Abstract

PURPOSE: To remove the influence of an interfering substance and measure a minute quantity of a substance in a living body fluid with high sensitivity, by using an antibody fragment which is obtained by decomposing an antibody immunoglobulin with a protease, and by adding a hydrophobic protein and salts to the antibody fragment.
CONSTITUTION: An antibody fragment 7 obtained by decomposing an antibody immunoglobulin with a protease is fixed in a solid phase 3 of silicone rubber or the like. On the other hand, a bonded substance comprising an enzyme 4 and an antibody fragment 7' bonded with each other is prepared. A living body fluid is brought into contact with the fragment 7 to bond a measured substance to the fragment 7. The bonded substance comprising the enzyme 4 and the fragment 7' is conjoined to the measured substance 1. The free bonded substance is removed by washing. The activity of the enzyme bonded to the solid phase is measured to determine the quantity of the substance 1. Even if there is a self-antibody such as a rheumatic factor 5, it is not bonded to any antibody fragment. Therefore, the self-antibody does not perform interferential action. Since a hydrophobic protein and salts are added to the antibody fragment, various factors originating from blood serum are also prevented from interfering with immunoreaction.
