Description
[발명이 속하는 기술분야][TECHNICAL FIELD OF THE INVENTION]
본 발명은 유기용매를 이용한 히알루론산 및 그 염의 입자 크기의 조절방법에 관한 것으로, 보다 상세하게는 유기용매의 첨가비를 조절하여 히알루론산 및 그의 염의 입자 크기를 조절하는 것을 포함하는 히알루론산 및 그의 염의 입자 크기 조절방법에 관한 것이다. The present invention relates to a method for controlling the particle size of hyaluronic acid and salts thereof using an organic solvent, and more particularly, to adjust the particle size of hyaluronic acid and salts thereof by adjusting the addition ratio of the organic solvent. A method for controlling particle size of salts.
[종래기술][Private Technology]
히알루론산은 보습효과를 가지며, 물리적 마찰에 대한 윤활효과 및 세균 등의 침입에 대한 보호효과가 우수하여 화장품 첨가제, 관절염치료제, 안과수술용 수술보조제 및 외과수술후의 유착저해제 등으로 광범위하게 사용되고 있다. 이러한 히알루론산은 소의 안구, 동물의 완충조직, 태반 및 닭 벼슬과 같은 생체조직에 다량 함유되어 있다.Hyaluronic acid has a moisturizing effect, has excellent lubricating effect against physical friction and protection against invasion of bacteria and the like, and is widely used as a cosmetic additive, an arthritis treatment agent, an ophthalmic surgery surgical aid, and an adhesive inhibitor after surgery. Such hyaluronic acid is contained in a large amount in biological tissues such as bovine eye, animal buffer tissue, placenta and chicken crest.
The method of obtaining hyaluronic acid is extracted from the above-mentioned biological tissues (US Pat. Nos. 4,141,973 and 4,303,676), or the fermentation of microorganisms is recovered as a fermentation product (Japanese Patent Laid-Open No. 62-2579, Japanese Patent Laid-Open No. 63). -141594, Republic of Korea Patent Registration No. 258237). However, hyaluronic acid obtained by the extraction method is mixed with impurities such as chondroitin sulfate, glycosamino glycansulfate, so as to require a complicated purification process to remove them, it is difficult to mass-produce at high cost. On the other hand, the production of hyaluronic acid using microorganisms is relatively low in production cost, and high molecular weight hyaluronic acid can be obtained by a relatively simple method. Therefore, in recent years, most hyaluronic acid has been produced commercially by microbial fermentation.
Various purification methods (Korean Patent Registration No. 0381407) have been reported according to the diversification of the use of hyaluronic acid, but there is no report on the control of the particle size of the powder after the final drying. Hyaluronic acid and its salts may be mixed with other raw materials to form a powder mixture or, depending on the intended use or application, there is an appropriate particle size. For example, Sweden's Q-med sells a mixture of hyaluronic acid as its main ingredient in fine line, restile and perlane, with fine lines in the upper dermis, restyrene in the middle of the dermis, and perlane in the dermis. Wrinkle improvement procedures that are injected deep or over the subcutaneous tissue are currently being performed in hospitals.
히알루론산의 입자 크기는 분쇄와 같은 물리적 처리를 통하여 조절할 수 있으나, 이 경우 제품의 손실, 흡습, 미생물을 비롯한 기타 오염 등의 문제점이 있어, 입자의 크기를 효율적으로 조절할 수 있는 방법에 대한 연구가 요구되고 있다.The particle size of hyaluronic acid can be controlled through physical treatment such as pulverization, but in this case, there are problems such as loss of product, moisture absorption, microorganisms and other contamination. It is required.
상기 종래기술의 문제점을 해결하기 위하여 도출된 것으로서, 본 발명은 히알루론산 정제 또는 추출과정에서 히알루론산 및 이의 염의 입자크기를 용이하게 조절할 수 있는 방법을 제공하는 것을 목적으로 한다.In order to solve the problems of the prior art, it is an object of the present invention to provide a method for easily adjusting the particle size of hyaluronic acid and salts thereof during the purification or extraction of hyaluronic acid.
또한 본 발명은 일정 크기의 히알루론산 및 이의 염 제조방법을 제공하는 것을 목적으로 한다.It is another object of the present invention to provide a hyaluronic acid and a method for preparing a salt thereof.
In order to achieve the above object, the present invention sequentially adds a salt and an organic solvent to a solution containing hyaluronic acid to precipitate hyaluronic acid and salts thereof, and recovers and dries the hyaluronic acid and salts thereof. As a particle size control method, the organic solvent is selected from the group consisting of methanol, ethanol, acetone and propanol, and provides a method of controlling the particle size of hyaluronic acid and salts thereof by adjusting the addition ratio of the organic solvent.
In another aspect, the present invention provides a method for controlling the particle size of hyaluronic acid and salts thereof, comprising the steps of sequentially adding a salt and an organic solvent to a solution containing hyaluronic acid to precipitate hyaluronic acid and salts thereof, and recovering and drying the precipitate. The organic solvent is selected from the group consisting of methanol, ethanol, acetone and propanol, and provides a method of controlling the particle size of hyaluronic acid and salts thereof by adjusting the concentration of hyaluronic acid contained in the solution.
이하, 본 발명을 상세히 설명한다. Hereinafter, the present invention will be described in detail.
본 발명은 히알루론산 및 이의 염의 입자 크기 조절방법에 관한 것이다. The present invention relates to a method for controlling particle size of hyaluronic acid and salts thereof.
본 발명의 입자 크기 조절방법은 히알루론산이 함유된 용액에 염을 첨가한 후 유기용매를 가하여 침전시키는 것을 포함한다. The particle size adjusting method of the present invention includes adding a salt to a solution containing hyaluronic acid, followed by precipitation by adding an organic solvent.
The solution containing hyaluronic acid may be any kind of liquid substance containing hyaluronic acid. For example, the solution is cultured by culturing the hyaluronic acid-producing microorganisms and filtering the culture medium to remove the cells, followed by a conventional hyaluronic acid purification method. It may be an aqueous solution before the salt is added to the hyaluronic acid, but is not limited thereto. The solution containing hyaluronic acid may contain hyaluronic acid as 0.1 to 2% (w / v), if the concentration of hyaluronic acid is higher than the 2% (w / v) can be adjusted to the range by adding distilled water have.
상기 염은, 나트륨, 칼륨, 마그네슘, 칼슘 등일 수 있으나, 나트륨이 바람직하다. 상기 염은 히알루론산이 함유된 용액에 0.5 내지 3 M로 첨가할 수 있다.The salt may be sodium, potassium, magnesium, calcium or the like, but sodium is preferred. The salt may be added at 0.5 to 3 M in a solution containing hyaluronic acid.
상기 유기용매는 메탄올, 에탄올, 아세톤 및 프로판올로 이루어진 군으로부터 선택될 수 있으나, 이에 한정되는 것은 아니다.The organic solvent may be selected from the group consisting of methanol, ethanol, acetone and propanol, but is not limited thereto.
The organic solvent is added to the solution containing hyaluronic acid at a predetermined ratio, and the particle size of hyaluronic acid and salts thereof precipitated is controlled according to the amount of the organic solvent added. That is, hyaluronic acid having a particle size of 20 to 300 μm and its salt may be prepared by adding the organic solvent in a volume ratio of 50 to 300 with respect to 100 volumes of the solution. In this case, the particle size may vary depending on the content of the hyaluronic acid or the type of the organic solvent in addition to the amount of the organic solvent added.
For example, when the organic solvent is added at a volume ratio of 50 to 100 with respect to 100 volumes of the hyaluronic acid-containing solution, hyaluronic acid having a particle size of 20 to 70 μm and a salt thereof are prepared, and the organic solvent is 100 to 200 volume ratio. When added, hyaluronic acid having a particle size of 60 to 100 µm and salts thereof is prepared, and when added at a volume ratio of 200 to 300, hyaluronic acid having a particle size of 70 to 120 µm and salts thereof is prepared. In this case, the concentration of hyaluronic acid may be included in the solution at 0.2 to 0.5% (w / v).
일예로, 유기용매으로 에탄올을 사용하는 경우, 히알루론산의 함유량에 따른 에탄올의 혼합비 및 제조되는 히알루론산 및 그의 염의 입자크기는 하기 표 1과 같다.For example, when ethanol is used as the organic solvent, the mixing ratio of ethanol according to the content of hyaluronic acid and the particle size of hyaluronic acid and salts thereof prepared are shown in Table 1 below.
(표 1)Table 1
또다른 일예로, 유기용매로 아세톤을 사용하는 경우, 히알루론산의 함유량에 따른 아세톤의 혼합비 및 제조되는 히알루론산 및 그의 염의 입자크기는 하기 표 2와 같다.As another example, when using acetone as an organic solvent, the mixing ratio of acetone according to the content of hyaluronic acid and the particle size of the prepared hyaluronic acid and salts thereof are shown in Table 2 below.
(표 2)Table 2
유기용매를 가하여 침전된 히알루론산 및 이의 염은 통상적인 여과 또는 원심분리 방법으로 용이하게 회수할 수 있으며, 침전물은 건조함으로써 히알루론산 및 이의 염을 수득할 수 있다.Hyaluronic acid and salts thereof precipitated by adding an organic solvent can be easily recovered by conventional filtration or centrifugation, and the precipitate can be dried to obtain hyaluronic acid and salts thereof.
본 발명의 히알루론산 및 이의 염의 입자 크기 조절방법은, 상온, 예컨대 10 내지 40 ℃에서 실시하며, 본 발명이 속하는 분야의 당업자라면 용이하게 변형할 수 있는 범위를 포함한다.The particle size control method of the hyaluronic acid and salts thereof of the present invention is carried out at room temperature, such as 10 to 40 ℃, includes a range that can be easily modified by those skilled in the art.
The method of controlling the particle size of hyaluronic acid and salts thereof of the present invention is not only easy and simple to implement, but also does not cause problems such as product contamination and quality deterioration, compared to conventional physical grinding methods. Thus, the method of the present invention can provide hyaluronic acid and its salts having various particle sizes, thereby expanding the field of application and efficacy of hyaluronic acid and its salts.
이하 본 발명의 실시예를 기재한다. 하기 실시예는 본 발명을 예시하기 위한 것일 뿐 본 발명이 하기 실시예에 한정되는 것은 아니다. Hereinafter, examples of the present invention will be described. The following examples are only for illustrating the present invention and the present invention is not limited to the following examples.
실시예 1 Example 1

Streptococcus sp KL0188 (Accession No. KCTC10248BP) was inoculated in 100 ml of Todd-Hewitt medium, incubated at 35 ° C. until log phase and used as primary seed culture. Primary seed cultures were inoculated in 1 L of Tryptic Soy Broth (Difco, USA), incubated at 35 ° C. to log phase and prepared as secondary seed cultures. Hyaluronic acid production medium (glucose 60g / L, yeast extract 5g / L, casein peptone 17g / L, glutamic acid 7g / L, magnesium sulfate 0.7g / L, dipotassium phosphate 2.5g / L, sodium chloride 5.0g / L, water ) 20 L was put into 30 L of Korean fermenter and sterilized, and 1 L of the secondary seed culture solution was inoculated. The cells were incubated at pH 6.95 to 7.05 at a temperature of 35 ° C. for 20 hours.
1-2. 균체 제거 및 배양액 희석 1-2. Cell removal and culture dilution
히알루론산 배양액에 0.05 %의 포르말린 용액을 가하여 3시간 동안 교반하여 균체를 불활화한 다음, 원심분리 또는 여과하여 균체를 제거하였다. 균체가 제거된 배양여액의 히알루론산 농도는 3 내지 7 g/L이다. 0.05% formalin solution was added to the hyaluronic acid culture and stirred for 3 hours to inactivate the cells, followed by centrifugation or filtration to remove the cells. The hyaluronic acid concentration of the culture filtrate from which the cells were removed was 3 to 7 g / L.
1-3. 에탄올을 이용한 히알루론산나트륨의 회수 1-3. Recovery of Sodium Hyaluronate Using Ethanol
Distilled water was added to the culture filtrate to adjust the hyaluronic acid concentration to 0.1%, 0.2% and 0.5%, respectively, and NaCl was added to each of the 1 M concentrations. Ethanol was added to 70%, 120%, 150%, 200% and 300% of the volume of the culture filtrate, and stirred for 30 minutes at 100 rpm, followed by centrifugation (500 xg, 10 minutes). The precipitated sodium hyaluronate was dried for 24 hours at 50 ° C. using a vacuum oven, and the sodium hyaluronate powder was dispersed in ethanol to measure the average particle size using a particle size analyzer. The particle size of the purified hyaluronic acid according to the ethanol treatment concentration is shown in Table 1. The results of Example 1 are shown in Table 1 below.
표 1에 나타낸 바와 같이, 히알루론산 나트륨 분말의 평균 입자 크기는 히알루론산 수용액의 히알루론산 농도와 투입된 에탄올의 농도가 낮을수록 작았으며, 히알루론산의 농도가 0.7 %이상에서는 에탄올의 처리 농도가 낮더라도 100 ㎛ 이상의 입자를 수득할 수 있었다.As shown in Table 1, the average particle size of the sodium hyaluronate powder was smaller as the hyaluronic acid concentration and the ethanol concentration of the hyaluronic acid aqueous solution were lower, and when the hyaluronic acid concentration was 0.7% or higher, Particles of 100 μm or more could be obtained.
따라서, 상기 결과로 히알루론산 정제시 에탄올을 처리함으로써 히알루론산의 입자 크기를 조절할 수 있음을 알 수 있었다. Therefore, it can be seen that the particle size of hyaluronic acid can be controlled by treating ethanol during hyaluronic acid purification.
실시예 2 Example 2
After the culture filtrate was prepared in the same manner as in Example 1, distilled water was added to adjust the hyaluronic acid concentration to 0.1%, 0.2% and 0.5%, and NaCl was added to each of 1M. Acetone was then mixed into 70%, 80%, 100%, 200% and 300% volumes of the NaCl-added liquid volume, stirred at 100 rpm for 30 minutes, and then centrifuged (500 xg, 10 minutes). . Precipitated sodium hyaluronate was dried for 24 hours at 50 ℃ using a vacuum oven, sodium hyaluronate powder was dispersed in ethanol and the average particle size was measured using a particle size analyzer. The results are shown in Table 2 below.
표 2에서, 아세톤을 사용하여 히알루론산의 입자 크기를 조절할 수 있음을 알 수 있다. 또한 100 ㎛ 이하의 입자크기의 히알루론산을 제조하는 경우에 유기용매로 에탄올보다 아세톤이 효과적임을 알 수 있다. In Table 2, it can be seen that acetone can be used to control the particle size of hyaluronic acid. In addition, it can be seen that acetone is more effective than ethanol as an organic solvent when preparing a hyaluronic acid having a particle size of 100 μm or less.
As described above, the present invention can prevent problems such as product contamination and deterioration by providing a method that can replace the hyaluronic acid and its salt size control method through physical grinding. In addition, with the present invention, hyaluronic acid having various particle sizes and salts thereof can be easily and simply prepared to meet the industrial demand for hyaluronic acid and salts thereof of a particular particle size.
