묘삼뿌리 표면에 AMF번식체(접종원)을 도포부착시키기 위해 효과적인 접착물질을 선발하기 위해 시판되고 있는 소맥분, 물엿, hydroxyethylcellulose(HEC)와 미역을 알칼리분해 추출하여 제조된 해조류 추출분해액을 대상으로 접착성(점도), 접종원 부착량, 묘삼의 표면 세포조직의 안전성 및 경제성 등을 기준으로 검정한 결과, 점도는 접착재료별로 152~161cps 범위 수준이었고, 묘삼의 표면 세포조직의 유지에는 문제가 없었으며, 비용 측면까지 고려할 때 해조류 추출분해액이 가장 바람직한 것으로 평가되었다.Adhesion to Seaweed Extract Degradation Solution Prepared by Alkalilysis Extraction of Commercial Wheat Flour, Starch Syrup, Hydroxyethylcellulose (HEC) and Wakame Seaweed to Select Effective Adhesion Materials for AMF Proliferation As a result of testing on the basis of sex (viscosity), inoculum adhesion amount, safety and economics of surface cell tissues of seedlings, the viscosity ranged from 152 to 161 cps for each adhesive material, and there was no problem in maintaining surface cell tissues of seedlings. 묘삼에 Arbuscular 균근균(AMF)의 접종효과를 확인하기 위하여 접착물질(해조류 + Hydroxycellulose, 150cps) 2.8g에 AMF 포자를 46±3개 수준으로 포함하는 접종원을 제조하고 전남 곡성 인삼재배지의 본 포장 이식시 이식직전에 묘삼에 일시 침지처리하여 접종원을 부착시키고 재식하여 이식 16주 후의 생육반응을 조사하였다. 표 2에서 보는 바와 같이 이식 16주 후 생장반응을 조사한 결과, AMF를 접종한 처리구(AMF+)는 AMF를 접종하지 않은 대조구(AMF-)에 비해 초장, 측근수, 엽면적, 생체중, 엽록소 함량 등이 높게 나타났고, 특히 지하부생체중과 엽록소 함량이 고도로 유의하게 증가되었다. 묘삼이식시 AMF 접종원 처리 후 16주경의 인삼 생육 진전도를 보여주고 있는 도 3에서도 AMF 처리시 초세의 양호함을 확인할 수 있었다.To confirm the inoculation effect of Arbuscular mycorrhizal fungi (AMF) on seedlings, an inoculum containing 46 ± 3 AMF spores in 2.8 g of adhesive material (seaweed + Hydroxycellulose, 150 cps) was prepared. 묘삼이식 16주 후 식물체를 채취하여 인삼의 경엽과 뿌리 중 무기성분을 분석한 결과 표 3과 같았다. AMF 접종처리시 인삼 경엽(지상부)에서는 칼리, 칼슘, 구리, 망간, 철, 아연 등의 함량이, 뿌리(지하부)에서는 인산, 칼리, 칼슘, 철, 아연 등의 함량이 유의하게 증가하는 것으로 나타났다.After 16 weeks of transplanting seedlings, plants were harvested and the inorganic components of the leaves and roots of ginseng were analyzed. AMF 접종원 접종 16주 후의 토양중 AMF 포자 밀도의 변화를 표 4에서 살펴보면, 묘삼이식시는 토양 30g당 75개 수준이었으나, 16주 후 조사시에는 접종원을 처리하지 않은 대조구(AMF-)에서는 186개 수준을 보였으나, AMF 접종원 처리구(AMF+)에서는 256개 수준을 나타내 대조구에 비해 37.6%가 더 증가된 수준이었다.The changes in AMF spore density in soil after 16 weeks of AMF inoculation were shown in Table 4, but seedlings were 75 levels per 30 g of soil during transplanting, but 186 in control (AMF-) treated at 16 weeks after inoculation.
