Abstract
PURPOSE: To obtain amorphous nicomol having remarkably high absorption efficiency compared with crystalline nicomol and hence effective at a considerably low dose, by dissolving crystalline nicomol useful as a medicine in a water-souble organic solvent, and precipitating from the solution with water, etc.
CONSTITUTION: Amorphous nicomol [general name of 2,2,6,6-tetrakis(nicotinoyloxymethyl)cyclohexanol] is prepared either by dissolving crystalline nicomol in a water-soluble organic solvent such as dimethylformamide and precipitating with water, or by dissolving in an acidic solvent such as hydrochloric acid, and precipitating by neutralizing with a base. The precipitated nicomol is dried at a low temperature (≤50°C) to prevent crystallization, by distilling off the solvent under reduced pressure or by spray-drying. As an alternative method, the objective compound is prepared by adding or dissolving a hydrophilic polymer, a sugar alcohol, bile acid, higher fatty acid, etc. to the nicomol solution and removing the solvent from the mixture.
USE: Medicine having lipometabolism exciting, peripheral circulation increasing activity.
