Abstract

Description
본 발명은 다브라페닙 메실레이트 또는 타핀라르®(Tafinlar®)로 알려지고 이하 화합물 A로 지칭되는, 다음의 화학식 I로 표시되는, N-{3-[5-(2-아미노-4-피리미디닐)-2-(1,1-디메틸에틸)-1,3-티아졸-4-일]-2-플루오로페닐}-2,6-디플루오로벤젠설폰아미드, 메탄설포네이트 염을 포함하는 경구 현탁액용 분산성 정제에 관한 것이다:

N-{3-[5-(2-아미노-4-피리미디닐)-2-(1,1-디메틸에틸)-1,3-티아졸-4-일]-2-플루오로페닐}-2,6-디플루오로벤젠설폰아미드(이하 화합물 B)는, 그 전체 개시 내용이 본원에 참조로서 포함되는 국제 출원일이 2009년 5월 4일이고; 국제 공개 번호 WO/2009/137391 및 국제 공개일이 2009년 11월 12일인, 국제 출원 번호 PCT/US2009/042682에서 BRAF 활성의 저해제로서, 구체적으로 암의 치료에 있어서 유용한 것으로, 유리 염기로서 이의 약학적으로 허용되는 염 및 용매화물과 함께 개시되고 청구된 화합물이다. 화합물 B는 실시예 58a의 화합물이다.

화합물 A 및 하이프로멜로스, 하나 이상의 충전제, 하나 이상의 붕해제, 및 하나 이상의 윤활제가 블렌더에서 함께 혼합된다. 마그네슘 스테아레이트를 이용한 활택 후, 혼합물은, 예를 들어 롤러 다짐 및 과립화 분쇄기를 사용하여, 건식 과립화를 위해 처리된다.
외부 상은 하나 이상의 약학적으로 허용되는 부형제를 포함하고, 예를 들어 확산 혼합기를 사용하여, 내부 상과 혼합된다. 바람직하게는, 하이프로멜로스, 하나 이상의 충전제, 및 하나 이상의 붕해제가 첨가된다. 가장 바람직하게는, 만니톨 및 미정질 셀룰로스가 외부 상에 충전제로서 첨가된다. 더욱 더 바람직하게는, 만니톨은 분산성 정제의 총 중량을 기준으로 약 12 중량% 내지 45 중량%의 범위로 외부 상에 첨가되고 미정질 셀룰로스는 분산성 정제의 총 중량을 기준으로 약 8 중량% 내지 20 중량%의 범위로 외부 상에 첨가된다. 가장 바람직하게는, 크로스포비돈은 붕해제로서 외부 상에 첨가된다. 더욱 더 바람직하게는, 크로스포비돈은 분산성 정제의 총 중량을 기준으로 약 1 중량% 내지 5 중량%, 보다 바람직하게는 5 중량% 미만의 범위로 외부 상에 첨가된다.
하이프로멜로스, 하나 이상의 충전제, 및 하나 이상의 붕해제를 포함하는 외부 상은, 예를 들어 확산 혼합기를 사용하여, 내부 상으로부터의 과립과 함께 혼합된다. 마그네슘 스테아레이트를 이용한 활택 후, 최종 블렌드를 적합한 회전식 프레스를 사용하여 압축하여 분산성 정제를 생산한다.
본 발명의 일 구현예에서, 분산성 정제의 제조 방법은
(a) 다음을 포함하는 내부 상을 형성하는 단계
(i) 화합물 A를 약학적으로 허용되는 부형제와 함께 혼합하는 단계,
(ii) 건식-과립화하는 단계,
(b) 다음을 포함하는 외부 상을 형성하는 단계
(i) 추가의 약학적으로 허용되는 부형제를 내부 상에 첨가하고 혼합하는 단계;
(c) 다음에 의해 분산성 정제를 형성하는 단계

"내부 상"은 활성 성분 화합물 A 및 하나 이상의 약학적으로 허용되는 부형제를 포함하는 과립 상(단계 (i) 및 (ii))을 의미한다.
"외부 상"은 내부 상(과립)에 첨가된 하나 이상의 약학적으로 허용되는 부형제를 의미한다(단계 (iii) 및 (iv)).
"분산성 정제의 총 중량"은 내부 및 외부 상인 정제의 중량을 의미한다.
물리적 및 화학적 안정성은 임의의 통상적인 방식으로 테스트될 수 있으며, 예를 들어, 분산성 정제는, 예를 들어 실온, 즉 25℃/60% RH에서의 저장 및/또는 40℃/75% RH에서의 저장 이후의, 용해, 파쇄성, 붕해 시간, 분산도, 화합물 A에 대한 분석, 분해 산물 및 외관의 측정에 의해 테스트될 수 있다.
분산성 정제의 형상 다양할 수 있으며, 예를 들어 원형, 타원형, 직사각형, 원통형 또는 임의의 다른 적합한 형상일 수 있다. 본 발명의 구현예에서, 상기 기재된 압축 방법에 의해 수득된 분산성 정제는 원형 또는 타원형이다. 분산성 정제의 가장자리는 경사지거나 둥글 수 있고 금이 그어질 수 있다. 가장 바람직하게는, 분산성 정제는 양면이 볼록한 경사진 모서리를 갖는 원형이다.
본 발명의 일 구현예에서, 분산성 정제는 활성 성분으로서 10 mg 내지 25 mg 용량의 화합물 A, 바람직하게는 활성 성분으로서 10 mg 용량의 화합물 A를 포함한다.
본 발명에 따른 분산성 정제는, 바람직하게는 경사진 모서리를 갖는 양면이 볼록한 원형이다. 분산성 정제는 5 mm 내지 10 mm, 바람직하게는 5 mm 내지 7 mm의 범위, 더욱 바람직하게는 6 mm의 직경을 갖는다.
본 개시 내용에 따른 정제의 경도, 또는 파쇄 저항성은 표준 테스트에 의해 결정될 수 있다. 정제 경도는, 바람직하게는 유럽 약전 2.9.8에 명시된 표준 테스트에 따라 결정된다. 크래머(Kraemer)® 3S 정제 테스트 장치와 같은 장치가 사용될 수 있다. 이 테스트는 정제의 파쇄에 대한 저항성을 결정하며, 파쇄함으로써 정제를 분쇄하는 데 필요한 힘으로 측정된다.

화합물 A, 약 5% 내지 10% w/w 크로스포비돈, 약 5% 내지 10% w/w 하이프로멜로스(여기서 하이프로멜로스는, 수 중 2 중량%에 대해 20℃에서 측정될 때, 4 mPa s 내지 6 mPa s, 바람직하게는 5 mPa s의 공칭 점도 및 28% 내지 30% 메톡실 치환 또는, 수 중 2 중량%에 대해 20℃에서 측정될 때, 80 mPa s 내지 120 mPa s, 바람직하게는 100 mPa s의 점도, 및 19% 내지 24% 메톡실 치환을 가짐), 및 약 2.5:1 내지 2:1의 중량비로 존재하는 충전제 만니톨 및 미정질 셀룰로스를 포함하는 제형은 낮은 파쇄성 값 및 짧은 붕해 시간을 갖는 분산성 정제를 생산하는 데 사용될 수 있으며, 이는 유럽 약전 규격을 준수함이 밝혀졌다.
본 발명의 분산성 정제에 또한 개별적인 외관을 부여하고 즉시 인식 가능하도록 하기 위하여 착색 및/또는 표시될 수 있다. 염료 또는 진홍색 색소의 사용은 분산성 정제 식별뿐만 아니라 외관 향상에 역할을 할 수 있다. 본 발명의 분산성 정제에 임프린트 코드를 사용하여 표시될 수 있다.
본 발명의 분산성 정제는 BRAF-돌연변이 양성 고형 종양의 치료에 유용하다.
본 발명의 분산성 정제의 활성 및 특징은 표준 임상 시험 및/또는 동물 시험에서 나타날 수 있다.
또한, 수득된 본 발명의 분산성 정제는 생산 공정까지 및 통상적인 포장, 예를 들어 밀폐된 알루미늄 블리스터 팩에서의, 예를 들어 2년 또는 심지어 3년 동안의 보관 동안 모두에서 안정하다. 통상적인 테스트에서 결정될 때 이 시간 동안 활성 성분으로서 약 5% 미만, 예를 들어 2 또는 3% 이하의 화합물 A가 분해될 수 있다. 예를 들어, HDPE 충전된 병 또는 블리스터에서 1년 내에 활성 성분으로서 1% 미만의 화합물 A가 분해된다.
본 발명은 또한 분산성 정제 형태의 화합물 A를 이러한 치료를 필요로 하는 포유동물, 바람직하게는 인간 대상체에 투여하는 방법에 관한 것이다. 본 발명은 또한 상기 언급된 질병 또는 장애 중 하나에 대한 포유동물, 바람직하게는 인간 대상체의 치료에서의 분산성 정제 형태의 화합물 A의 용도에 관한 것이다. 본 발명은 특히 활성 성분으로서 화합물 A의 4.5 mg/kg 내지 5.25 mg/kg 체중/일의 일일 투여량이 환자에게 투여되는 방법에 관한 것이다. 임의의 구체적인 환자에 대한 특정 투여량 수준은 연령, 체중, 전반적인 건강 상태, 하나 이상의 활성 약물과의 약물 병용, 질병의 유형 및 중증도를 포함하는 다양한 인자에 의존할 것이 이해될 것이다.
의약 포장은 본 발명에 따른 분산성 정제 및 하나 이상의 화합물 A 분산성 정제가 경구 투여되도록 권고하는 인쇄된 설명서를 포함한다.
본 발명의 또 다른 구현예에서, 치료법에 사용하기 위한 화합물 A의 분산성 정제가 제공된다.
또 다른 양태에서, 본 발명은 (i) 조성물을 수성 매질과 조합하는 단계, (ii) 조성물이 수성 매질에서 분산하여 분산액을 형성하도록 하는 단계 및 (iii) 분산액을 섭취하는 단계를 포함하는, 요법을 필요로 하는 환자에게 본 발명의 약학적 조성물을 투여하는 방법을 제공한다.
실시예
본원에서 이들 공정, 설계 및 실시예에서 사용된 기호 및 관례는 동시대 과학 문헌, 예를 들어, 문헌[Journal of the American Chemical Society] 또는 [Journal of Biological Chemistry]에서 사용된 것과 일치한다. 달리 표시되지 않는 한, 모든 온도는 ℃(섭씨 온도)로 표현된다.

표 1에 제시된 바와 같이, 화합물 A의 분산성 정제는 3분 이하의 붕해 시간 및 100턴 후 0.5% 미만의 낮은 파쇄성을 갖는 빠르게 분산되는 조성물을 제공한다. 다양한 크로스포비돈 대 하이프로멜로스 비율을 테스트하였다. 더 낮은 점도의 하이프로멜로스 등급을 갖는 제형은 더 긴 붕해 시간을 초래하였다. 하이프로멜로스 수준(7.5% w/w) 및 크로스포비돈 수준(7.5% w/w)은 동일하지만 공칭 점도 및 메톡실 치환 면에서 동일한 하이프로멜로스 등급이 아닌 제형(형태 2a 및 형태 5a)은 더 높은 점도의 하이프로멜로스 등급 제형(형태 5a)에서 더 짧은 붕해 시간을 초래했다. 더 낮은 하이프로멜로스 수준(5% w/w) 및 더 높은 크로스포비돈 수준(10% w/w)을 갖는 제형은 사용된 하이프로멜로스 등급의 점도 및 메톡실 치환 백분율에 관계없이, 유사한 붕해 시간(NMT 1분)을 나타냈다(형태 1a 및 형태 4a). 붕해 시간에 대한 하이프로멜로스 공칭 점도의 영향은 더 높은 하이프로멜로스 수준이 화합물 A의 분산성 정제 제형에 사용될 때 더욱 두드러지게 나타난다.

정제 제조 방법
블렌딩-체질-블렌딩
정제의 내부 상의 성분을 롤러 다짐을 위해 제조한다. 화합물 A, 미정질 셀룰로스, 아세설팜 포타슘, 크로스포비돈, 콜로이드 이산화 규소, 향미료, 하이프로멜로스, 및 만니톨을 적합한 크기의 블렌더에서 혼합하고 블렌딩한다. 블렌딩된 물질을 적합한 크기의 체에서 선별하고 적합한 mrl의 블렌더로 옮기고 블렌딩한다.
마그네슘 스테아레이트를 적합한 크기의 체에서 선별하고 블렌딩된 물질을 함유하는 적합한 크기의 블렌더로 옮긴 다음 추가 시간 동안 블렌딩한다.
롤러 다짐 및 분쇄
활택된 블렌드를 롤러 다짐기를 사용하여 리본으로 건식 과립화한다. 다져진 리본을 선별스크린으로 통과시켜 적합한 크기의 과립을 생산한다.
블렌딩-체질-블렌딩
정제의 외부 상의 성분을 정제화를 위해 제조한다. 추가 양의 미정질 셀룰로스, 만니톨, 하이프로멜로스 및 크로스포비돈을 적합한 크기의 블렌더에서 혼합하고 블렌딩한다. 블렌딩된 물질을 적합한 크기의 체에서 선별하고 내부 상의 과립과 함께 적합한 크기의 블렌더로 옮기고 블렌딩한다. 블렌드를 혼합하여 내부 상 및 외부 상 물질을 조합한다.
마그네슘 스테아레이트를 적합한 크기의 체에서 선별하고 블렌딩된 물질을 함유하는 적합한 크기의 블렌더로 옮긴 다음 추가 시간 동안 블렌딩한다.
압축
활택된 블렌드를 6 mm 원형, 경사진 모서리의 압형이 장착된 회전식 정제 프레스 상에서 목표 80 mg 중량으로 압축하여 10 mg 분산성 정제를 생산한다. 압축된 정제는 개별 중량 변동, 외관, 경도, 두께, 파쇄성 및 붕해 시간에 대한 공정-중 모니터링을 위해 샘플링한다.
정제는 필요에 따라 10 개 정제를 함유하는 건조제를 갖는 HDPE 용기 또는 블리스터(열 밀폐 가능한 래커 알루미늄 호일이 뒤를 받치는 PVC/PVDC) 내로 포장한다.
실시예 3
상기에 의해 본 발명의 바람직한 구현예가 예시되어 있지만, 본 발명은 본원에 개시된 특정 설명에 한정되지 않으며 다음의 청구 범위의 범주 내에 속하는 모든 변경에 대한 권리는 보유됨이 이해되어야 한다.
Claims (20)
- (a) 화합물 A, (b) 하이프로멜로스, 및 (c) 정제의 제조에 적합한 적어도 하나의 약학적으로 허용되는 부형제를 포함하는 분산성 정제로서, 화합물 A는 정제의 총 중량을 기준으로 5 중량% 내지 40 중량%의 양으로 존재하는, 분산성 정제.
- 제1항에 있어서, 하이프로멜로스는 정제의 총 중량을 기준으로 약 1 중량% 내지 약 13 중량%로 존재하는, 분산성 정제.
- 제2항에 있어서, 하이프로멜로스는 정제의 총 중량을 기준으로 약 5 중량% 내지 약 10 중량%로 존재하는, 분산성 정제.
- 제3항에 있어서, 하이프로멜로스는, 수 중 2 중량%에 대해 20℃에서 측정될 때, 4 mPa s 내지 6 mPa s, 바람직하게는 5 mPa s의 공칭 점도, 및 28% 내지 30% 메톡실 치환을 갖는, 분산성 정제.
- 제3항에 있어서, 하이프로멜로스는, 수 중 2 중량%에 대해 20℃에서 측정될 때, 80 mPa s 내지 120 mPa s, 바람직하게는 100 mPa s의 점도, 및 19% 내지 24% 메톡실 치환을 갖는, 분산성 정제.
- 제1항에 있어서, 정제는, 유럽 약전 2.9.1의 붕해 테스트에 따라 측정된, 15℃ 내지 25℃의 수 중에서의 정제의 붕해 시간인, 붕해 시간이 3 분 이하인, 분산성 정제.
- 제1항에 있어서, 정제는, 유럽 약전 2.9.8의 정제의 파쇄 저항성 테스트에 따라 측정된, 평균값 경도가 55 N 이하인, 분산성 정제.
- 제1항에 있어서, 약학적으로 허용되는 부형제는 다음을 포함하는, 분산성 정제:
(i) 총량이 정제의 총 중량을 기준으로 약 35 중량% 내지 70 중량%인 적어도 하나의 충전제,
(ii) 총량이 정제의 총 중량을 기준으로 약 2.5 중량% 내지 13 중량%인 적어도 하나의 붕해제,
(iii) 총량이 정제의 총 중량을 기준으로 약 0.1 중량% 내지 2 중량%인 적어도 하나의 활택제, 및
(iv) 총량이 정제의 총 중량을 기준으로 약 0.1 중량% 내지 2.5 중량%인 적어도 하나의 윤활제. - 제8항에 있어서, 충전제는 만니톨 및 미정질 셀룰로스인, 분산성 정제.
- 제9항에 있어서, 만니톨 및 미정질 셀룰로스는 약 2.5:1 내지 2:1의 중량 대 중량 비로 존재하는, 분산성 정제.
- 제8항 내지 제10항 중 어느 한 항에 있어서, 붕해제는 크로스포비돈인, 분산성 정제.
- 제11항에 있어서, 크로스포비돈은 정제의 총 중량을 기준으로 약 5 중량% 내지 10 중량%로 존재하는, 분산성 정제.
- 제8항 내지 제12항 중 어느 한 항에 있어서, 윤활제는 콜로이드 이산화 규소인, 분산성 정제.
- 제8항 내지 제13항 중 어느 한 항에 있어서, 활택제는 마그네슘 스테아레이트인, 분산성 정제.
- (i) 조성물을 수성 매질과 조합하는 단계, (ii) 조성물이 수성 매질에서 분산하여 분산액을 형성하도록 하는 단계, 및 (iii) 분산액을 섭취하는 단계를 포함하는, 제1항의 분산성 정제를 상기 조성물을 필요로 하는 환자에게 투여하는 방법.
- 제1항에 있어서, 암의 치료에 사용하기 위한, 분산성 정제.
- 제16항에 있어서, 암의 치료에 사용하기 위한 것이며, 암은 BRAF-돌연변이 양성 고형 종양인, 분산성 정제.
- (i) 화합물 A 및 적어도 하나의 약학적으로 허용되는 부형제를 혼합하는 단계;
(ii) (i)에서 수득된 혼합물을 과립화하는 단계;
(iii) (ii)에서 수득된 과립 형태를 적어도 하나의 약학적으로 허용되는 부형제와 혼합하여 혼합물을 형성하는 단계; 및
(iv) 단계 (iii)에서 수득된 혼합물을 압축하여 정제를 형성하는 단계를 포함하는, 제1항 내지 제14항 중 어느 한 항의 분산성 정제의 제조 방법. - 제18항에 있어서, 과립화 단계(ii)는 건식 과립화인, 방법.
- 제19항에 있어서, 건식 과립화는 롤러 다짐을 사용 과립화 분쇄기를 사용하는 단계인, 방법.
