Claims (1)
- 【特許請求の範囲】 1.被験体においてトランスフォーミング成長因子−βの活性を刺激する方法で あって、該被験体中の潜在型トランスフォーミング成長因子−βを、潜在型トラ ンスフォーミング成長因子−βを活性型トランスフォーミング成長因子−βに変 換するのに有効な量のトロンボスポンジンと接触させる工程を包含する、方法。 2.被験体においてトランスフォーミング成長因子−βの活性を刺激する方法で あって、該被験体中の潜在型トランスフォーミング成長因子−βを、潜在型トラ ンスフォーミング成長因子−βを活性型トランスフォーミング成長因子−βに変 換するのに有効な量の、トロンボスポンジンの精製された活性化ペプチドと接触 させる工程を包含する、方法。 3.被験体においてトランスフォーミング成長因子−βの活性を刺激する方法で あって、該被験体中の潜在型トランスフォーミング成長因子−βを、相当量の、 3から30アミノ酸を有する精製された活性化ペプチドと接触させる工程を包含 し、 ここで、該ペプチドはサブ配列R1−X1−X2−X3−R2を含み、X1はArgお よびLysからなる群から選択され、X2はProおよびPheからなる群から選択され、 X3はLysおよびArgからなる群から選択され、R1はH、アシル、または1から2 6アミノ酸のペプチド、R2はH、NH2、または1から26アミノ酸のペプチド であり、そして、 該ペプチドは、潜在型トランスフォーミング成長因子−βを活性型トランスフ ォーミング成長因子−βに変換する、方法。 4.被験体においてトランスフォーミング成長因子−βの活性を刺激する方法で あって、該被験体中の潜在型トランスフォーミング成長因子−βを、相当量の、 以下からなる群から選択される精製された活性化ペプチドと接触させる工程を包 含する、方法: 5.前記精製された活性化ペプチドが、アミノ酸配列RFK(配列番号18)か らなる、請求項2に記載の方法。 6.被験体においてトランスフォーミング成長因子−βの活性の刺激を阻害する 方法であって、該被験体中のトロンボスポンジンを、トロンボスポンジンに結合 して潜在型トランスフォーミング成長因子−βの活性型トランスフォーミング成 長因子−βへの変換を予防するのに有効な量の、トロンボスポンジンに特異的な 抗体と接触させる工程を包含する、方法。 7.被験体においてトランスフォーミング成長因子−βの活性の刺激を阻害する 方法であって、該被験体中の潜在型トランスフォーミング成長因子−βを、潜在 型トランスフォーミング成長因子−βの活性型トランスフォーミング成長因子− βへの変換を阻害するのに有効な量の、トロンボスポンジンの4つの連続したア ミノ酸の配列に対応する配列を有する、精製された阻害ペプチドと接触させる工 程を包含する、方法。 8.被験体においてトランスフォーミング成長因子−βの活性の刺激を阻害する 方法であって、該被験体中の潜在型トランスフォーミング成長因子−βを、相当 量の、以下からなる群から選択される精製された阻害ペプチドと接触させる工程 を包含する、方法: 9.被験体において創傷の治癒を増強する方法であって、該被験体の創傷部位に 、潜在型トランスフォーミング成長因子−βを活性型トランスフォーミング成長 因子−βに変換するのに有効な量のトロンボスポンジンを投与する工程を包含し 、トランスフォーミング成長因子−βの該活性化が結果として創傷の治癒を増強 する、方法。 10.被験体において創傷の治癒を増強する方法であって、該被験体の創傷部位 に、潜在型トランスフォーミング成長因子−βを活性型トランスフォーミング成 長因子−βに変換するのに有効な量の、TSP由来の精製された活性化ペプチドを 投与する工程を包含し、トランスフォーミング成長因子−βの該活性化が結果と して創傷の治癒を増強する、方法。 11.前記精製された活性化ペプチドが以下のアミノ酸配列からなる、請求項1 0に記載の方法: 12.前記精製された活性化ペプチドが、アミノ酸配列RFK(配列番号18) から実質的になる、請求項10に記載の方法。 13.被験体において病的状態でトランスフォーミング成長因子−βによって刺 激される線維症を予防する方法であって、該被験体の線維症を生じ得る部位に、 トロンボスポンジンに結合するのに有効な量の、トロンボスポンジンに特異的な 抗体を投与する工程を包含し、これによりTGF-βの活性化を予防して、結果とし て線維症が抑制される、方法。 14.被験体において病的状態でトランスフォーミング成長因子−βによって刺 激される線維症を予防する方法であって、該被験体の線維症を生じ得る部位に、 潜在型トランスフォーミング成長因子−βの活性型トランスフォーミング成長因 子−βへの変換を阻害するのに有効な量の、TSP由来の精製された阻害ペプチド を投与する工程を包含し、結果として線維症が抑制される、方法。 15.前記精製された阻害ペプチドが以下からなる群から選択される、請求項1 4に記載の方法: 16.前記精製された阻害ペプチドが、アミノ酸配列GGWSHW(配列番号3 )から実質的になる、請求項14に記載の方法。 17.被験体においてトランスフォーミング成長因子−β仲介型の内皮細胞の増 殖の阻害を遮断する方法であって、該被験体の内皮細胞を、トロンボスポンジン に結合するのに有効な量の、トロンボスポンジンに特異的な抗体と接触させる工 程を包含し、これによりトランスフォーミング成長因子−βの活性化を予防して 、結果として内皮細胞を増殖させる、方法。 18.被験体においてトランスフォーミング成長因子−β仲介型の内皮細胞の増 殖の阻害を遮断する方法であって、該被験体の内皮細胞を、潜在型トランスフォ ーミング成長因子−βの活性型トランスフォーミング成長因子−βへの変換を阻 害するのに有効な量の、TSP由来の精製された阻害ペプチドと接触させる工程を 包含し、結果として内皮細胞を増殖させる、方法。 19.前記精製された阻害ペプチドが以下からなる群から選択される、請求項1 8に記載の方法: 20.前記精製された阻害ペプチドが、アミノ酸配列GGWSHW(配列番号3 )から実質的になる、請求項18に記載の方法。 21.3から30アミノ酸を有する精製されたペプチドであって、 ここで、該ペプチドはサブ配列R1−X1−X2−X3−R2を含み、X1はArgお よびLysからなる群から選択され、X2はProおよびPheからなる群から選択され、 X3はLysおよびArgからなる群から選択され、R1はH、アシル、または1から2 6アミノ酸のペプチド、R2はH、NH2、または1から26アミノ酸のペプチド であり、そして、 該ペプチドは、潜在型トランスフォーミング成長因子−βを活性型トランスフ ォーミング成長因子−βに変換する、ペプチド 22.前記ペプチドが以下からなる群から選択される、請求項21に記載のペプ チド: 23.配列表において配列番号10で定義されるアミノ酸配列RWRPWTAW SEから実質的になる、ペプチド。 24.前記ペプチドが水溶性ポリマーに接合された、請求項21に記載のペプチ ド。 25.前記ペプチドが部分的および完全にレトロ−インベルソのペプチド配列か らなる群から選択される、請求項21に記載のペプチド。 26.配列表において配列番号28で定義されるアミノ酸配列アセチル-WHS WAA-NH2から実質的になる、精製されたペプチド。 27.前記ペプチドが部分的または完全にレトロ−インベルソのペプチド配列の 群から選択される、請求項26に記載のペプチド。
