The present invention also relates to microvessels formed in vitro, with smooth, continuous vascular interface, and methods for analyzing cancer metastasis mechanisms using the same. The present invention also relates to a method for screening drug candidates using a microfluidic chip.
Efforts have been made to elucidate the mechanism of each stage in cancer metastasis. Most of cancer metastasis mechanisms within (in includes cancer cells and endothelial cells (EC, endothelial cell) interaction, a number of test tubes vitro models were developed to mimic these interaction patterns. However, up to now, it has been shown that microvessels produced by the natural morphogenesis process can be used to substantially mimic cancer-endothelial cell interactions (e.g., angiogenesis, vascular and extravasation) during metastasis Appropriate extracellular models have not been reported.
11: Zabel et al. 2011).Cancer cells are known to continuously interact with surrounding blood vessels during cancer growth and metastasis (Non-Patent Document 1: Bergers and Benjamin 2003; Non-Patent Document 2: Carmeliet and Jain 2000). Cancer cell populations proliferate and secrete factors that stimulate the surrounding blood vessels to extend toward the cluster. This phenomenon is known as cancer angiogenesis. Cancer angiogenesis causes nutrients and oxygen to be supplied to cancer cells from blood vessels, causing cancer to grow into malignant tumors. Cancer cells composed of fully grown ellipsoids circulate around the blood vessels through the surrounding blood vessel cells and pass through the secondary site in the peripheral blood vessel region (Non-Patent Document 3: Sahai 2007). These extravasated cancer cells grow to form secondary cancer clusters. Most cancer deaths are caused by these recurrent cancer metastases. Therefore, as a stage involved in the progression of cancer metastasis, studies on cancer angiogenesis and migration through endothelial cells have been extensively conducted. Thus, an extracellular model capable of mimicking the metastasis stage within an easily controlled microenvironment is required for a deeper understanding of cancer metastasis. In the transwell experiment, cancer cells grew in the lower chamber and in the upper chamber endothelial cells were cultured in extracellular matrix such as collagen or Martrigel. Tube formation (Non-Patent Document 4: Tsujii et al. 1998) and migration of endothelial cells (Non-Patent Document 5: Abdollahi et al. 2005) can be observed in response to cancer-derived factors. In a model for migration of cancer cells through endothelial cells, the endothelial cells are cultured to confluence in a petri dish or a porous membrane, and the permeation of the endothelial monolayer is observed after several hours to several days of culture Non-Patent Document 6: Jin et al. 2012; Non-Patent Document 7: Kramer and Nicolson 1979; Non-Patent Document 8: Kusama et al. 2006; Non-Patent Document 9: Lee et al. 2003; : Roetger et al. 1998; Non-Patent Document 11: Zabel et al. 2011).
The development of micro-fabrication techniques has led to the development of extracellular models for cancer biology research. Microfluidic technology has a variety of advantages over conventional methods for precise regulation of microenvironmental factors, cell migration or high resolution imaging control of cell-cell interactions (Non-Patent Document 12: Chung et al. 2010; : Lee et al. 2014a). Many studies have modeled each stage of the cancer metastasis process using microfluidic technology. The cancer angiogenesis process was modeled by stimulating endothelial cells by attaching endothelial cells to the walls of hydrogels and co-culturing with cancer cells (Non-Patent Document 14: Kim et al. 2013). The disadvantage of these experiments is that the development begins from endothelial cell clusters deficient in appropriate cell-cell connections. Because angiogenesis originates from fully established blood vessels near the cancer community, this characteristic is in vivo ( in vivo ) is not physiologically related to angiogenesis. In another study, vessel walls were modeled by culturing a monolayer of endothelial cells in the microfluidic channel (Non-Patent Document 15: Jeon et al. 2013; Non-Patent Document 16: Zervantonakis et al. 2012). Migration through endothelial cells can be modeled by introducing cancer cells into the system. However, this vascular model involves attaching a single layer of endothelial cells to the hydrogel without an angiogenic or vasculogenic process. Recently, a microfluidic model has been developed to produce a vascular network by an angiogenic process in order to analyze extracellular efflux of cancer (Non-Patent Document 17: Chen et al. 2013). However, this model does not allow the concentration gradient direction of chemokines to induce chemotactic migration of cancer cells, and the unpredictable geometry of the vascular network has increased the complexity of the experiment.

In the microfluidic chip of the present invention, the height and width of the channel are not particularly limited and can be determined flexibly according to the kind of the material to be injected into the channel, and the purpose and condition of the experiment. In one embodiment, the height of the channel is from 10 탆 to 100 탆, preferably from 30 탆 to 80 탆, more preferably from 40 탆 to 60 탆.
따라서, 본 발명에 따른 미세혈관형성 시 마이크로포스트 사이의 갭에 존재하는 내피세포는 배지 채널이 있는 보다 더 낮은 방향으로 이동하려는 경향을 보이며, 이로 인해 평활한 혈관 경계면 형성이 이뤄진다 (도 2C).In another embodiment of the present invention, microvessels produced using the microfluidic chip are provided. Specifically, the production of microvessels provides a mixture of fibroblasts, for example, Lung Fibroblasts (LF) and fibrin, in a

In another embodiment, the present invention provides a method for screening anticancer drug candidate substances. Specifically, in the present invention, when a cell line having an angiogenesis-forming ability is treated together with a sample to induce cancer angiogenesis, if cancer angiogenesis is not generated, the sample may be discriminated as an anticancer drug candidate.
The present invention provides, in another embodiment, intracavitary infusion with the
As such, the microfluidic chip of the present invention can be used to model in vitro microvascular production, cancer angiogenesis, intracavitary inflow, and extravasation. The spatial co-culture flexibility of the microfluidic chip of the present invention allows pulmonary fibroblasts to stimulate endothelial cells to differentiate into microvessels and subsequently introduce cancer cells to model cancer-endothelial cell interactions. In one embodiment of the invention, the endothelial cells have appropriate cell-cell connections and are elongated as in vivo, the medium can be perfused through the lumen of the vessel, and two openings for the

To quantify the success rate of the smooth and continuous border of microvessels, the length of the vascular boundary was measured and compared with the linear length of the vascular channels according to microvessels using Image J. If the boundary length value was within ± 10% of the linear length of the vascular channel, the chip was considered successful. All microvessels with unconnected vessel walls were considered as failed.
Calculation of the permeability coefficient was performed using the method published in the prior art (non-patent document 19: Lee et al. 2014b). Briefly, FITC-dextran solution was introduced into microvessels and fluorescence imaged every 15 seconds using a multistep time-difference mode in Metamorph. The acquired time-difference image was analyzed with Image J and the permeability coefficient was calculated by [Equation 1]
Where lw is the length of the blood vessel wall that separates the peripheral blood vessel area and the microvascular area, li is the average light intensity in the microvascular region, and 1 is the total luminous intensity of the peripheral blood vessel region.

The present inventors have described in detail a microvascular network-manufacturing plan that can be perfused using a co-culture system of HUVEC and LF in a previous study (Non-Patent Document 14: Kim et al. 2013). The HUVEC sprout was stimulated by LF, opening the lumens to both sides of the channel and allowing fluid to flow through the lumen. However, the microvessel structure in the existing model can not be predicted, and other cell types could not be introduced into the peri-vascular region because the peripheral vascular region was filled with gel or PDMS walls after endothelial cell loading. Thus, the present inventors have produced microvessels with more predictable geometric features and peripheral vascular regions that can be filled with other cell types after preparation of microvessels. Although conventional studies have created microvessels that traverse through the vascular channels and connect the ends to the ends, the present inventors have arranged the two third channel (vascular channel) openings on the same side (lower side) The upper part of the third channel (blood vessel channel) was brought into contact with an empty channel (FIG. 1A). This structure creates microvessels in which two openings are disposed on the same side and the other side of the microvessel is in contact with a second channel, referred to as "bridge channel" in FIG. 1A. The first channel (upper channel) and the second channel (bridge channel) are empty during the microvascular growth, and the interface with the empty channel is formed such that the HUVEC in the upper part moves to the upper direction of the third channel Suppressed the generation and produced a smooth boundary parallel to the interface.
제 4 채널 (배지 채널)에 대한 2개의 개구부를 지닌 미세혈관이 7 ~ 8일 배양 후에 생성되었다(도 1B). 1B-1D schematically illustrate the microvessel generation and the performance of the cancer angiogenesis and intravascular infusion experiments using the
Next, cancer angiogenesis was modeled using a U87MG cell line known to have high angiogenic potential as a cancer inducing factor from microvessels (Non-Patent Document 20: Flament et al. 2013; Non-Patent Document 21: Yuan et al. 1996). We injected the U87MG-fibrin mixture into the first channel (upper channel) and then the fibrin into the second channel (bridge channel) (Fig. 1C). We observed the formation of cancer germs from pre-existing microvessels facing the cancer site, which was stimulated by the secretion of angiogenic precursors from U87MG cells. The present inventors adhered MDA-MB-231 cells to a fibrin gel exposed to a second channel (bridge channel) and supplied a medium to the first channel (upper channel) for intravascular infusion of cancer (FIG. 1D) . Growth factor-deficient medium (EBM) was fed to the first medium (upper channel) to induce chemotactic migration of cancer cells to the microvascular wall. After 2 to 3 days of culture, intravascularly inserted cancer cells were observed inside the microvessels, which suggests a successful modeling of the intravascular inflow of cancer.

The production of smooth, continuous vascular interfaces is important for microvascular applications in reproducible data analysis and further studies. Thus, the present inventors optimized the HUVEC inoculation concentration to produce microvessels with smooth, continuous boundaries. We tested three conditions (3, 6 and 9 x 10 6 HUVECs / ml and 7 x 10 6 LF / ml) using 4 to 5 chips per condition, and the results were obtained by three independent measurements As shown in Fig. Figure 2A shows microvascular morphology on
After formation of microvessels in the HUVEC channels (days 7-8), nuclear (blue) and CD31 (red) were immunostained and images were obtained by confocal microscopy (FIG. 3A). Microvessel cross-sectional images at various locations show an open three-dimensional lumen. The microvessel consists of an open lumen connecting two openings to the fourth channel (the delivery channel). The present inventors measured by introducing microbeads (Sigma) into microvessels. Microvessels opened to the culture channel were formed at 94% level (101 chips out of 108 chips), and the culture medium was perfused through the lumen Respectively.
The present inventors have demonstrated the openness of the lumen by introducing a 20 kDa FITC-dextran solution into the microvessel and visualized the operation of the lumen using a fluorescence microscope. FITC-dextrin filled the microvascular lumen without severe focal leakage through the wall (FIG. 3B). The permeability coefficient of microvessels was 1.58 +/- 0.32 x 10 -6 cm / s (mean ± standard deviation, n = 5), as measured by time difference acquisition of the FITC-dextran intensity profile. The permeability coefficient was within the range of values for other extracellular blood vessel models, indicating that microvessels contain appropriate cell-cell connections. We have analyzed the connections using fluorescent immunostaining for claudin-5 and ZO-I (Figures 3C and 3D). The fluorescence image of the two cell-cell connections shows a smooth, distinctly elongated morphology without any damaged or corrugated location, which is a fundamental feature of cell-cell connections in normal vascular vessels in vivo.
Cancer angiogenesis analysis
The present inventors analyzed the possibility of angiogenesis of cancer cells using microvessels having smooth blood vessel walls. After the microvascularization in the channel (7-8 days), we introduce the U87MG and fibrin mixture, a neuroblastoma cell line with high angiogenic potential, into the first channel (upper channel) and the second channel (bridge channel) Was filled with only fibrin gel. U87MG cells in the perivascular area secrete angiogenic factors to induce the production of angiogenic germs into microvessels, while microvessels are considered to already exist in the cancerous areas. Bevacizumab map was used for U87MG cell line (bevacizumab, bev) with or bevacizumab three concentrations without dole map (control of 0 / ml, 2.5 x 10 6 and 5 x 10 6), widely used for targeting angiogenesis Anti-VEGF antibody was used. Angiogenesis from microvessels was analyzed and the mean value of 6 chips was calculated for each condition.
On
Next, the number and distribution area of germination were quantified. Microvessels with cancer cells showed significantly increased number and area of germination compared to the control group (Figs. 4B and 4C). In addition, bevacizumab treatment weakened the angiogenic potential of U87MG cells and greatly reduced the number and distribution area of microvascular germs. The trends such as induction of germination by cancer cells and attenuation of germination by anti-VEGF treatment are consistent with the existing in vivo reports (Non-Patent Document 21: Yuan et al. 1996; Non-Patent Document 22: Pechman et al. ), Demonstrating that this microvessel model is suitable for the evaluation of anti-angiogenic drugs.
27: Horvath et al. 1988).The microvessels of the present invention were used as existing blood vessels for measuring the migration of MDA-MB-231 cancer cells through endothelial cells. The cells are commonly used for endothelial migration experiments and have malignant metastasis (Non-Patent Document 9: Lee et al. 2003; Non-Patent Document 23: Wu et al / 2001; Non-Patent Document 24: Zen et al., 2008). As shown in Figure 1D, the cancer cells were attached to the contact surface between the fibrin gel and the second channel (bridge channel) and allowed to pass through the microvascular wall through the cancer cells by incubating for 2-3 days. FIG. 5A shows fluorescence and interfering contrast microscopy (DIC) photographs of cancer cells (green) during intravascular inflow through the microvascular wall (red). Cancer cells entering the blood vessel into microvessels were directly counted. In this example, microvessels were treated with TNF-a at 5 ng / ml 24 hours before the introduction of cancer cells and the rate of intravascular inflow of cancer was compared with that of the control group. The TNF-a-treated adherent junction VE-cadherin showed collapsed continuity and corrugated morphology compared to the control microvasculature (Fig. 5B). The permeability of TNF-α treated microvessels over 24 h was 2.22 ± 0.67 × 10 -6 cm / s (mean ± SD, n = 5), which is 1.4 times greater than the value at normal conditions. These results demonstrate the TNF-alpha effect on endothelial cell binding and were consistent with previous reports (Non-Patent Document 25: Brett et al. 1989; Non-Patent Document 26: Burke-Gaffeyy and Keenan 1993; Document 27: Horvath et al. 1988).
In addition, the TNF-α effect on the intravascular inflow of cancer cells was analyzed. Intravenous inflow rates with and without TNF-α treatment were compared (n = 7 chips per condition). The rate of intravascular inflow of cancer into the microvessels treated with TNF-α was three times higher than that of the control group (FIG. 5C). These results indicate that TNF-a treatment has a significant effect on the intravascular inflow of cancer cells, consistent with previous reports (Non-Patent Document 28: Johrer et al. 2004; Non-Patent Document 29: Liang et al. 2007; Non-patent document 30: Zervantonakis et al. 2012).
