カンプトテシンの水溶性誘導体
本発明は、7位で置換された水溶性カンプトテシン(Camptothecin)誘導体、
腫瘍の治療におけるそれらの用途およびそれらの製造方法に関する。
発明の背景
天然細胞毒性アルカロイドであるカンプトテシンは、トポイソメラーゼI阻害
剤でありかつ有効な抗腫瘍剤である。それはWall,et al.(J.Am.Chem.Soc.,88,3
888(1966))により中国植物Camptotheca accuminataの葉および樹皮から初めて
単離された。
下記のように、カンプトテシンはキノリン(AおよびB)がピロリジン環(C
)に縮合し、これがα‐ピリドン環(D)に縮合し、これが更にラクトン環(E
)に縮合して構成される縮合環系である。
それは20位に不斉炭素を有し、2つのエナンチオマー形を可能にしている。し
かし、天然化合物は上記のように“S”配置で見出される。
細胞毒性剤は腫瘍を抑制または根絶するためにしばしば用いられ、即ちそれら
は化学療法剤である。カンプトテシンの細胞毒性活性は、トポイソメラーゼ阻害
剤としてのカンプトテシンの効力に直接関連していると考えられる〔トポイソメ
ラーゼ機能の詳細な説明に関しては、A.Lehninger,Principles of Biochemistry
,813,Worth Publishers,New York(1982);L.F.Liu,'DNA Topoisomerases',CRC C
ritical Review in Biochemistry,1-24,15(1983);H.Vosberg,'DNA Topoisomera
ses:Enzymes that Control DNA Conformation',Current Topics in Microbiolo
gy and Immunology,19,Springer-Verlag,Berlin(1985)参照〕。特に、カンプト
テシンは実験動物において白血病(L‐1210)およびある固形腫瘍の治療上
有効であることが示された。例えば、Chem.Rev.23,385(1973)およびCancer Trea
t.Rep.,60,1007(1967)参照。
残念ながら、臨床上、有効な抗腫瘍剤としての条件をカンプトテシンは完全に
は満たされなかった。カンプトテシンは生理学上適合する水性媒体に本質的に不
溶性であり、抗腫瘍治療にとり好ましい様式、即ち非経口投与上、それを十分に
可溶性にするよう変更されなければならない。それはそのナトリウム塩を形成す
ることにより、即ちラクトンを水酸化ナトリウムで開くことにより可溶性にでき
る(F.M.Muggia,et al.,Cancer Chemotherapy Reports,pt.1,56,No.4,515(1972)
参照)。しかしながら、M.C.Wani,et al.,J.Med.Chem.,23,554(1980)では、環E
のα‐ヒドロキシラクトン部分が抗腫瘍活性にとり絶対要件であることを報告し
ている。
水に対するその溶解度を改善するために作られたカンプトテシンの修正および
誘導例が知られている。これら誘導体の多くは白血病(L‐1210)モデルを
用いたインビトロおよび初期の動物研究で活性であったが、それらは移植固形腫
瘍を有する長期動物モデルで期待通りではなかった。
Miyasakaらの米国特許第4,399,282号は、特にヒドロキシメチルおよ
びアルコキシメチルにより7位で置換された一群のカンプトテシン誘導体につい
て開示している。更に、Miyasakaらの米国特許第4,399,276号は、カン
プトテシン‐7‐アルデヒドおよびある関連アルデヒド誘導体、例えばアセター
ル、オキシムおよびヒドラゾンについて開示している。更に最近では、Vishnuva
jjalaらの米国特許第4,943,579号は、Boehmらの欧州特許出願0 32
1 122 A2のように、A環上に置換基のある一連の水溶性カンプトテシン
誘導体を開示している。カンプトテシンの誘導体の他の例にはMiyasnkaらの米国
特許第4,473,692号および同4,545,880号並びにW.Kingsbury,
et al.,J.Med.Chem.,34,98(1991)がある。これらの参考文献はいずれも、カンプ
トテシン自体より強い抗腫瘍活性を有する化合物を報告していない。
Waniと共同研究者らは、10,11‐メチレンジオキシカンプトテシンが非置
換カンプトテシンよりも強力であることを報告した(M.C.Wani,et al.,J.Med.Ch
em.,29,2358(1986)および30,2317(1987)参照)。しかしながら、その水溶解度は
カンプトテシンと同様に乏しく、その臨床的有用性をかなり制限している。
我々は、今般、インビトロで良好なトポイソメラーゼI阻害活性を有し、かつ
インビボで顕著な抗腫瘍活性を有する、カンプトテシンの水溶性アナログを見出
した。
発明の概要
本発明の一の態様は、下記式(I)の水溶性カンプトテシンアナログ並びにそ
の薬学上許容される塩および溶媒和物である。

i)R1は水素、低級アルキル、(C3-7)シクロアルキル、(C3-7)シクロ
アルキル低級アルキル、低級アルケニル、ヒドロキシ低級アルキル、アミノ低級
アルキル、低級アルキルアミノ低級アルキル、アミノ低級アルキル、低級アルコ
キシ低級アルキル、または(CH2)tArを表し
(ここで、
tは0〜5であり、
Arは、フェニル、フリル、ピリジル、N‐メチルピロリル、イミダゾリルを
表し、またはヒドロキシ、メトキシ、ハロゲンおよびアミノから独立して選択さ
れる一以上の置換基を有するフェニル、フリル、ピリジル、N‐メチルピロリル
、イミダゾリルを表す)、
R2はジフェニルメチルまたは(CH2)tArを表すか、または
ii)R1およびR2は、それらが結合している窒素と一緒になって、N‐テトラ
ヒドロキノリルまたはN‐テトラヒドロイソキノリルを表す〕
薬学上許容される塩には、無機酸との塩、例えば塩酸、硫酸、リン酸、二リン
酸、臭化水素酸および硝酸塩、または有機酸との塩、例えば酢酸、リンゴ酸、マ
レイン酸、フマル酸、酒石酸、コハク酸、クエン酸、乳酸、メタンスルホン酸、
p‐トルエンスルホン酸、パルモ酸(palmoate)、サリチル酸およびステアリン
酸塩があるが、それらに限定されない。シュウ酸のような他の酸はそれら自体薬
学上許容されないが、本発明による化合物およびそれらの薬学上許容される塩を
得る上で中間体として有用である。
本発明のもう一つの態様は、トポイソメラーゼ阻害量の式(I)の化合物を患
者に投与することからなる、哺乳動物細胞でトポイソメラーゼI型を阻害する方
法、および有効抗腫瘍量の式(I)の化合物を腫瘍保有哺乳動物に投与すること
からなる、哺乳動物で腫瘍を治療する方法である。また、別の態様は活性成分と
して式(I)の化合物を含有した医薬処方剤に関する。式(I)の化合物と、こ
こで開示されたような合成に用いられる関連新規化学中間体の製造方法もまた、
本発明の範囲内にある。
発明の具体的説明 化合物
アルキルおよびアルコキシに関して本明細書において用いられる“低級”とい
う用語は1〜6の炭素、特に1〜3の炭素を意味し、アルケニルに関しては3〜
6の炭素を意味する(但し、二重結合は窒素に結合した炭素と結合していない)
。ここで用いられる“アリール”という用語は芳香環置換基、例えばフェニル、
ナフチル、フリル、ピリジル、N‐メチルピロリルまたはイミダゾリルを意味す
る。基“(CH2)t”には分岐が可能な分岐アルキレン鎖も含まれる。
本明細書において用いられる“N‐テトラヒドロキノリル”および“N‐テト
ラヒドロイソキノリル”という用語は下記のように定義される:
ラクトン環、即ち環Eは、式(I)の化合物の対応E開環形のアルカリ金属ま
たはアルカリ土類金属塩を形成するために、アルカリ金属またはアルカリ土類金
属塩基、例えば水酸化ナトリウムまたは水酸化カルシウムで開いてもよい。水に
対するその良好な溶解度のために、E開環形は慣用的な再結晶化技術で有利に精
製することができる。したがって、上記E開環形は、例えば酸、例えば塩酸との
処理により式(I)の化合物を形成して、精製形の式(I)の化合物を製造する
ために、中間体として使用できる。
前記のように、カンプトテシン部分は20位に不斉炭素原子を有し、2つのエ
ナンチオマー形、即ち“R”および“S”配置を可能にしている。本発明には双
方のエナンチオマー形およびこれら形の組合せが含まれる。簡便化のため、20
位で特定の配置が構造式中に示されていない場合には、双方のエナンチオマー形
とそれらの混合物が表されているとする。特に断らないかぎり、命名慣行“(R
,S)”はRおよびSエナンチオマーのラセミ(ほぼ等部分)混合物を表し、“
R”および“S”は本質的に光学上純粋なRおよびSエナンチオマーを各々表す
。本発明には式(I)の化合物の他の形、例えば溶媒和物、水和物、多形体等も
含まれる。
本発明による化合物の1つのサブグループは、式(I)において、
nが整数1または2を表し、
i)R1が水素、低級アルキル、(C3-7)シクロアルキル、(C3-7)シクロ
アルキル低級アルキル、低級アルケニル、ヒドロキシ低級アルキル、アミノ低級
アルキル、低級アルコキシ低級アルキル、または(CH2)tArを表し

tは0〜5であり、
Arは、フェニル、フリル、ピリジル、N‐メチルピロリル、イミダゾリルを

本発明による化合物のもう1つのサブグループは、式(I)において、

R2がジフェニルメチルまたは(CH2)tArを表し、

式(I)の化合物の一つの具体的なグループにおいて、Arはフェニル、フリ
ル、ピリジル、N‐メチルピロリル、イミダゾリルを表すか、またはヒドロキシ
、メトキシ、ハロゲンおよびアミノから独立して選択される一または二つの置換
基で置換されたフェニルを表す。典型的なAr基にはフェニルおよび置換フェニ
ル、2‐フリル、2‐ピリジル、4‐ピリジル、2‐N‐メチルピロリルと4‐
イミダゾリルがある。式(I)の化合物の具体的なサブグループにおいて、Ar
は置換フェニルまたは4‐ピリジルである。
化合物のもう一つのサブグループにおいて、R1は水素、低級アルキル、低級
アルキルアミノ低級アルキル、または‐(CH2)tArを表し、特に水素または
低級アルキルを表す。
式(I)の化合物のもう一つのサブグループにおいて、tは1、2または3、
特に1であり、nは1または2、特に2である。

一般化された方法(A)によれば、式(I)の化合物は、スキームIのSTE
P2で示された操作により製造される。
スキームIのSTEP1において、Xが脱離基(J.March,Advanced Organic C
hemistry,3rd.Ed.,page 179,John Wiley & Sons,New York(1985)で示されている
)、例えばクロロのようなハロゲンである式(II)の化合物は、引用することに
より本明細書の開示の一部とされる1990年1月16日付でWallらに発行され
た米国特許第4,894,456号(以下 456特許という)に示された方法
に従い式(III)の化合物と反応させられ、式(IV)の化合物を生じる。
STEP1の反応は、好ましくは酸または塩基触媒の存在下で行われる。酸触
媒は、好ましくは強鉱酸、例えば塩酸、硝酸、硫酸およびリン酸、あるいは強有
機酸、例えばC1-8アルカン酸およびC1-12アリールスルホン酸、特にp‐トル
エンスルホン酸である。塩基触媒は、好ましくは無機塩基、例えば炭酸ナトリウ
ムおよびカリウム、炭酸水素ナトリウムおよびカリウム、あるいは有機塩基、例
えば立体障害のある塩基、例えばトリエチルアミンおよびジイソプロピルアミン
である。
この反応は溶媒を用いず、あるいは極性または非極性溶媒の存在下で行っても
よい。好ましい極性溶媒はC1-6アルコール、C1-6エーテルおよびジメチルホル
ムアミドである。好ましい非極性溶媒は4〜10の炭素原子を有する分岐または
直鎖アルキル炭化水素と、6〜20の炭素原子を有する芳香族炭化水素、特にト
ルエンである。反応は通常加熱還流しながら行われる。
STEP2(方法A)において、式(IV)の化合物は、R1およびR2が式(I
)で定義された同義である式(V)の化合物による脱離基Xの置換により、式(
I)の化合物に変換される。この置換反応は、好ましくは、溶媒系、例えば水、
(C1-4)アルカノール、(C2-4)アルキレンジオール、1‐ヒドロキシ‐2‐
メトキシエタン、ジメチルアセトアミド(DMAC)、N‐メチルピロリジノン
、ジメチルホルムアミド(DMF)、テトラヒドロフラン(THF)、ジメチル
スルホキシド(DMSO)、トルエン、ジオキサンまたはこれら溶媒の組合せの
中で、過剰のアミン、即ち過剰の式(V)の化合物の存在下、塩基、例え
ば炭酸カリウムと共にまたはそれなしで、および/または触媒としてNaIと共
に実施される。
この方法は、R1が水素以外である式(I)の化合物を製造する上で、特に有
用である。
式(V)の化合物は市販され、化学文献で示されているか、あるいは有機化学
業界で知られる方法および物質を用いて有機化学業者により容易に製造される。
もう一つの一般化された方法(B)によれば、式(I)の化合物はスキームI
AのSTEP2aで示された操作により製造される。
STEP1aにおいて、式(II)の化合物は、R1およびR2が式(I)で定義
された同表である式(V)の化合物による脱離基X(経路Iの定義どおり)の置
換により、式(IIA)の化合物に変換される。この置換反応は、好ましくは、溶
媒系、例えば水、(C1-4)アルカノール、(C2-4)アルキレンジオール、1‐
ヒドロキシ‐2‐メトキシエタン、ジメチルアセトアミド(DMAC)、N‐メ
チルピロリジノン、ジメチルホルムアミド(DMF)、テトラヒドロフラン(T
HF)、ジメチルスルホキシド(DMSO)、トルエン、ジオキサンまたはこれ
ら溶媒の組合せ(混和しうる範囲内で)の中で、過剰のアミン、即ち過剰の式(
V)の化合物の存在下、塩基、例えば炭酸カリウムと共にまたはそれなしで、お
よび/または触媒としてNaIと共に実施される。
STEP2a(方法B)において、式(IIA)の化合物はスキームI、STE
P1で前記された場合と同様にして式(III)の化合物と反応させられ、式(I
)の化合物を生じる。
R1が水素である式(I)の化合物を製造する上で特に有用な、更に別の一般
化された方法(C)は、スキームIBのSTEP3bで示されている。
STEP1bにおいて、式(Va)の化合物(“Hal”はハロゲン、即ちフ
ルオロ、クロロ、ブロモまたはヨードである)、例えばトリフルオロアセトアミ
ドは極性非プロトン溶媒、例えばアセトニトリル中、極性非プロトン溶媒に可溶
性の塩基、例えば溶媒がアセトニトリルならば炭酸セシウムの存在下で式(II)
の化合物と反応させて、式(IIb)の化合物を生じる。
STEP2bにおいて、式(IIb)の化合物はスキームI、STEP1で示さ
れた場合と同様にして式(III)の化合物と反応させられ、式(IVb)の化合物
を生じる。
STEP3b(方法C)において、式(IVb)の化合物は鉱酸、例えば塩酸の
ような酸H+B-で処理されて、式(Ib)の化合物、即ち式(I)の化合物の塩
を生じる。式(Ib)の化合物は当業界の標準的な方法により水酸化または炭酸
アルカリ金属、例えば水酸化ナトリウムまたは炭酸カリウムのような塩基で処理
されて、対応する遊離塩基を生じる。例えば、式(Ib)の化合物は約5〜約1
00℃の温度範囲で約1〜約4時間にわたり炭酸カリウムの水溶液と共に攪拌さ
れる。次いで遊離塩基は必要であれば慣用的手段により薬学上許容される塩に変
換することができる。
式(II)および(III)の化合物は欧州特許公報EP0 540 099 A
1に記載された操作に従い製造される。
式(IIa)、(IIb)および(IVb)の新規中間化合物は本発明に含まれるも
のである。
本発明による式(I)の化合物は慣用的操作を用いて本発明の他の化合物に変
換してもよい。
このため、例えば、R1が水素原子を表す式(I)の化合物は慣用的技術を用
いてアルキル化される。反応は適切なアルキル化剤、例えばアルキルハライド、
アルキルトシレートまたはジアルキルサルフェートを用いて行われる。アルキル
化反応は、好ましくは、アミド、例えばジメチルホルムアミド、またはエーテル
、例えばテトラヒドロフランのような有機溶媒中、好ましくは塩基の存在下で行
われる。適切な塩基には、例えば水素化ナトリウムのような水素化アルカリ金属
、炭酸ナトリウムのような炭酸アルカリ金属、あるいはカリウムメトキシド、エ
トキシドまたはt‐ブトキシドがある。アルキル化反応は、好ましくは約25〜
約100℃の温度で行われる。
また、R1が水素原子を表す式(I)の化合物は還元アルキル化により式(I
)の他の化合物に変換してもよい。適切なアルデヒドまたはケトンによる還元ア
ルキル化は、水素化ホウ素または水素化シアノホウ素アルカリ土類金属を用いて
行われる。反応媒体は、好ましくは、場合により水の存在下における、アルコー
ル、例えばメタノールまたはエタノール、あるいはエーテル、例えばジオキサン
またはテトラヒドロフランである。反応は0〜100℃、好ましくは約5〜約5
0℃範囲の温度で行われることが好ましい。
また、R1が低級アルケニル基を表す式(I)の化合物は、R1が低級アルキル
基を表す式(I)の他の化合物に変換してもよい。還元は、好ましくは、水素と
、例えば炭の上に担持させてもよい、金属触媒、例えばラネーニッケル、あるい
は貴金属触媒、例えばパラジウム、白金、酸化白金またはロジウムの存在下で行
われる。反応はアルコール、例えばエタノールのような溶媒中、好ましくは約−
10〜約+50℃、好ましくは約20〜約30℃の温度で行われる。
本発明による式(I)の化合物またはその塩は、保護基を除去する反応に式(
I)の保護誘導体またはその塩を付すことにより製造される。
このため、式(I)の化合物またはその塩の製造における初期段階では、分子
中の1以上の感受性基を保護して望ましくない副反応を妨げることが必要および
/または望ましいであろう。
式(I)の化合物の製造に用いられる保護基は常法で用いられているものであ
ってよい。例えば、"Protective Groups in Organic Chemistry",Ed.J.F.W.McOm
ie(Plenum Press,1973)または"Protective Groups in Organic Synthesis",Theo
dora W.Greene(John Wiley and Sons,1981)参照。
慣用的なアミノ保護基には、例えばベンジル、ジフェニルメチルまたはトリフ
ェニルメチル基のようなアラルキル基、N‐ベンジルオキシカルボニルまたはt
‐ブトキシカルボニルのようなアシル基がある。このため、R1が水素を表す一
般式(I)の化合物は対応保護化合物の脱保護により製造してもよい。
ヒドロキシ基は、例えばベンジル、ジフェニルメチルまたはトリフェニルメチ
ル基のようなアラルキル基、アセチルのようなアシル基、、トリメチルシリルま
たはt‐ブチルジメチルシリル基のようなケイ素保護基により、あるいはテトラ
ヒドロピラン誘導体として保護される。
存在する保護基の除去は慣用的な操作により行われる。このため、ベンジルの
ようなアラルキル基は触媒(例えば、パラジウム炭)の存在下で水素付加分解に
より開裂され、N‐ベンジルオキシカルボニルのようなアシル基は例えば酢酸中
臭化水素での加水分解によるか、または還元、例えば接触還元により除去され、
ケイ素保護基は例えばフッ化物イオンでの処理または酸性条件下の加水分解によ
り除去され、テトラヒドロピラン基は酸性条件下で加水分解により開裂される。
明らかなように、前記いずれの方法においても、前記のように分子中でいかな

れかの後で実施される。
このため、本発明の別の態様によれば、下記反応:
(i)保護基の除去、および
(ii)式(I)の化合物またはその塩からその薬学上許容される塩への変換
が必要および/または所望であればいずれかの方法の後に適切な順序で実施され
る。
本発明の化合物を塩、例えば酸付加塩として単離することが望まれる場合には
、これは一般式(I)の遊離塩基を適切な溶媒(例えば、水性アルコール)中に
おいて、好ましくは当量で、適切な酸または硫酸クレアチニンで処理することに
より実施できる。
製造順序のうち最後の主要工程として用いられるだけでなく、本発明の化合物
の製造で前記された一般化された方法は、必要化合物の製造に際して中間段階で
望ましい基の導入に用いてもよい。したがって、このような多段階方法において
、反応の順序は反応条件が最終生成物で望まれる分子中に存在する基に影響を与
えないように選択されるべきであることは明らかであろう。
式(I)の化合物の生物活性はSエナンチオマーにあり、Rエナンチオマーは
ほとんどまたは全く活性を有しない。このため、式(I)の化合物のSエナンチ
オマーはラセミ混合物のようなRおよびSの混合物よりも通常好ましい。しかし
ながら、Rエナンチオマーが例えば他の化合物の対照試験または合成にとり望ま
れるのであれば、それは好ましくは’512特許の開示に従い製造された式(II
I)の化合物のRエナンチオマーを用いて前記操作により製造することができる
。

2000nM以上の濃度で観察しうる活性を示す化合物(表Aで“+”)は弱
〜中度の活性と考えられ、500nM以下の濃度で活性を示すもの(表Aで“+
+++”)は非常に活性である。“IC50”という用語は、DNA基質の50%
がトポイソメラーゼIにより捕捉された式(I)の化合物の濃度を意味する。
用途
このような活性に関して、式(I)の化合物は、口腔および咽頭(唇、舌、口
、咽頭)、食道、胃、小腸、大腸、直腸、肝臓および胆道、膵臓、喉頭、肺臓、
骨、結合組織、皮膚、結腸、***、子宮頸部、子宮体内膜、卵巣、前立腺、精巣
、膀胱、腎臓および他の尿組織、目、脳および中枢神経系、甲状腺および他の内
分泌腺、白血病(リンパ球、顆粒球、単球)、ホジキン病、非ホジキンリンパ腫
、多発性骨髄腫等の癌のような哺乳動物(ヒトを含む)腫瘍および癌性増殖の広
域スペクトルに対して活性である。ここで“腫瘍”、“癌”および“癌性増殖”
という用語は同義で用いられている。
抗腫瘍剤として有効であるために要求される式(I)の化合物の量は勿論治療
される個別の哺乳動物に応じて変わり、最終的には医療または獣医従事者の裁量
に委ねられる。考慮されるファクターには、治療される症状、投与経路、処方の
性質、哺乳動物の体重、表面積、年齢、一般的状態および投与される具体的な化
合物がある。しかしながら、適切な有効抗腫瘍用量は約0.1〜約200mg/kg
体重/日の範囲、好ましくは約1〜約100mg/kg/日の範囲である。全1日量
は1回分の用量、多数回分の用量、例えば2〜6回/日として、または選択され
た期間にわたり静脈内注入により与えられる。上記範囲より上または下の投与量
も本発明の範囲内であり、所望および必要であれば個別の患者に投与される。
例えば、75kg哺乳動物の場合、用量範囲は約75〜約7500mg/日であり
、典型的用量は約800mg/日である。別個の多数回分用量が指示されるならば
、治療では典型的には式(I)の化合物200mgが1日4回与えられる。処方
本発明の処方剤は、医療用の場合、活性化合物、即ち式(I)の化合物を、そ
のための許容されるキャリアおよび場合により他の治療活性成分と一緒に含んで
なる。キャリアは、処方の他の成分と適合してそのレシピエントにとり有害でな
いという意味で、薬学上許容されねばならない。
したがって、本発明は式(I)の化合物をそのための薬学上許容されるキャリ
アと一緒に含んでなる医薬処方剤を更に提供する。
処方剤には経口、直腸または非経口(皮下、筋肉内および静脈内を含む)投与
に適するものがある。経口または非経口投与に適したものが好ましい。
処方剤は好ましくは単位剤形で供与され、製薬業界で周知のいずれかの方法に
より製造される。すべての方法は活性化合物を1種以上の補助成分からなるキャ
リアと一緒にする工程を含む。一般的に、処方剤は活性化合物を液体キャリアま
たは微細固体キャリアと均一かつ完全に混合して、その後必要であれば製品を望
ましい単位剤形に成形することにより製造される。
経口投与に適した本発明の処方剤は、各々が既定量の活性化合物を含有したカ
プセル、カシェ剤、錠剤またはロゼンジのような別個の単位として;粉末または
顆粒として;あるいは水性液体または非水性液体中の懸濁液または溶液、例えば
シロップ、エリキシル、エマルジョンまたはドロート(draught)として供与さ
れる。
錠剤は、場合により1種以上の補助成分と共に圧縮または成形により作られる
。
圧縮錠剤は、場合により補助成分、例えば結合剤、滑沢剤、不活性希釈剤、界面
活性または分散剤と混合された活性化合物を易流動形、例えば粉末または顆粒と
して適切な機械の中で圧縮することにより製造される。成形錠剤は粉末活性化合
物といずれか適切なキャリアとの混合物を適切な機械の中で成形することにより
製造される。
シロップまたは懸濁液は糖、例えばスクロースの濃縮水溶液に活性化合物を加
えることにより製造され、それには補助成分も加えてよい。このような補助成分
には香味剤、糖の結晶化を遅らせる剤、または他の成分の溶解度を増加させる剤
、例えばグリセロールまたはソルビトールのような多価アルコールがある。
直腸または膣投与用の処方剤は、慣用的キャリア、例えばカカオ脂またはWite
psol S55(坐薬基剤に関するDynamite Nobel Chemical,Germanyの商標名)を含
有した坐薬として供与してもよい。
経皮投与の場合、本発明による化合物はクリーム、ゲル、軟膏、ローションま
たは経皮パッチとして処方される。このような組成物は、例えば適切な増粘剤、
ゲル化剤、乳化剤、安定剤、分散剤、懸濁剤および/または着色剤を添加して、
水性または油性基剤で処方される。
非経口投与に適した処方剤は、好ましくは受容者の血液と等張である活性化合
物の無菌水性製剤であることが好ましい。このような処方剤は、受容者の血液と
等張である、式(I)の化合物の薬学上および薬理学上許容される酸付加塩の溶
液または懸濁液であることが適切である。このため、このような処方剤は、蒸留
水、蒸留水または塩水中5%デキストロースと、これらの溶媒中で適度な溶解度
を有する式(I)の化合物の薬学上および薬理学上許容される酸付加塩、例えば
塩酸塩を含有していることが好ましい。有用な処方剤には式(I)の化合物を含

下記例は本発明を説明するものであるが、限定的に解釈されるべきではない。
これらの例で用いられる記号および慣例は現行化学文献、例えばthe journal of
the American Chemical Societyで用いられる場合と一致する。ここで用いられ
る“室温”という用語は約25℃を意味する。
一般操作
フラスコに(S)‐7‐クロロメチル‐10,11‐エチレンジオキシカンプ
トテシン(25〜150mg、0.06〜0.33mmol)、触媒ヨウ化ナトリウム
(1〜15mg、99.99%Aldrich)および無水1,4‐ジオキサン(2〜5
0ml)を入れる。アミンを加え(2〜5当量、溶媒なし)、攪拌スラリーを油浴
中75〜95℃で加熱した。反応は薄層クロマトグラフィーにより出発物質の消
失についてモニターした。混合液は、ロータリーエバポレーターで溶媒を除去し
て、残渣をエーテルで摩砕することにより後処理する。吸引濾過で集められた固
体物をシリカゲルクロマトグラフィーまたは逆相HPLC(Rainin Dynamax 60A
カラム、水中2%トリフルオロ酢酸(70〜80%)および4:1アセトニトリ
ル:THF(20〜30%)で溶出、254nmでモニター)により精製して、
各々遊離塩基またはトリフルオロ酢酸塩として生成物アミンを得る。
例1 7‐(N‐エチル‐N‐4‐ピリジルメチル)アミノメチレン‐10,11‐エ チレンジオキシ‐20(S)‐カンプトテシン(化合物1)
(A)6′‐アミノ‐3′,4′‐エチレンジオキシ‐2‐クロロアセトフェノ
ン
1リットル3首丸底フラスコに磁気攪拌棒、温度計、塩化カルシウム充填乾燥
管装備の還流コンデンサーおよび窒素導入口を供えた。反応容器に乾燥塩化メチ
レン(100ml)および1,4‐ベンゾジオキサン‐6‐アミン(15.12g
、100mmol)を入れた。反応容器を0℃に冷却し、その後内部温度を10℃以
下に維持しながら塩化メチレン中三塩化ホウ素の1M溶液400mlをゆっくり加
えた。塩化アルミニウム(13.34g、100mmol)を3回に分けて速やかに
加え、その後クロロアセトニトリル(7ml、110mmol)を加えた。反応液を0
℃で30分間攪拌し、その後40℃で16時間加熱した。反応液を熱源から除き
、室温まで冷却させ、その後氷1kg/1N HCl 500mlの混合液中にいれ
て反応停止させた。混合液を固体物が観察されなくなるまで攪拌した。塩化メチ
レン層を除去し、水層を塩化メチレンで2回抽出した。有機層を合わせ、塩水で
洗浄し、硫酸マグネシウムで乾燥し、濾過し、脱色炭で処理し、セライトパッド
で濾過し、固体残渣に濃縮した。固体物を酢酸エチル/ヘキサンから再結晶化さ
