Bremin, Germany)을 사용하여 분석하였으며, 당사슬들은 20 kV accelerating voltage에서 N2 laser UV light (337 nm)로 분석하였다. 중성 당사슬은 reflector positive mode에서 분석되고, 음 전하를 띄는 산성 당사슬은 linear negative mode에서 분석된다. 그리고 이미 질량 값을 알고 있는 peptide calibration standard (Cat. No. 206195, Bruker Daltonics)를 이용하여 보정하였다. 시료 분석 조건 설정 및 분석은 FlexControl 프로그램으로 이루어졌고, 데이터분석은 FlexAnaysis를 사용하였다. DHB (Cat. No. 206195, Bruker Daltonics) which already know mass value. Sample analysis conditions were set and analyzed using the FlexControl program, and data analysis was performed using FlexAnaysis.

No. 203074, 2,5-dihydroxybenzoic acid, Bruker Daltonics)를 매트릭스로 준비하였다. 그리고 인비트로 탈당화 반응(in vitro deglycosylation reaction)을 통해 얻어진 건조된 당사슬과 변형 과정을 거친 당사슬을 25% 아세토니트릴, 0.1% TFA로 녹인 후, 준비된 matrix 용액과 1:1 비율로 혼합하여 MALDI-TOF/TOF 분석용 타겟 (192 well maldi plate, Applied Biosystems, Foster City, CA, USA)에 1㎕를 올려 건조시켰다. 당사슬의 분석은 4700 Proteomics Analyzer (Applied Biosystems)를 이용하였으며, 이것들은 25kV accelerating voltage에서 MS-MS Positive Ion Mode를 사용하여 Nd:YAG laser UV light (355 nm)에서 분석하였다. 그리고 이미 질량 값을 알고 있는 6개의 스탠다드(Des-Arg-Bradykinin, Angiotensin 1, Glu-Fibrinopeptide B, ACTH (1-17 clip), ACTH (18-39 clip), ACTH (7-38 clip))를 이용하여 샘플의 질량 값을 보정하였다. 시료 분석 조건 설정 및 분석은 4000 series Explorer (TM) remote 프로그램으로 이루어졌다. PBS로 세정한 후 500배 희석한 Alexa 488로 표지된 항-IgM 항체를 중층하고, 실온에서 1시간동안 배양한다. 세정한 후 10mM 아세트산 트리에틸아민 용액 (buffer, pH 7.3)으로 세척하고, 당사슬 배열(glycan array)을 충분히 건조시킨 후 ScanArray 5000 (PerkinElmer) 공초점 스캐너(confocal scanner)를 사용하여 형광 신호를 스캐닝 하고 해석 소프트 IMAGENE 이미지 분석 소프트웨어로 해석하였다. 52-A11 항체가 각 당사슬에 결합하는 정도는 Alexa 488로부터 나오는 형광의 세기에 비례한다. 각 당사슬들은 칩 위에 6번씩 반복적으로 올라가 있으며 각 형광의 세기 중 제일 높은 것과 제일 낮은 것들은 제외되고, 4개의 형광의 세기만을 평균을 내어서 이를 RFU (relative fluorescence unit)로 명명하였다 (도 6).The glass chips of the slide glass (Consortium for Functional Glycomics, Version 3.1) containing 377 sugar chains are washed with PBS, and then washed with PBS by blocking at room temperature for 2 hours using 1% BSA / PBS. 52-A11 antibody diluted with 1% BSA / PBS is layered in a glycan array and incubated for one day at room temperature.
