調剤学的特性を有するニトロキシ化合物発明の利用分野
本発明は、医薬品の製造のために製薬工業において用されるニトロキシ化合物
に関する。発明の記載
本発明の対象は、式I(添付の式参照)の化合物であり、式中、それぞれの置
換基R1、R2、R3およびR4については、クマリン環において3、4、5、
6、7または8位の所望の炭素原子と結合することができ、その際、
R1は置換基:−Y−(CH2)m−A−(CH2)n−O−NO2を表し、
R2は水素、1−4C−アルキルまたは置換基:−Y−(CH2)m−A−(CH2
)n−O−NO2を表し、
R3は水素、1−4C−アルキル、1−4C−アルコキシ、1−4C−アルコキ
シ−1−4C−アルキル、1−4C−アルキルカルボニル、1−4C−アルキル
カルボニルオキシ、1−4C−アルキルカルボニル−1−4C−アルキル、カル
ボキシル、カルボキシ−1−4C−アルキル、1−4C−アルコキシカルボニル
、フェニル、フェニル−1−4C−アルキル、1−4C−フェニル−1−4C−
アルコキシ、ハロゲン、トリ
フルオロメチルまたはシアンを表し、
R4は水素、1−4C−アルキルを表し、
その際、置換基:−Y−(CH2)m−A−(CH2)n−O−NO2において、
YはO(酸素)、CO−O(カルボニルオキシ)またはCO−NH(カルボニル
アミノ)を表し、
mおよびnはそれぞれ0の数およびAは2−12C−アルキレン、または
mおよびnはそれぞれ1の数およびAはシクロヘキシレン、または
mおよびnはそれぞれ2の数およびAはO(酸素)またはO−CH2−CH2−O
(エチレンジオキシ)を表す。
1−4C−アルキルは、1から4個の炭素原子を有する直鎖または分枝鎖のアル
キル基を表す。たとえばブチル−、イソブチル−、s−ブチル−、t−ブチル−
、プロピル−、イソプロピル−、エチル−および特にメチル基が挙げられる。
1−4C−アルコキシ基は、酸素原子の他に前記の1−4C−アルキル基を有
している。メトキシ基が有利である。
1−4C−アルコキシ−1−4C−アルキルは、1−4C−アルコキシ基によ
り置換されている1−4C−アルキル基を表す。たとえばメトキシメチル−(C
H3OCH2−)およびメトキシエチル基(CH3OC
H2CH2−)が挙げられる。
1−4C−アルキルカルボニルは、前記した1−4C−アルキル基が結合して
いるカルボニル基を表す。たとえばアセチル基(CH3CO−)が挙げられる。
1−4C−アルキルカルボニルオキシ基は酸素原子の他に、前記した1−4C
−アルキルカルボニル基を有する。たとえばアセトキシ基(CH3CO−O−)
が挙げられる。
1−4C−アルキルカルボニル−1−4C−アルキルは1−4C−アルキルカ
ルボニル基により置換されている1−4C−アルキル基を表す。たとえば2−オ
キソプロピル−(CH3COCH2−)および3−オキソブチル基(CH3COC
H2CH2−)が挙げられる。
カルボキシ−1−4C−アルキルはカルボキシル基により置換されている1−
4C−アルキル基を表す。たとえばカルボキシメチル基(−CH2COOH)が
挙げられる。
1−4C−アルコキシカルボニルは前記した1−4C−アルコキシ基の内の一
つが結合しているカルボニル基を表す。たとえばメトキシカルボニル−(CH3
O−CO−)およびエトキシカルボニル基(CH3CH2O−CO−)が挙げられ
る。
本発明の範囲内でハロゲンは臭素、塩素およびフッ素である。
2−12C−アルキレンは、2〜12個の炭素原子
を有する直鎖または分枝鎖アルキレン基を表す。たとえば基:エチレン(−CH2
CH2−)、トリメチレン(−CH2CH2CH2−)、テトラメチレン(−CH2
CH2CH2CH2−)、ペンタメチレン(−CH2CH2CH2CH2CH2−)、ヘ
キサメチレン(−CH2−(CH2)4−CH2−)、オクタメチレン(−CH2−
(CH2)6−CH2−)、デカメチレン(−CH2−(CH2)3−CH2−)、ド
デカメチレン(−CH2−(CH2)10−CH2−)、1,2−ジメチルエチレン
[−CH(CH3)−CH(CH3)−]、1,1−ジメチルエチレン[−C(C
H3)2−CH2−]、イソプロピリデン[−C(CH3)2−]、2,2−ジメチ
ルプロピレン[−CH2−C(CH3)2−CH2−]、2−メチルプロピレン[−
CH2−CH(CH3)−CH2−]および2−メチルエチレン[−CH2−CH(
CH3)−]が挙げられる。
シクロヘキシレン基の内で、1,2−および特に1,4−シクロヘキシレン基
が挙げられる。シクロヘキシレン化合物において、置換基は相互にシス−または
トランス位置にあることができる。本発明は、シス位置およびトランス位置の化
合物ならびに全ての考えられるシス/トランス−混合物も包括している。
R1は置換基:−Y−(CH2)m−A−(CH2)n−O−NO2を表し、
R2は水素または1−4C−アルキルを表し、
R3は水素、1−4C−アルキル、1−4C−アルコキシ、1−4C−アルコキ
シ−1−4C−アルキル、ハロゲンまたはトリフルオロメチルを表し、およびR
4は水素または1−4C−アルキルを表し、その際、置換基:−Y−(CH2)m
−A−(CH2)n−O−NO2において
YはO(酸素)、CO−O(カルボニルオキシ)またはCO−NH(カルボニル
アミノ)を表し、
mおよびnはそれぞれ0の数およびAは2−12C−アルキレン、または
mおよびnはそれぞれ1の数およびAはシクロヘキシレン、または
mおよびnはそれぞれ2の数およびAはO(酸素)またはO−CH2−CH2−O
(エチレンジオキシ)を表す式Iの化合物が挙げられる。
特に、R1が[YがO(酸素)を表す場合]クマリン環の4位または7位に結
合している式Iの化合物が挙げられる。
さらに、特に、R1が[YがCO−O(カルボニルオキシ)またはCO−NH
(カルボニルアミノ)を表す場合]クマリン環の3位に結合している式Iの化合
物が挙げられる。

本発明による化合物の製造は、公知の方法により、たとえば:
a) R1および場合によりR2は置換基:−Y−(CH2)m−A−(CH2)n
−OHを表す式Iの化合物をニトロ化する、または
b) YがCO−O(カルボニルオキシ)を表す式Iの化合物の製造のために、
R1および場合によりR2は置換基:−CO−Zを表し、ZはOH(ヒドロキシ
)または適当な脱離可能基を表す式Iの化合物を、式:HO−(CH2)m−A−
(CH2)n−O−NO2の化合物と反応させる、または
c) YはCO−NH(カルボニルアミノ)を表す式Iの化合物の製造のために
、R1および場合によりR2は置換基:−CO−Zを表し、ZはOH(ヒドロキ
シ)または適当な脱離可能基を表す式Iの化合物を、式:H2N−(CH2)m−
A−(CH2)n−O−NO2と反応させる、または
d) R3がハロゲンを表す式Iの化合物の製造のために、R3が水素を表す式
Iの化合物をハロゲン化することにより行われる。
方法a)によるニトロ化は、公知の方法で、硝酸を用いて不活性溶剤(たとえ
ば酢酸エチルまたは酢酸)
中でまたは溶剤なしで、場合により水結合性または脱水性媒体(たとえば酢酸無
水物)の存在で、たとえば−20℃〜+50℃の温度で、有利に室温で行われる
。
方法b)またはc)による反応は、同様に当業者にその専門知識に基づきエス
テル化反応に関して公知であるような公知の方法により行われる。このエステル
化は不活性溶剤、たとえばジオキサンまたはテトラヒドロフラン中で、基:Zの
種類に応じて、脱水性もしくは水を化学的に結合する薬品、たとえばジシクロヘ
キシルカルボジイミド(Z=OHの場合)の存在で、または4−トルエンスルホ
ン酸の存在で、または補助塩基(たとえばトリエチルアミン)の存在で(Zが脱
離基、たとえばハロゲン原子、特に塩素を表す場合)行われる。カルボン酸(Z
=OH)から出発するエステル化の場合、たとえば、M.Bodanszky und A.Boda
nszky:”The Practice of Peptide Synthesis”(Springer-Verlag,Berlin,1
984,S.37)に記載されたように酸性の反応条件下で反応が行われる。
方法d)によるハロゲン化(これは有利に塩素化または臭素化として行われる
)は同様に公知の方法で、不活性溶剤、たとえばジクロロメタン中で行われる。
R1および場合によりR2が置換基:−Y−(CH2)m−A−(CH2)n−O
Hを表す式Iの化合物は、公知であり、これは当業者に公知の標準反応(たとえ
ば後に記載する実施例中に詳細に説明してあるように)
製造することができる。
次の例につき、本発明による化合物の製造を実験的に詳説する。名称により挙
げられた化合物およびその適用は、本発明の有利な対象である。実施例
出発化合物
A1. 4−(2−ヒドロキシエトキシ)クマリン
4−ヒドロキシクマリン10g、炭酸エチレン5.4gおよびテトラエチルア
ンモニウムブロミド4.3gからなる混合物を160℃で5時間加熱した。冷却
後に、この反応混合物を水および酢酸エチル中に溶かし、水相を酢酸エチルで抽
出した。有機相を硫酸マグネシウムで乾燥させ、濃縮させた。残留物を酢酸エチ
ルから再結晶させると、融点137.9〜141.4℃の目的化合物7.4g(
60%)が得られた。
A2. 7−(2−ヒドロキシエトキシ)−4−メチルクマリン
例A1と同様に、7−ヒドロキシ−4−メチルクマリンおよび炭酸エチレンか
ら、融点141.3〜145.2℃の目的化合物が得られた。
A3. 7−(2−ヒドロキシエトキシ)クマリン
例A1と同様に、7−ヒドロキシクマリンおよび炭酸エチレンから、融点86
.0〜89.2℃の目的化合物が得られた。
A4. 7−(2−ヒドロキシエトキシ)−4,4, 8−トリメチルクマリン
例A1と同様に、7−ヒドロキシ−3,4,8−トリメチルクマリンおよび炭
酸エチレンから、融点128.1〜131.8℃の目的化合物が得られた。
A5. 7−(2−ヒドロキシエトキシ)−4−トリフルオロメチルクマリン
例A1と同様に、7−ヒドロキシ−4−トリフルオロメチルクマリンおよび炭
酸エチレンから目的化合物が得られた。
A6. 7−(2−ヒドロキシエトキシ)−4−メトキシメチルクマリン
例A1と同様に、7−ヒドロキシ−4−メトキシメチルクマリンおよび炭酸エ
チレンから目的化合物が得られた。
A7. 4−(3−ヒドロキシ−2,2−ジメチルプポキシ)クマリン
4−ヒドロキシクマリン20.0g、3−ブロモ−2,2−ジメチルプロパノ
ール20.6gおよび水酸化カリウム7.0gをジメチルスルホキシド250m
1中に溶かした。この溶液を130℃で8時間加熱した。冷却後にこの反応混合
物を水中に注ぎ、酢酸エチルで抽出した。有機相を硫酸マグネシウムで乾燥させ
、濃縮させた。残留物をクロマトグラフィーにより精製した。
A8. 4−(3−ヒドロキシプロポキシ)クマリン
4−ヒドロキシクマリン16.2g、水酸化カリウム7gおよび3−クロロ−
1−プロパノール9.5gをジメチルホルムアミド中に溶かし、130℃で撹拌
した。反応の終了後(DC制御)に濃縮し、残留物を酢酸エチルに収容した。ク
ロマトグラフィーにより精製した後に、融点84〜90℃の目的化合物が得られ
た。
A9. 4−{2−[2−(2−ヒドロキシエトキシ)エトキシ]−エトキシ} クマリン
例A8と同様に、4−ヒドロキシクマリンおよび2−[2−(2−クロロエト
キシ)エトキシ]エタノールから、融点54〜57℃の目的化合物が得られた。
A10. 4−(6−ヒドロキシエチルオキシ)クマリン
例A8と同様に、4−ヒドロキシクマリンおよび6−ブロモ−1−ヘキサノー
ルから、80℃の反応温度で、水中に残留物を溶解させ、酢酸エチルで抽出し、
クロマトグラフィーにかけることにより目的化合物が得られた。
A11. 7−(3−ヒドロキシプロポキシ)−4−メチルクマリン
例A8と同様に、7−ヒドロキシ−4−メチルクマリンおよび3−クロロ−1
−プロパノールから、融点95〜96.5℃の目的化合物が得られた。
A12. 7−[2−(2−ヒドロキシエトキシ)エ トキシ]−4−メチルクマリン
例A8と同様に、7−ヒドロキシ−4−メチルクマリンおよび2−(2−クロ
ロエトキシ)エタノールから、融点91〜92℃の目的化合物が得られた。
A13. 7−[4−(ヒドロキシメチル)[トランス]シクロヘキシルメトキ シ]−4−メチルクマリン
例8Aと同様に、7−ヒドロキシ−4−メチルクマリンおよび4−ブロモメチ
ル[トランス]シクロヘキシルメタノールから、目的化合物が得られた。
A14. 4−[4−(ヒドロキシメチル)[トランス]シクロヘキシルメトキ シ]クマリン
水素化ナトリウム分散液(60%)2.5gをジメチルホルムアミド中の4−
ヒドロキシクマリン10gの溶液に注ぎ、室温で30分間撹拌した。4−ブロモ
メチル[トランス]シクロヘキシルメタノール12.5gを添加した後、この反
応混合物を120℃で2.5時間加熱した。冷却し、濃縮した後、残留物を酢酸
エチル中に溶かした。水で洗浄し、硫酸マグネシウムで乾燥させた後、有機相を
濃縮した。残留物を酢酸エチル/ジエチルエーテルから再結晶させた。最終生成物
1. 4−(2−ニトロキシエトキシ)クマリン
硝酸1.6mlおよび無水酢酸5.0mlからなる混合物を、酢酸/酢酸エチ
ル中の4−(2−ヒドロキシエトキシ)クマリン5.5gの溶液に添加した。数
時間後に、酢酸エチルおよび水を添加した。水相を炭酸ナトリウム溶液でアルカ
リ性にし、酢酸エチルで抽出した。有機相を硫酸マグネシウムで乾燥させ、濃縮
させた。エタノールから再結晶させた後に、融点119.2〜120.7℃の目
的化合物が得られた。
2. 4−メチル−7−(2−ニトロキシエトキシ)クマリン
7−(2−ヒドロキシエトキシ)−4−メチルクマリンのニトロ化により例1
と同様に、融点97.7〜98.3℃の目的化合物が得られた。
3. 7−(2−ニトロキシエトキシ)クマリン
7−(2−ヒドロキシエトキシ)クマリンのニトロ化により、例1と同様に、
融点111.2〜112.3℃の目的化合物が得られた[酢酸エチル/石油エー
テル(60〜80)から]。
4. 7−(2−ニトロキシエトキシ)−3,4,8−トリメチルクマリン
7−(2−ヒドロキシエトキシ)−3,4,8−トリメチルクマリンのニトロ
化により、例1と同様に、融点160.9〜161.8℃の目的化合物が得られ
た(メタノールから)。
5. 7−(2−ニトロキシエトキシ)−4−トリフルオロメチルクマリン
7−(2−ヒドロキシエトキシ)−4−トリフルオロメチルクマリンのニトロ
化により、例1と同様に、
融点90.0〜91.1℃の目的化合物が得られた。
6. 4−メトキシメチル−7−(2−ニトロキシエトキシ)クマリン
7−(2−ヒドロキシエトキシ)−4−メトキシメチルクマリンのニトロ化に
より、例1と同様に、融点110.7〜112.3℃の目的化合物が得られた。
7. 4−(2,2−ジメチル−3−ニトロオキシプロポキシ)クマリン
4−(3−ヒドロキシ−2,2−ジメチルプロポキシ)クマリンのニトロ化に
より、例1と同様に、融点117.8〜118.3℃の目的化合物が得られた。
8. 4−(3−ニトロキシプロポキシ)クマリン
4−(3−ヒドロキシプロポキシ)クマリンのニトロ化により、例1と同様に
、融点96〜98℃の化合物が得られた(エタノールから)。
9. 4−{2−[2−(2−ニトロキシエトキシ)エトキシ]エトキシ}クマ リン
4−{2−[2−(2−ヒドロキシエトキシ)エトキシ]エトキシ}クマリン
のニトロ化により、例1と同様に、融点55.3〜57.1℃の目的化合物が得
られた(エタノールから)。
10. 4−(6−ニトロキシヘキシルオキシ)クマリン
4−(6−ヒドロキシヘキシルオキシ)クマリンのニトロ化により、例1と同
様に、融点94.5〜97.
6℃の目的化合物が得られた(エタノールから)。
11. 4−メチル−7−(3−ニトロキシプロポキシ)クマリン
7−(3−ヒドロキシプロポキシ)−4−メチルクマリンのニトロ化により、
例1と同様に、融点80〜83℃の目的化合物が得られた(エタノールから)。
12. 4−メチル−7−[2−(2−ニトロキシエトキシ)エトキシ]クマリ ン
7−[2−(2−ヒドロキシエトキシ)エトキシ−4−メチルクマリンのニト
ロ化により、例1と同様に、融点63〜64℃の目的化合物が得られた。
13. 4−メチル−7−[4−(ニトロキシメチル)[トランス]シクロヘキ シルメトキシ]クマリン
7−[4−(ヒドロキシメチル)[トランス]シクロヘキシルメトキシ]−4
−メチルクマリンのニトロ化により、例1と同様に、融点116〜120℃の目
的化合物が得られた。
14. 4−[4−(ニトロキシメチル)[トランス]シクロヘキシルメチルオ キシ]クマリン
4−[4−(ヒドロキシメチル)[トランス]シクロヘキシルメチルオキシ]
クマリンのニトロ化により、例1と同様に、融点110〜112℃の目的化合物
が得られた(メタノールから)。
15. 3−ブロモ−4−メチル−7−(2−ニトロキシエトキシ)クマリン
臭素3.0gのジクロロメタン中の溶液を、4−メチル−7−(2−ニトロキ
シエトキシ)クマリン5.0gのジクロロメタン中の溶液に、撹拌しながらゆっ
くりと滴加した。さらに1時間撹拌した後、沈殿物を濾別し、メタノール/アセ
