Abstract
본 발명은 대식세포 유래 소포로 코팅된 지질 나노입자 및 이의 제조 방법에 관한 것이다. 본 발명의 대식세포 유래 소포로 코팅된 지질 나노입자를 사용하면 타겟 세포 내부에 염증성 사이토카인과 함께 siRNA를 전달할 수 있으므로 유전자 치료와 면역 치료를 동시에 수행할 수 있다. 또한, 대식세포 유래 소포로 코팅된 지질 나노입자는 T 세포와 NK 세포에는 유전자를 전달하지 않고, 염증성 사이토카인만을 전달하여 세포 활성화를 일으켜 면역 반응을 활성화한다.
Description
본 발명은 대식세포 유래 소포로 코팅된 지질 나노입자 및 이의 제조 방법에 관한 것이다.
기존에 암 환자에게 시행되어 오던 수술, 방사선 치료, 항암화학요법, 면역요법 등과는 달리, 유전자 치료는 이미 발생된 암이라는 현상을 치료의 대상뿐만 아니라 암이 유발된 원인 중의 하나를 직접 교정한다는 점에서 제5세대 암 치료법으로 여겨진다. 현재 유전자 치료를 통한 암 치료는 새로운 치료 유전자를 암세포에 도입하여 특정 유전자를 과발현 또는 억제함으로써 보다 효과적인 전략으로 발전하고 있다. 이러한 치료용 핵산을 효과적으로 전달하기 위해서는 유전자 전달 기술이 중요하며, 현재 효율성이 가장 높다고 알려져 사용되는 바이러스 벡터는 면역원성을 유발할 수 있다는 단점이 있다. 따라서 유전자 전달 기술은 생체 내 유전자 전달을 나노입자와 같은 비바이러스 기반 벡터를 이용하는 방향으로 이동하고 있는 추세다.
코로나19 팬데믹으로 등장한 지질 나노입자가 유전자 백신으로 임상에서 그 유효성이 입증된 바 있으나, 암 치료 등의 유전자 치료 플랫폼으로 사용되기 위해서는 생체 내 빠른 제거 및 분해로 지속적인 유전자 제공이 어려운 한계를 개선할 필요성이 있다. 따라서, 핵산 입자를 세포에 안정적으로 전달할 수 있는 개선된 지질 나노입자에 대한 개발이 필요한 실정이다.
본 발명자들은 봉입된 핵산 분자를 안정적으로 세포에 전달함과 동시에 면역세포를 활성화시킬 수 있는 지질 나노입자를 개발하고자 예의 연구 노력하였다. 그 결과 염증성 대식세포 유래 소포를 지질 나노입자에 코팅하여 제작한 나노입자를 사용하는 경우 대상 세포에 핵산 분자를 효과적으로 전달하고 면역 세포를 활성화시켜 유전자 치료와 면역치료를 동시에 수행할 수 있다는 것을 규명하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 지질 성분을 포함하는 나노입자 코어; 및 상기 나노입자 코어의 표면에 코팅된 대식세포 유래 소포를 포함하는 나노입자를 제공하는 것이다.
본 발명의 다른 목적은 지질 성분을 포함하는 나노입자 코어; 및 상기 나노입자 코어의 표면에 코팅된 대식세포 유래 소포를 포함하는 나노입자의 제조방법을 제공하는 것이다.
본 발명의 또 다른 목적은 지질 성분을 포함하는 나노입자 코어; 상기 나노입자 코어의 표면에 코팅된 대식세포 유래 소포; 및 상기 나노입자 코어에 탑재된 세포사멸 유도 핵산 분자 또는 소수성 약물을 포함하는 나노입자를 포함하는 암 예방 또는 치료용 약제학적 조성물을 제공하는 것이다.
본 발명의 일 양태에 따르면, 본 발명은 a) 지질 성분을 포함하는 나노입자 코어; 및 b) 상기 나노입자 코어의 표면에 코팅된 대식세포 유래 소포를 포함하는 나노입자를 제공한다.
본 발명자들은 봉입된 핵산 분자를 안정적으로 세포에 전달함과 동시에 면역세포를 활성화시킬 수 있는 지질 나노입자를 개발하고자 예의 연구 노력하였다. 그 결과 염증성 대식세포 유래 소포를 지질 나노입자에 코팅하여 제작한 나노입자를 사용하는 경우 대상 세포에 핵산 분자를 효과적으로 전달하고 면역 세포를 활성화시켜 유전자 치료와 면역치료를 동시에 수행할 수 있다는 것을 규명하였다.
"나노입자"라는 용어는 직경이 약 1nm 내지 약 300 nm (예컨대, 1nm 내지 300 nm, 1nm 내지 200 nm, 1nm 내지 50nm, 1nm 내지 30 nm, 1nm 내지 20 nm, 1nm 내지 15 nm, 100 nm 내지 300 nm, 및 150 nm 내지 300nm, 200nm 내지 300nm, 또는 250nm 내지 300nm)인 물체를 의미한다.
본 발명의 일 구현예에 있어서, 상기 나노입자는 구형인 것이다.
본 발명의 일 구현예에 있어서, 상기 대식세포는 염증성 대식세포(M1 macrophage)인 것이다.
본 명세서의 용어 "대식세포(macrophage)"란, 선천 면역을 담당하는 주요한 면역 세포를 의미한다. 대식세포는 온 몸에 정착성으로 존재하는 것이 대부분이나, 일부는 혈액 내에서 단핵구의 형태로 존재한다. 대식세포는 세포 조직이나 이물질, 미생물, 암세포 등 건강한 몸에 존재하는 비단백질 분자를 흡수하고 소화시키는 식세포 작용을 한다.
대식세포는 비활성화 대식세포(M0), 전염증성 대식세포(M1), 또는 항염증성 대식세포(M2)로 분류될 수 있다. 대식세포에 LPS나 IFN-gamma와 같은 염증성 자극을 주게 되면 Th1의 활성화가 유도되며, 항박테리아를 위한 염증성 대식세포(M1)의 형태를 띠게 된다. 반면, M2 대식세포는 IL-4, IL-13, IL-10에 의하여 활성화가 되는 형태를 의미한다.
본 발명의 일 구현예에 있어서, 상기 지질 성분은 콜레스테롤, 이온화 가능한 지질, 양이온성 지질, 인지질, PEG-지질 또는 이들의 조합을 포함하는 것이다.
본 명세서에서 용어 "지질"은 지방산의 에스테르를 포함하지만 이에 제한되지 않고, 물에 불용성이지만 많은 유기 용매에 가용성임을 특징으로 하는 유기 화합물의 군을 지칭한다.
본 명세서에서 용어 "콜레스테롤" 및 그의 유도체는 예를 들어, 콜레스타놀, 콜레스타논, 콜레스테논, 코프로스타놀, 콜레스테릴-2'-하이드록시에틸에테르, 콜레스테릴-4'-하이드록시부틸 에테르, 및 이들의 혼합을 포함하지만 이에 제한되지 않는다.
본 명세서에서, 용어 "이온화 가능한 지질(ionizable lipid)"은 산 성 pH에서는 양전하를 띠나, 생체 내 pH에서는 중성인 지질을 의미한다. 이온화 가 능한 지질은 산성 pH에서 양전하를 띰으로써 지질 나노입자 내에 RNA 등 음전하를 띠는 입자를 응축할 수 있으며, 생체 내 pH에서는 중성을 유지함으로써 독성을 최 소화할 수 있다.

본 명세서 상의 용어, "양이온성 지질"은 선택된 pH, 예컨대 생체 내 pH에서 양전하를 갖는 지질을 의미한다. 양이온성 지질은 일반적으로 음으로 하전된 핵산 분자의 정전기적 복합체화를 유도하기 위해 포함된다. 양으로 하전된 나노입자는 음으로 하전된 핵산 분자와 정전기적으로 상호작용하는 것으로 여겨지며, 이는 치료제의 복합체화를 촉진할 뿐만 아니라, 핵산 분자를 효소적 분해로부터 보 호할 수 있다.
본 발명의 일 구현예에 있어서, 상술한 양이온성 지질은 1,2- dioleoyloxy-3-(trimethylammonium)propane (DOTAP), N41-(2,3- dioleyloxy)propyl]-N,N,N-trimethylammonium chloride (DOTMA), 5- carboxyspermylglycinedioctadecylamide (DOGS), 2,3-dioleyloxy-N-[2(sperminecarboxamido)ethyl]-N,N-dimethyl-l-propanaminium (DOSPA) 및 1,2-dioleoyl-3- dimethylammonium-propane (DODAP)으로 이루어지는 군으로부터 선택되는 1종 이상의 양이온성 지질일 수 있다.
본 발명의 일 구현예에 있어서, 상술한 인지질은 핵산을 안정적으로 유지하고 세포막(플라즈마 막 및 소기관 막)과의 융합을 억제하지 않는 한 특별히 제한되지 않으며, distearoylphosphatidylcholine (DSPC), 1,2-dioleoyl-snglycero-3-phosphocholine (DOPC), 1,2-dipalmitoyl-sn- glycero-3-phosphocholine (DPPC), 1,2-Dipalmitoyl-sn-glycero-3-Phospho-rac-(1-glycerol) (DPPG), 1,2- dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), 1-palmitoyl-2-oleoyl-snglycero-3-phosphocholine (POPC), 1-palmitoyl-2-oleoyl-sn-glycero-3- phosphoethanolamine (POPE) 및 dioleoyl-sn-glycero-3-phosphoethanolamine-N- (maleimidomethyl) (DOPE-mal)으로 이루어지는 군으로부터 선택되는 1종 이상의 인지질일 수 있으며, 더욱 구체적으로는 DSPC(distearoylphosphatidylcholine)일 수 있으나, 이에 한정되는 것은 아니다.
본 명세서 상의 용어, "PEG 지질(pegylated lipid)"은 지질 모이어 티에 컨쥬게이션된 폴리에틸렌글라이콜(Poly(ethylene glycol))의 유도체로서, 지 질 부분 및 폴리에틸렌 글라이콜 부분을 모두 포함하는 분자를 의미한다.
본 발명의 일 구현예에 있어서, 상술한 PEG 지질은 페길화 다이아실 글리세롤 지질 (pegylated diacylglycerol, PEG-DAG), 페길화 세라마이드 지질 (pegylated ceramide lipid, PEG-Cer), 페길화 포스파티딜에탄올 아민 지질 (pegylated phosphatidylethanoloamine lipid, PEG-PE), 페길화 석시네이트 디아실 글리세롤 지질 (pegylated succinate diacylglycerol lipid, PEG-S-DAG), 페길화 다이알콕시프로필카바메이트 지질(pegylated dialkoxypropylcarbamate lipid), 1,2-dimyristoyl-rac-glycero-3-methoxypolyethylene glycol("PEG-DMG" 또는 "DMGPEG") 및 1,2-dicapryl-rac-glycero-3-methylpolyoxyethylene glycol(C10 다이아실 글리세롤 PEG)으로 이루어진 군으로부터 선택되는 1종 이상의 PEG 지질일 수 있고, 보다 구체적으로 1,2-dimyristoyl-rac-glycero-3-methoxypolyethylene glycol-2000 (DMG-PEG 2000)일 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 일 구체예에 있어서, 상기 지질 성분은 다음과 같은 것이다:
a) 상기 이온화 가능한 지질은 4-(dimethylamino)butanoate (D-Lin MC3-
DMA 또는 MC3) 인 것;
b) 상기 인지질은 DSPC(distearoylphosphatidylcholine)인 것; 또는
c) a) 및 b)의 조합.
본원에서 사용된 용어 "4-(dimethylamino)butanoate (D-Lin MC3-DMA 또는 MC3)"는 이온화지질의 하나로 지질 나노입자 형성에 주로 사용되며, "DSPC(Distearoylphosphatidylcholine)"는 인지질의 일종인 포스파티딜콜린으로 세포막의 구성 성분이다.
본 발명의 일 구현예에 있어서, 상기 나노입자 코어에 대한 대식세포 유래 소포의 개수 비는 1.0 내지 3.0인 것이다. 예를 들어, 1.0 내지 2.5, 1.0 내지 2.0, 1.0 내지 1.5, 1.5 내지 3.0, 2.0 내지 3.0, 2.5 내지 3.0, 또는 1.5 내지 2.5일 수 있으나, 이에 제한되지는 않는다.
본 발명의 일 구현예에 있어서, 상기 나노입자의 크기는 100nm 내지 300nm인 것이다. 예를 들면, 100nm 내지 300nm, 100nm 내지 270nm, 100nm 내지 240nm, 100nm 내지 210nm, 100nm 내지 180nm, 100nm 내지 150nm, 100nm 내지 120nm, 130nm 내지 300nm, 160nm 내지 300nm, 190nm 내지 300nm, 220nm 내지 300nm, 250nm 내지 300nm, 280nm 내지 300nm, 130nm 내지 270nm, 160nm 내지 240nm, 또는 190nm 내지 210nm 일 수 있으나 이에 제한되지 않는다.

본 발명의 일 구현예에 있어서, 상기 나노입자 코어는 소수성 약물 또는 핵산 분자를 포함할 수 있는 것이다.
상기 나노입자 코어에 탑재할 수 있는 약물의 종류는 예를 들면, 항암제, 항생제, 항염증제, 당뇨병 치료제일 수 있다. 구체적으로, 독소루비신(doxorubicin), 파클리탁셀(paclitaxel), 도세탁셀(docetaxel), 펨브롤리주맙(pembrolizumab), 니볼루맙(nivolumab), 암포테리신 B(amphotericin B), 그라미시딘(gramicidin), 덱사메타손(dexamethasone), 이부프로펜(ibuprofen), 인슐린(insulin), 또는 GLP-1 길항제(glucagon-like peptide-1 agonist) 일 수 있으나, 이에 제한되지는 않는다.

본 발명의 약학 조성물은 약학 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 또는 희석제를 추가로 포함할 수 있다. 예를 들면, 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 만니톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이다.
본 발명에 따른 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에 있어서, 약학적으로 유효한 양은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 상기 조성물의 사용량은 환자의 나이, 성별, 체중에 따라 달라질 수 있으나, 펩타이드가 암의 치료에 유용한 혈중 농도가 되도록 충분한 양을 1일 1회 내지 수회 투여할 수 있다.
상기 조성물의 투여량은 투여 경로, 질병의 정도, 성별 체중, 나이 등에 따라 증감될 수 있다. 따라서, 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 약학적 조성물의 투여량은 바람직하게는 1일 당 0.001 내지 100mg/kg(체중)이고 상술한 투여량의 약학적 조성물을 적용을 원하는 부위에 대해, 적용 목적에 따라 국소적으로 적용할 수 있다.
핵산(예를 들어, 간섭 RNA 또는 mRNA)의 "유효량" 또는 "치료적 유효량"은 원하는 효과, 예를 들어, 간섭 RNA의 부재에서 검출된 정상 발현 수준과 비교하여 표적 서열의 발현 억제; 또는 단백질이 발현되는 유기체에서 바람직한 생물학적 효과를 유발하는 양의 단백질의 mRNA-유도된 발현을 발생시키기에 충분한 양을 의미한다. 표적 유전자 또는 표적 서열의 발현의 억제는 대조군에 비해 간섭 RNA로써 획득한 값이 약 90%, 85%, 80%, 75%, 70%, 65%, 60%, 55%, 50%, 45%, 40%, 35%, 30%, 25%, 20%, 15%, 10%, 5%, 또는 0%인 경우에 달성된다. 다른 실시양태에서, 발현된 단백질은 체내 세포 유형에서 정상적으로 발현되는 단백질의 활성 형태이고, mRNA의 치료적 유효량은 건강한 개체의 세포 유형에서 정상적으로 발현되는 단백질의 양의 최소 50%(예를 들어, 최소 60%, 또는 최소 70%, 또는 최소 80%, 또는 최소 90%)인 인코딩된 단백질의 양을 생성하는 양이다. 표적 유전자 또는 표적 서열의 발현 측정을 위한 적합한 분석은, 예를 들어, 도트 블랏, 노던 블랏, 제자리 혼성화, ELISA, 면역침전, 효소 기능과 같은 당업자에게 공지인 기술을 사용하는 단백질 또는 RNA 수준의 검사 뿐만 아니라 당업자에게 공지인 표현형 분석을 포함한다.
본 발명의 약학적 조성물은 개체에게 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 뇌내 투여, 경구 복용, 피하 주사, 복강 투여, 정맥 주사, 근육 주사, 척수 주위 공간(경막내) 주사, 설하 투여, 볼 점막 투여, 직장 내 삽입, 질 내 삽입, 안구 투여, 귀 투여, 비강 투여, 흡입, 입 또는 코를 통한 분무, 피부 투여, 경피 투여 등에 따라 투여될 수 있다. 바람직하게는, 정맥 주사일 수 있다.
본 발명에 따른 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 본 발명이 속하는 기술분야에 통상의 기술자에 의해 용이하게 결정될 수 있다.
본 발명의 일 구현예에 있어서, 상기 세포사멸 유도 핵산 분자는 Bcl2, Bcl-xl, Bcl-w, Mcl-1, A1/BFL-1, XIAP, Survivin, 표피 성장 인자 수용체(Epidermal growth factor receptor, EGFR), 혈관 내피 성장 인자(Vascular endothelial growth factor¸VEGF), KRAS(Kirsten rat sarcoma virus), c-Myc 및 이들의 조합으로 이루어진 군에서 선택되는 유전자를 표적하는 siRNA 인 것이다.
본 명세서에서 용어 "BCL2"는 BCL2 유전자에 의해 코딩되는 단백질을 의미하며, 세포 사멸을 억제하거나 유도하여 세포 사멸을 조절한다. Bax 및 Bak를 포함하는 BCL2 계열의 전-세포사멸 단백질은 일반적으로 미토콘드리아 막에 작용하여 세포사멸 캐스케이드에서 중요한 신호인 시토크롬 c 및 ROS의 투과 및 방출을 촉진한다. 이러한 전-세포사멸 단백질은 BH3-단독 단백질에 의해 차례로 활성화되고, BCL-2 및 그의 상대적인 BCL-X1에 의해 조절된다.

본 명세서의 용어 "혈관 내피 성장 인자(VEGF)"는 혈관의 성장을 촉진하는 단백질로 종양의 성장과 생존에 필수적이며. VEGF 유전자를 표적으로 하는 siRNA는 암의 전임상 모델에서 혈관 신생(새로운 혈관 형성)과 종양 성장을 억제하는 것으로 밝혀졌다.
본 명세서의 용어"KRAS"는 폐암과 췌장암을 포함한 여러 유형의 암에서 돌연변이가 발생하는 단백질로, 접근 가능한 결합 부위의 부족으로 인해 siRNA로 KRAS를 표적으로 삼는 것이 어려웠지만, 최근의 발전으로 KRAS 발현과 종양 성장을 억제할 수 있는 가능성이 발견되었다.
본 명세서의 용어 "c-Myc"는 여러 유형의 암에서 과발현되는 전사인자로 세포 성장과 증식을 조절하는 데 중요한 역할을 수행하며, siRNA로 c-Myc를 표적으로 삼으면 암세포 성장을 억제하고 세포 사멸을 유도하는 것으로 밝혀졌다,
본 발명의 일 구현예에 있어서, 상기 암은 유방암, 난소암, 위암, 폐암, 간암, 간세포암, 기관지암, 비인두암, 후두암, 췌장암, 방광암, 대장암, 결장암, 직장암, 자궁경부암, 뇌암, 전립선암, 골암, 두경부암, 피부암, 갑상선암, 부갑상선암, 편평세포암, 복막암, 신장암, 고환암, 식도암 및 요관암으로 이루어진 군에서 선택되는 것이다. 다만 이에 제한되지는 않으며, 암은 예를 들면, 암종, 림프종, 모세포종, 육종, 신경내분비 종양, 종피종, 슈반세포종, 수막종, 선암종 또는 흑색종일 수 있다.
본 발명의 일 구현예에 있어서, 상기 약제학적 조성물은 종양 조직에 국소 투여되는 것이다.
본원에서 사용된 "국소 투여"은 유기체 내에서 표적 부위에 직접적으로 siRNA 또는 mRNA와 같은 핵산을 전달하는 것을 지칭한다. 예를 들어, 상기 약제학적 조성물은 종양과 같은 질환 부위 또는 염증 부위와 같은 다른 표적 부위 또는 간, 심장, 췌장, 신장 등과 같은 표적 기관에 직접 주사함으로써 국소적으로 투여될 수 있다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:
(a) 본 발명은 지질 성분을 포함하는 나노입자 코어; 및 상기 나노입자 코어의 표면에 코팅된 대식세포 유래 소포를 포함하는 나노입자를 제공한다.

대식세포 유래 소포로 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)를 제작하기 위해 지질 나노입자(LNP)를 제작하였다. 유효 성분을 포함한 지질 나노입자의 합성 과정은 도 2a에 나타내었다. 유효 성분으로는 siRNA를 사용하였으며, 지질은 PEG-DMG 2000, MC3, DSPC 및 콜레스테롤을 사용하였다. 지질 성분은 MC3: DSPC: cholesterol: PEG를 50:10:38.5:1.5의 몰 비율로 사용하였다. 지질나노입자는 자가조립 (self-assembly) 방법으로 제조하였다. 여러 종류의 지질을 상기 비율로 조합하여 에탄올에 용해시켜 준비하고, 지질과 유전자의 질량비는 20으로 구성하여 pH5.2의 25mM 아세트산나트륨(sodium acetate)용액에 siRNA를 넣고, 에탄올과 아세트산나트륨용액의 부피비를 1:3으로 구성하여 두 용액을 섞어 지질 나노입자가 자가조립(self-assembly)되도록 하였다. 에탄올을 제거하기위해 분자량 10K 센트리콘(centricon)을 이용하여 PBS에 10배 희석하여 3000rpm, 20분, 4℃조건에서 3번의 원심분리기를 수행하여 지질나노입자를 수득했다. 수득된 지질나노입자는 4℃에서 보관했다.
제작한 지질 나노입자(LNP)의 물리적 특성을 검증하였다. 결과는 도 2b, 도 2c 및 도 2d에 나타내었다. 도 2의 b)에 나타낸 바와 같이, siRNA가 봉입된 지질 나노입자(LNP)는 198nm의 크기를 가지고, 0.24mV의 중성의 전하를 가짐을 확인할 수 있었다. 도 2c에 나타낸 바와 같이 siRNA가 봉입된 지질 나노입자(LNP)는 구형의 형태를 가짐을 확인하였다. 도 2d에 나타낸 바와 같이 지질 나노입자(LNP)는 물, 인산 완충 생리식염수 및 세포 배양액에서의 지질 나노입자(LNP) 크기를 시간 흐름에 따라 확인하였을 때, 96시간이 경과하였을 때도 크기가 일정하게 유지됨을 확인할 수 있었으며, 이는 제작한 지질 나노입자(LNP)가 안정성이 높음을 의미한다.
제작한 지질 나노입자(LNP)의 유효성분 봉입율 및 암세포 전달 효율을 확인하였다. 결과는 도 2e 및 도 2f에 나타내었다. 도 2e에 나타낸 바와 같이, 지질 나노입자(LNP) 내부의 siRNA 봉입율을 확인 시, 64%의 봉입율을 가짐을 확인할 수 있었다. 도 2f에 나타낸 바와 같이, 대장암세포주 CT26에 siRNA가 봉입된 지질 나노입자(LNP)를 처리하였을 때, siRNA를 나타내는 녹색 형광이 대장암세포 내부에서 확인되는 바, 봉입된 siRNA가 대장암세포로 전달됨을 확인할 수 있었다.
1-2. 대식세포 유래 소포의 제작
대식세포 유래 소포로 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 제작을 위해 대식세포 유래 소포를 제작하였다. 대식세포는 면역세포 중 부착 단백질의 발현이 높고, 다른 면역세포와 상호작용이 활발하여, 암세포와 결합력이 높고 다른 면역세포를 활성화시킬 수 있을 것으로 판단하였다. 도 3a는 대식세포 유래 소포의 제작 과정을 나타낸다. 대식세포주에 LPS를 처리하여 염증성 대식세포로 극성화하고 초음파분쇄기로 세포에 물리적 자극을 가한 후, 원심분리를 통해 핵을 제거하고 세포막을 회수하여 압출기를 통해 10 ㎛, 5 ㎛, 1 ㎛, 0.4 ㎛의 미세공극을 가지는 막을 다수 통과하여 염증석 대식세포 유래 소포를 제작하였다.
제작한 대식세포 유래 소포(M0-C-LNP, M1-C-LNP)의 물리적 특성을 검증하였다. 결과는 도 3b, 도 3c 및 도 3e에 나타내었다. 도 3b에 나타낸 바와 같이, 비염증성 및 염증성 대식세포 유래 소포(M0-C-LNP, M1-C-LNP)의 크기를 확인하였을 때, 각각 205, 187nm의 크기를 가짐을 확인할 수 있었다. 또한 도 3c에 나타낸 바와 같이, 비염증성 및 염증성 대식세포 유래 소포(M0-C-LNP, M1-C-LNP)의 표면전하를 확인하였을 때, 각각 -24.35, -24.42 mV로 표면전하에 큰 차이가 없음을 확인할 수 있었다. 도 3e에 나타낸 바와 같이, 소포의 형태를 확인하였을 때 염증성 대식세포 유래 소포(M0-C-LNP, M1-C-LNP)는 구형의 형태를 가짐을 확인하였다.
도 3d에 나타낸 바와 같이 비염증성 및 염증성 대식세포 유래 소포(M0-C-LNP, M1-C-LNP)의 단백질에서 소포 마커 확인 시, 각각의 입자에서 소포 마커의 발현을 확인할 수 있었다. 도 3h에 나타낸 바와 같이, 비염증성 및 염증성 대식세포 유래 소포(M0-C-LNP, M1-C-LNP)의 단백질에서 염증 관련 마커의 발현 확인 시, 염증성 대식세포 유래 소포에서 염증 관련 마커의 발현이 높게 유지되고 있음을 확인할 수 있었다. 도 3f에 나타낸 바와 같이, 염증성 대식세포 유래 소포를 염색하여 대장암세포주 CT26에 처리하였을 때, 대식세포 유래 소포를 나타내는 붉은 형광이 염증성 대식세포 유래 소포가 암세포 표면에서 관찰되는 바, 대식세포 유래 소포가 암세포막에 융합됨을 확인할 수 있었다.
1-3. 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 제작
대식세포 유래 소포(M1-EV)로 지질 나노입자(LNP)를 코팅하여 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)를 제작하였다. 염증성 대식세포 유래 소포가 코팅된 지질 나노입자의 제작 과정은 도 4a에 나타내었으며, 형광 발현을 통해 나노입자 합성 비율을 확인하였다. 염증성 대식세포 유래 소포체와 siRNA가 봉입된 지질 나노입자를 형성하고, 각 입자를 동일 입자 수로 맞추어 섞은 후 압출기를 통해 100 nm의 구멍을 가지는 막을 다수 통과하여 염증성 대식세포 유래 소포체로 코팅된 지질 나노입자를 제작하였다.
대식세포 유래 소포(M0-C-LNP, M1-C-LNP)와 지질 나노입자(LNP)의 입자 수 비율에 따른 대식세포 유래 소포가 코팅된 지질 나노입자의 합성 효율을 검증하였다. 결과는 도 4b에 나타내었다. 도 4b에 나타낸 바와 같이, 대식세포 유래 소포(M0-C-LNP, M1-C-LNP)와 지질 나노입자(LNP)를 1:1 비율로 하였을 때, 대식세포 유래 소포가 코팅된 지질 나노입자의 합성이 가장 효율적임을 확인할 수 있었다.
제작한 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 특성을 검증하였다. 결과는 도 4d, 도 4e, 도 4f 및 도 4g에 나타내었다. 도 4d에 나타낸 바와 같이, 비염증성 또는 염증성 대식세포 유래 소포로 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 크기를 확인하였을 때, 각각 193, 203nm의 크기임을 확인할 수 있었다. 또한 도 4e에 나타낸 바와 같이, 비염증성 및 염증성 대식세포 유래 소포로 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 표면전하를 확인하였을 때, -24.35, -24.42 mV로 표면전하에 큰 차이가 없음을 확인할 수 있었다. 도 4f에 나타낸 바와 같이, 염증성 대식세포 유래 소포로 코팅된 나노 지질입자(M1-C-LNP)는 구형의 형태를 가짐을 확인하였다. 도 4g에 나타낸 바와 같이, 비염증성 및 염증성 대식세포 유래 소포로 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 단백질에서 소포 마커 확인 시, 각각의 입자에서 소포 마커를 발현하고 있음을 확인할 수 있었다.
실시예 2: 염증성 대식세포 유래 소포로 코팅된 지질 나노입자의 siRNA 전달 효율 검증
대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 약물 전달체로서의 가능성을 검증하기 위해 siRNA 봉입율을 확인하였다. 결과는 도 5a에 나타내었다. 도 5a에 나타낸 바와 같이, 지질 나노입자(LNP), 비염증성 또는 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 siRNA 봉입율 확인 결과, 소포 코팅 이후에도 나노입자 내부에 siRNA가 봉입되어 있음을 확인할 수 있었다.
대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 siRNA 전달 효율을 검증하였다. 결과는 도 5b 및 도 5c에 나타내었다. 도 5b에 나타낸 바와 같이, 대장암세포에 대한 지질 나노입자(LNP), 비염증성 또는 염증성 대식세포 유래 소포가 코팅된 지질 나노입자의 siRNA 전달능력 검증 결과, 비염증성 또는 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)는 같은 입자 수에서 지질 나노입자(LNP)보다 적은 유전자 양을 봉입하고 있음에도 암 세포 내부에 붉은 형광이 더욱 많이 관찰되므로 더 많은 유전자를 대장암세포에 전달함을 확인할 수 있었다.
또한, 도 5c에 나타낸 바와 같이, 나노입자 처리시 대장암세포에서 타겟 유전자 Bcl2의 RNA발현을 확인하였을 때, 타겟 유전자의 발현이 억제됨을 확인할 수 있었는데, 이는 대식세포 유래 소포가 코팅된 나노입자(M0-C-LNP, M1-C-LNP)가 암세포에 siRNA를 효과적으로 전달함을 나타낸다. 또한, 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 siRNA의 봉입율이 지질 나노입자(LNP)에 비해 낮았으나, 타겟 유전자 Bcl2의 발현 억제 효율은 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)에서 더 높음을 확인할 수 있었다.
제작한 나노입자가 siRNA의 전달을 통해 실제 세포 사멸 효과를 발휘하는 지 여부를 대장암세포에서 검증하였다.
결과는 도 5d, 도 5e 및 도 5f에 나타내었다.
도 5d 및 도 5e에 나타낸 바와 같이, 세포 사멸 관련 단백질의 발현 정도를 확인한 결과, 지질 나노입자(LNP)와 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP) 처리군의 BCL2 단백질의 발현량이 낮음을 확인할 수 있었다. 세포 사멸 효과를 확인한 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)에서 지질 나노입자 대비 세포 사멸 효과가 약 5 내지 8배로 매우 높게 나타났는데, 이는 지질 나노입자와 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)와 지질 나노입자(LNP)의 BCL2의 단백질 발현량의 차이보다 매우 큰 차이로 확인된다. 이는, 단순히 타겟 유전자 Bcl2의 발현 억제뿐아니라, 대식세포 유래 소포의 코팅으로 인한 상승효과가 작용했음을 나타낸다. 이는 도 5f의 세포 증식/사멸 염색 이미지에서도 확인할 수 있다.
실시예 3: siRNA를 포함하는 염증성 대식세포 유래 소포로 코팅된 지질 나노입자의 항암 효과 검증
동물 실험을 통해 siRNA를 포함하는 대식세포 유래 소포로 코팅된 지질 나노입자(M1-C-LNP)의 항암효과를 검증하였다. 동물 실험 스케쥴은 도 6a에 나타내었다.
효율적인 암 치료 및 부작용 감소의 가능성을 확인하기 위해, 제작한 각 나노입자들의 종양 체류 시간을 검증하였다.
결과는 도 6b, 도 6c, 도 6d 및 표 1에 나타내었다.
도 6b에 나타낸 바와 같이, 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP), 대식세포 유래 소포로 코팅된 지질 나노입자(M1-C-LNP)의 종양 체류 기간을 형광 발현 및 형광값을 정량해 확인하였을 때, 염증성 대식세포 유래 소포(M1-EV)와 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)의 종양 체류 기간이 길게 유지됨을 확인할 수 있었다. 이는 도 6c 및 도 6d에 나타낸 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)에서의 부착 단백질 발현을 검증 결과에서 확인할 수 있듯이, 염증성 대식세포 유래 소포(M1-EV)와 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)에서 부착 단백질의 발현이 높은 것에 기인한 것으로 판단된다. 나노입자의 종양 체류 기간이 길다는 것은, 나노입자 투여시 효과적으로 종양을 치료할 수 있다는 것, 나노입자가 다른 세포로 유입되어 세포 사멸 등의 부작용을 유발할 가능성이 낮다는 것을 의미할 수 있다.

도 6e에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스의 종양에서의 세포 사멸 관련 마커 및 종양 미세환경 관련 마커의 단백질 발현 확인 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP) 처리군에서 종양 내 세포 사멸 마커의 발현이 증가함을 확인할 수 있었다.
도 6f에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자가(M1-C-LNP) 처리된 마우스의 종양에서의 세포 사멸 관련 마커 및 종양 미세환경 관련 마커의 단백질 발현 정량 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP) 처리군에서 세포 사멸 유도가 통계적으로 유의미하게 가장 높은 것을 확인할 수 있었다. 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP) 처리군에서 세포 사멸 유도 정도는 대조군 대비 약 3배 이상이었다.
도 6g에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스 종양의 성장을 확인한 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP) 처리군에서 마우스 종양의 성장억제가 통계적으로 유의미하게 효과적임을 확인할 수 있었다. 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP) 처리군은 유일하게 처리 후 23일 경과시 대조군 대비 종양 크기가 50% 이하로 감소하였다.
도 6h에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스 종양의 조직 구조와 세포 증식 마커의 발현 확인 시, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자 처리군에서, 종양 조직 밀도가 낮아지며, 세포 증식 마커의 발현이 감소함을 확인할 수 있었다.
실시예 4: siRNA를 포함하는 염증성 대식세포 유래 소포로 코팅된 지질 나노입자의 독성 검증
제작한 나노입자의 생체 적합성을 확인하기 위해 마우스에서 독성 여부를 검증하였다.
결과는 도 7a, 도 7b 및 도 7c에 나타내었다.
도 7a에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스의 혈액 독성 평가 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자의 독성은 대조군과 비교 시 큰 차이가 없음을 확인하였다.
도 7b에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스의 주요 장기 독성 평가 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)의 독성은 대조군과 비교 시 큰 차이가 없음을 확인할 수 있었다. 특히 염증성 대식세포 유래 소포(M1-EV)의 경우 특히 간에서 독성을 보였으나, 이를 지질 나노입자에 코팅하면 독성이 완화 또는 제거됨을 확인할 수 있었다.
도 7c에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스의 몸무게 변화를 확인하였을 때, 대조군과 큰 차이가 없음을 확인할 수 있었다.
염증성 대식세포 유래 소포(M1-EV)의 독성이 코팅으로 인해 완화 및 제거됨을 확인하기 위해 코팅 전후의 독성을 검증을 다시 검증해보았다.
결과는 도 8에 나타내었다.
도 8에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV)와 지질 나노입자(LNP)의 혼합물과 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스의 혈액 독성 평가 결과, 염증성 대식세포 유래 소포(M1-EV)와 지질 나노입자(LNP)의 단순 혼합물은 혈액 독성을 보인 반면, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)는 혈액 독성이 없음을 확인할 수 있었다.
실시예 5: 염증성 대식세포 유래 소포로 코팅된 지질 나노입자의 면역 세포로의 약물 전달성 검증
제작한 나노입자의 높은 암세포 특이성 및 면역 치료 활용 가능성을 확인하기 위해 면역 세포에 siRNA의 전달 여부를 확인하였다.
결과는 도 9의 a), 도 9의 b), 도 9의 c) 및 도 9의 d)에 나타내었다.
도 9의 a)에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 T세포 및 NK세포에서의 세포 사멸율 확인 시, 80% 이상의 생존율을 가짐을 확인할 수 있었다.
도 9의 b)에 나타낸 바와 같이, 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 T세포 및 NK세포에서의 전달된 유전자의 위치를 확인했을 때, 유전자가 세포막에 존재함을 확인할 수 있었다.
도 9의 c)에 나타낸 바와 같이, 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 대장암세포, T세포 및 NK세포에서의 전달된 유전자의 루시퍼레이즈 발현 확인 결과, T세포 및 NK세포에서는 암세포인 CT26세포와 달리 루시퍼레이즈의 발현 정도가 미약했으므로, 대장암세포로는 유전자가 잘 전달되지만 T세포 및 NK세포에는 유전자가 전달되지 않음을 확인할 수 있었다.
또한, 도 9의 d)에 나타낸 바와 같이, 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 T세포 및 NK세포에서의 전달된 유전자의 타겟 단백질의 발현 확인 시, 타겟 유전자 발현의 변화를 확인할 수 없었다.
이는 염증성 대식세포 유래 소포로 코팅된 지질 나노입자(M1-C-LNP)는 siRNA를 면역 세포로 전달하지 않음을 나타내고, 따라서 면역 세포의 사멸을 유도하지 않음을 의미한다.
실시예 6: 염증성 대식세포 유래 소포로 코팅된 지질 나노입자의 면역 세포 활성화 검증
면역 세포에 siRNA를 전달하지 않는 것에 그치지 않고, 제작한 나노입자의 면역 치료 효과를 확인하기 위해, 나노입자 처리시 면역세포를 활성화 여부를 검증하였다.
결과는 도 10a, 도 10b, 도 10c, 도 10d 및 도 10e에 나타내었다.
도 10a에 나타낸 바와 같이, 비염증성 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 RNA에서의 염증 관련 마커 확인 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)에서 염증성 사이토카인의 발현이 높게 유지됨을 확인하였다.
도 10b에 나타낸 바와 같이, 비염증성 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M0-C-LNP, M1-C-LNP)의 단백질에서 염증 관련 마커를 확인 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)에서 염증 관련 마커의 발현이 높게 유지되고 있음을 확인하였다.
도 10c에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 T세포 및 NK세포에서 과립 매개 사멸 관련 마커의 단백질 발현을 확인 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리되었을 때 과립 매개 사멸 관련 마커의 발현이 증가함을 확인하였다.
도 10d에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 T세포 및 NK세포에서 과립 매개 사멸 관련 마커의 단백질 발현을 정량 결과, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리되었을 때 과립 매개 사멸 관련 마커의 발현이 통계적으로 유의미하게 증가함을 확인하였다.
제작한 나노입자로 인해 활성화된 면역 세포의 항암 효과를 검증하였다.
결과는 도 10e 및 도 11의 a) 및 도 11의 b)에 나타내었다,
도 10e에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 T세포 및 NK세포와 공배양한 대장암세포의 세포 사멸율 확인 시, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 T세포 및 NK세포와 공배양한 대장암세포의 대부분이 사멸함을 확인하였다.
도 11의 a)에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자(LNP) 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스 종양 내 T세포 및 NK세포의 분포 확인 시, T세포 및 NK세포의 분포의 차이가 거의 없음을 확인할 수 있었다. 반면 도 11의 b)에 나타낸 바와 같이, 염증성 대식세포 유래 소포(M1-EV), 지질 나노입자 및 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 마우스 종양 내 T세포 및 NK세포 내 세포 활성 마커 확인 시, 염증성 대식세포 유래 소포가 코팅된 지질 나노입자(M1-C-LNP)가 처리된 종양 내 T세포와 NK세포에서의 세포 활성 마커가 통계적으로 유의미하게 증가함을 확인할 수 있었다.
즉, 본원에서 제작한 염증성 대식세포 유래 소포로 코팅된 지질 나노입자(M1-C-LNP)는 암세포에 siRNA를 효과적으로 전달하는 것뿐만 아니라, 면역 세포를 활성화시켜 면역 치료 효과를 가짐을 확인하였다.
Claims (18)
- a) 지질 성분을 포함하는 나노입자 코어; 및 b) 상기 나노입자 코어의 표면에 코팅된 대식세포 유래 소포를 포함하는 나노입자.
- 제1항에 있어서, 상기 대식세포는 염증성 대식세포(M1 macrophage)인 것인, 나노입자.
- 제1항에 있어서, 상기 지질 성분은 콜레스테롤, 이온화 가능한 지질, 양이온성 지질, 인지질, PEG-지질 또는 이들의 조합을 포함하는 것인, 지질 나노입자.
- 제3항에 있어서, 상기 지질 성분은 다음과 같은 것인, 지질 나노입자:
a) 상기 이온화 가능한 지질은 4-(dimethylamino)butanoate (D-Lin MC3-
DMA 또는 MC3) 인 것;
b) 상기 인지질은 DSPC(distearoylphosphatidylcholine)인 것; 또는
c) a) 및 b)의 조합.
- 제1항에 있어서, 상기 나노입자 코어에 대한 대식세포 유래 소포의 개수 비는 1.0 내지 3.0인 것인, 나노입자.
- 제1항에 있어서, 상기 나노입자의 크기는 100nm 내지 300nm인 것인, 나노입자.
- 제1항에 있어서, 상기 나노입자 코어는 소수성 약물 또는 핵산 분자를 포함할 수 있는 것인, 나노입자.
- 제7항에 있어서, 상기 핵산 분자는 DNA, 상보성 DNA(complementary DNA, cDNA), RNA, 메신저 RNA(messenger RNA, mRNA), 운반 RNA(transfer RNA, tRNA), 라이보좀 리보핵산(rRNA), 마이크로 RNA(micro RNA), 작은 간섭 RNA(small interference RNA, siRNA), 짧은 헤어핀 RNA(short hairpin RNA, shRNA), 앱타머(aptamer), 안티 센스 올리고뉴클레오타이드, 리보자임(ribozyme), 펩타이드 핵산(Peptide nucleic acid, PNA), DNAzyme 및 단일 가이드 RNA(single guide RNA, sgRNA)로 이루어진 군에서 선택되는 것인, 나노입자.
- 제1항에 있어서, 상기 대식세포 유래 소포는 염증성 사이토카인을 포함하는 것인, 나노입자.
- 제9항에 있어서, 상기 염증성 사이토카인은 IL-6(interleukin-6), iNOS(induced nitric oxide synthase), TNF-α(tumor necrosis factor- α), IL-1β(interleukin-1β), 및 이들의 조합으로 이루어진 군에서 선택되는 것인, 나노입자.
- 제1항에 있어서, 상기 나노입자는 다음과 같은 특성을 갖는 것인 나노입자:
(a) 표적 세포에 약물을 전달하는 것;
(b) 종양 체류 시간이 향상된 것;
(c) 면역 세포의 활성화를 유도하는 것; 및
(d) (a) 내지 (c)에서 선택되는 2이상의 것으로 이루어진 조합.
- 제11항에 있어서, 상기 면역 세포는 NK 세포, T 세포, 또는 이들의 조합인 것인, 나노입자.
- 다음 단계를 포함하는, 지질 성분을 포함하는 나노입자 코어; 및 상기 나노입자 코어의 표면에 코팅된 대식세포 유래 소포를 포함하는 나노입자의 제조 방법:
(a) 지질 나노입자 및 대식세포 유래 소포를 준비하는 단계;
(b) (a) 단계에서 제작한 지질 나노입자 및 대식세포 유래 소포의 균질화 단계; 및
(c) (b) 단계에서 균질화 된 지질 나노입자 및 대식세포 유래 소포를 융합시키는 코팅 단계.
- 제13항에 있어서, 상기 (b) 균질화 단계는 압출(extrusion)을 통해 90nm 내지 110nm의 구멍을 갖는 막을 통과시키는 과정을 포함하는 것인, 나노입자의 제조 방법.
- a) 지질 성분을 포함하는 나노입자 코어; b) 상기 나노입자 코어의 표면에 코팅된 대식세포 유래 소포; 및 c) 상기 나노입자 코어에 탑재된 세포사멸 유도 핵산 분자 또는 소수성 약물을 포함하는 나노입자를 포함하는 암 예방 또는 치료용 약제학적 조성물.
- 제15항에 있어서, 상기 세포사멸 유도 핵산 분자는 Bcl2, Bcl-xl, Bcl-w, Mcl-1, A1/BFL-1, XIAP, Survivin, 표피 성장 인자 수용체(Epidermal growth factor receptor, EGFR), 혈관 내피 성장 인자(Vascular endothelial growth factor¸VEGF), KRAS(Kirsten rat sarcoma virus), c-Myc 및 이들의 조합으로 이루어진 군에서 선택되는 유전자를 표적하는 siRNA인 것인, 암 예방 또는 치료용 약제학적 조성물
- 제15항에 있어서, 상기 암은 유방암, 난소암, 위암, 폐암, 간암, 간세포암, 기관지암, 비인두암, 후두암, 췌장암, 방광암, 대장암, 결장암, 직장암, 자궁경부암, 뇌암, 전립선암, 골암, 두경부암, 피부암, 갑상선암, 부갑상선암, 편평세포암, 복막암, 신장암, 고환암, 식도암 및 요관암으로 이루어진 군에서 선택되는 것인, 암 예방 또는 치료용 약제학적 조성물.
- 제15항에 있어서, 상기 약제학적 조성물은 종양 조직에 국소 투여되는 것인, 암 예방 또는 치료용 약제학적 조성물.
