PURPOSE:To preclude following unnecessary measurement due to a failure in preliminarily adjustment and to improve the accuracy and efficiency of measurement by deciding whether or not the quantity of light in a specific wavelength range of a light source is a proper value at the time of calibration and performing the measuring operation only when the quantity of the light is the proper value. CONSTITUTION:When processing is set in calibration mode, a control arithmetic circuit 21 operates light sources 4 and 5 and a photodetecting means 6 while a sample is not inserted or while a filter for calibration is inserted and measures the quantity of the light of the specific wavelength of the light sources 4 and 5. Then the arithmetic circuit 21 decides whether or not the measured quantity of light is within a predetermined proper range. When it is judged that the quantity is the proper value, the control circuit 21 enables following measuring operation and stores the quantity of light measured at the time of calibration in a storage means 26. Then, the quantity of light stored in the storage means 26 and the quantity of sample transmitted light which is measured next are used to calculate the density of a specific substance in the sample, e.g. chlorophyll.
