Abstract
본 발명은 카페인 외에 디메틸글리신 및/또는 디메틸글리신의 염 역시도 함유하는 조성물에 관한 것이다. 또한, 본 발명은 모발, 두피 및/또는 피부 트리트먼트를 위한 이 조성물의 용도(화장용 및/또는 의료용)에 관한 것이다.
Description
본 발명은 카페인 외에 디메틸글리신 및/또는 디메틸글리신의 염 역시도 함유하는 조성물에 관한 것이다. 또한, 본 발명은 모발, 두피 및/또는 피부 트리트먼트를 위한 이 조성물의 용도(화장용 및/또는 의료용)에 관한 것이다.
인간의 모발은 신체에 대한 보호 기능으로서의 중요성을 크게 상실했다. 그러나, 건강한 모발은 전세계적으로 여성과 남성에게 큰 문화적 의미를 있으며 종종 부와 건강의 표시로 여겨진다. 결과적으로, 예를 들어 탈모에 의해 야기되는 모발 가늘어짐은 종종 삶의 질에 부정적 영향을 미친다. 안드로겐과 같은 호르몬에 의해 유발되는 탈모는 안드로겐 탈모증(androgenetic alopecia 또는 AGA)로 알려져 있으며, 특히 남성에 있어서는 물론이고 여성에 있어서도 탈모의 가장 흔한 원인이다. 남성과 여성의 탈모 패턴은 구분될 수 있다.
모발 성장과 모발 재성장은 혈액에 의해 제공되는 모낭에 대한 적절한 영양 공급에 달려 있다. 따라서, 두피에서의 혈류(미세 순환) 감소는 탈모를 촉진하거나 심지어 탈모를 유발할 수 있다. 예를 들어, 대머리 남성은 정상 두피 모발을 가진 남성보다 두피의 정점 영역으로의 혈류가 훨씬 적다. 따라서, 이러한 미세 순환의 현저한 감소는, 예를 들어 안드로겐 탈모증에서, 탈모 및 모발 재성장(휴지기에서 성장기로의 전환) 부족에 대한 설명이 될 수 있다. 대체로, 탈모는 말초 혈관에서의 혈류 감소를 나타내는 첫 번째 임상 징후들 중 하나로 볼 수 있다.
카페인은 아프리카, 동아시아 및 남미에 자생하는 다양한 식물들의 씨앗, 견과류 및 잎에서 얻어지며, 가장 잘 알려진 카페인 소스는 소위 커피 빈(coffee bean), 즉 커피 식물의 씨앗이다. 카페인은 다양한 약리학적 효과를 갖는 것으로 알려져 있다. 이것은 중축 신경계에 대한 자극 효과를 갖는 것으로 알려져 있다. 이것은 세계에서 가장 일반적으로 사용되는 향정신성 물질로 여겨지며, 다른 향정신성 물질들과는 달리 합법적이어서 사실상 전세계적으로 제한이 없다. 예를 들어, 이것은 커피나 차와 같은 음료를 통해 경구로 섭취될 수 있을 뿐만 아니라, 카페인 경구 섭취가 특정 기능 장애들에 대해 긍정적인 영향을 미침을 시사하는 증거가 점점 늘어나고 있다.
또한, 카페인은 피부 장벽을 비교적 쉽게 통과할 수 있기 때문에 국소 적용에 적합한 화합물이다. 따라서, 유럽 출원 EP 1 396 261 A1은 스킨 케어 조성물의 유효 성분으로서 카페인의 사용, 특히 남성 피부의 케어를 위한 카페인-함유 스킨 케어 조성물의 사용, 및 그러한 스킨 케어 조성물의 제조방법을 기술하고 있다. 카페인의 사용은 특히 남성 피부에서 표피 재생을 지원한다. 안드로겐 탈모증 치료에 카페인을 사용하는 것은 특히 Daniels, G., Akram, S., Westgate, G. 현재까지 안드로겐 탈모증과 같은 탈모용으로 승인된 약학적 치료법은 거의 없으며, 기존 치료법은 효과가 충분하지 않거나 불쾌한 부작용을 일으킨다. 염증으로 인한 급성 원형 탈모(원형 탈모증(alopecia areata) 또는 줄여서 AA로 알려져 있기도 함) 및 그 밖의 원인으로 인한 탈모에 대한 치료 옵션과 관련해서도 마찬가지로 불만족스러운 상황이다.
소위 피부 부속물로서의 모발 외에도 피부의 기능 역시 건강한 외모에 매우 중요하다. 모발 또는 모근에 영향을 미치는 생리학적 효과(예를 들어, 염증) 역시도 피부에 유사한 방식으로 영향을 미친다. 따라서, 피부 트리트먼트는 보호 기능 향상 및 차단 특성 강화와 같은 의학적 이유 및 더욱 매끄럽고 단순히 더 아름다운 안색과 같은 순전히 시각적 및 심미적 이유 모두를 위해 대단히 중요하다.
따라서, 개선된 트리트먼트 방법에 대한 필요성, 특히 탈모 방지를 위해 국소적으로 사용될 수 있으며 부작용이 없거나 무시할 수 있을 정도에 불과한 조성물에 대한 필요성이 여전히 존재한다. 특히, 본질적으로 약이며 어떠한 부작용도 나타내지 않는 조성물 또는 본질적으로 화장품인 조성물에 대한 필요성이 존재한다.
또한, 피부의 느낌과 외관을 모두 향상시킴과 동시에 피부의 보호 기능 및 차단 기능을 강화하는 개선된 스킨 케어 조성물에 대한 필요성도 존재한다.
이에 기초하여, 본 발명의 목적은 모발, 두피 및 피부의 트리트먼트를 위한, 특히 탈모 예방 및/또는 치료를 위한 우수한 내약성의 조성물을 제공하는 것이다. 또한, 이 조성물은 국소적으로 적용될 수 있어야 하고 선행기술로부터 공지된 조성물들의 단점들을 극복할 수 있어야 한다.
놀랍게도, 이 목적은 청구항 제1항에 따른 조성물 및 청구항 제7항에 따른 그 용도에 의해 달성되었다. 종속항들에는 바람직한 실시예들이 명시되어 있다.
본 발명에 따르면, 상기 목적은 (A) 디메틸글리신 및/또는 디메틸글리신의 염 및 (B) 카페인을 포함하는 조성물을 제공함으로써 달성된다.
놀랍게도, 본 발명에 따른 유효 성분 조합(카페인과 디메틸글리신 및/또는 디메틸글리신의 염)은 탈모, 특히 유전성 탈모 및 노화 관련 탈모의 치료에 있어서 카페인 또는 디메틸글리신 및/또는 디메틸글리신의 염이 단독으로 갖는 효능에 비해 개선된 효능을 갖는 것으로 나타났다. 상기 유효 성분들의 조합은 피부 및 두피에서의 미세 순환을 기대 이상으로 증가시키며, 피부와 모근으로의 영양분 및 산소 공급을 크게 향상시킨다. 또한, 의학적으로나 미용적으로도 피부에서 내약성이 매우 좋다.
디메틸글리신(N,N-디메틸글리신)은 식물, 동물 및 인간에서 발견되지만, 인간에서는 극히 소량만 형성된다. 이것은 베타인 호모시스테인 메틸라제(betaine homocysteine methylase)를 이용한 베타인의 아미노기 전이반응(transamination)에 의해 콜린(choline)으로부터 글리신(glycine)이 다단계 생합성되는 과정에서 중간체로서 형성된다.
(디메틸아미노)아세트산이기도 한 N,N-디메틸글리신은 다음의 화학식 1로 표시된다:

디메틸글리신의 사용은 물론이고 그 염, 용매화물 및 수화물의 사용 역시도 독창적인 것이다. 이들은 바람직하게는 디메틸글리신의 약학적으로 또는 미용적으로 허용 가능한 염이다. 상기 염은 20℃에서 적어도 10g/l의 수중 용해도(solubility in water)를 갖는 수용성 염인 것이 특히 바람직하다.
바람직한 실시예에서, 상기 디메틸글리신의 염은 디메틸글리신의 알칼리 염, 알칼리 토금속 염 또는 암모늄 염이다.
예로는 나트륨, 칼륨, 칼슘, 마그네슘 및 암모늄 염들이 있다. 암모늄 염에서 암모늄 양이온은 1 내지 4개의 탄소 원자를 각각 독립적으로 갖는 1 내지 4 개의 알킬기를 캐리(carry)한다. 디메틸글리신의 나트륨 염 및 칼륨 염이 바람직하며, 디메틸글리신의 나트륨 염, 즉 소듐-N,N-디메틸글리신이 특히 바람직하다.
대안적으로 바람직한 실시예에서, 상기 디메틸글리신의 염은 디메틸글리신과 무기 및/또는 유기 산의 염일 수 있다.
디메틸글리신과 무기산의 염의 예로는 디메틸글리신의 염산염, 브롬화수소산염, 요오드화수소산염, 황산수소염, 황산염, 아황산수소염, 아황산염, 탄산수소염, 탄산염, 일인산염(monophosphate), 이인산염(diphosphate) 및 삼인산염(triphosphate), 그리고 이들의 혼합물이 있다. 디메틸글리신의 염산염이 특히 바람직하다.
디메틸글리신과 유기산의 염의 예로는 디메틸글리신의 아세테이트, 락테이트(lactate), 시트레이트(citrate), 숙시네이트(succinate), 푸마레이트(fumarate), 말리에이트(maleate) 및 벤조에이트(benzoate), 그리고 이들의 혼합물이 있다.
본 발명에 따른 디메틸글리신 및/또는 디메틸글리신의 염은 케라티노사이트(keratinocytes)에서 산소 전환(oxygen turnover) 및 세포 활성을 향상시키고, 따라서 피부 및 모낭에서 세포 활성을 촉진하는 것으로 추정된다. 또한, 피부 장벽을 강화하고, 상처 치유를 지원하며, 피부를 매끄럽게 한다. 본 발명에 따르면, 디메틸글리신 및/또는 디메틸글리신의 염은 모근 및 피부를 강화하는 상당한 효과를 달성하고, 특히, 일상적으로 또는 노화와 관련되어 스트레스를 받거나 약해진 피부 및 탈모(예를 들어, 유전성 탈모 및 노화 관련 탈모)를 치료함에 있어서 본 발명에 따른 카페인의 효과를 놀라울 정도로 명확히 지원한다.
카페인의 IUPAC명은 1,3,7-트리메틸-3,7-디하이드로-1H-퓨린-2,6-디온이다. 대안적으로, 카페인은 1,3,7-트리메틸크산틴으로 지칭되기도 한다. 카페인은 아래의 화학식 2로 표시된다.

카페인은 메틸크산틴 계열에 속하는 메틸크산틴 알칼로이드이다. 이것은 쓴 맛이 나는 결정질 물질이고, 퓨린 유도체로 간주될 수 있으며, 화학적으로 디옥시리보핵산 및 리보핵산의 아데닌 염기 및 구아닌 염기와 관련이 있다.
본 발명에 따른 조성물은 디메틸글리신 및/또는 디메틸글리신의 염을 바람직하게는 상기 조성물의 총 중량을 기준으로 0.001 중량% 내지 10.0 중량%의 비율로 함유한다. 바람직한 실시예에서, 본 발명에 따른 조성물은 디메틸글리신 및/또는 디메틸글리신의 염을 0.01 내지 8.0 중량%, 더욱 바람직하게는 0.1 내지 6.0 중량%, 더욱 바람직하게는 0.3 내지 5.0 중량%, 특히 0.5 내지 3.0 중량%의 비율로 함유하는데, 각각의 경우에서 상기 비율은 상기 조성물의 총 중량을 기준으로 한 것이다. 본 발명의 바람직한 실시예에서, 본 발명에 따른 상기 조성물은 0.1 중량%, 0.2 중량%, 0.3 중량%, 0.4 중량%, 0.5 중량%, 0.6 중량%, 0.7 중량%, 0.8 중량%, 0.9 중량%, 1.0 중량%, 1.1 중량%, 1.2 중량%, 1.3 중량%, 1.4 중량%, 1.5 중량%, 2.0 중량% 또는 2.5 중량%의 디메틸글리신 및/또는 디메틸글리신의 염을 함유할 수 있는데, 각각의 경우에서 상기 비율은 상기 조성물의 총 중량을 기준으로 한 것이다. 본 발명에 따른 상기 조성물은 바람직하게는 디메틸글리신 및/또는 디메틸글리신의 염을 각각의 용매화물 및 수화물(예를 들어, 소듐 디메틸글리시네이트의 이수화물)을 포함하는 순수한 화학 물질로서 포함하는데, 이는 이것이 조성물의 순도를 증가시키고 바람직하지 않은 부작용 발생을 감소시키기 때문이다.
바람직한 실시예에서, 본 발명에 따른 조성물은 카페인을 0.001 내지 3.0 중량%, 더욱 바람직하게는 0.005 내지 2.50 중량%, 더욱 바람직하게는 0.01 내지 2.0 중량%, 특히 0.1 내지 1.5 중량%의 비율로 함유하는데, 각각의 경우에서 상기 비율은 상기 조성물의 총 중량을 기준으로 한 것이다. 본 발명의 바람직한 실시예에서, 본 발명에 따른 상기 조성물은 0.01 중량%, 0.05 중량%, 0.1 중량%, 0.2 중량%, 0.3 중량%, 0.4 중량%, 0.5 중량%, 0.6 중량%, 0.7 중량%, 0.8 중량%, 0.9 중량%, 1.0 중량%, 1.1 중량%, 1.2 중량%, 1.3 중량%, 1.4 중량%, 또는 1.5 중량%의 카페인을 함유할 수 있는데, 각각의 경우에서 상기 비율은 상기 조성물의 총 중량을 기준으로 한 것이다.
디메틸글리신 및/또는 디메틸글리신의 염과 카페인 사이의 중량비는 바람직하게는 10:1 내지 1:10, 바람직하게는 6:1 내지 1:6, 더욱 바람직하게는 4:1 내지 1:4, 특히 바람직하게는 2:1 내지 1:2의 범위이다. 본 발명의 바람직한 실시예에서, 디메틸글리신 및/또는 디메틸글리신의 염과 카페인 사이의 중량비는 0.5; 0.6; 0.7; 0.8; 0.9; 1.0; 1.1; 1.2; 1.3; 1.4; 1.5; 1.6; 1.7; 1.8; 1.9 또는 2.0이다.
바람직한 실시예에서, 본 발명에 따른 조성물은 적어도 하나의 다른 유효 성분을 함유할 수 있는데, 상기 적어도 하나의 다른 유효 성분은 멘톨(menthol), 비오틴(biotin), 아연 PCA(zinc PCA), 니아신아미드(niacinamide), 판테놀(panthenol), 엑토인(ectoine), 유비퀴논-10(ubiquinone-10), 타우린(taurine), 판토락톤(pantolactone), 에키네시아(echinacea), 카르니틴(carnitine), 토코페릴 아세테이트(tocopheryl acetate) 및 이들의 조합들로부터 선택된다.
멘톨은 모노사이클릭 모노테르펜알코올(monocyclic monoterpenealcohol)이며, 혈류를 자극하는 유효 성분으로서 본 발명에 따른 조성물에 첨가될 수 있다. 또한, 멘톨은 두피에 상쾌한 감각 자극을 유발할 수 있다.
비타민 B7 또는 비타민 H로도 알려져 있는 비오틴은 B군(B complex)의 수용성 비타민이다. 본 발명에 따르면, 비오틴은 탈모를 더욱 감소시키고 피부를 강화시킬 수 있다.
아연 PCA는 L-피롤리돈 카르복실레이트의 아연 염이며, 본 발명의 조성물에 항균 효과를 갖는 물질로서 첨가될 수 있다.
니아신아미드(또한, 니코틴아미드)는 니코틴산의 아미드이며, 비타민 B3로도 알려져 있다. 본 발명에 따른 니아신아미드는, 산화 스트레스(oxidative stress) 감소와 같은 다른 특성들에 더하여, 모발 성장 촉진 효과를 갖는다.
판테놀은 체내에서 판토텐산(pantothenic acid)(비타민 B5)으로 변환되는 프로비타민이다. 판토텐산은 조효소 A(coenzyme A)의 일부이며, 따라서 피부 대사에 중요하다. 본 발명에 따르면, 판테놀의 작용에 의해 피부의 탄력과 수분이 더욱 향상된다. 또한, 가려움증과 염증이 완화되고 상처 치유가 촉진된다.
엑토인은 사이클릭 아미노산이며, 공명-안정화된(resonance-stabilized) 양쪽성 이온으로 수용액에 존재한다. 엑토인은 보습 효과를 가지며, 본 발명에 따르면, 모발의 자연 구조를 더욱 안정화시킨다. 엑토인은 자외선으로부터의 보호를 수행하고 염증성 질환 치료에 도움이 될 수 있다는 것도 밝혀졌다.
유비퀴논-10(Q10 또는 조효소 Q10)은 퀴논 유도체이다. 유비퀴논 군에 속하는 Q10은 항산화제로 간주되며, 본 발명에 따르면, 모발, 특히 모근에 대해 안정화 효과가 있다.
본 발명에 따르면, 타우린 또는 2-아미노에탄설폰산 역시도 항산화제로서 피부 및 모발, 특히 모근에 대해 추가적인 안정화 효과를 갖는다.
판토락톤은 치환된 락톤 군으로부터 나오며, 본 발명에 따르면, 모근의 성장 인자를 더욱 자극한다.
본 발명에 따르면, 에키네시아는 피부와 두피에 대해 진정 효과가 있고, 가려움증 및 긴장 발생을 완화시킨다. 또한, 에키네시아는 두피의 혈액 순환을 촉진함으로써 산소와 영양분이 풍부한 혈액을 모낭에 공급하는데, 이것은 모발, 특히 모근에 대해 추가적인 안정화 효과를 가져다 준다.
카르니틴은 아마도 모발 성장-감소 인자의 형성을 억제하고, 본 발명에 따르면, 이러한 방식으로 탈모에도 작용한다.
토코페릴 아세테이트는 항산화 특성을 나타내며, 본 발명에 따르면, 피부 및 모발, 특히 모근에 대해 추가적인 안정화 효과를 갖는다.
본 발명의 바람직한 실시예에서, 본 발명에 따른 조성물은 메탄올, 비오틴, 아연 PCA, 니아신아미드, 판테놀, 엑토인, 유비퀴논, 타우린, 판도락톤, 에키네시아, 카르니틴, 토코페릴 아세테이트 및 이들의 조합들로부터 선택되는 적어도 하나의 다른 유효 성분을 상기 조성물의 총 중량을 기준으로 0.001 내지 10.0 중량%, 더욱 바람직하게는 0.005 내지 7.50 중량%, 더더욱 바람직하게는 0.01 내지 5.0 중량%, 특히 0.1 내지 3.0 중량%의 비율로 각각 함유한다.
또한, 본 발명에 따른 조성물은 바람직하게는 수성(water-based)이다. 이것은 상기 조성물이 바람직하게는 45.0 내지 85.0 중량%의 물을 함유한다는 것을 의미한다.
특히 피부 적용/피부 트리트먼트를 위해, 상기 조성물은 바람직하게는 800 내지 6000 mPa·s, 특히 바람직하게는 1000 내지 5700 mPa·s, 매우 특히 바람직하게는 2500 내지 5500 mPa·s의 점도를 갖는데, 각 경우에 있어서 상기 점도는 Haake RheoStress1 유량계(ThermoFisher Scientific)를 이용하여 플레이트-플레이트 형상(rotary body PP60 Ti)에서 10/s의 전단 속도로 20℃에서 DIN 53019-1:2008-09에 따라 측정되는 값이다.
일 실시예에서, 상기 조성물은 계면활성제를 포함한다. 상기 계면활성제는 음이온성, 비이온성, 양이온성 또는 양쪽성 계면활성제일 수 있다. 상기 계면활성제는 바람직하게는 음이온성 또는 비이온성 계면활성제이며, 특히 바람직하게는 가능한 한 순한(즉, 특히 피부에 좋은) 음이온성 또는 비이온성 계면활성제이다. 본 발명에 따르면, 비이온성 계면활성제는 특히 매우 우수한 유화 특성 및 우수한 스킨 케어 특성을 갖기 때문에 사용된다. 음이온성 계면활성제는 세정 기능이 특히 높다는 점에서 바람직하다. 따라서, 이들은 샴푸와 같은 세정 조성물에 특히 유용하다. 양이온성 계면활성제는 우수한 헤어 케어 특성을 가지며, 본 발명에 따라 특히 컨디셔너, 샴푸 및 트리트먼트와 같은 헤어 케어 조성물에 사용된다.
본 발명에 따른 조성물은 계면활성제를 2 내지 40 중량%, 특히 5 내지 30 중량%, 바람직하게는 7 내지 20 중량%, 특히 바람직하게는 10 내지 17 중량%의 양으로 함유하는 것이 바람직한데, 각각의 경우에서 상기 양은 상기 조성물의 총 중량을 기준으로 한 것이다. 계면활성제의 적합한 양은 8 중량%; 9 중량%; 10 중량%; 11 중량%; 12 중량%; 13 중량%; 14 중량%; 15 중량%; 16 중량%; 17 중량%; 18 중량%; 19 중량%; 20 중량%; 21 중량%; 22 중량%; 23 중량%; 24 중량%; 25 중량%인데, 각각의 경우에서 상기 양은 상기 조성물의 총 중량을 기준으로 한 것이다. 본 발명의 국소 조성물(topical composition)은 하나 이상의 음이온성 계면활성제를 0.1 내지 20 중량%, 바람직하게는 1 내지 17 중량%, 특히 바람직하게는 5 내지 15 중량%의 양으로 함유하는 것이 특히 바람직한데, 각각의 경우에서 상기 양은 상기 조성물의 총 중량으로 기준으로 한 것이다. 음이온성 계면활성제의 적합한 양은 1 중량%; 2 중량%; 3 중량%; 4 중량%; 5 중량%; 6 중량%; 7 중량%; 8 중량%; 9 중량%; 10 중량%; 11 중량%; 12 중량%; 13 중량%; 14 중량%; 15 중량%; 16 중량%; 17 중량%; 18 중량%; 19 중량%; 20 중량%인데, 각각의 경우에서 상기 양은 상기 조성물의 총 중량을 기준으로 한 것이다. 이러한 양에서 상기 계면활성제는 특히 높은 세정 성능을 가지며 피부, 두피 및 모발에 대해 내약성이 매우 뛰어나다.
본 발명의 계면활성제는 특히 책 “Surfactants and interfacial phenomena”(저자: Milton Rosen 및 Joy Kunjappu, 출판사: John Wiley & Sons, Inc., 2012, 제4판)에 기재되어 있다.

헤어 컨디셔너는 모발 표면 상의 전하를 중화시킴으로써 모발의 정전기를 줄일 수 있다. 헤어 컨디셔닝제의 예로는 4차 암모늄 화합물이 있다.
지질 보충 물질 또는 과지방제(superfatting agents)로도 알려져 있는 지질 보충제는 표피 장벽 기능에 파괴적 영향이 미치는 것을 방지할 수 있는 친유성 물질이다. 지질 보충제의 예로는 울 왁스(wool wax), 스쿠알렌, 액체 파라핀, 식물성 오일, 실리콘 및 세틸 팔미테이트(cetyl palmitate)가 있다.

항산화제 또는 항-산화제는 본 발명의 조성물에서 다른 성분들의 산화를 지연시키거나 완전히 없애는 화합물이다. 사용 가능한 항산화제의 예로는 시트르산(citric acid), 아스코르브산(ascorbic acid) 및 부틸화 하이드록시아니솔(butylated hydroxyanisole)이 있다.
레올로지 조정제 및 증점제는 본 발명의 조성물의 적용 특성을 개선하는데 도움이 될 수 있다. 정제염(염화 나트륨)의 첨가는 레올로지 조정제 및 증점제로 여겨질 수 있다. 염화 나트륨을 첨가함으로써 본 발명에 따른 조성물의 유동성이 소정 한계 내에서 영향을 받을 수 있고 요구 수준으로 조정될 수 있다. 셀룰로오스 유도체 또는 폴리아크릴레이트 역시도 증점제로 사용될 수 있다.
본 출원의 맥락에서, 케어 제품(care products)은 모발 및/또는 (두피) 피부를 케어하는 물질을 의미하는 것으로 이해되어야 한다. 가수분해 밀 단백질(hydrolyzed wheat protein) 및 알란토인(allantoin)은 두피와 모발에 영양을 공급한다. 가수분해 밀 단백질은 주로 보습 특성을 갖고 있다.
본 발명의 조성물에 특징적 색상을 부여함으로써 다른 제품들로부터 쉽게 구별될 수 있도록 하기 위하여 염료가 선택적으로 사용될 수 있다. 그러나, 본 발명의 범위 내에서, 인간의 모발을 착색시키기 위하여 염료가 사용될 수도 있다.

본 발명에 따르면, 상기 조성물은 국소적으로 적용된다. 국소적 적용은 외부 적용(external application), 특히 국부적 외부 적용(local, external application)을 의미한다. 본 발명에 따른 조성물은 바람직하게는 피부, 두피 및 모발에 대한 케어 제품(즉, 샴푸, 컨디셔너, 트리트먼트, 에멀젼, 로션, 샤워 젤, 데이 크림, 페이스 크림, 페이스 플루이드 및/또는 토닉과 같은 트리트먼트제)이며, (순수한) 스타일링 제품(styling products)은 아니다. 다시 말해, 본 발명의 국소 조성물은, 효과가 케라틴화된 모발 자체에 미칠 뿐 두피(즉, 대사 활성 모근)에는 미치지 않는 헤어 스타일링에 그 일차적 목적이 있는 것이 아니다. 오히려, 피부, 두피 및 모발의 의학적/약학적 또는 미용적 트리트먼트에 중점을 둔다.

일 실시예에서, 본 발명에 따른 조성물은 리브-온 제형(leave-on formulation) 또는 린스-오프 제형(rinse-off formulation)의 형태일 수 있다.

연구 설계 및 결과
1. 체외 연구의 일환으로, 인간의 뿔-형성 각질 세포를 이용한 세포 배양 모델에서 카페인 또는 Na-디메틸글리네이트 그리고 이들의 조합의 효과를 시험하였다. 이를 위해, HaCaT 세포를 DMEM 배지(소 태아 혈청 및 항생제-항진균제 혼합물 포함)에서 1일, 3일, 5일, 및 7일 동안 배양하였고, 무엇보다도 세포의 생존력(viability), 증식(proliferation) 및 이동(migration)을 적절한 측정 방법을 이용하여 측정하였으며, 세포 성장과 관련된 성장 인자의 발현을 측정하였다.

인간 표피 각질 세포(HaCaT)를 96-웰 플레이트에 5,000 세포/웰의 세포 밀도로 시딩하고 배지(10% FBS, 1% 페니실린-스트렙토마이신 및 0.5% 펀지존(Fungizone)이 가해진 DMEM)에서 배양하였다. 다음 날에 이어 추가로 24시간, 48시간 및 72시간 동안 37℃ 및 5 부피%의 CO2에서 배양한 후, 세포 배양 배지를 상기 유효 성분 농도의 소듐 디메틸글리시네이트(DMG) 및 카페인(caff) 또는 이들의 조합으로 처리하거나 이것들 없이(대조군) 처리하였다. 측정은 이전에 B. I. Tth, N. Dobrosi, A. Dajnoki, G. Czifra, A. Olh, A. G. Szllsi, I. Juhsz, K. Sugawara, R. Paus, T. Br, J Invest Dermatol 2011 , 131, 1095-1104에서 발표된 연구와 유사하게 수행되었다.
도 1은 24시간, 48시간 및 72시간의 배양 후의 세포 생존력을 보여주는데, 72시간 후의 경우에 있어서 DMG와 카페인의 조합 첨가로 인한 증가를 볼 수 있다. 세포 생존력은 MTT 분석을 이용하여 측정되었다. 흡수(absorption)는 4회 반복하여 측정되었고 24시간에서의 대조군에 대해 정규화되었다(normalized). 표준편차와 함께 평균값이 표시된다.
증식의 증거:
증식을 측정하기 위해 소위 CyQUANT 분석이 수행되었다. 이 형광-기반 분석(fluorescence-based assay)에서는, 사용된 형광 염료가 DNA(데옥시리보핵산)에 결합하며, 세포 DNA의 함량이 샘플의 세포 수에 대한 직접적 측정이 된다.

도 2는 24시간, 48시간 및 72시간의 배양 후의 세포 증식을 보여주는데, 72시간 후의 경우에 있어서 DMG와 카페인의 조합 첨가로 인한 명확한 파격적 증가를 볼 수 있다. 세포 증식은 CyQUANT 분석을 이용하여 측정되었다. 형광(fluorescence)은 3회 반복하여 측정되었고 24시간에서의 대조군에 대해 정규화되었다. 표준편차와 함께 평균값이 제공된다.
이동의 증거:
이동 측정을 위해, 이전에 T. Kawabata, T. Otsuka, K. Fujita, G. Sakai, R. Matsushima-Nishiwaki, O. Kozawa, H. Tokuda, International journal of molecular medicine 2018, 42, 3149-3156에서 발표된 연구를 기반으로 소위 상처 치유 분석(wound healing assay)이 수행되었다. 이 원리는, 시간의 경과에도 아직 서식하지 않는 배양 표면으로의 세포 이동을 측정하는 것을 기반으로 한다. 이를 위해, 실리콘 삽입물(표준화된 폭을 갖는)에 의해 분리된 2개의 인접한 웰 또는 캐비티에 20,000개의 세포들을 각각 시딩하고 배지(5% FBS, 1% 페니실린-스트렙토마이신 및 0.5% 펀지존이 가해진 DMEM)에서 48시간 동안 37℃ 및 5 부피%의 CO2에서 배양하였다. 이어서 상기 플라스틱 삽입물을 제거하였고(“상처 생성”), 시간의 경과에도 세포가 서식하지 않은 배양 표면을 이미지를 사용하여 측정함으로써 세포의 이동을 문서화하였다. 동시에, 각 경우에 이미 존재하는 상기 유효 성분 농도의 소듐 디메틸글리시네이트(DMG) 및 카페인 또는 이들의 조합을 갖거나 갖지 않는(대조군) 세포 배양 배지가 상기 플라스틱 삽입물을 제거한 직후(0h) 및 추가로 24시간이 경과한 후에 체인지되었다. 이미지 평가 및 세포가 서식하지 않는 배양 표면에 대한 측정이 소프트웨어(ImageJ)를 사용하여 수행되었다.
도 3은 서로 다른 농도들의 DMG 및 카페인을 이용한 시간의 경과에 따른 이동을 보여준다. 대조군 및 DMG 또는 카페인의 개별 적용에 비해 DMG와 카페인의 조합에서 이동 또는 상처 봉합이 파격적으로 빠르게 일어난다. 시간 0h에 상처가 생성되었고, 16시간, 20시간 및 24시간의 배양 후에 이미지를 촬영하였다. 이 이미지들을 기초로, 아직 세포가 서식하지 않는 배양 표면을 ImageJ 소프트웨어를 이용하여 측정하였다. 측정 값들은 0h 시점(최대 크기의 상처)에 대해 정규화되고 %로 주어진다.
VEGF 유전자 발현 검출:
VEGF(Vasular Endothelial Growth Factor)는 새로운 혈액 및 림프관의 성장과 형성을 촉진한다. 유전자 발현은 정량적 실시간 PCR(qRT-PCR)로 알려진 표준 방법을 사용하여 측정되었다.
인간 표피 각질 세포(HaCaT)를 6-웰 플레이트에 140,000 세포/웰의 세포 밀도로 시딩하고 배지(10% FBS, 1% 페니실린-스트렙토마이신 및 0.5% 펀지존이 가해진 DMEM)에서 배양하였다. 다음 날, 각 경우에 이미 존재하는 상기 유효 성분 농도들을 갖거나 갖지 않는(대조군) 세포 배양 배지가 체인지되었고, 24시간 후에 세포가 체취되었다. 유전자 발현은 이전에 B. V. Diaz, M.-C. Lenoir, A. Ladoux, C. Frelin, M. Demarchez, S. Michel, Journal of Biological Chemistry 2000, 275, 642-650에서 발표된 연구에 기반하여 qRT-PCR을 이용하여 측정되었다.
도 4는 24시간의 배양 후의 VEGF 유전자 발현을 보여주는데, DMG를 카페인과 조합하여 첨가함으로써 유전자 발현이 상당히 증가한 것을 볼 수 있다. qRT-PCR에 의해 측정된 상대적 VEGF 유전자 발현(3회 측정, 항시적 발현 유전자(GAPDH - glyceraldehyde-3-phosphate dehydrogenase)의 각 유전자 발현에 대해 정규화 됨)이 나타나 있다. 표준편차와 함께 평균값이 제공된다.
요약하면, 특히 카페인과 DMG의 조합이 성장과 관련된 HaCaT 세포 파라미터에 긍정적 영향을 미치고 카페인 또는 DMG 단독으로 HaCaT 세포를 처리하는 것에 비해 성장 인자 VEGF 발현이 상당히 증가하였음이 놀랍게도 밝혀졌다.
2. 무작위 배정 및 위약-대조군 연구 설계를 통한 이중-맹검 연구의 일부로서, 실시예 1의 본 발명에 따른 샴푸 및 참조예 1a, 1b 및 1c의 샴푸들이 유전성 탈모가 있는 남녀에 의해 6개월 동안 사용되었다. 처음과 6개월 후에 탈모율이 피부과 감독 하에 측정되었고, 피험자들은 적용 종료 시 탈모 감소에 대해 주관적으로 평가를 하고 그 결과를 설문지를 이용하여 기록하도록 요청 받았다.
실시예 1의 본 발명에 따른 샴푸를 사용한 피험자들이 가장 현저한 탈모 감소를 결정할 수 있었던 것으로 밝혀졌다. 이것은, 같은 그룹의 피험자들에서 3가지 참조 삼푸들 중 하나를 사용한 피험자들에 비해 현격히 증가된 탈모율 감소가 측정된 결과와 일치한다.
Claims (10)
- (A) 디메틸글리신(dimethylglycine) 및/또는 디메틸글리신의 염, 그리고
(B) 카페인(caffeine)

상기 조성물은 0.001 내지 10.0 중량%의 디메틸글리신 및/또는 디메틸글리신의 염을 함유하는,

상기 조성물은 0.001 내지 3.0 중량%의 카페인을 함유하는,
조성물. - 제1항 내지 제3항 중 어느 한 항에 있어서,
상기 조성물은 멘톨(menthol), 비오틴(biotin), 아연 PCA(zinc PCA), 니아신아미드(niacinamide), 판테놀(panthenol), 엑토인(ectoine), 유비퀴논-10(ubiquinone-10), 타우린(taurine), 판토락톤(pantolactone), 에키네시아(echinacea), 카르니틴(carnitine), 토코페릴 아세테이트(tocopheryl acetate) 및 이들의 조합들로부터 선택되는 적어도 하나의 다른 유효 성분을 함유하는,
조성물. - 제1항 내지 제4항 중 어느 한 항에 있어서,
상기 조성물은 헤어 컨디셔닝제(hair conditioning agents), 지질 보충제(lipid replenishers), 방부제(preservatives), 안정제(stabilizers), 방향제(fragrances), 항산화제(antioxidants), 레올로지 조정제(rheology modifiers), 증점제(thickeners), 컨디셔닝제(conditioning agents), 염료(dyes), 진주 광택제(pearlescent agents), 미백제(lightening agents), 용제(solvents), pH 조절제(pH modifiers), 및 이들의 조합들로부터 선택되는 적어도 하나의 첨가제를 함유하는,

상기 조성물은 국소적으로 적용되고/적용되거나, 바람직하게는 피부 및 모발의 신진대사를 촉진하기 위한 케어 조성물(care composition)인,

상기 모발 및 두피의 트리트먼트는 탈모의 치료 및/또는 예방을 위한 것이고, 상기 피부의 트리트먼트는 표피 장벽 기능(epidermal barrier functions) 및 표피 장벽 완전성(epidermal barrier integrity)을 향상시킴으로써 상기 피부의 외관을 개선하고 상기 피부의 수분 함량을 증가시키며 상처 치유를 향상시키기 위한 것인,
조성물의 용도. - 제7항 및/또는 제8항에 있어서,
상기 탈모는 원형 탈모증(alopecia areata)[원형 탈모(circular hair loss)], 여성과 남성에 있어서의 안드로겐 탈모증(alopecia androgenetica)[유전성 탈모(hereditary hair loss)], 미만성 탈모증(diffuse alopecia)[미만성 탈모(diffuse hair loss)], 연령-관련 탈모[노화 탈모증(senescent alopecia)] 및 항암화학요법(chemotherapy)으로 인한 탈모로부터 선택되는,
