본 발명은 부유미생물 측정 장치 및 측정 방법에 관한 것으로서, 더 구체적으로는 실시간으로 부유미생물의 포집과 정량 분석을 하나의 장치에서 연속적으로 수행할 수 있는 측정 장치 및 방법에 관한 것이다. 공기 질 유지 수준과 관련하여 국가(대한민국)에서는 미세먼지, 이산화탄소, 포름알데히드, 일산화탄소, 이산화탄소와 같은 실내공기 내 유해물질을 실시간으로 측정하도록 하고 있다. 그러나 총 부유 세균이나 총 부유 곰팡이와 같이 부유미생물 입자의 농도는 실시간으로 측정하지 않고 1년에 1회 또는 2년에 1회 측정하도록 하고 있어서 측정의 실효성이 매우 낮은 문제점이 있다. 일반적으로 부유미생물의 농도 측정을 위해서는 최소 24시간 이상의 배양 시간이 필요하므로, 실시간으로 부유미생물의 농도를 정량화 하기에는 어려움이 따른다.Regarding the level of air quality maintenance, the country (Korea) requires real-time measurement of harmful substances in indoor air such as fine dust, carbon dioxide, formaldehyde, carbon monoxide, and carbon dioxide. 현재까지 부유미생물(airborne microorganism)을 탐지 및 측정하는 기술과 관련해서 다양한 입자 포집 장치들이 개발됐다. 대표적인 예로, 일반 미세먼지 포집 방법과 유사한 필터 집진 방법(Filtration)과 입자가 포함된 공기를 충돌판에 충돌시켜 관성력에 의해 입자를 포집하는 관성 충돌법(inertial impaction)이 있다. 공기 중 부유미생물을 필터 집진 방식이나 관성 충돌법을 사용하여 포집하는 경우, 분석 장비나 센서를 이용하여 측정하기 위해, 멸균 증류수나 인산완충생리식염수(Phosphate buffered saline, PBS) 등을 이용하여 포집 된 시료를 액상 추출하는 과정이 필요하다. 따라서, 필터 집진 방법과 관성 충돌법을 통한 부유미생물의 농도 변화 관측은 연속적이고 실시간으로 이루어질 수 없다.Up to now, various particle collection devices have been developed in connection with technology for detecting and measuring airborne microorganisms. 또한, 현재 보편적으로 사용되는 미생물 탐지방법은 콜로니계수법(Colony count)과 중합효소연쇄반응(Polymerase Chain Reaction, PCR)이다. 특히, 콜로니계수법은 실내외 부유미생물의 농도를 정량화할 때 사용하는 대표적인 방법이다. 콜로니계수법은 시료를 평판 형태로 24시간 이상 적절한 온도와 습도에서 배양하여 나타난 미생물 군락의 수를 계수하는 방법이다. 그러나 콜로니계수법은 특정 균류만 배양이 가능하며 배양 시간이 길다는 단점이 있다. 한편, 중합효소연쇄반응은 생물의 유전물질을 증폭하여 분석하는 방법이다. 중합효소연쇄반응은 포집한 시료를 높은 정확도로 구체화 및 정량화할 수 있다. 그러나 중합효소연쇄반응은 많은 시간과 정교함, 전문가의 운용을 요구하기 때문에 콜로니계수법 방식과 마찬가지로 실시간으로 부유미생물의 농도 추이를 관측하기가 어려운 문제점이 있다. 또한, 이러한 장비는 현장이나 실험실에서 연구자가 직접 구동해야 하는 수동형 장치로서 연속적인 부유미생물 포집 및 탐지에 관한 전반적인 장치의 개발에는 해결해야 할 난제가 많이 남아있는 상황이다.In addition, currently commonly used microbial detection methods are colony count and polymerase chain reaction (PCR).
