Abstract
PURPOSE: To prepare the titled enzyme usable for the determination of creatinine and creatine in a large quantity, by cultivating Cylindrocarpon didymum M-1.
CONSTITUTION: A microorganism which belongs to the genus Cylindrocarpon and has the ability to produce sarcosine oxidase, e.g., cylindrocarpon didymum M-1, is cultivated in an ordinary culture medium containing a carbon and a nitrogen sources, an inorganic salt, etc. The addition of choline, dimethylglycine or sarcosine to the culture medium further improves the productivity of the sarcosine oxidase. After the aerobic cultivation, the aimed substance is obtained by the centrifugation, ultrasonic treatment, milling in a homogenizer, fractionation with ammonium sulfate, ion exchange chromatography, gel filtration method, etc.
