Abstract

PURPOSE: To prepare cholesterol in high purity, with extremely simple operation, by extracting the organs from a mouse afflicted with hyperliposis caused by a novel mutant gene, and extracting the organ with a solvent.
CONSTITUTION: A mouse afflicted with hyperliposis caused by a novel munant gene (lpd) is grown, and slaughtered. The organs are extracted from the body and homogenized. Cholesterol is extracted from the product and purified by conventional method. For example, a normal female mouse is transplanted with the ovary of the lpd homozygote, and is mated with a lpd/+male mouse to obtain the lpd/lpd bodies and the lpd/+bodies at a ratio of 1:1. The process is repeated to produce the lpd homozygote. Thereafter, the acetone-soluble fraction is extracted by acetone fractionation process, purified, by silica gel column chromatography, and recrystallized to obtain cholesterol having a purity of ≥99%.
