Claims (1)
- 【特許請求の範囲】 1.真菌であって、該組換えDNA技術によるareA遺伝子が、機能的なareA活性 化因子を供給するようには発現され得ない方法によって修飾された真菌。 2.前記不活性化がareA遺伝子のすべてまたは一部の欠失によって得られたも のである、請求項1記載の真菌。 3.前記不活性化がareA遺伝子自身の発現を調節する発現シグナルの調節を妨 げることによって得られたものである、請求項1記載の真菌。 4.前記不活性化がアンチセンス技術を用いて得られたものである、請求項1 記載の真菌。 5.前記不活性化がareA遺伝子中に内部的に余分なDNAを挿入することにより 得られたものである、請求項1記載の真菌。 6.該真菌が糸状菌、好ましくは、アスペルギルス、トリコデルマ、フミコー ラ、カンジダ、アクレモニウム、フサリウム及びペニシリウムを含む群から選択 された属に属するものである、請求項1〜5のいずれか1項に記載の真菌。 7.真菌が、A.オリザエ、A.ニガー、A.アワモリ、A.ホエニシス、A .ジャポニクス、A.フォエチダス、A.ニドラン、T.リーセイ、T.ハルチ アヌム、H.インスレンス、H.ラヌギノサ、F.グラミネアルム、F.ソラニ 、P.クリソゲヌム等を含む群から選択される種に属するものである、請求項6 に記載の真菌。 8.請求項1に記載の真菌の生産する法であって、前記不活性化がAreA遺伝子 の欠失により得られたものであって、該方法は i)着目の真菌からのareA遺伝子をクローン化し、 ii)内部の部分が置換もしくは欠失されるか、または余分なDNAが挿入されたa reA遺伝子を含む、DNA構築体を生産し、 iii)該構築体を有する真菌を形質転換し、 iv)areA-である形質転換体を選択すること含む、生産方法。 9.請求項1に記載の真菌の生産方法であって、前記不活性化がアンチセンス 技術により得られたものであって、 i)areA遺伝子から転写されたmRNAに相補的なRNAの合成を生じさせる発現プ ラスチドの構築、 ii)前記発現プラスミド及び異なるプラスミドまたは同じプラスミドのいずれ かにくっついている適当な標識での宿主真菌の形質転換、 iii)前記標識を用いる形質転換体の選択、及び iv)AreA産物の合成における低下を示す株についての選択された形質転換体の スクリーニングを含む、生産方法。 10.所望の遺伝子産物の生産方法であって、請求項1〜7のいずれか1項に記 載の真菌を適当な条件で適切な増殖培地で培養し、所望の遺伝子産物を回収し、 精製することによる方法。 11.所望の遺伝子産物の生産方法であって、請求項1〜7のいずれか1項に記 載の真菌であって、所望の遺伝子産物をコードするDNA配列を真菌のゲノムに、 機能的な仕方で組込まれるように形質転換された真菌を適当な条件で適切な増殖 培地で培養し、所望の遺伝子産物を回収し、精製することによる方法。 12.所望のポリペプチドを生産する方法であって、真菌を適当な増殖培地で培 養し、前記ポリペプチドを前記培養から回収する方法を含んでなり、前記真菌は 前記真菌中に前記ポリペプチドまたはその前駆体の発現を引き起こし得る組換え DNA構築体を担持し、前記真菌はさらに前記真菌の野生型よりも少量の機能的なA reAを生産す ることにより特徴づけられる、前記方法。 13.請求項12に記載の方法であって、前記真菌のゲノムに組込まれたとき、機 能的なAreAの生産の減少を引き起こし得るDNA構築物で、前記真菌の親を形質転 換することを含む方法により、前記真菌は野生型よりも少量のAreAを生産するよ うに修飾されている、前記方法。 14.請求項12に記載の方法であって、前記真菌により前記ポリペプチドが細胞 外培地に分泌される方法。 15.請求項12に記載の方法であって、前記真菌が同様の真菌よりも多量の前記 ポリペプチドを生産し、前記同様の真菌は、野生型の前記真菌により生産される AreAと同様な量のAreAを生産し、前記同様な真菌は前記真菌と他のすべての点で 同一である、方法。 16.前記遺伝子産物が分泌されたタンパク質である、請求項10または11〜15記 載の方法。 17.請求項10〜16のいずれか1項に記載の方法であって、所望の遺伝子産物が 、産業用ペプチドまたはタンパク質、好ましくは酵素である、方法。 18.請求項17に記載の方法であって、前記酵素は、プロテアーゼ、リパーゼ、 クチナーゼ、セルラーゼ、キモシンを含む群から選択される、方法。 19.請求項10〜116のいずれか1項に記載の方法であって、所望の遺伝子産物 が、治療上活性なペプチドまたはタンパク質である、方法。 20.請求項19に記載の方法であって、前記治療上活性なペプチドまたはタンパ ク質が、インスリン、成長ホルモン、グルカゴン、ソマトスタチン、インターフ ェロン、PDGF,VII因子、VIII因子、ウロキナーゼ、tPA,EPOまたはTPOを含む群 から選択される、方法。 21.10〜20の方法のいずれか1つの方法により生産された遺伝子産物。 22.A.オリザエからのareA遺伝子(配列番号1)またはその機能的な対立遺 伝子をコードする、DNA配列。 23.A.オリザエからのAreA活性化因子(配列番号2)。
