実用化されている主な遺伝子検査方法として、生体試料から核酸を抽出し、PCR(Polymerase Chain Reaction:ポリメラーゼ連鎖反応)法やNASBA(Nucleic Acid Sequence−Based Amplification:核酸配列に基礎をおいた増幅法)法などを用いて放射性同位元素や、蛍光色素を標識された核酸の増幅を行ってターゲット遺伝子を増幅し、ターゲット遺伝子の塩基配列またはその濃度を測定する方法などが知られている。
遺伝子の発現量の検査や突然変異などの解析には、電気泳動法が用いられている。しかし、電気泳動法は、測定に手間と時間がかかり、また一度に行える測定に制限があるなどの問題点があった。
そこで、最近では、蛍光標識された核酸を複数のキャピラリー内で反応させ、多くの試料を一度に迅速に処理できるキャピラリー電気泳動法が広く用いられている。キャピラリー電気泳動法によれば、従来の電気泳動法を用いた方法に比べて、より短時間で簡便に測定ができる。
さらに、最近になって、同時に複数の遺伝子検査を行うことができるDNAチップを用いた検査方法が新しく開発されている。DNAチップには、ガラス基板の表面に多数のDNAプローブを固定化するDNAチップや半導体製造工程を応用してシリコンウエハ上の微小な領域に合成した多数のオリゴプローブなどを付着させたDNAチップがある。
上記のようなDNAチップを用いた検査方法では、サンプル中のDNAの塩基配列や発現量を複数、同時に決定できる。また、DNAチップの応用によって多くの遺伝子発現量や複数の突然変異の解析など、多様な検査を行うことが可能となっている。さらには、DNAチップを用いて得られたデータから、多くの遺伝子を複数のグループに分類して、発生や分化に伴う遺伝子の変動に関する情報なども得ることができる。
ところが、DNAチップを用いた遺伝子解析法は、次のような問題を有する。一度に多数の検査を行える利点があるものの、長い検査時間を必要とする。全体を通して検査工程が多く、且つ煩雑な操作が必要であるため、再現性のよい検出結果が得られにくい。
上記のような問題を克服するために、DNAチップの担体として、再現性がよく、且つ短時間でDNAチップと同様の検査を行うことができる多孔質のフィルタを用いる方法や、ハイブリダイゼーション反応を電気的な力によって行う方法が開発されている(例えば、特表平9−504864号公報、特表平2000−515251号公報および特表平2001−501301号公報参照)。
さらに、最近になって、DNAマイクロアレイを用いた遺伝子検査装置が考えられている。この装置では、DNAマイクロアレイの反応槽内で起こった反応生成物から発せられた蛍光を用いて遺伝子解析を行う装置である。この装置は、顕微鏡をベースとした装置である。DNAマイクロアレイの反応槽内で予め蛍光標識された標的核酸と核酸プローブとの間でハイブリダイゼーション反応を起こさせ、この反応によりDNAマイクロアレイに捕捉された蛍光物質からの蛍光信号を取得し、これを基に蛍光画像を得るようにしている。ここで、DNAマイクロアレイは、反応槽(液体収容部)を有している。
このようなDNAマイクロアレイを用いた遺伝子検査方法では、蛍光物質を効率よく励起して、DNAマイクロアレイの反応槽内で起こった反応生成物から安定した蛍光を発生させることが、精度の高い遺伝子解析を行う上で極めて重要である。
従来、光源としてLED(Light Emitting Diode)光源(この場合は励起光)を用いて蛍光物質に励起光を照射して蛍光物質を励起し、励起された蛍光物質が発する蛍光に対して入射光の波長を選択的に透過するフィルタ手段を通して固体撮像素子で蛍光信号を受光し、この蛍光信号に基づいて蛍光画像を得るようにした装置が提案されている(例えば、特開平10−132744号公報参照)。
LEDからの光をダイクロイックミラー、対物レンズを介して励起光としてウェルに照射し、試料から発光された蛍光を対物レンズ、ダイクロイックミラーを介しピンホールユニットを介して光電子増倍管で検出するようにした装置も提案されている(特開2002−116148号公報参照)。
半導体光源を2次元的に配列して面光源としてコンデンサレンズを介して標本を照明し、標本を透過した光を対物レンズに入射するようにした装置、つまり、2次元的に配列した半導体光源からの照明光を光軸に沿って送り、対物レンズの光軸と一致させた状態で、対物レンズに入射し、拡大した画像が得られるようにした装置が提案されている(特公平7−122694号公報参照)。
半導体励起光源を用いて、対物レンズを介して試料に光を当てて試料内部の蛍光色素を励起し、試料からの蛍光を入射光と同じ光学系を通して光検出器で検出するようにした装置も提案されている(USP6154282号参照)。

Furthermore, recently, a test method using a DNA chip capable of performing a plurality of gene tests at the same time has been newly developed. DNA chips include a DNA chip that fixes a large number of DNA probes on the surface of a glass substrate, and a DNA chip in which a large number of oligo probes synthesized on a small area on a silicon wafer are applied by applying a semiconductor manufacturing process. is there.
In the inspection method using a DNA chip as described above, a plurality of DNA base sequences and expression levels in a sample can be determined simultaneously. In addition, various tests such as analysis of many gene expression levels and multiple mutations can be performed by applying a DNA chip. Furthermore, from data obtained using a DNA chip, it is possible to classify many genes into a plurality of groups and to obtain information on gene fluctuations accompanying development and differentiation.
However, the gene analysis method using a DNA chip has the following problems. Although there is an advantage that a large number of inspections can be performed at one time, a long inspection time is required. Since there are many inspection processes throughout and a complicated operation is required, it is difficult to obtain a detection result with good reproducibility.
In order to overcome the above-described problems, a method using a porous filter that can perform the same test as a DNA chip in a short time as a carrier of a DNA chip, and a hybridization reaction are performed. Methods using electric force have been developed (see, for example, JP-T 9-504864, JP-T 2000-515251, and JP-T 2001-501301).
Furthermore, recently, a genetic testing apparatus using a DNA microarray has been considered. In this apparatus, gene analysis is performed using fluorescence emitted from a reaction product generated in a reaction chamber of a DNA microarray. This device is a microscope-based device. In the reaction chamber of the DNA microarray, a hybridization reaction is caused between the target nucleic acid and the nucleic acid probe that have been fluorescently labeled in advance, and the fluorescence signal from the fluorescent substance captured by the DNA microarray is obtained by this reaction. In such a genetic testing method using a DNA microarray, a fluorescent substance is efficiently excited, and stable fluorescence is generated from a reaction product generated in the reaction chamber of the DNA microarray. It is extremely important to do.
Conventionally, an LED (Light Emitting Diode) light source (excitation light in this case) is used as a light source to irradiate the fluorescent material with excitation light to excite the fluorescent material, and the incident light is irradiated with respect to the fluorescence emitted by the excited fluorescent material. An apparatus has been proposed in which a fluorescence signal is received by a solid-state imaging device through filter means that selectively transmits wavelengths, and a fluorescence image is obtained based on the fluorescence signal (see, for example, Japanese Patent Laid-Open No. 10-132744). ).
Light from the LED is irradiated to the well as excitation light via a dichroic mirror and objective lens, and fluorescence emitted from the sample is detected by a photomultiplier tube via a pinhole unit via the objective lens and dichroic mirror. 以下、本発明の実施の形態を図面に従い説明する。なお、以下の実施の形態においては、励起光を蛍光物質に照射して、蛍光物質からの蛍光を検査する蛍光検出装置について説明するが、標識物質に光を照射し、標識から発せられる散乱光や反射光を検出する検出装置にも適用可能であることはもちろんである(以下、これらを総称して「光検出装置」と称する)。蛍光物質としては、種々の蛍光色素、蛍光ガラス粒子も用いることができる。なお、散乱光や反射光で検出を行う場合の標識物質としては、金属粒子や誘電体粒子を用いる。例えば、金属粒子の他には、銀、白金、シリコンなどの微粒子やラテックス粒子を用いることができる。特に、金、銀、白金などの金属の微粒子は、粒径が10〜100nmのものが、運動状態にある粒子の速さが最適となるため、特に好ましい。また、ラテックス粒子は、粒径が0.1〜1μmのものが、同様に、運動状態にある粒子の速さが最適となるため、特に好ましい。適切な粒径は、粒子の比重とブラウン運動の速さにより決定される。ここで、粒子の運動状態は、例えば、ブラウン運動や振動などがあげられる。
なお、本明細書で使用する「特異的結合物質」とは、ホルモン類、腫瘍マーカー、酵素、抗体、抗原、アブザイム、その他のタンパク、核酸、cDNA、DNA、RNA、PNAなどであって、生体由来物質と特異的に結合可能な物質を意味し、プローブと呼ばれる。
また、「生体由来物質」とは、プローブを固相化する基材上の所定の位置に配置された既知の特異的結合物質と特異的に結合する物質であって、生体から抽出、単離等された物質を意味するが、生体から直接抽出されたものだけでなく、これらを化学処理、化学修飾等したものも含まれる。例えば、ホルモン類、腫瘍マーカー、酵素、抗体、抗原、アブザイム、その他のタンパク、核酸、cDNA、DNA、RNA、PNAなどの物質である。
「生体由来物質」と「特定結合物質」が特異的に結合するとは、例えば、DNAやRNA等で見られる相補的なヌクレオチド配列の間に不安定な二重鎖が形成されるような場合(ハイブリダイゼーション)や、抗原と抗体、ピオチンとアピジン等のように、特定の物質とのみ選択的に反応するきわめて特異性の高い結合を意味する。
(第1の実施の形態)
図1は、本発明の第1の実施の形態に係る光検査装置の概略構成を示す図である。本発明の各実施の形態に係る光検査装置は、集光レンズと、試料の保持手段と、光源と、光検出器を有する。
図1において、光検査装置は試料台1を備え、試料台1上には、被検体としてのDNA反応容器2が載置されている。
図2は、DNA反応容器2の具体例としてのDNAスライドガラス反応容器3を示す図である。DNAスライドガラス反応容器3は、スライドガラス状の反応容器中に配列された試料槽301を有する。試料槽301の中には、DNAマイクロアレイ302が敷設されている。
上記の構成において、試料槽301内に試料溶液(試験サンプル)が注入されると、DNAマイクロアレイ302内で予め蛍光標識された標的核酸と核酸プローブとの間でハイブリダイゼーション反応が生じる。このハイブリダイゼーション反応によりDNAマイクロアレイに捕捉された蛍光物質、例えば蛍光色素物質が蛍光を発する。この場合、反応に寄与しなかった試料溶液は緩衝液と共に洗浄される。

対物レンズ5は、対物レンズ保持機構6に保持されている。対物レンズ保持機構6は、筒状の保持部材601を有している。保持部材601の内部には、対物レンズ5の基端部が嵌め込まれている。保持部材601の周面には、円周方向に沿って等間隔に複数のネジ穴602が形成されている。保持部材601の周面が当接するように固定部材603が設けられている。固定部材603は、図示しない装置本体に固定され、対物レンズ5の光軸方向に沿った長穴604が固定部材603に形成されている。長穴604を介して保持部材601周面のネジ穴602に位置調整用ネジ605がねじ込まれている。位置調整用ネジ605のねじ込みを緩めることで、対物レンズ5が保持部材601とともに、長穴604に沿って光軸5a方向に移動する。これにより、フォーカスを調整できる。
対物レンズ5の光軸5a上には、対物レンズ5とともに光検出手段を構成するフィルタ7、結像レンズ8および光検出器9が配置されている。フィルタ7は、試料202に標識された蛍光物質が励起されて発せられた蛍光を選択的に透過する。結像レンズ8は、フィルタ7で選択された蛍光を光検出器9の検出面に結像する。光検出器9は、検出面に集光された蛍光の強度を検出して、電気信号に変換し、後述するコンピュータ10に出力する。ここで、結像レンズ8は、素材として通常のレンズに用いられているBK7などのガラスレンズでも良いが、結像レンズ8として、石英ガラス、あるいはプラスチックレンズ、または回折光学素子、液晶レンズなど、通常の可視光に対して集光作用を施すことができる素子や素材を用いることができる。
対物レンズ5の周囲には、半導体光源手段として複数のLED光源ユニット11(例えば、4個のLED光源ユニット11、図1では3個のLED光源ユニット11を図示)が配置されている。
LED光源ユニット11は、筒状のLED光源ホルダー1101を有している。LED光源ホルダー1101は、その内部に配置されたLED光源1102を有している。LED光源ホルダー1101の内部に配置されたLED光源1102から発する光の光路上には、バンドパスフィルタ1103が配置されている。バンドパスフィルタ1103は、LED光源1102から発する光の波長域近傍を主に通過させる。
複数のLED光源ユニット11がそれぞれ有するLED光源1102の発光波長のスペクトル特性は同一である。
LED光源ユニット11からの光は、試料台1上の試料槽201の試料202に対して励起光として照射される。この場合、試料202に対する各LED光源ユニット11は、試料202面を均一の明るさで照明する位置に取り付けることが必要である。すなわち、試料202に照射される光の入射角度に起因して試料202面で励起ムラが生じず、一部または全面に立体部分を有する試料202に対しても影や励起ムラが生じないように、各LED光源ユニット11が取り付けられる。
具体的には、図4A及び図4Bに示すように、各LED光源ユニット11(合計4個)の光の中心軸が、試料202面の周縁a、b、c、dを通り、且つ試料202面の略中心軸上の垂直上方または下方の空間位置の1点e(又はf)で交差するように、各LED光源ユニット11の取り付け位置が設定されている。この場合において、試料202面で励起ムラが生じないようにできるならば、試料202面の中心軸上からずれた位置であっても、試料202面の垂直上方または下方の空間位置の1点で交差するように各LED光源ユニット11の取り付け位置を設定することも可能である。
なお、LED光源ユニット11から試料台1上の試料202に照射される励起光は、試料202上面に限らず、試料202内部の対物レンズ5の光軸と直交する面に照射する場合も考えられる。
このように構成された光検査装置の主要部は、遮光ボックス12内に設置され、外部から遮光されている。
図5は、光検査装置全体のブロック図を示している。
図5において、コンピュータ10は、表示手段としてのモニタ13を備えている。コンピュータ10には、図1に記載の装置の主要部を含む蛍光検出ユニット14と各LED光源ユニット11を駆動する駆動手段としてのLED光源ユニット駆動回路15とが接続されている。LED光源ユニット駆動回路15は、コンピュータ10からの制御指令を受けると、蛍光検出ユニット14内のLED光源ユニット11のうちから駆動するLED光源ユニット11を決定すると同時に、LED光源ユニット11の駆動電流の大きさを設定してLED光源ユニット11に駆動電流を供給する。
上記のように構成した実施の形態の動作を説明する。
いま、コンピュータ10がLED光源ユニット駆動回路15に制御指令を送ると、LED光源ユニット駆動回路15は、この時の指令内容に応じて、蛍光検出ユニット14内のLED光源ユニット11のうちから駆動するLED光源ユニット11を決定し、同時に、LED光源ユニット11の駆動電流の大きさを決定してLED光源ユニット11に駆動電流を供給する。
ここで、以下の説明においては、蛍光検出ユニット14内の全て(4個)のLED光源ユニット11が同時に駆動されたものとして説明する。
LED光源1102より光が発せられると、バンドパスフィルタ1103を通過した所定波長域の光が試料台1上の試料槽201の試料202に向けて励起光として照射される。
この場合、各LED光源1102からの光は、試料202面で励起ムラが生じないように、図4A及び図4Bに示すように、それぞれの光の中心軸が、試料202面の周縁a、b、c、dを通り、且つ試料202面の略中心軸上の垂直上方(または下方)の空間位置の1点e(又はf)で交差するように、各LED光源1102が配置されている。
LED光源1102が励起光を試料202に照射することによって、試料202内の蛍光物質が蛍光を発する。この蛍光は、対物レンズ5により集光され、フィルタ7、結像レンズ8を透過して光検出器9の検出面に結像される。光検出器9は、蛍光の強度を検出して、電気信号に変換した後に、当該電気信号をコンピュータ10に出力する。
コンピュータ10は、輪郭強調、コントラスト補正、色補正などの画像処理や信号解析などを行い、蛍光画像としてモニタ13上に表示する。
上記のように、第1の実施の形態では、同一発光波長のスペクトルを有するLED光源1102を有する複数のLED光源ユニット11(例えば4個のLED光源ユニット11)を対物レンズ5の周囲に並べて配置し、複数のLED光源ユニット11で同時に励起光を試料202に照射するようにしたので、試料202内の蛍光物質を励起するのに十分な強度の励起光を効率よく得ることができる。
また、複数のLED光源1102からの光が試料202面で均一になるように、それぞれの光の中心軸が、試料202面の周縁を通り、且つ試料202面の略中心軸上の垂直上方または下方の空間位置の1点で交差するように設定したので、試料202面での影や励起ムラを無くすことができ、試料槽201内で生成される蛍光物質より安定した蛍光を発生させることができる。
さらに、LED光源1102からの光は、対物レンズ5の光軸から所定の角度を持った斜め上方から試料202面に向けて照射されるので、試料202表面からの鏡面反射光が、そのまま対物レンズ5側の受光用光路を通過して光検出器9にノイズ光として入ることがなく、ノイズ光を低減することができる。
さらに、蛍光物質を励起するための励起光源としてLEDを用いているので、安価で、長寿命で、発熱が少なく、しかも安全である。さらに消費電力も少なくできる。また装置構成を小型で携帯性に富むものにできる。
さらに、LED光源ユニット11は、LED光源1102以外の光学素子として、バンドパスフィルタ1103のみを有しているので、構成が簡素であり、組立てや光軸の調整が容易であるばかりでなく、コストの低減にもつながり、有用である。
さらに、LED光源ユニット11を対物レンズ5の周囲に配置することによって、励起光の光路と蛍光検出のための対物レンズ5を有する光路とを分離しているので、ダイクロイックミラーのような余分な光学素子を設置する必要もなく、この点でも簡単な装置構成にできる。またダイクロイックミラーなどの光学素子がないために、これらの光学素子表面で発生する反射光や散乱光、またこれらの光学素子を光が通過することによって生じる蛍光強度の減衰などが生じないので、光検出器9で検出される光強度の損失を最小限に抑えることができる。しかも、励起光が通る励起用光路と蛍光検出手段の光路が完全に分離されているので、双方の光軸を合わせるための調整が不要であり、装置全体の光軸調整が簡素化できる。従って、これらのことから、常に信頼性の高い蛍光検出を行うことができる。
(第1の変形例)
上述したLED光源ユニット11として、例えば、図6に示すようなLED光源ユニット16を使用することができる。
図6は、第1の実施の形態の第1の変形例に係るLED光源ユニット16の概略構成を示す図である。
LED光源ユニット16は、筒状のLED光源ホルダー1601を有している。LED光源ホルダー1601は、その内部に配置されたLED光源1602を有している。また、LED光源ホルダー1601の内部に配置されたLED光源1602から発する光の光路上には、バンドパスフィルタ1603、拡散板1604、集光レンズ1605が配置されている。ここで、バンドパスフィルタ1603は、LED光源1602から発する光のうち波長域近傍を主に透過させる。拡散板1604は、バンドパスフィルタ1603を透過したLED光源1602からの光の強度の不均一性(強度ムラ)を抑えるために使用される。拡散板1604として、例えば“すりガラス”や半透明のプラスチック板などが用いられている。集光レンズ1605は、拡散板1604で拡散された光を集光し、焦点距離で決まる位置に当該拡散光をフォーカスする。この場合、バンドパスフィルタ1603と拡散板1604の位置は、拡散板1604、バンドパスフィルタ1603および集光レンズ1605の順でもよい。
第1の変形例に係るLED光源ユニット16は、LED光源1602から発する光の光路上に集光レンズ1605を有するので、試料202面に集光ビームを照射することができる。これにより、例えば集光レンズ1605のNAが0.9以上の大きい集光レンズ1605を用いれば、試料202面の極めて小さい領域、例えば直径0.5μm程度の範囲に、集光させることができ、特定の極めて狭い範囲の励起を行うことができる。また、LED光源1602からの励起光が集光ビームとなって、照射断面の面積が小さくなっているので、試料槽201の側壁など、試料202面以外の部位に励起光が照射され、ノイズ光となって、試料202からの蛍光に混入する要因となることを防ぐことができる。さらにLED光源1602からの励起光を試料202面内の1点に集光させるように各LED光源1602の方向を調整してやれば、LED光源1602からの励起光を試料202面の中の所望の特定部分に限定して照射することができる。これにより、効率的にLED光源1602からの励起光を試料202面内の所望の部位に照射することができる。しかも、LED光源1602からの励起光が集光されているので、効率良く蛍光物質を励起することができる。また光が拡散していないので、試料202面以外の試料槽201内の部分、例えば側壁などを含めて光を誤照射することなどを防止することができる。
(第2の変形例)
上述したLED光源ユニット11として、例えば、図7に示すようなLED光源ユニット17を使用することができる。
図7は、第1の実施の形態の第2の変形例に係るLED光源ユニット17の概略構成を示す図である。
LED光源ユニット17は、筒状のLED光源ホルダー1701を有している。LED光源ホルダー1701は、その内部に配置されたLED光源1702を有している。また、LED光源ホルダー1701の内部に配置されたLED光源1702から発する光の光路上には、バンドパスフィルタ1703、拡散板1704、コリメートレンズ1705が配置されている。ここで、バンドパスフィルタ1703および拡散板1704は、図6で説明したものと同様である。また、コリメートレンズ1705は、拡散板1704で拡散された光を平行な光に変換し、コリメート光を出射する。この場合、バンドパスフィルタ1703と拡散板1704の位置は、拡散板1704、バンドパスフィルタ1703およびコリメートレンズ1705の順でもよい。
第2の変形例に係るLED光源ユニット17は、LED光源1702から発する光の光路上に配置されたコリメートレンズ1705を有するので、試料202面にムラのないコリメート光を照射することができる。これにより、試料202面内の照射位置の特定が行いやすくなり、照射位置をコンピュータ制御により自動調整する上で容易にプログラムを組むことができるとともに、照射面位置とその照度などの予測も立てやすく、有益である。さらに手動により、励起光を照射する位置を設定するような場合においても、試料202面内での所望の照射位置に的確に励起光の操作を行うことができる。また、光が拡散していないので、試料202面以外の試料槽201内の部分、例えば側壁などを含めて光を誤照射することなどを防止することができる。
(第3の変形例)
上述したLED光源ユニット11として、例えば、図8に示すようなLED光源ユニット18を使用することができる。
図8は、第1の実施の形態の第3の変形例に係るLED光源ユニット18の概略構成を示す図である。
LED光源ユニット18は、筒状のLED光源ホルダー1801を有している。LED光源ホルダー1801は、その内部に配置されたLED光源1802を有している。また、LED光源ホルダー1801の内部に配置されたLED光源1802から発する光の光路上には、バンドパスフィルタ1803、拡散板1804が配置されている。ここで、バンドパスフィルタ1803および拡散板1804は、図6で説明したものと同様である。この場合、バンドパスフィルタ1803と拡散板1804の位置は、拡散板1804、バンドパスフィルタ1803の順でもよい。
第3の変形例に係るLED光源ユニット18は、LED光源1802から発する光の光路上に配置された拡散板1804を有するので、試料202面にムラのない拡散光を照射することができる。これにより、試料202面全体に渡ってほぼ一様な励起光をムラなく照射することができ、安定した、再現性の優れた蛍光を受光することができる。
なお、これらLED光源ユニット11、16、17、18は、試料202や試料槽201の大きさや構造などに応じて使い分けることができる。つまり、LED光源ユニット11、16、17、18は、LED光源にバンドパスフィルタ、レンズ、拡散板などの光学素子を組み合わせて一体化に構成しているので、各LED光源ユニット単位で、励起光を集光したり、コリメート光としたり、拡散光とすることができる。これにより、試料202面への励起光のビームパターンをさまざまに制御することが可能であり、多彩な形状の試料槽201はもとより、凹凸のある試料にも対応することができる。また、LED光源ユニット16(17)として、用途に応じてバンドパスフィルタ1603(1703)、拡散板1604(1704)のいずれか一方を用いてもよいし、両方とも使わなくともよい。さらにLED光源ユニット11、16、17、18のバンドパスフィルタ1103、1603、1703、1803は、試料として用いる蛍光色素の発光波長のスペクトル領域に応じて、ハイパスフィルタあるいはローパスフィルタとしても良いことは勿論である。集光レンズ1605やコリメートレンズ1705は素材として通常のレンズに用いられているBK7などのガラスレンズでも良いが、集光レンズ1605やコリメートレンズ1705として、石英ガラス、あるいはプラスチックレンズ、または回折光学素子、液晶レンズなど、通常の可視光に対して集光作用を施すことができる素子や素材を用いることができる。また、第1の実施の形態では、対物レンズ5周囲に、複数のLED光源ユニット11が配置される場合を述べたが、試料202が極めて小さい場合には、図1において対物レンズ5周囲に配置されるLED光源ユニット11を1個のみとすることもできる。このようにしても、上述したと同様な効果を期待できる。
(第2の実施の形態)
第2の実施の形態は、LED光源ユニットの試料面に対する傾き角を変化させて、光の照射角度を調整できるようにした実施の形態である。
図9は、第2の実施の形態の概略構成を示す図である。図9において、対物レンズ21は、対物レンズ保持機構22に保持されている。対物レンズ保持機構22は、筒状の保持部材2201を有している。保持部材2201の内部に対物レンズ21が嵌め込まれている。保持部材2201の周囲には、半導体光源手段保持部材としてLED光源ユニットホルダー23が設けられている。
LED光源ユニットホルダー23には、周面方向に沿って均等な間隔で複数(例えば、8〜12個程度)のLED光源ユニット収納孔2301が設けられている。これらLED光源ユニット収納孔2301は、試料台1上の試料202面に向かって傾斜して配置されている。傾斜角度は、対物レンズ21の光軸21aに軸に対して45°〜60°程度となっている。また、LED光源ユニットホルダー23の周面には、LED光源ユニット収納孔2301に貫通するネジ穴2302が設けられている。ネジ穴2302は、各LED光源ユニット収納孔2301に沿った方向に所定間隔をおいて2個所づつ形成されている。
LED光源ユニット収納孔2301には、LED光源ユニット24が収容されている。LED光源ユニット24は、筒状のホルダー2401を有し、ホルダー2401の内部にLED光源2402が配置されている。そして、このようなLED光源ユニット24は、Oリング25を介在させてLED光源ユニット収納孔2301に収容されている。この場合、Oリング25は、2個所のネジ穴2302の間に位置する。
LED光源ユニット収納孔2301には、LED光源ユニット24のLED光源2402から発する光の光路上に、バンドパスフィルタ2403が配置されている。ここで、バンドパスフィルタ2403は、LED光源2402から発する光の波長域近傍を主に通過させる。
また、LED光源ユニット24のホルダー2401とバンドパスフィルタ2403の間には、スペーサ26が配置されている。スペーサ26は、LED光源2402とバンドパスフィルタ2403の位置決めを行う。スペーサ26の素材としてはアルミニューム、真鍮などの金属あるいはプラスチックが用いられる。
各LED光源ユニット収納孔2301に貫通する2個所のネジ穴2302には、位置調整手段としての煽りネジ27がねじ込まれている。煽りネジ27は、LED光源ユニット24のホルダー2401側面のOリング25を挟んだ2点を押圧する。煽りネジ27の押圧力をねじ込み量により加減して、試料202面に対するLED光源ユニット24の傾き角を変化させることにより、光の照射角度を調整できる。この場合、これら2個の煽りネジ27により調整されるLED光源2402から発せられる光の照射角度は、±2〜3°程度となっている。
一方、保持部材2201の周面には、円周方向に沿って等間隔に複数のネジ穴2202が形成されている。また、保持部材2201の周面が当接するように固定部材2203が設けられている。固定部材2203は、装置本体28側に固定されており、対物レンズ21の光軸方向に沿った長穴2204が形成されている。そして、長穴2204を介して保持部材2201周面のネジ穴2202に位置調整用ネジ2205がねじ込まれている。これにより、位置調整用ネジ2205のねじ込みを緩めることで、対物レンズ21は、保持部材2201とともに、長穴2204に沿って光軸21a方向に移動する。これにより、フォーカスを調整できる。
上記の構成において、まず、各LED光源ユニット24のLED光源2402を点灯し、これらLED光源2402から発せられる励起光を試料台1上の試料202面に照射する。この場合、LED光源2402から発せられる光の発光スペクトルは、全て同一である。
この状態で、対物レンズ保持機構22の位置調整用ネジ2205を緩め、対物レンズ21を保持部材2201とともに、光軸21a方向(図示矢印A方向)に移動させることでフォーカス調整を行う。
次に、LED光源ユニットホルダー23の煽りネジ27を緩めて、LED光源ユニット24をLED光源2402の光軸方向(図示矢印B方向)に移動させる。これにより、LED光源ユニットホルダー23の高さ方向の位置調整を行い、試料202面上での明るさを調整する。続けて、煽りネジ27によるねじ込み量を加減して、試料202面に対するLED光源ユニット24の傾き角(図示矢印C方向)を調整することにより、試料202に対する光の照射位置の微調整を行う。
このようにして、試料202面上での光の明るさを調整するとともに、光の照射角度の微調整を行い、図4A及び図4Bで述べたように、それぞれのLED光源ユニット24から発せられる光の中心軸が、試料202面の周縁を通り、且つ試料202面の略中心軸上の垂直上方または下方の空間位置の1点で交差するように設定する。これにより、試料202面をムラなく均一の明るさで照明することができる。
また、こうすることで、試料202面の所望する特定の箇所に光を集光させることもできるので、蛍光物質の効率的な励起を行うことができると共に、試料槽201の側壁などに励起光が当たり、ノイズ光となることなども極力抑えることができる。
なお、第2の実施の形態では、煽りネジ27のねじ込み量の調整を手動で行っているが、例えば煽りネジ27のねじ込み量の調整に相当する箇所にモータを使用し、このモータをコンピュータに連動させて、自動的に操作できるようにしてもよい。また、LED光源ユニット24は、高さ方向の位置調整および傾き角を少なくとも一方の調整のみできるようにしたものであってもよい。
(変形例)
ところで、試料面をムラなく均一の明るさの光で照明するには、事前にこの状態を確認する必要がある。
図10は、第2の実施の形態の変形例に係る光検査装置の概略構成を示す図であり、図1と同一部分には、同符号を付している。また、本変形例では、図5で述べた光検査装置のブロック図を援用するものとする。
本変形例では、複数のLED光源ユニット11からの光が照射される試料台1上には、試料槽201に代えて光強度検出手段として光検出器31が配置されている。ここで、光検出器31としては、固体撮像素子(CCDカメラやCMOSセンサー)や撮像管などが用いられる。
光検出器31は、各LED光源ユニット11からの光の強度を個別に検出する。光検出器31の検出出力は、図5で述べたコンピュータ10に取り込まれる。
コンピュータ10は、各LED光源ユニット11からの光の強度に応じた光検出器31の出力から光強度のバラツキを解析して、LED光源ユニット駆動回路15に信号を送り、各LED光源ユニット11のLED光源1102に供給する電流を個別に制御する。
本変更例では、各LED光源1102に供給する電流の大きさを試料面近傍で得られる光強度に基づいて制御できるので、各LED光源1102からの光を同一の明るさに揃えることができ、試料202面を均一の明るさで照明することができる。このため、試料202面での照明ムラを抑えることができ、試料槽201内の蛍光物質をほぼ一様に励起することができ、信頼性の高い蛍光検出を行うことができる。
本変形例では、コンピュータ10を用いたて各LED光源1102に供給する電流の大きさを調整したが、光検出器31で得られた各LED光源ユニット11のLED光源1102からの光の強度に基づいて手動でLED光源ユニット駆動回路15を調整して、LED光源1102からの光を同一の明るさに揃え、その後、光検出器31を取り外し、この位置に試料槽201を設置し直すようにしてもよい。
一方、光検出器9に代えて撮像手段(例えばCCDカメラやCMOSセンサー(共に図示せず)を使用し、試料台1上に試料槽201をそのまま載置しておいて、各LED光源ユニット11のLED光源1102からの光を試料槽201に個別に照射し、試料槽201からの蛍光を対物レンズ5、フィルタ7、結像レンズ8をそれぞれ通過させ、撮像手段で撮像し、撮像画像をコンピュータ10に導きモニタ13に表示させる。そして、モニタ13の蛍光画像の各画素の明るさをコンピュータ10で解析することにより、各LED光源1102からの光の強度のバラツキを判断し、この結果に基づいてLED光源ユニット駆動回路15を調整して、それぞれのLED光源1102に供給する電流を個別に制御するようにもできる。
このようにしても、各LED光源1102からの光を同一の明るさに揃えることができるので、試料202面での照明ムラを抑えることができ、試料槽201内の蛍光物質をほぼ一様に励起することができる。
(第3の実施の形態)
第3の実施の形態は、ピーク発光波長の異なる2個以上のLED光源を用いて複数種類の蛍光色素を励起し、蛍光信号を検出できるようにした実施の形態である。
図11A及び図11Bは、第3の実施の形態の概略構成を示す図で、図9と同一部分には、同符号を付している。また、第3の実施の形態では、図5で述べた光検査装置のブロック図を援用するものとする。
LED光源ユニットホルダー23には、ピーク発光波長の異なる3種類のLED光源を有する第1乃至3のLED光源ユニット41、42、43が装着されている。一般にLED光源の発光スペクトルは山型構造となっており、1つのピーク波長を有する。ここでは、例えば、第1のLED光源ユニット41は、発光のピーク波長が490nmのLED光源が用いられ、第2のLED光源ユニット42は、発光のピーク波長が520nmのLED光源が用いられ、第3のLED光源ユニット43は、発光のピーク波長が630nmのLED光源が用いられている。
第1乃至3のLED光源ユニット41、42、43は、LED光源ユニットホルダー23周囲に沿って、それぞれ4個ずつ対物レンズ21を挟んで対称な位置に配置されている。つまり、これらの第1乃至3のLED光源ユニット41、42、43は、それぞれ発光波長のスペクトル特性の異なるLED光源を有しており、これら異なるスペクトル特性を有する第1乃至3のLED光源ユニット41、42、43を順番にLED光源ユニットホルダー23の周囲に沿って等間隔で配置している。ここでは、第1のLED光源ユニット41、第2のLED光源ユニット41、第3のLED光源ユニット43の順に繰り返し配置されている。また、これら第1乃至3のLED光源ユニット41、42、43は、対物レンズ21を挟んで対向する位置に、同一スペクトル特性のものが来るように配置されている。
第1乃至3のLED光源ユニット41、42、43のそれぞれのLED光源の前方に設置されているバンドパスフィルタ(図面では、第1のLED光源ユニット41のLED光源4101前方に設置されているバンドパスフィルタ4102のみ示している)は、それぞれのLED光源の発光のピーク波長近傍を最も良く透過させる特性を持つものとなっている。すなわち、第1のLED光源ユニット41のバンドパスフィルタ4102は、最も良く透過させる波長が490nm付近に設定され、第2のLED光源ユニット42のバンドパスフィルタは、最も良く透過させる波長が520nm付近に設定され、第3のLED光源ユニット43のバンドパスフィルタは、最も良く透過させる波長が630nm付近に設定されている。
図12は、このように構成された装置の光検出光学系の概略構成を示すもので、ここでは、上述した3種類の異なる波長をピーク発光波長に持つLED光源ユニット41,42,43により試料への励起光を照射する場合を示している。
図12において、対物レンズ21上方の光軸上に2個のダイクロイックミラー45,46が配置されている。
ダイクロイックミラー45は、対物レンズ21の光軸に対して略45°の方向に反射光が進むように設置されており、蛍光色素FITCのピーク発光波長520nmよりも少し長い波長、例えば550nm以下の波長の光を反射し、それよりも長い波長の光を透過させるような透過反射スペクトル特性を有している。また、ダイクロイックミラー46は、対物レンズ21の光軸に対して略45°の方向に反射光が進むように設置されており、蛍光色素Cy3のピーク発光波長565nmよりも少し長い波長、例えば620nm以下の波長の光を反射し、それよりも長い波長の光を透過させるような透過反射スペクトル特性を有している。
そして、ダイクロイックミラー45の反射光路には、集光レンズ47を介してCCDカメラ48が配置されている。また、ダイクロイックミラー46の反射光路には、集光レンズ49を介してCCDカメラ50が配置され、透過光路には、集光レンズ51を介してCCDカメラ52が配置されている。
CCDカメラ48、50、52の出力は、コンピュータ10に送られる。
上記のような構成において、いま、1つのサンプルに対して、ターゲットDNAを3種類設定し、これらにそれぞれ異なる蛍光色素FITC(Fluorescein−isothiocyanate)、Cy3、Cy5を標識する。次に、これらの蛍光色素で標識されたDNAを含んだサンプル溶液を試料槽に滴下し、DNAハイブリダイゼーションを行わせ、反応に寄与しなかった標識物質を緩衝液(PBS(リン酸緩衝液)、EDTA(エチレンジアミンテトラ酢酸二ナトリウム)、NaClの混合液:PH7.4)等で洗浄する。
このようにして取得された試料を図11A及び図11Bに示す試料台1上にセットする。そして、各LED光源ユニット41、42,43からの励起光を試料槽201に照射する。このとき、3種類の異なる波長をピーク発光波長に持つLED光源からの光を全て同時に試料202面に照射する。
すると、DNAハイブリダイゼーションによってDNAプローブと反応したサンプルに標識されている蛍光色素から蛍光が発せられ、これらの光は、図12に示す対物レンズ21を通過して、ダイクロイックミラー45に到達する。この場合、蛍光色素FITCによる蛍光はダイクロイックミラー45で反射され、レンズ47を通過してCCDカメラ48に入り、コンピュータ10に導かれ、FITCによる緑色の蛍光画像として得られる。また、蛍光色素Cy3による蛍光は、ダイクロイックミラー45を透過し、ダイクロイックミラー46で反射され、レンズ49を通過してCCDカメラ50に入り、コンピュータ10に導かれ、Cy3による橙色の蛍光画像として得られる。さらに蛍光色素Cy5による蛍光は、ダイクロイックミラー45、ダイクロイックミラー46をそれぞれ透過し、レンズ51を通過してCCDカメラ52に入り、コンピュータ10に導かれて、Cy5による赤色の蛍光画像が得られる。コンピュータ10では、これらCCDカメラ48、50、52による3種類の蛍光色素からの蛍光画像を合成してモニタ13上に出力するか、あるいは、3種類の蛍光色素からの蛍光画像を別々にモニタ13上に表示する。
図13は、第1乃至3のLED光源ユニット41、42、43の駆動回路のブロック図を示す。
この場合、第1乃至3のLED光源ユニット41、42、43の駆動回路として、それぞれLED光源ユニット駆動回路54、LED光源ユニット駆動回路55、LED光源ユニット駆動回路56が設けられている。第1のLED光源ユニット41には、LED光源ユニット駆動回路54が接続され、第2のLED光源ユニット42には、LED光源ユニット駆動回路55が接続され、第3のLED光源ユニット43には、LED光源ユニット駆動回路56が接続されている。また、これらLED光源ユニット駆動回路54、LED光源ユニット駆動回路55およびLED光源ユニット駆動回路56には、共通の電源装置53が接続され、また、図5で述べたコンピュータ10が接続されている。
LED光源ユニット駆動回路54、LED光源ユニット駆動回路55、LED光源ユニット駆動回路56は、共通の電源装置53より電源の供給を受ける。また、LED光源ユニット駆動回路54、LED光源ユニット駆動回路55、LED光源ユニット駆動回路56は、コンピュータ10からの指令に基づいて制御され、第1のLED光源ユニット41、第2のLED光源ユニット42、第3のLED光源ユニット43の各LED光源に駆動電流を供給し、3種類の蛍光色素の励起光を発生させる。
この場合、これらの励起光により得られた3種類の蛍光信号は、図12に示すように、それぞれダイクロイックミラー45,46によりCCDカメラ48、50、52に別々に到達し、これらCCDカメラ48、50、52からの画像出力信号がコンピュータ10に導かれ、画像解析が行われ、3種類の蛍光色素による合成画像が生成され出力される。
図14は、電源装置53とLED光源ユニット駆動回路54、55、56の具体的な回路構成を示している。
この場合、電源装置53は、100Vの交流電源57からの出力を、トランスから構成される変圧回路58に導びき電圧調整を行う。変圧回路58からの出力は、ダイオード4個から成るブリッジ整流回路59に送って全波整流した後、平滑用コンデンサー60によって平滑化する。平滑化された出力は、ダーリントン接続した2個のパワートランジスタ61、62およびOPアンプ63より構成される定電圧回路64に導びかれ、一定電圧として出力される。この場合、ツェナーダイオード65の端子電圧を基準電圧にとり、OPアンプ66や抵抗などの回路素子より構成される可変基準電圧生成回路67により可変基準電圧を生成している。そして、OPアンプ63に接続された可変基準抵抗68の値を変化させることにより、可変基準電圧生成回路67の可変基準電圧との関係から定電圧回路64の電圧出力を調整し、LED光源ユニット駆動回路54、LED光源ユニット駆動回路55、LED光源ユニット駆動回路56に対して駆動電流を供給する。つまり、可変基準抵抗68の値を変化させることによりLED光源ユニット駆動回路54、LED光源ユニット駆動回路55、LED光源ユニット駆動回路56への駆動電流を同時に調整することができる。ここで、FET(field effect transistor:電界効果トランジスタ)70は、万一、電流がショートした場合に急激な電流がOPアンプ63に流れ、OPアンプ63を損傷する不測の事態を防ぐ働きを担う。また、電源スイッチ69により電源オンオフすることができる。電源ステッチ69は、コンピュータ10と接続されており、コンピュータ10によっても、電源入力を調節する。なお、電源スイッチ69は、手動により切り替えても良い。
このような電源装置53には、LED光源ユニット駆動回路54、55、56が並列に接続されている。LED光源ユニット駆動回路54は、図11A及び図11Bで述べた4個分の第1のLED光源ユニット41の駆動回路として、可変抵抗71とスィッチ72の直列回路が各別に用意されている。同様に、LED光源ユニット駆動回路55も、4個分の第2のLED光源ユニット42の駆動回路として、可変抵抗71とスィッチ72の直列回路が各別に用意され、さらにLED光源ユニット駆動回路56も、4個分の第3のLED光源ユニット43の駆動回路として、可変抵抗71とスィッチ72の直列回路が各別に用意されている。
これらLED光源ユニット駆動回路54、55、56では、それぞれの可変抵抗71、73、75の抵抗値を調整することにより、第1乃至3のLED光源ユニット41、42、43のそれぞれのLED光源に供給する駆動電流を調節し、その出力光強度を制御することができる。この出力光強度の調節は、手動で行ってもよいし、あるいは電気的にコンピュータで自動調整を行っても良い。これにより、第1乃至3のLED光源ユニット41、42、43の各LED光源の駆動電流を個別に制御することができる。
また、LED光源ユニット駆動回路54、55、56では、それぞれのスイッチ72、74、76のオンオフを制御することで、第1乃至3のLED光源ユニット41、42、43のそれぞれのLED光源を全て独立に点灯、消灯することができる。これらのオンオフ制御は、コンピュータ制御により、自動的に行っても良いし、手動で操作しても良い。
なお、コンピュータ制御により、可変抵抗71、73、75の抵抗値を調整する場合は、上述の図10で説明したように第1乃至3のLED光源ユニット41、42、43の個々のLED光源からの光強度を検出し、この検出信号をコンピュータに導き、コンピュータで光強度を解析して、可変抵抗71、73、75の抵抗値を調整する。または、この検出で得られた個々のLED光源からの光の強度を基に、手動で可変抵抗71、73、75の抵抗値の調整を行っても良い。これにより、DNAマイクロアレイの反応槽の内部にほぼ均一にLED光源からの光を照射するように調整できる。また試料槽201内の所望の箇所へ励起光を集中させることなど、光強度の部分的な加減を行うことができる。さらに、可変抵抗71、73、75は、全てのLED光源に流れる駆動電流の大きさを制御することができるので、励起光の強度を一斉に強くしたり、弱くしたりすることができる。これにより、個々の試料に対して、的確な強さの励起光を照射することができ、効率良く蛍光を検出することができる。
従って、このようにすれば、ピーク発光波長の異なる3種類のLED光源を有する第1乃至3のLED光源ユニット41、42、43を用いて3種類の蛍光色素を同時に励起できるようにしたので、これら3種類の蛍光色素からの蛍光画像を取得し、これらを合成したり、個別に表示することができる。
また、第1乃至3のLED光源ユニット41、42、43を構成する個々のLED光源は、励起する蛍光色素に応じたものの点灯、消灯を簡単に行うことができ、しかも、可変抵抗の可変操作により駆動電流の大きさを個別に調整することもできるので、個々の蛍光色素に対して的確な強さの励起光を照射することができ、効率良く蛍光を検出することができる。
なお、上述の第3の実施の形態では、蛍光色素については3種類としたが、これに限ることなく、2種類でも良いし、4種類以上についても同様な方法で蛍光画像を得ることができる。すなわち、ダイクロイックミラー、レンズ、CCDカメラを蛍光色素1種類ずつについて、それぞれ対応して追加すれば良い。また、蛍光色素についても実施例で示したFITC、Cy3、Cy5に限らず、ローダミン・グリーン(Rhodamine G)、テキサス・レッド(Texas Red)、RITC(Rhodamine B−isothiocya)などを用いても良い。
また、LED光源ユニットの数には特に制限はなく、複数のLED光源ユニットを複数の駆動回路により個別に制御することができる。また、複数のLED光源ユニットの出力波長は、同じでもよく、異なっていても本発明を適用することができる。また、異なるピーク発光波長を持つLED光源の光を1種類ずつ試料に照射し、それを異なる波長の数繰り返す構成とすることも可能である。例えば、透過波長の異なるフィルタをセットしたターレット等を用いて、異なる波長のLED光源と連動させた切り換えをコンピュータで自動的に行うことも可能である。
(第4の実施の形態)
第4の実施の形態は、LED光源ユニットから発する光の光路上に反射部材を配置し、光路の光を反射させることで、励起光の光軸角度を、より対物レンズの光軸角度に近付けるようにした実施の形態である。
図15は、第4の実施の形態の概略構成を示す図で、図1と同一部分には、同符号を付している。
この場合、対物レンズ5の周囲には、半導体光源手段としての複数のLED光源ユニット81(図示例では4個のLED光源ユニット81、うち1個は図示せず)が、対物レンズ5の光軸を中心に放射状に配置されている。
LED光源ユニット81は、筒状のLED光源ホルダー8101を有している。LED光源ホルダー8101は、その内部に配置されたLED光源8102を有している。また、LED光源ホルダー8101の内部のLED光源8102から発する光の光路上には、バンドパスフィルタ8103、拡散板8104、集光レンズ8105が配置されている。ここで、バンドパスフィルタ8103は、LED光源8102から発する光の波長域近傍を主に通過させる。
LED光源ユニット81から発する光の光路上には、反射部材として反射板82がそれぞれ配置されている。反射板82は、LED光源ユニット81からの光を反射させ、試料台1上の試料槽201の試料202に対して励起光として照射する。反射板82は、同時に、対物レンズ5の光軸に対して励起光の光軸がなす角度θを小さくして、励起光の光軸角度を対物レンズの光軸角度に近付けるようにもしている。
ここで、反射板82のそれぞれの取付位置は、LED光源ユニット81からの光を反射して試料202面に均一の明るさで照射させるとともに、照射される光の入射角度に起因して試料202面で励起ムラが生じないように、さらに、一部または全面に立体部分を有する試料202に対しても影や励起ムラが生じないように調整されている。
その他については、図1と同様である。
従って、このようにしても、第1の実施の形態と同様な効果を期待することができる。さらに、LED光源ユニット81から発する光の光路上に反射板82が配置され、光路の光を反射させるようにしたので、励起光の光軸角度を対物レンズ5の光軸角度に近付けることができる。このことは、試料202面の真上に近いところ(対物レンズ5の観察視野に入らない限界)から励起光を照射できるようになるので、LED光源ユニット81から反射板82を介して試料202面までの光路の最も短い距離aと最も長い距離bの差を小さなものにできる。これにより、励起光の強度は、光路の距離に反比例することから、試料202面上での励起光の強度差を小さくできることになり、試料202面を効率的に、均一に励起することができる。
なお、第4の実施の形態についても、第2の実施の形態の変形例および第3の実施の形態で述べるような試料をムラなく均一の明るさの光で照明するための制御方法を採用することができる。
(第1の変形例)
第4の実施の形態では、LED光源ユニット81から発する光の光路上に反射部材として反射板82を配置し、光路の光を反射させるようにしたが、このような方法を用いれば、LED光源ユニット81の配置の自由度が増すことから、対物レンズ5の周囲に多数のLED光源ユニット81を配置することができる。
図16は、第4の実施の形態の第1の変形例に係る光検査装置の概略構成を示す図で、図15と同一部分には、同符号を付している。
対物レンズ5の周囲には、半導体光源手段としての複数個のLED光源ユニット81が多層(図示例では2層)に配置されている。また、これらLED光源ユニット81から発する光の光路上には、各層のLED光源ユニット81に対応させて反射部材として反射板82、82’(図示例では2層に対応)が各別に配置されている。反射板82、82’は、各層のLED光源ユニット81からの光を反射させ、試料台1上の試料槽201の試料202に対して励起光として照射する。
この場合、反射板82、82’のそれぞれの取付位置についても、LED光源ユニット81からの光を反射して試料202面に均一の明るさで照射させるとともに、照射される光の入射角度に起因して試料202面で励起ムラが生じないように、さらに、一部または全面に立体部分を有する試料202に対しても影や励起ムラが生じないように調整されている。
その他については、図15と同様である。
第4の実施の形態の第1の変形例によれば、反射板82、82’を用いて、各層の光源ユニット81からの光を反射させるようにしたので、各層のLED光源ユニット81の配置する位置の自由度を増すことができる。そして、これらLED光源ユニット81の配置する位置の自由度が増すことにより、第4の実施の形態の場合よりさらに多くのLED光源ユニット81を使用することが可能となる。これにより、試料202面を照射するのに必要な光量をより簡単に得ることができるとともに、試料202面を光量ムラなく照明することができる。
また、LED光源ユニット81の数を大幅に増やすことができるので、これらLED光源ユニット81を複数の組に分けて、それぞれの組ごとに発光波長のスペクトル特性の異なるLED光源を用いることにより、第3の実施の形態で述べたと同様に複数種類の蛍光色素を励起し、蛍光信号を検出できるようにすることも可能となる。
(第2の変形例)
第4の実施の形態では、LED光源ユニット81から発する光の光路上に反射部材として反射板82を配置し、光路の光を反射させるようにしたが、反射部材としてダイクロイックミラーを用いることができる。
図17は、第2の変形例の概略構成を示す図で、図15と同一部分には、同符号を付している。
LED光源ユニット81は、筒状のLED光源ホルダー8101の内部に配置されたLED光源8102を有している。LED光源ホルダー8101の内部のLED光源8102から発する光の光路上には、拡散板8104、集光レンズ8105が配置され、上述したバンドパスフィルタ8103が省略されている。
LED光源ユニット81から発する光の光路上には、反射部材としてダイクロイックミラー83が配置されている。ダイクロイックミラー83は、LED光源ユニット81のLED光源8102から発する光の波長域近傍を主に反射させるような特性を有する。また、ダイクロイックミラー83は、LED光源ユニット81からの光を反射し、試料台1上の試料槽201の試料202に対して励起光として照射する。この場合も、対物レンズ5の光軸に対して励起光の光軸がなす角度を小さくして、励起光の光軸角度を対物レンズの光軸角度に近付ける。
その他については、図15と同様である。
このように反射部材として反射板に代えてダイクロイックミラー83を用いるようにしても、第4の実施の形態と同様な効果を期待することができる。さらに、ダイクロイックミラー83として、LED光源ユニット81のLED光源8102から発する光の波長域近傍を主に反射させるような特性を有するものを用いることにより、LED光源ユニット81に内蔵されていたバンドパスフィルタを省略することができる。これによりLED光源ユニット81のコストを引き下げることができ、且つ小型化も実現できるという効果がある。
(第5の実施の形態)
第5の実施の形態は、LED光源ユニットから発する光の光路上に導光部材を配置し、光路を曲げることで、励起光の光軸角度を、より対物レンズの光軸角度に近付けるようにした実施の形態である。
図18は、第5の実施の形態の概略構成を示すもので、図1および図15と同一部分には、同符号を付している。
この場合、対物レンズ5の周囲には、半導体光源手段としての複数(図示例では4個、うち1個は図示せず)のLED光源ユニット81が配置されている。
LED光源ユニット81は、筒状のLED光源ホルダー8101を有している。LED光源ホルダー8101は、その内部に配置されたLED光源8102を有している。また、LED光源ホルダー8101の内部のLED光源8102から発する光の光路上には、バンドパスフィルタ8103、拡散板8104、集光レンズ8105が配置されている。ここで、バンドパスフィルタ8103は、LED光源8102から発する光の波長域近傍を主に通過させる。
これらLED光源ユニット81から発する光の光路上には、導光部材として、オプチカルファイバ84の光入射端84aが各別に配置されている。オプチカルファイバ84は、LED光源ユニット81からの光の光路を自在に曲げることができる。これにより、光出射端84bからの光を試料台1上の試料槽201の試料202に対して励起光として照射し、同時に、対物レンズ5の光軸に対して励起光の光軸がなす角度を小さくして、励起光の光軸角度を対物レンズの光軸角度に近付けることができる。
この場合、これらオプチカルファイバ84の光出射端84bの位置は、出射光を試料202面に均一の明るさで照射させるとともに、照射される光の入射角度に起因して試料202面で励起ムラが生じないように、さらに、一部または全面に立体部分を有する試料202に対しても影や励起ムラが生じないように調整されている。
その他については、図1および図15と同様である。
従って、第5の実施の形態においても、第1の実施の形態と同様な効果を期待することができる。さらに、LED光源ユニット81から発する光の光路上に配置されたオプチカルファイバ84で光路を曲げるようにしたので、励起光の光軸角度を対物レンズの光軸角度に近付けることができる。これにより、第4の実施の形態と同様に、試料202面上での励起光の強度差を小さくできることになり、試料202面を効率的に、均一に励起することができる。
なお、本実施の形態では、導光部材として、オプチカルファイバ84を適用した例を述べたが、例えばガラス棒やアクリル材を成形加工したようなものを適用することもできる。
なお、第5の実施の形態についても、第2の実施の形態の変形例および第3の実施の形態で述べるような試料をムラなく均一の明るさの光で照明するための制御方法を適用することができる。
(変形例)
第5の実施の形態では、LED光源ユニット81から発する光の光路上に導光部材として、オプチカルファイバ84を配置し、光路を曲げるようにしたが、このような方法を用いれば、LED光源ユニット81の配置の自由度が増すことから、対物レンズ5の周囲に多数のLED光源ユニット81を配置することができる。
図19は、第5の実施の形態の変形例に係る光検査装置の概略構成を示す図で、図18と同一部分には、同符号を付している。
図19において、対物レンズ5の周囲には、半導体光源手段としての複数個のLED光源ユニット81が多重(図示例では2重)に配置されている。また、これらLED光源ユニット81から発する光の光路上には、導光部材として、オプチカルファイバ84の光入射端84aが各別に配置されている。オプチカルファイバ84は、LED光源ユニット81からの光の光路を自在に曲げることを可能にしたもので、光出射端84bからの光を試料台1上の試料槽201の試料202に対して励起光として照射する。
本変形例の場合も、オプチカルファイバ84の光出射端84bの位置は、出射光を試料202面に均一の明るさで照射させるとともに、照射される光の入射角度に起因して試料202面で励起ムラが生じないように、さらに、一部または全面に立体部分を有する試料202に対しても影や励起ムラが生じないように調整されている。
その他については、図18と同様である。
本変形例では、オプチカルファイバ84を用いることで、光源ユニット81からの光の光路を曲げるようにしたので、各LED光源ユニット81の配置する位置の自由度を増すことができる。LED光源ユニット81の配置する位置の自由度が増すことにより、第5の実施の形態の場合よりさらに多くのLED光源ユニット81を使用することが可能となる。これにより、試料202面を照射するのに必要な光量を簡単に得ることができるとともに、試料202面を光量ムラなく照明することができる。
また、LED光源ユニット81の数を大幅に増やすことができるので、LED光源ユニット81を複数の組に分けて、それぞれの組ごとに発光波長のスペクトル特性の異なるLED光源を用いることにより、第3の実施の形態と同様に複数種類の蛍光色素を励起し、蛍光信号を検出できるようにすることも可能となる。
なお、本変形例でも、導光部材として、オプチカルファイバ84を適用した例を述べたが、例えばガラス棒やアクリル材を成形加工したようなものを適用することができる。
本発明は、上記実施の形態に限定されるものでなく、実施段階では、その要旨を変更しない範囲で種々変形することが可能である。例えば、上述した各実施の形態において適用する試料には、DNAマイクロアレイの一例に限定されず、いわゆるDNAマイクロアレイと呼ばれる反応容器の全てに適用可能である。また、DNAに限らず、それ以外の種々の生物学的材料を扱う検査や測定に広く適用可能である。
さらに、上記実施の形態には、種々の段階の発明が含まれており、開示されている複数の構成要件における適宜な組み合わせにより種々の発明が抽出できる。例えば、実施の形態に示されている全構成要件から幾つかの構成要件が削除されても、発明が解決しようとする課題の欄で述べた課題を解決でき、発明の効果の欄で述べられている効果が得られる場合には、この構成要件が削除された構成が発明として抽出できる。
本発明の実施の形態によれば、試料の標識した物質に光を照射するための光路と集光レンズを有する蛍光検出手段の光路が分離されているので、ダイクロイックミラーなどの光を減衰させる光学素子を省略でき、検出される光強度の損失を最小限に抑えることができる。
また、本発明の実施の形態によれば、半導体光源手段を集光レンズの周囲に沿って複数個配置し、これらより同時に光を照射するようにしたので、標識物質に光を照射するのに十分な強度の光を効率よく得ることができる。
さらに、本発明の実施の形態によれば、半導体光源手段から発せられる光の方向および集光レンズの光軸に対する傾き方向をそれぞれ調整可能としているので、試料の所望する特定箇所に光を集光させることができ、標識物質への効率的な光の照射を行うことができる。
さらにまた、本発明の実施の形態によれば、試料近傍で得られる光強度に基づいて半導体光源手段への駆動電流を各別に制御できるので、半導体光源手段からの光を同一の明るさに揃えることができ、試料を均一の明るさで照明することができる。
以上述べたように本発明の実施の形態によれば、十分な光を効率よく照射でき、信頼性の高い光検出を行うことができる光検出装置を提供することができる。Hereinafter, embodiments of the present invention will be described with reference to the drawings.
