Abstract

PURPOSE: To make it possible to separate a long-chain dicarboxylic acid advantageously, by incubating a microorganism capable of producing the long-chain dicarboxylic acid in a culture medium of a straight chain hydrocarbon as a substrate, allowing the fermentation broth thus obtained to stand in a specific pH range, and centrifuging the broth to separate the microbial cell.
CONSTITUTION: A microorganism capable of producing a dicarboxylic acid is incubated in a culture medium of a straight chain hydrocarbon, e.g. n-tridecane, as a substrate, and the fermentation broth thus obtained is adjusted to pH9W12 with an alkili, e.g. NaOH or KOH, and allowed to stand at ordinary temperature for about 10hr. The broth is then centrifuged to separate the microbial cell from the hydrocarbon attached to the cell and other components. The separated fluid thus obtained is filtered through diatomaceous earth as a filter aid to give a clear filtrate, which is fed to the deposition step of the long-chain dicarboxylic acid. The separated solid content (mainly the diatomaceous earth) is circulated through the centrifuging step and reused. 95% or more of the microbial cell is separated to facilitate the purification of the long-chain dicarboxylic acid.
