NEW MATERIAL:A novel bicyclooctane derivative of formula I (R1 is H, or lower alkyl group; X is vinylene or ethylene chain; R2 is 4W7C alkyl group; R3 is lower alkyl group).
EXAMPLE: 2β-( 3'-Methyl-3'-hydroxy-trans-1'-octenyl )-3α-hydroxy-7-( 4'-methoxycarbonyl butylidene)-bicyclo[3.3.0]octane.
USE: Prostaglandin I2 analogs having powerful inhibitory actions on blood platelet aggregation.
PROCESS: A compound of formula II (Y is H or OH) is reacted with a Wittig reagent of formula III (M is H or alkali metal; R1 is aryl group) in the presence of a solvent, e.g. a dialkyl sulfoxide, and the OH-protecting group, if present, is eliminated and esterified to form the compound of formula I stabler than prostaglandin I2.
