 생성세포와를 혼합 배양하는 것을 특징으로 하는 사람이나 원숭이 이외의 동물을 면역시키기 위해 사용할 성인 T세포 백혈병 관련 세포주의 제조 방법.
- 제1항에 있어서, 사람이나 원숭이 이외의 상기 동물인 토끼인 것을 특징으로 하는 성인 T세포 백혈병 관련 세포주를 제조하는 방법.
- 제1항 또는 제2항에 있어서, 상기 성인 T세포를 백혈병 관련 항원 생성세포가 MT-2 세포인 것을 특징으로 하는 성인 T세포를 백혈병 관련 세포주를 제조하는 방법.
- 사람이나 원숭이 이외의 동물의 백혈구와 성인 T세포 백혈병 관련 항원 생성세포의 혼합배양으로부터 얻은 성인 T세포 백혈병 관련 세포주를 상기 동물에 접종하는 것을 특징으로 하는 성인 T세포 백혈병 관련 항혈청을 제조하는 방법.
- 제4항에 있어서, 사람이나 원숭이 이외의 상기 동물이 토끼인 것을 특징으로 하는 성인 T세포 백혈병 관련 항혈청의 제조방법.
- 제4항 또는 제5항에 있어서, 상기 성인 T세포 백혈병 관련 항원 생성세포가 MT-2세포인 것을 특징으로 하는 성인 T세포 백혈병 관련 항혈청의 제조방법.
