Abstract
Description
Claims (22)
- 1.人工マトリックスを用いて制御された細胞の移植のための人工マトリックス を形成する方法であって、以下の工程a.多孔性で非毒性のマトリックスを患者 に移植する工程であって、該マトリックスが、体内で耐圧縮性または耐引っ張り 性を有し、細胞をマトリックス内に導入するのに適しており、そして血管新生さ れ、生存可能な結合組織で浸潤された10%と90%の間の部位を生成するため に血管組織および結合組織の内部成長に適している、工程およびb.該マトリツ クスの10%と90%の間のものが血管新生されるまで待ち、次いで生存可能な 細胞を血管新生されたマトリックスに導入する工程、 を包含する方法。
- 2.前記マトリックスの孔サイズが、約100ミクロンと300ミクロンの間で あり、血管の内部成長を可能にし、そして細胞または患者に損傷を与えることな くマトリックス内に細胞を注入することを可能にする、請求項1に記載の方法。
- 3.前記部位が、腸間膜、皮下組織、筋膜下および腹膜上から成る群から選択さ れる、請求項1に記載の方法。
- 4.患者に門脈下大静脈吻合を行う工程をさらに包含する、請求項1の方法。
- 5.マトリックスの原料として、ポリ無水物、ポリオルトエステル、ポリグリコ ール酸、ポリ乳酸、コポリマーおよびその混合物およびコラーゲンから成る群か ら、マトリックスを形成するための生体分解性ポリマーを選択する工程をさらに 包含する、請求項1に記載の方法。
- 6.栄養物、補因子、成長因子、脈管形成を刺激する化合物、免疫モジュレータ ー、炎症の阻害剤、退行因子、細胞の分化を刺激する因子、リンパ管網の内部成 長を刺激する生物学的活性分子、神経成長を増大する因子および薬物から成る群 から選択された化合物をマトリックス内に与える工程をさらに包含する、請求項 1に記載の方法。
- 7.前記マトリックスが、移植時にマトリックス内に細胞を導入する手段を含む 、請求項1に記載の方法。
- 8.前記マトリックスが、細胞を導入するための分布チャンネルを含む、請求項 7に記載の方法。
- 9.前記マトリックスが、細胞を導入するためのカテーテルを含む、請求項7に 記載の方法。
- 10.前記細胞が、肝細胞である、請求項1に記載の方法。
- 11.前記細胞を、副甲状腺細胞、甲状腺細胞、副腎−視床下部−下垂体軸の細 胞、神経細胞、骨および軟骨を形成する細胞、および平滑筋および骨格筋を形成 する細胞から成る群から選択する工程をさらに包含する、請求項1に記載の方法 。
- 12.多孔性で弾力性のある非毒性のマトリックスであって、該マトリックスが 、患者に移植した後のマトリックスに細胞を導入するための手段との組合せにお いて、体内で耐圧縮性または耐引っ張り性を有し、マトリックスヘの細胞の導入 に適しており、そして高度に血管新生された部位を生成するための血管組織およ び結合組織の内部成長に適しており、患者への移植後、マトリックス内に細胞を 導入する手段と組み合わせる、マトリックス。
- 13.栄養物、成長因子、補因子、脈管形成を刺激する化合物、免疫モジユレー ター、炎症の阻害剤、退行因子、分化を刺激する因子、リンパ管網の内部成長を 刺激する生物学的活性分子、神経成長を増大する因子、薬物、およびそれらの組 合せから成る群から選択された化合物をさらに含有する、請求項12に記載のマ トリックス。
- 14.前記マトリックスが、異なる起源の細胞に対して付着する別々の区域を与 えるために形造られる、請求項12に記載のマトリックス。
- 15.前記導入する手段が、マトリックス内に形成されるチャンネルである、請 求項12に記載のマトリックス。
- 16.前記導入する手段が、カテーテルである、請求項12に記載のマトリック ス。
- 17.前記マトリックスの孔サイズが、約100ミクロンと300ミクロンの間 であり、血管の内部成長を可能にし、そして細胞または患者に損傷を与えること なくマトリックス内に細胞を尊人することを可能にする、請求項12に記載のマ トリックス。
- 18.前記マトリックスが、該マトリックスの材料として、ポリ無水物、ポリオ ルトエステル、ポリグリコール酸、ポリ乳酸、コポリマーおよびその混合物およ びコラーゲンから成る群から選択された、生体分解性ポリマーから形成される、 請求項12に記載のマトリックス。
- 19.前記マトリックスが、エチレンビニルアセテート、ポリビニルアルコール の誘導体、テフロン、ナイロンおよびシリコンから成る群から選択された非分解 性ポリマーから形成される、請求項12に記載のマトリックス。
- 20.胆管細胞、副甲状腺細胞、甲状腺細胞、副腎−視床下部−下垂体軸の細胞 、心筋細胞、腎上皮細胞、腸管細胞、腎基底膜細胞、神経細胞、血管細胞、腸管 細胞、骨および軟骨を形成する細胞、平滑筋および骨格筋を形成する細胞から成 る群から選択された細胞をさらに含有する、請求項12に記載のマトリックス。
- 21.分離された肝細胞をさらに含有する、請求項12に記載のマトリックス。
- 22.ポリビニルアルコールの誘導体から形成される、請求項12に記載のマト リックス。
