본 발명은 약제 및 기타 작용제를 신체에 전달시키기 위한 의료구에 관한 것으로, 특히 피부침투 약제 전달시스템에 요구되는 침투력, 점도 및 접착력의 특성들을 가진 매트릭스 조성물에 관한 것이다.
표피에 생리적 작용제(이후부터는 약제라함)를 투여하는 여러가지 형태의 시스템이 알려졌는데 이러한 의료구들은 안기나(angina)의 치료를 위한 니트로글리세린 연고와 같이 표피에 직접 사용하는 단순히 약제가 첨가된 크림, 연고 및 겔로부터, 미국특허 제 3,923,939호에 기술된 바와 같이 고정된 배열의 매트릭스를 통하여 약제가 분산되는 보다 정확히 조절할 수 있는 시스템 및, 미국특허 제 4,031,894호와 4,201,211호에 기술된 바와 같이 장시간 표피를 통하여 투여되는 약제의 양을 정확히 측정하기 위하여 속도조절막이나 기타 다른 구조를 사용한 보다 이상적 시스템까지 있다. 어떤 특별한 시스템의 실제 구조에 관계없이 이러한 모든 시스템들은 분산시키고자 하는 약제가 분산될 수 있게 하는 어떤 형태의 약제 저장소를 이용하고 이러한 저장소는 투여시스템에 적합한 점도, 침투력 및 접착력의 특성들을 가져야만 한다. 이것은 특히 상기 두 특허에 기술된 바와 같이 봉합변이 없는 적층된 시스템에서 접착층과 약제 저장층이 충분한 점도를 가지고 있어서 층들이 스며나오는 것을 방지할 수 있도록 하는 것이 중요하다. 상기의 두개 미국특허들은 클로리딘과 스코폴아민을 분산시키는데 사용하기 위하여 광물질오일-폴리이소부티렌(MO-PIB) 매트릭스를 기술하였는데 이러한 매트릭스는 또한 어떤 광물질오일 가용성 약제를 표피에 분산시키는 데 유용하다. 클로리딘과 스코폴아민 이외에도 예컨대, 에스트라디올, 페닐프로판올아민, 프로판올올, 오우아바인, 살부타몰, 과나벤즈, 라베톨올, 아트로핀, 할로페리돌, 브로모크리프틴, 클로로페니라민, 메트리포네이트, 이소소르비드, 디나이트레이트 및 니트로글리세린 같이 광물질 오일내에서의 용해도가 5mg/ml를 초과하지 않는 약제들이 특히 적당하다.
일차적 약제 또는 약제에 부가하여 조성물은 또는 표피 침투력을 개량하기 위한 침투 강화제, 표피 자극을 감소시키기 위한 세포보호제, pH조절을 위한 완충액 및 당해분야에 공지된 다른 물질들을 포함할 수 있다.
상기 2개의 미국특허에 기술된 바와 같이 전형적인 MO/PIB 매트릭스 조성물은 PIB의 혼합물과 혼합된 25℃에서 5-100cp 점도의 광물질오일을 포함한다. MO는 보통혼합물의 중량으로 35-65%로 구성되었고 PIB는 혼합물의 35-65%로 구성되었다. PIB 혼합물은 보통 저분자량(LMW) PIB(35,000-50,000 점도 평균 분자량)와 고분자량(HMW) PIB(1,000,000-1,500,000 점도 평균 분자량)를 포함한다. 적당한 혼합물은 35-65%의 광물질 오일, 10-40%의 LMW PIB 및 10-40%의 HMW PIB를 포함한다.
어떤 저장조성물의 형태는 일반적으로 최종제품의 디자인에 요구되는 점도, 약제 침투력 및 접착력과 같은 특성들이 제공되도록 조절된다. 일반적으로 PIB는 농후제(濃厚劑 : thickener)로서 작용하고 MO는 약제에 대한 용매로서 작용한다. 일반적으로 MO/PIB 비를 증가시키면 침투력이 증가하고 점도가 감소하는 반면 MO/PIB 비를 감소시키면 반대효과를 갖는다.
상기 두개의 미국특허에 기술된 바와 같이 동일한 일반적인 MO-PIB 혼합물은 약제 저장소로서 사용되던가 표피에 부착시키기 위한 접촉 접착면으로서 사용될 수 있으며 접착층은 약제물질을 포함시켜도 되고 포함시키지 않아도 된다.
전형적으로 분산시키고자 하는 약제는 용해되며, 저장소가 용해 및 분산상에 함유된 포화상태보다 많은 양으로 매트릭스 물질을 통하여 분산된다. 분산상은 매트릭스내의 약제농도가 요구되는 분산기간동안 포화 또는 포화 이상의 농도를 유지하기에 충분한 양으로 존재한다. 중량으로 40%의 약제를 함유시킬때 약제저장소로서 사용하기 위한 매트릭스는 중량으로 약제의 20%이상까지 함유하며 기본 투약량과 함께 접착제로서 사용할때에는 중량으로 약제의 10%까지 함유한다.
최적의 매트릭스 조성물을 만들기 위하여서는 조성물이 1.5×107포이즈 (pois e) 보다 적지않은 점도를 가져야 하고 약제가 적당한 크기의 표피부착물로부터 적당한 유리속도로 투여되도록 하기 위하여 충분한 높은 침투력 DCs를 갖도록 결정하여야 한다. 이러한 특성들을 조합하여 약제투여시스템은 그들의 구조적 일체성을 유지 즉, 스며나오거나 흘러나오지 않게 하며 이들이 들어 있는 포장으로부터 용이하게 꺼낼수 있으며, 적당한 크기로 만들수 있고, 적층시스템을 시스템으로부터 약제유리속도를 조절하기 위하여 속도조절막을 이용하여 충분히 높은 침투력을 갖게 하는 탁월한 물리적 특성을 갖도록한다. 매트릭스 조성물의 점도는 MO/PIB 비를 변형시킴에 의하여 변형될 수 있으나 PIB의 비율을 증가시킴에 의한 점도의 증가는 시스템의 침투력을 원하지 않는 낮은 준위까지 감소시키는 결과를 가져온다. 또한 침투력을 상승시키기 위한 광물질오일함량의 증가는 냉각흐름의 경향이 있고 나쁜 구조적특성을 갖는 낮은 점도 조성물을 이루게 된다. 본 발명전에는, 예컨대, 1.0의 MO/PIB 비는 시판될 수 있는 피부침투시스템에서 사용하는 최고로 가능한 준위였다.
카보트사제(Cabot Corporation製)인 Cab-O-Sil같은 콜로이드성 이산화실리콘(CSD)과 기타 유사한 콜로이드성 실리카물질이 광물질오일에 대한 농후제 (thick-enor)로 공지되었다(미합중국, 메사추세츠주 02110, 보스톤, 하이스트리트 125 소재 카보트사제 Cab-O-Sil의 성질 및 작용참조). 또한 유익한 약제를 투여하기 위한 새로운 밴데이지(Bandage)란 제목으로 게일(Gale) 등이 1981년 1월 29일자로 출원한 미국특허 명세서 일련번호 278,364호에 기술된 바와 같은 다른 형태의 약제 매트릭스를 농후시키기 위하여 CSD를 사용하는 것이 공지되었다. CSD는 국부조제약내에 포함시켜도 안전한 물질이라는 것이 FDA에 의하여 입증되었다.
따라서, MO-PIB 매트릭스 조성물의 점도를 증가시키기 위하여 CSD를 사용하기로 결정하였다. CSD를 어떤 MO-PIB 조성물에 첨가할때 첨투력을 감소시킴이 없이 점도가 증가됨이 발견되었는데, 실제로, 매트릭스 조성 혼합물의 여러가지 성분들의 어떤 조성범위내에서, 점도를 증가시킬뿐만 아니라 약제 침투력도 증가된 MO-PIB 조성물을 제조할 수 있음이 발견되었다. 어떤 범위내에서, 이러한 조성물을 사용하여 피부침투 치료시스템의 다른 성질에도 예상외의 개량이 이루어졌다.
종래의 시스템에서는 시스템의 역학적 성질과 확산성을 독립적으로 변화시킬수 없었다(다시 말하면, 침투력의 증가는 시스템내 점도의 감소 및 부작용을 가져온다). 그러나 본 발명에 따르면, 어떤 형태에 CSD의 사용은 이러한 성질을 독립적으로 변화시킬 수 있으며 높은 침투력과 높은 점도를 함께 얻을 수 있다.
따라서 본 발명의 목적은 개량된 성질을 가진 약제투여시스템을 위한 매트릭스 조성물을 제공하는데 있다.
본 발명의 다른 목적은 높은 점도와 높은 약제침투력을 함께 가진 매트릭스 조성물을 제공하는데 있다.
본 발명의 또 다른 목적은 적어도 1.5×107포이즈(poise)의 점도를 가진 약제가 부가된 MO-PIB 조성물을 제공하는데 있다.
본 발명의 다른 목적은 광물질오일, PIB, CSD와 포화 이상의 농도로 분산되는 적당한 광물질오일-가용성 약제 조성물로부터 형성된 약제가 부가된 매트릭스를 제공하는데 있다.
본 발명의 또 다른 목적은 원하는 점도, 침투력 및 접착력을 갖는 MO-PIB 매트릭스로 이루어진 속도가 조절되는 피부침투치료시스템을 제공하는데 있다.
본 말명의 목적 및 기타 다른 목적들은 첨부도면을 참조로 한 본 발명의 하기 기술로부터 보다 명백하게 될 것이다.
본 발명을 상술하면, 본 발명은 1.5×107포이즈의 점도와 통상의 광물질오일 가용성 약제조성물에 대한 높은 침투력을 가진 MO-PIB 매트릭스 조성물을 제조할 수 있음을 나타낸 것이다. 본 발명에 사용된 바와 같이 광물질오일 가용성약제는 광물질오일내에서의 용해도가 적어도 10㎍/㎖이고 5mg/ml을 초과하지 않는 약제이다. 이러한 약제의 제한하지 않은 예로는, 스코폴아민, 클로니딘, 에스트라디올, 페닐프로판올아민, 프로프란올올, 오우아바인, 살부탐올, 과나벤즈, 라베톨올, 아트로핀, 할로페리돌, 브로모크리프틴, 클로로페니라민, 메트리포네이트, 이소소르비드디나이트레이트, 및 니트로글리세린을 들 수 있다. 본 발명에 따른 상기의 점도와 침투력 특성은 MO/PIB 비가 1.0보다 크고 바람직하게는 1.4-1.8의 범위내이고, 조성물의 적어도 5%이며 바라직하게는 7.5-10%의 콜로이드성 이산화실리콘 (CSD)을 함유할때 얻을 수 있다. 이러한 매트릭스 조성물은 광물질오일-가용성 약제조성물의 약 40중량 %까지 첨가시킬 수 있다. 그러나 실제로 약제저장소로서 사용시 첨가량은 20%를 거의 초과하지 않으며 접착제로서 사용시 10%를 거의 초과하지 않는다.
제1도를 참조하면, 본 발명에 따른 전형적인 피부침투 치료시스템은 불투과성 배면(11)으로 형성된 밴데이지(10), 포화농도 이상의 농도로 분산되는 약제(14)를 가진 매트릭스 물질로 형성된 저장층(13), 이것을 통하여 약제가 공지의 속도로 분산되는 투과성이나 미공성(微孔性)막 같은 약제 유리속도 조절층(15), 약제(17)를 포함한 접착층(16) 및 보호물을 제거할 수 있는 코팅(18)을 포함한다. 이러한 층들은 당해 분야에 공지된 예정된 크기와 형상을 가진 밴데이지로 적층되거나 조립된다. 제1도는 적당한 실시예를 기술한 것이나 하나나 그 이상의 층들은 삭제될 수도 있고 반복시킬 수도 있는데, 기본적인 피부침투시스템은 표피와의 약제전달관계에서 매트릭스를 유지하기 위한 수단을 제공하는 약제를 함유하는 매트릭스에 있다.
다음 실시예에서, 본 발명에 따른 매트릭스 조성물을 가진 접착제와 저장소를 종래의 시료와 비교하였다. 모든 실시예에서 MO와 PIB는 초기에 주위조건하에서 함께 혼합하여 겔(gel)상으로 만든 다음, 약제조성물과 CSD(Cab-O-SilM-5 및 M-7)를 첨가하여 균일 분산제로 만든다. 이러한 조성물로부터 제조된 직경이 1.78㎝(넓은 2.5㎠)의 피부침투치료제 시료는 약 50㎛ 두께의 저장층, 약 50㎛ 두께의 접촉 접착층, MO로 포화된(0.9㎎ MO/㎠, 25마이크론 두께로 된 미공성(微孔性) 폴리프로피렌 막(Celgard2400이란 상표로 시판)의 속도 조절막, 미국특허 제 4,031,894호와 4,201,211호에 기술된 바와 같은 불투과성 배면층 및 제거할 수 있는 라이너를 함께 적층시켜 이루어진다. 접착제 조성물이 유리라이너에 너무나 견고하게 접착되었을 경우 53% PIB/47% MO의 5-10마이크론 두께의 피복층을 접착제와 라이너 사이에 만들어준다. 사용된 MO는 25℃에서 7CP의 점도를 가진 경질 광물질오일이고 LMW PIB는 약 35,000의 평균 분자량을 갖고, HMW PIB는 약 1,200,000의 평균 분자량을 갖는다.
다음 실시예에서, 접착 및 저장 매트릭스의 조성물은 침투투여시스템을 만드는데 사용된 조성물이다. 접착과 저장 매트릭스에 대한 MO/PIB 비가 다르고, Celgard층은 광물질오일로 포화되는 한편 ; 시스템은 높은 MO/PIB를 가진 조성물로부터 낮은 쪽으로 광물질오일이 전달되기 때문에 결과적으로 평형이 이루어진다. 그러나 접착층과 저장층은 둘다 포화이상이고 매트릭스내 과잉의 용해되지 않은 약제는 용이한 전이가 예상될 수 없으므로 약제의 상당한 전이는 이루어질 수 없다. 그러므로 침투 전달시시템의 MO/PIB의 전체값은 초기 MO/PIB 비의 중간치가 될 것이나 정확한 값은 저장소와 접착층의 각각에 사용된 물질의 상대적양 뿐만 아니라, 셀가드 막 내에 있는 광물질오일의 양과 저장시간 및 온도에 따른다.
다음 기술에서 조성물은 배면부, 유리라이너 및 속도조절막 같은 TTS의 구성요소에 관계없이 매트릭스 겔의 중량%로 정의한다.
[실시예 1]
종래의 방법에 따른 접촉 접착조성물을 47%의 MO, 27.8%의 LMW PIB, 22.2%의 HMW PIB(MO/PIB=0.94) 및 3%의 클로니딘으로부터 제조한다. 종래의 방법에 따른 약제 저장조성물은 47%의 MO, 22.2%의 LMW PIB, 17.8%의 HMW PIB(MO/PIB=1.18) 및 약 13%의 클로니딘을 포함하도록 제조한다. 클로니딘 피부침투시스템은 상술한 바와 같은 물질들로부터 제조하며, 이렇게 제조된 시스템은 40℃에서 약 8×106포이즈의 겉보기 점도를 나타낸다.
[실시예 2]
실시예 1의 클로니딘 TTS는 낮은 점도를 나타내므로 실시예 1의 조성물 이외에 1%, 2%, 5% 및 7.5%의 CSD를 첨가함에 따른 일련의 접착 및 저장조성물로부터 시스템을 제조한다. TTS의 점도에 대한 CSD 첨가효과가 제2도에 기술되었다.
[실시예 3]
종래의 방법에 따른 스코폴아민을 함유한 접촉 접착 및, 저장조성물은 46.1%의 MO, 28.9%의 저분자량 PIB 23.0%의 고분자량 PIB 및 2.0%의 스코폴아민과 41.7%의 MO, 26.2%의 LMW PIB, 20.8%의 HMW PIB 및 11.3%의 스코폴아민으로부터 각각 제조한다. 상기 성분 이외에 1%, 2%, 5% 및 7.5%의 CSD를 확대 첨가함에 따른 일련의 접착 및 저장조성물을 제조하는 것도 적당하다. 스코폴아민 침투시스템의 점도에 대한 CSD 농도의 효과가 또한 제2도에 도시되었다.
[실시예 4]
CSD 부재하에, 실시예 1의 PIB 분율을 삭감한 나머지를 7.5% CSD로 그리고 실시예 1의 모든 다른 성분들을 삭감시킨 나머지를 7.5%의 CSD로 첨가하여 MO/PIB 비를 변화시킨 접촉 접착 및 저장조성물을 제조하여 피부침투시스템으로 만든다. PIB의 성분에서 CSD분을 첨가함에 의하여 아래의 표 1에 표시된 바와 같이 MO/PIB의 값을 증가시킬 수 있다. 이러한 조성물로부터 제조된 피부침투시스템으로부터 클로니딘 유리속도에 대한 MO/PIB(셀가드층내 MO 함유) 변화효과가 제3도에 도시되었다. 제3도에 도시된 바와 같이, PIB분율의 일부로서 CSD의 첨가에 의하여 얻을수 있는 보다 높은 MO/PIB 값은 지금까지 얻을 수 없었던 보다 높은 유리속도를 나타냈다.
표1은 실시예 1,2 및 4에 따라 제조된 시스템에서 얻은 데이타를 요약한 것이다.

표 1에 표시된 바와 같이 실시예 2와 4에서 제조된 제품들은 둘다 실시예 1에서 제조된 제품보다 약 2배의 점도 개량이 나타났다. 그러나 실시예 4의 제품은 농후제(thickener)로서 사용된 PIB의 일부를 제거하였어도 점도감소를 나타내지 않고 실시예 2의 제품의 약제 유리속도보다 탁월한 개량을 나타냈다.
[실시예 5]
클로니딘과 스코폴아민을 분산시키기 위한 피부침투시스템을 실시예 1-4에 기술된 선택된 저장 및 접착조성물로부터 상술한 바와 같이 제조하여 포장 통합성 시험을 받는다. 이 시험에서 시스템은 봉해진 금박포내에 포장되어 평형조건에 도달될때까지 주위 조건하에서 최소한 1개월간 방치한다. 이 저장기간중 시스템은 금박포로부터 제거시 손상되는 노출변으로부터 접착 및 저장층들의 냉각흐름에 대한 취약점이 있다. 그러므로 금박포를 통상방법으로 열때 TTS에 의하여 받은 손상을 관찰한다. 보통 사용자가 임의로 금박포를 열어서 아래로 뒤집고 흔들었을때 관찰되는 가능한 조건은 다음과 같다.
(a) TTS가 금박포로부터 자유롭게 떨어진다.
(b) TTS가 금박포의 상단면(유리라이너 반대면)에 접착되어 금박포로부터 TTS를 자유롭게 유리시키기 위하여서는 유리라이너상을 벗겨내야 하므로서 다음의 결과를 초래한다 : (1) 사용할 수 있는 형태로 금박포로부터 본래대로 TTS를 분리 (2) 유리라이너가 상단면과 함께 나와서 금박포에 아직 부착되어 사용할 수 없는 형태로 됨.
(c) TTS의 유리라이너가 금박포에 부착되어 유리라이너상을 벗겨내면 본래대로의 TTS의 금박포로부터 제거될 수 있음.
(d) 유리라이너가 금박포의 한면에 접착되었고 상단은 포의 다른면에 접착되어 금박포를 열었을때 TTS에 고칠수 없는 손상을 일으킴.
제4도는 여러가지 클로니딘과 스포콜아민시스템에 대한 시험결과를 나타낸 것이다.
결과에 나타난 바와 같이 CSD 농도에서 적어도 1.5×107포이즈의 점도에 대하여 약 5%이상의 감소가 있음이 확인되었다(제2도 참조).

MO/PIB 비를 변화시켜 일련의 MO-PIB 조성물을 제조하여 클로니딘과 스코폴아민에 대한 조성물의 침투력을 측정한 결과가 제5도에 도시되었다. 도시된 바와 같이 침투력에 상당한 개량이 보다 높은 MO/PIB 비에서 수득되었다. 클로니딘에 대하여 1.2 이상의 값은 속도조절막이 유리속도의 우수한 조절 메카니즘을 제공하도록 설계되었음을 나타내는 1×10-4㎍/㎝ sec 이상의 침투력을 제공한다.
[실시예 7]
CSD 첨가량을 변화시킨 프로프탄올올 염기가 첨가된 MO-PIB 매트릭스를 제조하여 시험관내에서 침투력에 대한 CSD의 효과를 측정한다. 표 2는 프로프탄올올 염기의 전달성에 대한 MO/PIB 단일 시스템에서 다양한 백분율로 Cab-O-Sil의 첨가효과를 나타낸다.

결과의 고찰
제2도에 나타난 바와 같이 MO-PIB 시스템에 CSD의 첨가는 겔(gel)의 점도를 증가시킨다. 약 3%의 CSD에서 스코폴아민 같이 유동시스템일지라도 적당한 점도를 얻을 수 있으며, 이러한 점도는 약 5%의 CSD에서 최대가 되는 경향이 있다. CSD의 더 많은 첨가는 점도의 비례적 증가를 가져오지 못한다. 피부침투 시스템 제조의 물리적 성질은 보다 높은 CSD 준위에서 더 많은 개량이 기대되지 않는다. 그럼에도 제4도에 기술된 포장을 여는 시험의 결과 5% 이상의 CSD에서 포장으로부터 자유낙하 되는 것이 상당히 개량되었음을 나타낸다. 단순히 CSD 함량을 5%로부터 7.5%까지 상승시킴에 의하여 자유낙하 퍼센트가 클로니딘시스템에 대하여서는 17.5%로부터 90%로, 그리고 스콜폴아민시스템에 대하여서는 10%로부터 79%로 증가되었다.
본 발명의 목적에 적당한 점도를 가진 MO-PIB 조성물에서 얻을 수 있는 본 발명전의 최대 MO/PIB 비는 약 1.00이다. 제5도에 도시된 바와 같이 이러한 비에서 클로니딘 같은 약제에 대한 침투성은 원하는 것보다 낮다. PIB는 MO-PIB 겔 내에서 농화제(濃化劑)이므로 어떤 주어진 시스템에서 MO/PIB 비가 증가할 때 점도는 감소한다는 것을 예상할 수 있다. 또한 MO/PIB 겔의 점도가 CSD의 첨가로서 증가될때 일정한 MO/PIB 비에서 겔내 약제 침투력은 감소한다는 것을 예상할 수 있다. 그러나 표 1에 나타난 바와 같이 PIB분율내의 양으로서 CSD의 첨가는 모든 성분내의 양으로서 CSD의 첨가에 의하여 얻는 결과와 비교할때 점도의 상당한 감소는 일어나지 않는다. 또한 표 2에 나타난 바와 같이 약 10% CSD까지의 첨가에 의하여 MO-PIB 매트릭스의 침투력은 증가한다(일정한 MO/PIB 비에서).
그러므로 본 발명에 따르면 광범위한 광물질오일에 가용성인 약제를 분산시기 위한 매트릭스로서 사용하기에 적당한 성질을 가진 약 6% 이하의 CSD(약제-유리 기준으로) 특히 6-11%의 CSD를 가진 MO-PIB-CSD 혼합물을 제공한다. 또한 본 조성물이 1.0 이상의 MO/PIB 비를 이룰때 지금까지 얻을 수 없었던 약제 침투력을 얻을 수 있었다. 1.2 이상의 MO/PIB 비를 가진 본 발명에 따른 조성물은 이때까지 얻을 수 없었던 클로니딘 침투력을 가진 클로니딘 함유 매트릭스를 제조하는데 사용할 수 있다. 미공성(微孔性)속도 조절막이 MO로 포화된 적층형 TTS's를 제조하는데 사용할때 시스템의 전체 MO/PIB는 실제로 1.45를 초과하는 한편 원하는 구조적 특성을 유지한다.
본 발명의 기술한 바에 대한 여러가지 변형이, 하기의 청구범위에 의하여서만 한정되는 본 발명의 범위로부터 이탈됨이 없이 당해 분야의 기술자들에 의하여 이루어질 수 있음은 자명한 일이다.
Claims (14)
- MO/PIB 비가 적어도 1.0이고 적어도 1.5×107포이즈의 점도를 가짐을 특징으로 하는 광물성오일, 폴리이소부틸렌 및 적어도 5%의 콜로이드성 이산화실리콘으로 구성된 약제 전달 기구내의 매트릭스로서 사용하기에 적당한 조성물.
- 제1항에 있어서, 광물성 오일에 가용성인 분산될 약제를 40%까지 포함하는 바의 조성물.
- 제2항에 있어서, 상기 약제가 조성물내에 약제의 포화 농도와 약 20% 사이의 준위로 존재하는 바의 조성물.
- 제3항에 있어서, 광물성오일 대 폴리이소부틸렌의 비가 적어도 1.2이고 조성물이 적어도 7.5 중량%의 클로이드성 이산화실리콘을 포함하는 바의 조성물.
- 제4항에 있어서, 상기 약제가 클로니딘, 스코폴아민, 프로프탄올올, 에스트라디올, 페닐프로판올아민, 오우아바인, 살부탐올, 과나벤즈, 라베톨올, 아트로핀, 할로페리돌, 브로모크리프린, 클로로페니라민, 메트리포네이트, 이소소르비드디나이트레이트 및 니트로글리세린으로 구성된 그룹으로부터 선택한 바의 조성물.
- 제1항에 있어서, 상기 약제가 클로니딘이고 클로니딘에 대한 조성물의 침투력이 적어도 1.0×10-4㎍/㎝ sec인 바의 조성물.
- 제6항에 있어서, MO/PIB의 값이 적어도 1.2인 바의 조성물.
- 분산시키고자 하는 광물성오일에 가용성인 약제를 함유한 광물성오일과 폴리이소부틸렌의 겔로 구성된 조성물에서 이 조성물이 분산될 5%의 콜로이드성 이산화실리콘을 갖고, WO/PIB 비가 적어도 1.0이며 점도가 1.5×107포이즈로 개량된 약제 함유 매트릭스 조성물.
- 제8항에 있어서, 상기 MO/PIB 비가 적어도 1.2이고, 콜로이드성 이산화실리콘이 6-10%의 양으로 존재하며 상기 약제가 포화농도 이하의 양 및 40%보다 많지 않은 양으로 존재하는 바의 매트릭스 조성물.
- 제8항에 있어서, 상기 약제가 클로니딘이고 클로니딘에 대한 매트릭스의 침투력이 적어도 1×10-4㎍/㎝ sec인 바의 조성물.
- 약제 저장층, 접착층 및 이들 사이에 오는 약제 유리속도 조절막으로 구성되어 있으며 적어도 저장층내에는 적당한 광물성오일에 가용성인 약제가 동저장층내에서의 동약재 포화농도 이상의 농도로 퍼져있고, 저장층과 접착층이 광물성오일과 폴리이소부틸렌의 혼합물을 혼합하는 기구에서, 저장 및 접착층이 적어도 5%의 콜로이드성 이산화실리콘을 포함하고 적어도 1.5×107포이즈의 점도를 갖으며, 저장 및 접착층 내 광물성 오일 대 폴리이소부틸렌의 비가 적어도 1.2로 개량됨을 특징으로 하는 투약기구.
- 제10항에 있어서, 상기 유리속도 조절막이 광물성오일을 포함하고 피부침투치료시스템내 MO/PIB의 전체 비율이 적어도 1.4인 바의 피부침투 투약기구.
- 제11항에 있어서, 상기 약제를 클로니딘, 스코폴아민, 프로프란올올, 에스트라디올, 페닐프로판올아민, 오우아바인, 살부탐올, 과나벤즈, 라베롤올, 아트로핀, 할로페니돌, 브로모크리프틴, 클로로페니라민, 메트리포네이트, 이소소르비드디나이트레이트 및 니트로글리세린으로부터 선택한 바의 피부침투 투약기구.
- 제9항에 있어서, 상기 약제가 클로니딘이고 기구로부터 환부로의 시험관적 약제 유리속도는 적어도 2.0㎛/㎠hr인 바의 피부침투 투약기구.
