ロールは、トコフェロールとコハク酸がエステル結合し
た化合物である。コハク酸トコフェロールのトコフェロ
ール部分は、天然品、合成品のいずれであってもよく、
また、α、β、γ、δ型のいずれでも、また、d体、l
体、dl体のいずれであっても使用できる。さらにコハク
酸トコフェロールはカルシウム塩等の無機塩、あるいは
有機塩であっても差し支えない。d−α−コハク酸トコ
フェロール、dl−α−コハク酸トコフェロール、dl−α
−コハク酸トコフェロールカルシウムは市販されてお
り、容易に入手可能である。本発明においてコハク酸ト
コフェロールの粒子径は、250μm以下であることが好ま
しい。この粒径のものを得るためには、ハンマーミル、
ボールミル、ジェット粉砕等、公知の粉砕方法によるこ
とができるが、粉砕中に熱を発生することがあるため、
冷却下で行うことが好ましい。ここで粒子径の値は、た
とえば顕微鏡法のように直接計測したものを意味する。
粉砕したコハク酸トコフェロールを篩過することによ
り、粒子径が250μm以下のコハク酸トコフェロールの重
量比が99%以上であるコハク酸トコフェロール粉末を得
ることができる。コハク酸トコフェロールは、以下に例
示する一種以上の製剤用担体と混合および練合後、造粒
機で造粒し粒剤を製造する。造粒方法としては、押し出
し造粒法、攪拌転動造粒法、流動層造粒法、流動攪拌転
動造粒法、遠心流動転動造粒法、噴霧造粒法、破砕造粒
法等があり、いずれの方法でも差し支えないが、押し出
し造粒法を用いる場合に本発明の効果が最も顕著に現れ
る。その後、公知の方法で乾燥、整粒および篩過等の操
作を行う。押し出し造粒機等に装填するスクリーンの径
は特に限定されないが、好ましくは1.5mm以下、さらに
好ましくは0.7mm以下である。本発明における粒剤と
は、顆粒剤または細粒剤と称されるものであり、通常そ

リンナトリウム、グリチルリチン酸ジカリウムなどが用
いられる。着色剤、着香料としては医薬品に添加するこ
とが許可されているものであればよく、特に限定されな
い。また、粒剤は必要に応じてコーティングを施しても

間混合した。その後ヒドロキシプロピルセルロース600g
をアルコール3200gに溶解した結合剤溶液を添加し、15
分間混練合した。この連合物を径0.5、0.6または0.7mm
のスクリーンを装填した押し出し造粒機で造粒し、顆粒
剤を得た。この顆粒剤を55±5℃にセットした棚式乾燥
機を使用して、乾燥減量が1%以下になるまで乾燥し

を通過し、40号篩に残留した部分を分析用試料とした。

顆粒および残留物中のコハク酸トコフェロール含量をH
PLC法によって測定し、その結果を表1に示した。測
定値は試料1250mg中にコハク酸トコフェロールが100mg
相当含まれていた場合の含量を100%として表記した。
含量が95〜105%の範囲のときに適合として○を付し、
それ以外は不適合として×を付した。また、顆粒中含量
および造粒機残留物中の含量が共に適合であったもの
を、最終的な適合品として判定した。その結果、押し出
し造粒機のスクリーン径が0.7mm以下で、コハク酸トコ
フェロールが60、100号全通品および粉砕品、すなわち
