Description
Claims (1)
- 【特許請求の範囲】 1.ヘルパーT細胞エピトープ(Th)および黄体形成ホルモン放出ホルモン(LHRH )よりなるペプチドであって、該LHRHが該ペプチドのカルボキシル末端にあるこ とを特徴とする当該ペプチド。 2.該ペプチドが式: (A)n-(Th)m-(B)o-LHRH [式中、Aは、独立dw、アミノ酸、α-NH2、トリパルミトイルシステイ ン、脂肪酸、インベーシンドメインまたは対応するインベーシンドメインの免疫 刺激アナログ; Bはアミノ酸; 各Thは、独立して、ヘルパーT細胞エピトープ、またはその免疫増強アナ ログもしくはセグメントからなるアミノ酸配列であり; LHRHは、黄体形成ホルモン放出ホルモンまたはその免疫原性アナログ; nは1から約10; mは1から約4;および oは0から10である] で表される請求項1記載のペプチド。 3.該ペプチドが哺乳動物のLHRHに対する免疫応答を剌激する請求項2記載の ペプチド。 4.該ペプチドでの免疫化が血清のテストステロンを正常値の10%未満に軽減 させ得る請求項2記載のペプチド。 5.該ペプチドでの免疫化が前立腺の萎縮を生ずるかまたはその成長を妨げる 請求項2記載のペプチド。 6.該LHRHが配列番号:1のアミノ酸配列を有する請求項2記載のペプチド。 7.該Thが配列番号:2-9または42-52、またはそのアナログもしくはセクメン トの任意の1つから選択されるアミノ酸配列を有する請求項2記載のペプチド。 8.該ペプチドが配列番号:10-41の何れか1つのアミノ酸配列を有する請求項 2記載のペプチド。 9.少なくとも1つのAがインベーシンドメインである請求項2記載のペプチド 。 10.nが4で、かつAが次の順序でα-NH2、インベーシンドメイン、グリシンお よびグリシンである請求項9記載のペプチド。 11.該インベーシンドメインが配列番号:53のアミノ酸配列を有する請求項2ま たは10記載のペプチド。 12.配列番号:10、13、16、18、19、32または38のアミノ酸配列からなるペプ チド。 13.免疫学的に有効な量の請求項1、2、8、9または12記載のペプチドおよび医 薬上許容される担体よりなるワクチン組成物。 14.該ペプチドの免疫学的有効量が投与用量当り体重キログラム当り約0.5μg から約1mgまでである請求項13記載のワクチン組成物。 15.哺乳動物に請求項13記載のワクチン組成物を該哺乳動物の不妊症状態を作 り出す時間の間かつ条件下で投与することからなる該哺乳動物に不妊症を誘発さ せる方法。 16.アンドロゲン依存型癌腫を治療する方法であって、該癌腫の抑制を発効さ せるかまたはその成長を防ぐ時間の間かつ条件下で哺乳動物に請求項13記載のワ クチン組成物を投与することからなる該アンドロゲン依存型癌腫を治療する方法 。 17.哺乳動物のLHRHの活性を抑制する方法であって、該LHRHの血清濃度を減ず るに十分な時間の間かつ条件下で請求項1、2、8、9または12記載のペプチドを該 哺乳動物に投与することからなる哺乳動物のLHRHの活性を抑制する方法。 18.該LHRH活性の抑制は、前立腺増殖、アンドロゲン依存性癌腫、前立腺癌腫 、精巣癌腫、子宮内膜症、良性の子宮腫瘍、再発性の機能的卵巣嚢腫、(重症の )月経前症候群、もしくはエストロゲン依存胸部腫瘍に対する治療法;エストロ ゲン依存胸部がん腫に対する治療法;もしくは不妊症を誘発させるための治療法 である請求項17記載の方法。 19.該LHRH活性の抑制は、豚のテンジクネズミの病毒を軽減し、犬および猫 を免疫去勢し、かつ雄の成熟馬を去勢するためのものである請求項17記載の方法 。 20.請求項1、2、8、9または12記載の2つ以上のペプチドの混合体からなるペ プチド組成物。 21.該混合体が配列番号:13、16、18および19のアミノ酸配列を有するペプチ ドの組合せからなる請求項20記載の組成物。 22.該混合体が配列番号:13、16、19および32のアミノ酸配列を有するペプチ ドの組合せからなる請求項20記載の組成物。 23.免疫剌激インベーシンドメイン、ヘルパーT細胞(Th)エピトープおよびペ プチドハプテンを含んでなる、約30ないし約90個のアミノ酸の合成ペプチド。 24.該ペプチドハプテンが前記ペプチドハプテンに対する免疫応答を剌激する 請求項23記載のペプチド。 25.該インベーシンが配列番号:53のアミノ酸配列かまたは該配列に対応する 免疫刺激性アナログである請求項23または24記載のペプチド。 26.前記Thエピトープが配列番号:2-9、42-52の何れか1つから選択されたアミ ノ酸配列か、もしくは該配列に対応する免疫増強アナログかまたはセグメントを 有する請求項23ないし25の何れか1つに記載のペプチド。 27.該ペプチドハプテンが配列番号:1、66-91の何れかから選択されたアミノ 酸配列か、もしくは該配列に対応する免疫原性アナログかを有する請求項23ない し26の何れか1つに記載のペプチド。 28.該ペプチドのアミノ末端に共有結合された(A)nを有するペプチドであって 、Aが、独立して、アミノ酸、α-NH2、トリパルミトイルシステインまたは脂肪 酸であり;nが1から約10までである請求項23ないし27の何れか1つに記載のペプ チド。 29.該インベーシンドメイン、該Thエピトープおよび該ペプチドハプテンが任 意の順序で共有的に結合されたアミノ酸の基を表すペプチドであって、実質的に 免疫反応性を保持しかつ該基の任意の2つの間にスペーサー(B)oを有し、Bが独立 して任意のアミノ酸でかつoが0から約10までである請求項23ないし28の何れか1 つに記載のペプチド。 30.oが2であってBがグリシンとグリシンである請求項29記載のペプチド。 31.該ペプチドが配列番号:92-114の何れか1つのアミノ酸配列を有するペプチ ドか、またはペプチド115を有する請求項23または24記載のペプチド。 32.該ペプチドハプテンがアミリンであるかまたはその免疫原性アナログであ る請求項24記載のペプチド。 33.該ペプチドハプテンが配列番号:66-68の何れか1つのアミノ酸配列を有す る請求項32記載のペプチド。 34.免疫学的に有効量の請求項32または33記載のペプチドおよび医薬上許容さ れる担体よりなるワクチン組成物。 35.糖尿病の治療において、循環アミリン濃度または血中グルコース濃度を下 げるため哺乳動物に請求項34記載のワクチン組成物を投与することを特徴とする 非インスリン依存型糖尿病の治療方法。 36.該ペプチドハプテンが、ガストリン34、カストリン17、またはその免疫原 性アナログである請求項24記載のペプチド。 37.該ペプチドハプテンが配列番号:69-76の何れか1つのアミノ酸配列を有す る請求項36記載のペプチド。 38.免疫学的に有効量の請求項36または37記載のペプチドと医薬上許容される 担体よりなるワクチン組成物。 39.請求項38のワクチン組成物を哺乳動物に投与してガストリン濃度を下げる ことからなる消化性漬瘍またはガストリン刺激腫瘍を治療する方法。 40.該ペプチドハプテンがガストリン放出ペプチドであるかまたはその免疫原 性アナログである請求項24記載のペプチド。 41.該ペプチドハプテンが配列番号:77-78の何れか1つのアミノ酸配列を有す る請求項40記載のペプチド。 42.免疫学的に有効量の請求項40または41記載のペプチドと医薬上許容される 担体よりなるワクチン組成物。 43.請求項42のワクチン組成物を哺乳動物に投与してガストリン放出ペプチド の濃度を下げることからなる消化性潰瘍、ガストリン放出ペプチド剌激腫瘍ま たは肺癌を治療する方法。 44.該ペプチドハプテンがIgE分子のCH4領域由来のペプチドであるかまたはそ の免疫原性アナログである請求項24記載のペプチド。 45.該ペプチドハプテンが配列番号:79のアミノ酸配列を有する請求項44記載 のペプチド。 46.免疫学的に有効量の請求項44または45記載のペプチドと医薬上許容される 担体よりなるワクチン組成物。 47.請求項46のワクチン組成物を哺乳動物に投与してマスト細胞または好塩基 細胞のヒスタミン濃度を減ずるかまたはIgE媒介活性化を遮断することからなる アレルギーを治療する方法。 48.該ペプチドハプテンがクラミジア・トラコマティス(Chlamydia trachomat is)主外膜タンパク質(MOMP)の可変領域(VDI-IV)であるかまたはその免疫原性ア ナログである請求項24記載のペプチド。 49.該ペプチドハプテンが配列番号:80-89の何れか1つのアミノ酸配列を有す る請求項48記載のペプチド。 50.免疫学的に有効量の請求項48または49記載のペプチドおよび医薬上許容さ れる担体よりなるワクチン組成物。 51.請求項50のワクチン組成物を哺乳動物に投与してクラミジア・トラコマテ ィスマストに対する中和抗体を生成することからなるクラミジアに対して哺乳動 物を免疫感作する方法。 52.該ペプチドハプテンがHIV V3の主中和領域であるかまたはその免疫原性ア ナログである請求項24記載のペプチド。 53.該ペプチドハプテンが配列番号:90のアミノ酸配列を有する請求項52記載 のペプチド。 54.免疫学的に有効量の請求項52または53記載のペプチドと医薬上許容される 担体よりなるワクチン組成物。 55.請求項54のワクチン組成物を哺乳動物に投与してヒトの免疫不全ウイルス に対する中和抗体を生成することからなる、後天性免疫不全症候群(エイズ) を治療するかまたはヒトの免疫不全ウイルス感染を防ぐ方法。
