Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.抗原を粘膜部位に送達して、分泌性および全身性免疫グロブリンの産生を刺 激する方法であって、 (a)抗原を溶媒希釈微小担体に組み込んで、溶媒希釈微小担体統合抗原を形 成し; (b)免疫グロブリン産生を刺激するのに充分な投与量で、該溶媒希釈微小担 体統合抗原を粘膜表面に接触させる ことを含む方法。 2.溶媒希釈微小担体統合抗原を投与のためにエアゾル化する請求項1記載の方 法。 3.該エアゾル化された溶媒希釈微小担体統合抗原を吸入させる請求項2記載の 方法。 4.該溶媒希釈微小担体統合抗原を経口的に投与する請求項1記載の方法。 5.該抗原が病原のサブユニットである請求項1記載の方法。 6.体内の粘膜部位に抗原を送達して、分泌性および全身性免疫グロブリン応答 を開始させるためのワクチン送達系であって、 溶媒希釈微小担体内で担送される抗原 を含む送達系。 7.該ワクチン送達系が投与のためにエアゾル化される請求項6記載の送達系。 8.該ワクチン送達系が経口的に投与される請求項6記 載の送達系。 9.予防接種投与のための病原サブユニットの免疫原複合体を調製する方法であ って、 (a)下記の段階: (i)両親媒性材料を、該両親媒性材料を可溶化するのに適した第一量の 非水性溶媒中で可溶化して、第一混合物を形成し; (ii)ある量の水を該第一混合物に、混濁懸濁液を形成するのに充分な量 で添加し;そして (iii)第二量の適切な非水性溶媒を該混濁懸濁液に、室温で光学的清澄 さを有し、室温で単相であることを特徴とする前駆溶液を含む第二混合物を形成 させるのに充分な量で添加する段階 によって溶媒希釈微小担体前駆溶液を調製し、そして (b)第三混合物を形成するために、水性媒体に希釈した病原のサブユニット を含む、小胞への該第三混合物の組織化を実施するのに適した所定量の抗原溶液 を該前駆溶液に添加する ことを含む方法。 10.該所定量の該抗原溶液を2またはそれ以上のアリコート中の該前駆溶液に 添加して、該サブユニットの段階的添加を実施する請求項9記載の方法。 11.該第三混合物を渦流に付す追加的段階を更に含む請求項9記載の方法。 12.該抗原溶液の各アリコートを加えた後に第三混合 物を渦流に付す追加的段階を更に含む請求項10記載の方法。 13.該抗原溶液の1アリコートあたり500μgの該病原サブユニットを添加 する請求項10記載の方法。 14.該前駆溶液5容あたり1容の該抗原溶液を加える請求項10記載の方法。 15.形成された小胞が約200nm〜約400nmの直径である請求項10記載の 方法。 16.活性成分を脂質小胞中に封入する方法であって、 活性成分を前駆溶液と混合して、装荷された前駆体混合物を形成する段階を含 み、 該前駆溶液は、下記の段階: (i)両親媒性材料を、該両親媒性材料を可溶化するのに適した第一量の 非水性溶媒中で可溶化して、第一混合物を形成し; (ii)ある量の水を該第一混合物に、混濁懸濁液を形成するのに充分な量 で添加し;そして (iii)第二量の適切な非水性溶媒を該混濁懸濁液に、室温で光学的清澄 さを有し、室温で単相であることを特徴とする前駆溶液を含む第二混合物を形成 させるのに充分な量で添加する段階 に従って作成されている方法。 17.該装荷された前駆体混合物を過剰な水に希釈して、該活性成分が装荷され た溶媒希釈微小担体という賦 形剤を形成する段階を更に含む請求項16記載の方法。 18.該装荷された前駆体混合物を空気と混合して、該活性成分が装荷された溶 媒希釈微小担体という賦形剤を形成する段階を更に含む請求項16記載の方法。 19.下記の段階: (a)両親媒性材料を、該両親媒性材料を可溶化するのに適した第一量の非水 性溶媒中で可溶化して第一混合物を形成し; (b)ある量の水を該第一混合物に、混濁懸濁液を形成するのに充分な量で添 加し;そして (c)第二量の適切な非水性溶媒を該混濁懸濁液に、室温で光学的清澄さを有 し、室温で単相であることを特徴とする前駆溶液を含む第二混合物を形成させる のに充分な量で添加する段階 に従って調製された前駆溶液。 20.該前駆溶液を活性成分と混合して溶媒希釈微小担体という賦形剤を形成す るための指示を与えるラベルと接着されるか、またはそれを随伴する、該前駆溶 液を保持する容器を更に含む請求項19記載の前駆溶液。
