Abstract
Description
The present invention relates to a cosmetic composition containing phytoene and phytofluene, which is a type of carotenoid, together with retinol, which is a conventional cosmetic ingredient, and more particularly, phytoene, which is a type of carotenoid, together with retinol, which is a conventional cosmetic ingredient. ) And phytofluene, which have no skin deterioration and discoloration of retinol from light, temperature, oxygen, etc. during long-term storage, and effectively maintains its activity, resulting in excellent cosmetic effects such as wrinkle improvement with retinol It is about.
일광 중 자외선에 의한 영향은 매우 큰데 자외선은 생체에 미치는 영향에 따라 UVA(320-400nm), UVB(280-320nm), UVC(200-280nm)로 분류된다. In today's industrially developed world, people are frequently exposed to environmental hazards such as dangerous pollutants, free radicals and harmful oxidation processes. In particular, short-term exposure to sunlight may cause sunburn, skin blackening, etc. In the long-term exposure, photoaging of the skin, genetic mutations caused by DNA destruction, and effects on Langerhans cells Immune system abnormalities, skin cancer, etc. may occur. The effect of ultraviolet rays during daylight is very large. UV rays are classified into UVA (320-400nm), UVB (280-320nm), and UVC (200-280nm) according to their effects on living organisms. The UV rays that reach the double indicator are UVA and UVB, which are about 95% UVA and 5% UVB.
Since UVA has a longer wavelength than UVB, it penetrates into the dermis of the skin and promotes skin aging by protein denaturation, capillary expansion and DNA destruction in the dermis. UVB, on the other hand, has a short wavelength, reaching only the epidermal layer of the skin, but has about 1000 times stronger energy than UVA, which can mainly cause skin tissue destruction such as erythema and sunburn.
In most people's growing seasons, the skin responds easily to various sun protection systems, but as the age increases, the antioxidant system is destroyed by active oxygen species produced by UV light, which lowers the defenses and oxidizes the skin. Under stress, damage to biological components such as lipid peroxidation, protein oxidation, chain breakdown and abnormal crosslinking of collagen and elastin, elastic fibers, cleavage of hyaluronic acid chains, promotion of melanin production reactions, and oxidation of DNA Various skin aging phenomena such as wrinkle formation, skin thickening, blemishes, freckles, sagging skin, loss of skin elasticity, skin roughness, skin dryness, and skin stains appear. To prevent such skin aging, develop an antioxidant system that protects the skin from UV rays, inhibits the generation of free radicals, and removes the free radicals generated. It is necessary to delay or prevent skin aging by maintaining the activity of substances such as vitamin-E, vitamin-C, catalase and ubiquinol.
Particularly in the cosmetics industry, there is an increasing demand for methods and compounds for solving the photoaging problem of skin caused by free radicals induced by ultraviolet rays. Retinoids and carotenoids can be considered as representative materials for this purpose. Existing reported examples include US Pat. Nos. 4603,146 and 4877,805 using retinoids.
Retinoids used in skin care cosmetics include retinol, retinyl, retinyl ester, retinic acid and the like. Retinol is an endogenous compound that exists naturally in the human body and is essential for the differentiation and growth of epithelial tissues. It has excellent skin wrinkle improvement, prevention, and acne healing. In addition, retinol has very high safety compared to other retinoids such as retinic acid, and thus can be said to be suitable as a raw material for skin improvement cosmetics. However, when retinol is prescribed in the product, it becomes discolored or discolored due to light, heat, temperature, moisture, etc., or when time passes, the potency decreases as an active ingredient of cosmetics, and also causes skin irritation due to the deteriorated substance. do.
Therefore, various studies have been conducted to stabilize retinol from light, heat, temperature, moisture, and time-lapse.
So far, examples of stabilizing retinol include a patent for an oil-in-water (O / W) type of emulsifier that stabilizes retinic acid using an oil-soluble oxidizing agent, BHT, dl-a-tocopherol and a chelating agent EDTA (US Pat. No. 3,906,108). There is a patent (US Pat. No. 4,2247,547) for an oil-in-water emulsifier type in which retinol is stabilized using an oil soluble tocopherol and citric acid. The present invention also relates to an oil-in-water (W / O) type of emulsifier that stabilizes retinol using butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA). Patent (US Pat. No. 4,826,828), and water-in-oil type patent (US Pat. No. 4,720,353) that stabilizes retinol using antioxidants BHA, ascorbic acid, tocopherylolinate, and more than one type to improve chemical stability of retinoids An oil-in-water emulsion composition (European Patent 0440,398BI) containing water-soluble, oil-soluble antioxidants and chelating agents has been published.
European Patent No. 1,568,106A1 discloses an oil-in-water type emulsion containing a chelating agent and an oil-soluble antioxidant, a chelating agent and a water-soluble antioxidant, an antioxidant and a free base imidazole simultaneously present in the aqueous phase and the oil phase of the emulsion. Compositions have been reported.
In the case of the various patents, it can be seen that the type of oil / water-soluble antioxidant or chelating agent is used according to the emulsification form to stabilize the retinol and various derivatives. As such, various antioxidants are used to stabilize retinol, and BHT and BHA are the most widely used, and these are radical scavengers, which strongly inhibit radical chain reactions, thereby having antioxidant activity.
In general, the generation of radicals that cause discoloration of cosmetics, odor, deactivation of active ingredients (lower potency), and lower emulsification stability are promoted by metal ions. Thus, by using chelating agents for metal ions with antioxidant May increase the efficacy of antioxidants. In addition, antioxidants should be stable and effective over a wide range of pH (pH), and should not show discoloration or odor by any reaction, and should be compatible with other cosmetic ingredients without irritating the skin. In addition, it must have an effective antioxidant capacity against substances that are highly unstable and easily oxidized such as retinol such as light, temperature, heat and moisture.
However, most of the antioxidants including BHT used in cosmetics have many problems in terms of stability. In the case of BHT, which is the most widely used antioxidant, when the formulation containing the same is manufactured at a process of 75 ° C. or higher, the titer of BHT drops so that it does not function properly as an antioxidant when the product is used for a long time. This is because BHT has a low molecular weight and high volatility. In addition, since BHT itself is very inferior to photooxidation, it may be discolored to yellow when exposed to light, oxygen, or other chemical reactions. This is because BHT is photooxidized and converted to stilbene quinone, which is yellow in color. Therefore, there are many limitations in using BHT to secure the stability of products used for a long time.
Therefore, the present inventors conducted a study on how to overcome the shortcomings of the conventional BHT, using phytoene and phytofluene together with retinol, thereby maintaining the retinol activity for a long time to improve the effect of retinol The present invention has been completed by discovering that it increases the stability of the product.
Accordingly, an object of the present invention is to provide a cosmetic composition capable of maximizing the skin beauty effect having retinol by effectively maintaining the activity of retinol from light, temperature, oxygen, etc., without deterioration and discoloration during long-term storage.

Hereinafter, the present invention will be described in detail with reference to Examples and Test Examples, but the present invention is not limited thereto.
For the experimental example, as shown in Table 1, according to the present invention, a composition containing phytoene and phytopluene together with retinol (example) and a composition containing BHT (comparative example) were prepared. The raw materials of phase A and phase B of Table 1 were uniformly mixed, and then heated to 75 ° C., emulsified in two phases, and cooled to 45 ° C., followed by addition and mixing of retinol, flavor, and preservative to form a final product.
Experimental Example 1. Product Discoloration Experiment
The Examples and Comparative Examples of Table 1 were stored in an opaque glass container in a constant temperature bath maintained at 45 ° C. for 8 weeks, and then the degree of separation and discoloration was measured and the results are shown in Table 2 below.
The stability at this time was evaluated by classifying product separation and discoloration into six grades.
Product separation and discoloration evaluation criteria

Experimental Example 2. Titer Test of Retinol
After the compositions of Examples and Comparative Examples were stored at 45 ° C. for 4 weeks, the content of retinol in each content was analyzed by high performance liquid chromatography. Each content was diluted in a mixed solution of methanol and ethyl acetate (50 wt% each) at a concentration of 200 mg / mL, and then eluted with tetrahydrofuran / acetonitrile (5:95) and 0.05 N ammonium acetate solution. . A reversed phase column (C-8) was used and the retinol content was analyzed at 340 nm. Immediately after the preparation of each content, the content of retinol was stored for 1 to 4 weeks on the basis of 100, and then the content of the retinol was calculated and the results are shown in Table 3 below.
As a result of Experimental Examples 1 and 2, the Example showed only a slight discoloration that is difficult to distinguish with the naked eye even after 8 weeks, whereas the Comparative Example shows a discoloration phenomenon that can be visually identified after 4 weeks. It is very high. In addition, the retinol of the Example was almost unchanged for two weeks and can be seen to fall slightly after four weeks. These results show that phytoene and phytofluene inhibit the oxidation of retinol very effectively compared to BHT, thereby preserving the titer.
Experimental Example 3. Skin wrinkle improvement
Twenty women over 30 years old (average age 40 years) were divided into two groups (A and B), and the examples for group A and the comparative examples for group B were formulated according to Table 1, respectively. After applying for 12 weeks (2 times / day, 0.2g / time) on the area of upper arm 2 × 2 cm 2 , the replica of skin wrinkles is floated using a transparent silicone solution (Skin Visiometer) SV400, C + K Electronic Gmbh, Germany) was used to measure skin wrinkles. 3D analysis of the replica image with a CCD camera, and the wrinkle improvement effect is analyzed by the average wrinkle roughness (Rz) of the following equation, which is the sum of the roughness of each wrinkle (Rm: m is an integer of 1 or more) divided by the number of wrinkles. The experimental results are shown in Table 4 below.
As a result, after 12 weeks, the Example was lowered by 34.6 μm (p <0.01), and the wrinkles were improved by about 62.4% or more compared with the Comparative Example. Therefore, it can be seen that phytoene and phytofluene effectively preserve the titer of retinol, which effectively improves the wrinkle improvement effect of retinol compared to the comparative example.
Experimental Example 4. Skin Stability Test
Thirty subjects (average age 26 years, age distribution 19-34 years) were divided into two groups (A, B), with examples for group A and comparative examples for group B, respectively. After two months at room temperature, the formulation was subjected to a patch test on the upper arm using a Haye's TestChamber. Only psoriasis, eczema, other skin lesion holders, pregnant women, nursing mothers, contraceptives and antihistamines were excluded.
The test site was wiped with 70% ethanol and dried, and the Example was added to the A group, and the Comparative Example to the B group, respectively, was added dropwise to the chamber by 15 µg. The patch was applied for 24 hours, and after removing the patch, the test site was marked with a marking pen, and the test site was observed after 24 hours, 48 hours, and 72 hours, respectively.
Determination was carried out 24 hours, 48 hours, 72 hours after patch removal, skin reaction was determined according to the regulations of the International Contact Dermatitis Research Group (ICDRG) Table 5 below. The experimental results are shown in Table 6 below.
As a result of the skin safety test, it was found that the Example had a safe cosmetic having no irritation to the skin with an average degree of stimulation of 0. In the comparative example, after 24 hours, it showed mild irritation, and after 48 hours, it was weak but irritating. Therefore, in the formulation of this experiment containing retinol and antioxidant, it was found that phytoene and phytofluene effectively inhibited the retinol oxidation in the case of the example, and that the retinol was oxidized and skin irritation was induced in the comparative example over time. .
