Abstract

PURPOSE: A Chaetomium bacterium, producing trehalase is cultured and trehalase is extracted and collected from the culture medium, thus producing efficiently trehalase.
CONSTITUTION: A trehalase-producing bacterium, which belongs to Chaetomium, is aerobically cultured in a liquid medium or on the surface of a solid medium at a temperature of 25W35°C, preferably near 30°C, under slightly acidic conditions of about 5 pH at the beginning of the incubation for over 3 days, thus accumulating trehalase. The culture in the case of liquid incubation is filtered to separate cells and then subjected to salting out or precipitation with an organic solvent or in the case of solid incubation is extracted and then treated in the same manner to give crude trehalase enzyme powder. The resulting crude enzyme is further purified by desalting with molecular sieve or by pH treatment.
