Abstract
PURPOSE: To collect a human β-interferon, by administering a human β-interferon antigen consisting of a composite of a N-terminated peptide with of a human β- interferon with a carrier to a mammal.
CONSTITUTION: An N-terminated peptide of a human β-interferon expressed by the formula (R is H, H-Phe-, H-Leu-Gly-Phe-, etc.) is reacted with a carrier in an amount of 2W6 times that of the peptide and a haptene-carrier linking reagent in a molar amount of 5W10 times that of the peptide in a buffer solution at a pH of 7W10 at 0W40°C to give a human β-interferon antigen, which is then administered to a mammal, e.g. a rabbit or guinea pig, to collect an antibody produced in the living body. The resultant antibody has improved specificity for the human β-interferon and high potency and sensitivity, and can be obtained at a high potency and sensitivity, and canb be obtained at a high reproducibility. The antibody can be reacted with a well-known insolubilizing substance to give an insolubilized antibody.
