Description
本発明は、Daedalea dickinsii KSDD6、
FERM−P No.3993又はLentinus edodes
KSLE7、FERM−P No.3994をデイステイラー
ズ・ソルブルスに接種して得られる菌糸体由来の
多糖体でKS物質と命名されるインターフエロン
誘起剤に関する。
本発明者等はすでにLentinus edodesの芽胞お
よびその培養菌体より強力なインターフエロン誘
起剤である2本鎖RNAが抽出されることを見出
している(F.Suzuki、T.Koide、A.Tsunoda、&
N.Ishida:Mushroom Science IX(Part )、
Proceedings of the Ninth International
Scientific Congress on the Cultivation of
Edible Fungi、Tokyo、1974、p509)。
更にLentinus edodesからレンチナンと称され
る抗腫瘍物質が調製されうることも知られている
(G.Chihara、Y.Maeda、J.Hamuro、T.Sasaki

本発明者らは、同様担子菌類についての培養
を、特殊な培地(後記)で更に研究を行なつてい
たところ、培養菌糸体よりレンチナンとは異なる
糖組成を有する多糖体の抽出に成功し、この多糖
体にインターフエロン誘起能を見出し、その結果
発現されるインフルエンザ感染に対する防御作用
を見出し、本発明を完成した。
本発明によれば、インターフエロン誘起作用を
有する多糖体は、次のようにして培養した
Daedalea dickinsii KSDD6、FERM−P No.
3993又はLentinus edodes KSLE7、FERM−P
No.3994の菌糸体より抽出される。
(1) 菌体の培養
本発明物質を調製するには、上記担子菌を約
3〜5゜Pに調節したデイステイラーズ・ソル
ブルズを主成分とする培地に無菌的に接種し、
通常の通気培養条件下、たとえば15〜35℃、好
ましくは20〜30℃、0.01〜2.5//分、好
ましくは0.5〜1.5//分の通気を行ないつ
つ、約1〜30日間、好ましくは約15〜25日間培
養を行なう。
(2) 本発明物質の抽出
上記培養混合物を過または遠心分離により
菌糸体を分離し、次いで菌糸体を所望により適
宜細断し、熱水または塩類希水溶液、好適には
60〜130℃、特に80〜100℃の約5%NaCl含有
水を用いて抽出を行なう。抽出は撹拌下約5分
〜3時間、特に30〜60分間行なえば充分であ
る。
抽出液について所望により固形物を分離後、
水と任意に混和する有機溶媒、たとえばメタノ
ール、エタノールを用いて沈澱処理を行ない、
得られた沈澱を過または遠心分離により集
め、凍結乾燥によりインターフエロン誘起作用
を有する多糖体が得られる。なお上記有機溶媒
沈澱操作の代りに硫安等を用いた塩析法によつ
ても、同様の目的を達することができる。な

3993をデイステイラーズ・ソルブルズ単独培地で
24℃で14日間振とう培養して得られた培養菌を、
デイステイラーズ・ソルブルズ(3゜P)に接種
し、24℃で1.8//分の通気を行ないつつ11
日間培養した。培養混合物を遠心分離することに
より菌糸体38gを得た。
得られた菌糸体に熱5%食塩水10を加え、60
分間煮沸後上澄液と沈澱を分離した。同様の操作
を更に2回繰返した。このようにして得られた上
澄液を合せ、これに最終エチルアルコール濃度が
70%になるようにエチルアルコールを加え、冷暗
所に一夜放置した。生成した沈澱を集め、凍結乾
燥して目的とするKS物質4gを得た。
得られた物質の物性を調べた結果を下記に示

インフルエンザ感染防御試験
平均体重14〜16gのDDI系マウス40匹を対照群
としてマウスにインフルエンザウイルスを経鼻噴
霧感染させた。次に同様に感染されたマウスに例
2で得たKS物質を投与し、また陽性対照群には
ヴイラゾールを投与し、生存日数と生存率を調べ
た。KS物質とヴイラゾールは生理食塩水に溶解
したのち、ウイルス感染前1時間、同時、感染後
1、3、6時間目の各時間、以後4日間は1日2
回腹腔に計15回投与した。またKS物質を経口に
より、同様の手順で投与した群も試験群とした。
また同様マウス40匹に対し腹腔より0.5mlの生理
食塩水を投与した群を対照群とした。
使用ウイルスはインフルエンザA2/熊本株
で、マウスに充分馴化した株であり、感染経路は
噴霧感染による鼻腔接種で、ウイルス接種量は

例 5
平均体重22gのDDI系マウス40匹に腹腔より例
2で得られたKS物質200mg/Kg体重を投与した群
()、同40匹に経口によりKS物質500mg/Kg体重
を投与した群()を処理群とし、動物体内に誘
起されるインターフエロン(interferon、ウイル
ス感染阻止物質)力価を検討した。この際同様マ
ウス40匹に対してキナクリン(quinacrin)200
mg/Kg体重を腹腔より投与した群()を陽性対
照群とし、更に同様マウス40匹に対し腹腔より
0.5mlの生理食塩水を投与した群()を対照群
とした。
各群とも投与後4、8、12、16、20、24、28、
32時間後に40匹より任意に抽出した5匹のマウス
を屠殺し、採取した血液を3000r.p.m.で10分間遠
心し、血清を分離した。
誘起されたインターフエロンは、マウスL細胞
由来の一変異株でチミジン・キナーゼ欠損のL−
1D細胞に対する水胞性口内炎ウイルス(VSV)
の感染を、段階的に希釈したマウス血清がどこま
で阻止するかにより定量し、最終的に米国立予防
衛生研究所(National Institute of Health
(NIH))より供与された国際マウスインターフエ
ロンを用いて国際単位に換算した。
その結果を第5図に示す。
例 6
KS物質によつてマウス血清中に誘起されたイ
ンターフエロンについて、以下の検討を行なつ
た。
例1により得られたKS物質200mg/Kg体重を、
平均体重21gのDDI系マウスの腹腔に投与し、20
時間後に屠殺し、採取した血液を3000r.p.m.にお
いて10分間遠心し、血清を分離し、この血清を56
℃で1時間加熱処理したもの、0.1N塩酸でPH2.0
に調整し、4℃に18時間放置したもの、更に最終
濃度1000mcg/mlとなる量のトリプシンで37℃に
おいて3時間処理したものについて例5と同様に
インターフエロン力価を検討した。

KS物質により誘起されたインターフエロンの
種特異性を検討するため以下の試験を行なつた。
平均体重2Kgの家兎10羽、平均体重20gのマウ
ス30匹、平均体重800gのニワトリ10羽の腹腔に
例1で得られたKS物質200mg/Kg体重を各々投与
し、20時間後にそれぞれの群より採取した血液
を、3000r.p.mで10分間遠心分離し、これら3群
の動物の血清を分離した。
次に、あらかじめ用意したウサギ由来の樹立
RK−13細胞、マウス由来のL−1D細胞および初
代培養のニワトリ繊維芽細胞を、これら3群の血
清の希釈液で20時間37℃で前処理し、例5で示し
た方法でウイルス感染阻止効果を調べた。

ついても全く同様であつた。
従つてKS物質によつて誘起されるインターフ
