Abstract

PURPOSE: To make it possible to measure the MAO activity accurately and readily, by determining the oxidation product, formed through the enzyme reaction of an azo derivative of a specific formula or its salt as a substrate, optically.
CONSTITUTION: A solution of an azo derivative, e.g. 2-(3'-nitro-4'-aminomethylphenylazo)-m-cresol hydrochloride, of formula I: (X is aromatic group except unsubstituted phenyl group; R is group forming a chromophore with the adjacent -N=N-, e.g. unsubstituted phenyl group) or its salt as a substrate in tris (hydroxymethyl)aminomethane-HCl buffer is subjected to the enzyme reaction with blood serums, warmed and extracted with cyclohexame. The absorbance of the extract is measured by spectroscopy at a peak of 435nm due to the formed oxidation porduct, 2-(3'-nitro-4'-benzaldehydoazo)-m-cresol, and the ratio of the absorbance to that of the standard solution provides the activity of enzyme unit of monoamine oxidase (MAO) in the serums.
