본 발명은 피부 주름개선효과를 갖는 기능성 조성물에 관한 것으로써, 더욱 상세하게는 나노화된 금 입자를 포함하는 피부 주름개선효과를 갖는 기능성 조성물에 관한 것이다. The present invention relates to a functional composition having an anti-wrinkle effect, and more particularly, to a functional composition having an anti-wrinkle effect comprising nanoparticles of gold.
금(gold)은 반자성물질로서 이온작용으로 인한 혈액순환을 촉진시켜 피부노화방지 및 기미제거에 좋고, 호르몬 분비촉진으로 인한 피부활성화로 피부주름방지 및 주근깨제거 등에 효과가 있으며, 해독작용으로 인한 상처치유로 인하여 피부염증방지 및 여드름치유에 효과가 있고, 면역기능을 강화시키며, 심장, 비장, 췌장, 근육에서의 손상된 세포를 재생시키는 효능이 있고, 뇌하수체 내분비선을 자극하여 영양성분 흡수를 개선하는 효과가 있는 것으로 알려져있음(대한민국특허 제10-0539111호). 이러한 이유로 최근 들어 금을 함유한 화장품제품들이 등장하고 있으며, 대한민국특허출원 제10-2004-26472호 에서와 같이 금분을 첨가한 화장품 제조방법이 개시된 바 있으나 주로 마이크로단위로 가공된 금을 사용하고 있으며, 이 경우 피부의 보 습성 및 흡수성이 약한 단점이 있음. 최근 활발한 연구가 진행되고 있는 나노기술은 각종 소재분야에 적용되어 그 기존의 단점을 개선하는 획기적인 대안으로 제시되고 있는데, 대한민국특허 10-0586273호는 기존의 반자성 금입자를 상자성 나노입자 분말로 가공하여 흡수율을 개선시킬 수 있는 가능성을 보고하였고, 대한민국특허 제10-0732249호에서는 금과 은 합금 나노입자를 이용한 안료조성물로서 사용 가능성을 제시한바 있음. (실시예1) 세포에서 주름개선효과 Example 1 Wrinkle Improvement Effect in Cells
아데노신 0.04중량%에 평균 50nm 크기의 금입자를 0.001중량%부터 0.1중량%까지 세포배양액에 희석하여 시료를 준비하였다. 섬유아세포를 2x105/ml로 96 well에 접 종하고 다음날 준비된 시료를 10 ul 씩 각 well에 첨가하여 48시간 동안 배양하였다. 각 시간대 별로 배양된 세포 상층액을 취해 세포 밖으로 유리된 MMP-1의 양을 MMP-1 ELISA kit(Amersham)를 사용하여 측정하였다. 표 1에 나타난 바와같이 나노화된 금입자 함량 0.01중량%를 첨가한 조건에서 70% 정도로 가장 높은 MMP-1 저해효과를 나타냈고, 그 이상의 농도에서는 비슷한 효과를 나타냈으며, 대조군1의 아데노신만 첨가시에는 80%저해 효과를 나타냈다. 이것은 나노골드가 MMP-1 발현저해 효과가 있으며 아데노신의 항주름 효과가 한층 증가키는 시너지효과를 통해 항주름 효과를 나타내는 것으로 사료되었다. 나노화된 금입자의 주름개선효과를 알아보기 위해 상기 실시예 2의 나노골드 에센스의 화장품 시료를 성인 여성(나이 20-50세) 60명을 대상으로 사용전후의 설문 조사를 6개월간 실시하였다. 효과에 대한 유효성은 미국 특허 No.5126135의 방법에 따라 이것에 의해 효과가 있을 때 2점, 조금 있을 때 1점, 효과가 없을 때 0점으로 그 점수를 산출해서 유효성을 [합계점/(조사 인원수 X 2)]*100으로 표시한 결과는 표 2와 같다. 나타난 바와 같이 주름개선 효과에 대한 유효성이 90이상으로 매우 우수하게 나타났다.
