Abstract

PURPOSE: To obtain, in high yields, a water-soluble chitosan derivative useful as a film-forming agent or excipient for medical use, by reacting chitosan with an alkylene oxide in the presence of an alkali in a mixed system containing a specified organic solvent and water.
CONSTITUTION: A slurry is formed by adding, with agitation, 100pts.wt. chitosan to 250W500pts.wt. difficultly water-soluble organic solvent (e.g., aliphatic or aromatic hydrocarbon), an organic solvent miscible with water, (e.g., 3W4C aliphatic alcohol) or a mixture thereof. Then, to this slurry are added, with agitation, 5W 50pts.wt. alkali (e.g., NaOH) and 10W150pts.wt. water. Next, an alkylene oxide (e.g., ethylene oxide, propylene oxide) is added in an amount corresponding to the desired substitution degree, and the resulting mixture is reacted at 20W120°C for 5W10hr to obtain a water-soluble chitosan derivative. A film can be formed by dissolving the derivative in water or the like, spreading the solution over a smooth plate and drying the spread solution.
