Abstract
NEW MATERIAL:A prostaglandin derivative expressed by formula I [R1 is H, alkyl, cycloalkyl, cation, etc.; W is -CH2CH2- or -CH=CH-; A and B are H or OH; A+B is OXO; X is O, NH or valence bond; R2 is -C(=0) NHR3 (R3 is alkyl, phenyl, etc.) when X is O or NH, or -R3 when X is valence bond]. EXAMPLEMethyl (Z)-7-[(1'S, 4'R, 5'S, 6'R)-5'-(S)-(α-hydroxybenzyl)-2'-oxabicyclo[2.2.1] heptan-6'-yl]-5-heptenoate.
USE: A compound having an anticoagulation action on platelets, and effective for preventing and treating cerebral thrombosis and myocardial infarction.
PROCESS: For example, the compound of formula I (X is valence bond) is obtained as follows: The compound of formula II is reacted with a Grignard reagent of the formula R2MgX (X is Cl, Br or I) to give compounds of formulas III and IV, which are converted into compounds of formulas VI and VII. Alternatively, the compound of formula III or IV is oxidized to give a compound of formula V, which is then hydrolyzed to afford a compound of formula VIII.
