제1도는 에피더민(Epidermin)의 완전한 산 가수분해물의 아미노산 크로마토그램이다(닌히드린 염색, λ=570㎜).
제3도는 에피더민의 UV 스펙트럼이다.
제10도는 본 발명의 배지에 대하여 생존하는 박테리아의 수 및 마이크로코쿠스루테우스에 대한 활성을 비교한 그래프이다.

Claims (2)

  1. 스타필로코쿠스 에피더미디스(Staphylococcus epidermidis) DSM 3095를 2 내지 4%의 육즙 등의 질소원, 1 내지 3%의 당 또는 당알코올의 탄소원 및 0.25 내지 1%의 탄산염 및/또는 0.25 내지 0.5%의 알칼리토금속의 수산화물로 구성되는 복합 영양배지 용액중에서 34 내지 37℃의 호기 조건하에 성장시킨 후, 세포 및 무기염을 제거하고; 배양모액을 pH 8에서 n-부탄올로 추출하고, 부탄올 추출물을 증발시켜 그 잔류물을 메탄올에 용해시킨 다음 이 용액을 에테르 침전에 의해 지방성분을 제거하거나, 배양모액을 아크릴레이트 또는 폴리스티렌을 주재로 하는 중합체로 처리하고, 흡착된 에피더민을 메탄올/진한염산(99:1)에 의해 수지로 부터 유리시킨 다음 이 용액을 암모니아로 중화시키고, 진공중에서 증발 농축시켜, 생성된 활성물질을 단리시키고; 상기에서 단리한 물질을 겔크로마토그래피[메탄올/아세트산(95:5)를 사용하여 세파덱스 LH-20에서]하여 다른 저분자량 펩티드, 아미노산 및 염을 분리 제거하고; 일차적으로, n-부탄올/에틸아세테이트/0.1N 아세트산(3:1:3)의 시스템을 사용하여 액체-액체 분배를 수행하고, 연속하여, 이차적으로, 기선상에 남아있는 에피더민을 2-부탄올/0.05N암모늄 아세테이트(1:1) 중성 시스템을 사용하여 분배하는 크레이그의 다중 역류 분배를 수행하고; 용출물을 동결건조시켜 무색분말 형태의 순수한 에피더민을 단리시킴을 특징으로 하는, 하기의 일차구조를 가지며, 그 아미노산 조성이 Asn(1), Pro(1), Gly(2), Ala(2), He(2), Phe(2), Lys(2), Lan(2), β-Me-Lan(1), Dhb(1), Tyr(1) 및 S-(2-아미노비빌)-D-시스테인(1)인 항생물질 에피더민의 제조, 단리 및 정제방법.
    상기 구조식에서, N-말단에 인접한 개개의 티오에테르아미노산의 반이 D배치이다.
  2. 제1항에 있어서, 하기의 추가 특성을 갖는 에피더민의 제조방법.
    a. 형태:무색분말, b. 용해도:물/빙초산 또는 메탄올/빙초산의 혼합물에 매우 쉽게 용해; 저급 알코올에 용해; 클로로포름, 아세톤, 디에틸에테르, 석유 에테르에 불용. c. 실리카겔 플레이트 상에서 염색 반응;닌히드린, 염소/TDM[TDM=4,4'=비스-(디메틸아미노)-디페닐메탄], 오르신/술푸르산, 아니스알데히드/술푸르산. 254㎜의 UV에서, 물을 분무하여 비-파괴검출. d. 박층 크로마토그래피:실리카겔 플레이트 상품 60F254(Merck)를 사용하였다. 시스템 A:클로로포름/메탄올/17% 암모니아 (2/2/1) Rf=0.73
    시스템 B:클로로포름/메탄올/17% 암모니아 시스템 C:n-부탄올/빙초산/물
    (70/35/10) Rf=0.30 (4/1/1) Rf0.73
    e. HPLC:제7도 참조. f. 안정성 pH2 내지 pH7에서 안정. 그이상의 pH에서 활성이 급강하. g. 분자량:약 2160(음이온부재). h. U.V.흡수스펙트럼:수용액에서 λmax×267nm(제3도 참조). i. IR흡수스펙트럼:브롬화 칼륨 펠렛상 시료의 IR 스펙트럼(제4도). i. NMR 스펙트럼 1H-NMR 스펙트럼:제5도 13C-NMR 스펙트럼:제6도.
