Abstract
Description
Claims (22)
- 1.成熟DF3抗原に比して、非グリコシレートDF3抗原へ優先的に結合する 単クローン抗体であって、該抗体は、T7R8P9A10P11G12S13( SEQIDNO:3)のアミノ酸配列に従ったエピトーブに対して特定のもので あり、該エピトーブは位置11にプロリンを含むことを特徴とする単クローン抗 体。
- 2.請求項1に記載の抗体において、前記エピトーブは、R8P9A10P11 G12(SEQ ID NO:5)のアミノ酸配列中に存在することを特徴とす る単クローン抗体。
- 3.請求項1に記載の抗体において、前記抗体は、それが成熟DF3抗原に対し て有する親和性の少なくとも10倍の親和性をもって非グリコシレートDF3抗 原に結合することを特徴とする単クローン抗体。
- 4.請求項1に記載の抗体において、前記抗体は、D6R8P9A10P11G 12S13(SEQ ID NO:4)のアミノ酸配列を有するペプチドに結合 することを特徴とする単クローン抗体。
- 5.請求項1に記載の抗体において、前記抗体は、ヒトガン細胞の細胞質の成分 と反応することを特徴とする単クローン抗体。
- 6.請求項5に記載の抗体において、前記抗体は、前記細胞のフォルマリン処理 後であっても、前記成分と反応することを特徴とする単クローン抗体。
- 7.請求項1に記載の抗体において、前記抗体は、DF3−P単クローン抗体が 結合する同じエピトーブへ結合することを特徴とする単クローン抗体。
- 8.請求項1に記載の抗体において、前記抗体は、ハイブリドマDF3−Pによ って生成されることを特徴とする単クローン抗体。
- 9.請求項1に記載の抗体において、前記抗体は、DF3−P単クローン抗体が 前記非グリコシレートエピトープに結合するときと同じまたはそれよりも高い親 和性をもって前記エピトーブと結合することを特徴とする単クローン抗体。
- 10.請求項1に記載の抗体において、前記抗体は、浸透性管内癌細胞を含む胸 細胞部には選択的に結合するが、すべてが正常細胞からなる胸組織部には結合し ないことを特徴とする単クローン抗体。
- 11.生物学的サンプル中の未成熟DF3抗原を検出する方法において、前記生 物学的サンプルを、請求項1に記載の抗体を含む試薬のアリコートに接触させる ステップと; 前記抗体と前記生物学的サンプルの構成成分との間に免疫複合体の形成を検出す るステップであって、前記免疫複合体の形成は前記生物学的サンプル中に未成熟 DF3抗原が存在することを示すステップと、を含むことを特徴とする生物学的 サンプル中におけるDF3抗原検出方法。
- 12.請求項11に記載の方法において、前記免疫複合体の形成は、ELISA によって検出されることを特徴とする生物学的サンプル中におけるDF3抗原検 出方法。
- 13.請求項11に記載の方法において、前記生物学的サンプルは、ヒト血液、 血清、または尿であることを特徴とする生物学的サンプル中におけるDF3抗原 検出方法。
- 14.請求項11に記載の方法において、前記抗体は、ハイブリドーマDF3− Pによって生成されることを特徴とする生物学的サンプル中におけるDF3抗原 検出方法。
- 15.請求項11に記載の方法において、さらに、非グリコシレートDF3抗原 の標準料を含む制御サンプルまたはそのフラグメントを供給するステップと;前 記制御サンプルを前記試薬の第2のアリコートと接触させるステップと;前記生 物学的サンプル中における免疫複合体形成の量を前記制御サンプル中における免 疫複合体形成量と比較するステップと;を含むことを特徴とする生物学的サンプ ル中におけるDF3抗原検出方法。
- 16.請求項1に記載の抗体を含む免疫毒素、または細胞毒素媒体に結合された その抗原結合フラグメント。
- 17.請求項16に記載の免疫毒素において、前記細胞毒素媒体は、前記抗体ま たは前記抗原結合フラグメントに化学的に共役されていることを特徴とする免疫 毒素。
- 18.請求項16に記載の免疫毒素において、前記細胞毒素媒体は、ペプチド結 合によって前記抗原結合フラグメントに結合されたポリペプチドであって、前記 免疫毒素は、遺伝子工学的に開発されたハイブリッドDNA分子の発現によって 生成されることを特徴とする免疫毒素。
- 19.検出可能なラベルに結合された、請求項1の抗体を含む撮像媒体またはそ の抗原結合フラグメント。
- 20.請求項19に記載の撮像媒体において、前記ラベルは放射性核種であるこ とを特徴とする撮像媒体またはそのフラグメント。
- 21.請求項1に記載の単クローン抗体を含む免疫アッセイキット及び該キット を使用するための指示書。
- 22.生理学的に受容可能なキャリア内におけるアミノ酸配列RPAPGから成 る分子を含むワクチン。
