【発明の詳細な説明】
腫瘍性細胞増殖の治療用のサイトカイン受容体を標的とした分子発明の背景
本発明は、腫瘍性細胞の増殖、例えば、癌の治療に関する。
腫瘍性細胞を標的とした薬物を用いての腫瘍性細胞増殖を制限しようとする試み
はこれまでに多くあった。モノクローナル抗体および受容体リガンドは共に標的
化剤として示唆されてきた。
マーフィー (Murpt+y) (米国特許第4675382号)は、サイト
カイン/インターロイキン−2ハイブリツド蛋白が、免疫系に関与する癌の治療
で用いられ得ることを示唆している。
レメストル(LeMBistre)ら(ランセット(Lancet)337 :
1124.1991)は、ジフテリアトキシン/インターロイキン−2ハイブ
リッド分子を用いCの慢性リンパ球白血病を持つ患者の治療を記載している。
発明の概要
一般に、本発明は、患者において腫瘍性細胞増殖を治療する方法をその要旨とし
、ここに、該腫瘍性細胞は非リンパ球かつ非単球起源である。該方法は、腫瘍性
細胞で発現されたサイトカイン受容体に特異的に結合できる分子を患者に投与す
ることを含み、該分子は腫瘍性細胞の活性を減少させることができる。該サイト
カイン受容体はリンパ球または単球起源の細胞で通常発現されている受容体であ
る。
種々の好ましい具体例において、該腫瘍性細胞は造血幹細胞由来ではな(:該腫
瘍性細胞は肉腫であり:該腫瘍性細胞は骨肉腫であり、該腫瘍性細胞は繊維肉腫
であり:該腫瘍性細胞は平滑筋肉腫であり、該腫瘍性細胞はカルシノーマであり
:および該腫瘍性細胞は横絞筋肉腫である。より好ましい具体例において、該カ
ルシノーマは肺カルシノーマであり、および該カルシノーマは肝細胞カルシノー
マである。
好ましい具体例において、該サイトカイン受容体はインターロイキン受容体であ
る。より好ましい具体例において、該インターロイキン受容体はインターロイキ
ン−2受容体であり:該インターロイキン受容体はインターロイキン−4受容体
であり、および該インターロイキン受容体はインターロイキン−6受容体である

和性インターロイキン−2受容体である。もう1つの好ましい具体例において、

の部分はサイトカイン受容体に特異的に結合できる分子を含む。より好ましい具
体例において、該第2の部分はサイトカイン受容体に特異的な抗体のすべてまた
はその結合部位を包含し、該第2の部分はサイトカイン受容体についてのりガン
トのすべてまたはその結合部位を包含し、および該第1の部分は細胞毒を含み。
および該抗体は補体活性化抗体である。さらに、より好ましい具体例において、
該サイトカイン受容体はインターロイキン受容体である。なおさらにより好まし
い具体例において、該リガントはインターロイキンである。
もう1つの好ましい具体例において、該細胞毒は、酵素的に活性であるが、−膜
化された真核細胞受容体結合活性を有しないペプチドトキンンの断片である。
より好ましい具体例において、ベブチドトキンンの該断片はジフテリアトキシン
のフラグメントAおよびサイドシルへのトロンスローケーンランを容易にするた
めの十分なジフテリアトキシンのフラグメントBを含む。
他の好ましい具体例において、該分子はDABsa。IL−2であり:該分子は
DA83sslL 4てあり、該分子はDABsselL 6であり:該分子は
DAB+5g1L 2てあり、該分子はDA B4@611− 4であり;およ
び該分子はDA84sslL 6である。
「非リンパ球および非単球起源」とは、リンパ系幹細胞または単球からの子孫で
はないべての細胞、例えば、Tリンパ球、8928球、およびマクロファージ以
外の細胞を意味する。「リンパ球および単球起源」とは、リンパ系幹細胞または
単球の子孫の細胞、例えば、Tリンパ球、8928球、形質細胞、およびマクロ
ファージを意味する。「特異的に結合する」とは、当該分子が他の細胞表面受容
体に実質的に結合しないことを意味する。「造血幹細胞起源でない」とは、すべ
ての細胞は多能性幹細胞からの子孫でないすべての細胞を意味する。
本発明の他の特徴および利点はその好ましい具体例の以下の記載、ならびに請求
の範囲より明らかであろう。
本発明者らによるin vitro毒性実験は、ある種の癌細胞はインターロイ
キン−4またはインターロイキン−2受容体を標的とする細胞毒に対して感受性
である。
従って、これらの後記にて詳細に記載するインターロイキン受容体標的化細胞毒
は癌細胞の活性を減少させる手段を提供する。
後記する実験において、種々の癌細胞系は、ジフテリアトキシンの受容体結合ド
メイン(細胞内に存在する場合、その単一の分子は蛋白合成をブロックできる)
がヒト・インターロイキン−4の一部で置き換えられている融合蛋白(DAB3
.。
TL−4)に感受性であることが示される。
種々の細胞系をDAB3891 L 4に暴露し、蛋白合成のレベルを後記のご
とくに測定した。この分析の結果を表1に示すが、そこでは、IC6゜は蛋白合
成で50%の減少を引き起こすのに要する薬剤濃度と定義されている。横絞筋、
平滑筋、肝臓、骨、および肺からの癌細胞系はDA83gelL 4に感受性で
あり;これらの細胞系についてのICUは10 10Mないし10′□8Mであ
ることが判明した。

の感受性は、多数のインターロイキン−4受容体を発現することが期待されるT
HLV−1形質転換T−リンパ球および植物性赤血球凝集素活性化T−リンパ球
のごとき細胞の感受性に匹敵する(表2)。対照的に、ヒト・インターロイキレ
−4に認識されないインターロイキン−4受容体を発現する活性化ラント・T−
リンパ球は、ジフテリアトキシンに感受性であるに拘わらず、D/’1131@
911− 4に対してほとんど感受性でない(表2)。この結果は、DABss
elL 4はインターロイキン−4受容体発現細胞を選択的に中毒化することを
示す。

同様の一連の実験は、ヒト・横絞筋肉腫細胞系A673 (ATCC受託番号C
RL1598)は、ジフテリアトキシンの受容体結合ドメインがヒト・インター
ロイキン−2の一部で置き換えられた融合蛋白(DAB4861 L 2)に感
受性であることを示した。7.6X10−’MにおけるDAB41161L 2
は蛋白合成を50%減少させた。
インターロイキン受容体標的化細胞毒に対する腫瘍性細胞の感受性についてのア
ンセイ
蛋白合成を測定することによって細胞毒性を評価した。DA Bsge I L
4(またはDABasa I L 2)の存在下および不存在下における[1
4C]ロイシンの取込は以下の通りである。細胞を細胞タイプに適した培養培地
中にて96ウエルマイクロタイタープレートにて平板培養した。DABssjL
4 (または1)A B41g I L 2)を種々の濃度で添加し、[14
C]ロイシンでのパルス標識化に先立って20時間、該培養をインキュベートし
た。次いで、細胞をトリプシン処理し、ガラス繊維製のフィルターマットに収穫
し、計数した。IC,。は[+4(:]ロインンの取込の50%減少を導く細胞
毒の濃度である。
轡者から得た組織試料(例えば、バイオプシー)から単離した細胞を含めた他の
細胞タイプの感受性は前記したごとくに評価できる。また、活性は、調べようと
する細胞タイプに適したいずれの標準的な活性アッセイを用いても測定できる。

本発明の方法で有用な分子
本発明の方法で有用な分子はガイカイン受容体に標的化される。一般に、本発明
で有用な分子を実施し得る3つの方法がある= (1)該分子は細胞毒性ドメイ
ンを有するゆえ細胞を殺すことができる。(2)該分子(抗体)は補体固定を誘
導することによって細胞溶解を引き起こすことができる:および(3)該分子は
受容体のりガントの結合または摂取をブロックすることができる。すべての3つ
の場合において、該分子は受容体担持細胞に対して標的化されていなければなら
ない、これは、受容体のりガント(またはその誘導体の部分)あるいは当該分子
の一部としての抗−受容体抗体を含ませることによって達成される。
インターロイキン−2(IL−2)受容体標的化分子はこれらの3つのアプロー
チの各側を提供する。II、−2のIL−2受容体結合部分および細胞毒を包含
する融合分子はインターロイキン−2受容体を担持する腫瘍性細胞を殺すのに使
用できる。同様に、前記した分子の第2のタイプ、補体固定抗体は、IL−2受
容体に向けられたこの例において、IL−2受容体担持細胞を排除することがで
きる。この例において、分子の第3のタイプはその受容体に対するIL−2の結

持する腫瘍性細胞が増殖シグナルをIL−2から受け取るのを妨げるであろう。
腫瘍性細胞増殖を治療するのに有用な分子は多数の形態を採ることができる。
IL−2それ自体が標的化剤である場合、該分子は細胞毒性ハイブリッド分子で
あり得、そこでは、IL−2はトキシン分子、好ましくはポリペプチドトキシン
に融合している。I L−2Rに結合し、IL−2活性を欠き、かつボナフィド
(bona fide) I L −2の結合および/または摂取をブロックす
るll−2の誘導体は本発明の方法で有用である。何故ならば、それはIL−2
R担持細胞のIL−2誘導増殖を妨げるであろうからである。抗−II、−2R
抗体が標的化剤である場合、細胞毒性ハイブリッド分子は抗体のすべてまたは一
部を細胞毒に融合させることによって形成できる。IL−2/トキシンハイブリ
ツドのようなかかる抗体/トキシンハイブリッドの効率は、そわが結合する細胞
によって摂取されるハイブリッド分子に依存している。IL−2の結合および/
または摂取をブロックする抗−IL−2R抗体もまた本発明の方法で有用である
。溶解性抗−IL−2R抗体は、IL−2R担持細胞の補体媒介溶解を引き起こ
すので、本発明で有用である。
いつくかの分子は2種またはこれを超える分子のすべてまたは一部の融合によっ
て形成されるハイブリッド分子であり得る。該ハイブリッド分子は組換えDNA
分子によってコードされるハイブリッド蛋白であり得るが、この場合、2つのド
メインはペプチド結合によって(直接あるいは中間ドメインを通じて)連結され
ている。別法として、2つのドメインは別に生産でき、また別の化学的連結工程
における共有結合によって連結できる。いくつかの場合、ハイブリッド分子の細
胞毒ドメインはそれ自体2つの別々の分子から誘導してもよい。
標的化剤としてのインターロイキン−2インターロイキン−2(IL−2)また
はそのいずれかのIL−2受容体結合誘導体を細胞毒についての標的化剤として
使用できる。IL−2のDNAおよびアミノ酸配列は公知であり(タダッグ(T
adatsugu)ら、ネイチャー (Nature) 302 : 305.
1983)、その構造はX線結晶解析によって予測されている(プラントツーバ
ー(Brandhuber)ら、サイエンス(Science) 238 :
1707.1987)。IL−2の遺伝工学により作製した変異体の分析により
、いずれの残基がIL−2R結合(コリンズ(Collins)ら、プロ/−デ

および生物活性(ニーエン(Cohen)ら、勺イエンス(Science)
234 : 349.1989 :コリンズら、前掲)に重要かに関する情報が
いくらか提供されている。本発明で有用なI L−2の変異体は、残基74およ
び残基79(ウィリアムス(Williams)ら、ヌクレイツク・アンズ・リ
サーチズ(Nucl、 Ac1ds Res、 )]、 6 : 1045.1
988によるナンバリング)の間の領域において1個またはそれを超えるアミノ
酸残基を欠く欠失突然変異体を含む(ここに、参照して一体化させるゲンバウフ
s (Genbauffe)ら、USSN388゜557)。これらの突然変異
体はトキシンを効果的にIt、−2R担持細胞に対して標的化する(ケンハウフ
ェ(Genbauffe’)ら、前掲)。一般に、細胞毒を標的化4−るのに有
用な1f−2変異体は!1.−2Rに効果的に結合し、飲食作用をうけなければ
ならない。IL−2受容体に結合する種々の変異体の能力は後述する+ 1.−
2 R結合アッセイでテストできる。
IL−2受容体を担持する細胞に対して標的化された分子をデザインするにおい
て、他の受容体と同様に、11、−2受容体はいくつかの形態を有し:1の形態
を担持し、他の形態を担持しない細胞を標的化するのが望ましいであろうことを
認識する必要がある。ヒト・インターロイキン−2受容体は高−1中−1および
低−親和性形態を有する。高親和性受容体は〜1.Q−10Mの見掛けのに、を
有し、2つのサブユニットp55および(p70とも呼ばれる)p75よりなる
。細胞表面で発現されると、p75およびp55の両サブユニットはIL−2に
結合できる。該p75サブユニットは、生親和性受容体(K、〜8.2X10−
’°M)に対応し、p55サブユニットは低親和性受容体(K、〜1−3X10
−εM)に対応する。該p75サブユニットは休止状態のT細胞、ナチュラルキ
ラー細胞、単球/マクロファージ、およびリンホカイン−活性化キラー(1,、
AK)細胞前駆体の表面に発現さね、一方、該高親和性受容体は活性化されたT
−およびB−細胞に発現される。
本発明の方法においては、高親和性受容体を担持する細胞のみを標的化するのが
望ましいであろう。これらの状況において、有用な分子は、中または低親和性受
容体を担持する細胞にほとんど影響しない濃度にて、高親和性IL−2受容体を
排除または中和する。IL−2それ自体と同様に、分子がTL−2受容体のすべ
ての3つのクラスにつき親和性を有する場合、低親和性受容体を担持する細胞へ
の有意な結合を許容しない濃度にて該分子を投与することによって選択性は達成
される。ハイブリッド分子はIL−2と比較して受容体親和性を変更するかも知
れない。かかるハイブリッド分子は、多かれ少なかわ、高親和性IL−2受容体
を担持する細胞につき親和的であり得る。例えば、高親和性受容体を有する細胞
は、生親和性受容体を担持する細胞よりも、ジフテリアトキシンおよびIL−2
の一部よりなる融合蛋白DA B411111−−2に対し500〜1000倍
以上感受性である(ウォーターズ(Waters)、ニーロビーアン・ジャーナ
ル・オブ・イミュノロンー(Eur、 J、 Immunol、、 )、20
: 785.1990)。
細胞毒は多くの方法でIL−2誘導体に付着させることができる。好ましくは、
IL−2/トキンンハイブリツドは、融合遺伝子の発現によって産生されるハイ
ブリッド蛋白である。別法として、細胞毒およびIL−2誘導体は別々に生産し
て、非ペプチド結合によって後にカップリングさせることができる。連結方法は
後記する。
好ましくは、有用な細胞毒は細胞内に存在する場合にのみ有意に細胞毒性であっ
て、標的化ドメインの不存在下でいずれの所与の細胞からも実質的に排除される
。
ペプチドトキンンはこれらの両基準を満たし、容易にハイブリッド分子に一体化
される。混合細胞毒、すなわち、2種またはそれを超えるトキシンのすべてまた
は一部よりなる細胞毒を用いることもできる。いくつかの有用なトキシンは後期
にて詳細に記載する。
標的化剤としてのインターロイキン−4およびインターロイキン−6インターロ
イキンー4 (IL−4)は種々の細胞タイプに作用するサイトカインである。
その受容体はCD4+ T細胞および単球を含めた多数の細胞タイプに発現され
る。I L−4はT細胞増殖因子として作用でき、IL−2誘導リンパ球増殖に
対して影響を有すると考えられる。
IL−4受容体担持細胞またはIL−6受容体担持細胞に向けられた細胞毒はI
L−2R担持細胞に向けられる分子の効果を増強し得る。IL−4およびIL−
6の蛋白およびDNA配列は公知である(リー(Lee)ら、ジャーナル・オブ
・バイオロジカル・ケミストリー(J、Biol、Chet)263 + 10
817.1988 ヒラメ(hirano)ら、ネイチ+ −(Nature)
324 : 73.1986)。これらのリンホカインはIL−2/トキシンハ
イブリツド分子と同様のハイブリッドリンホカイン/トキシン分子を創製するの
に使用できる。
標的化剤としてのモノクローナル抗体
選択したリンホカイン受容体に向けられたモノクローナル抗体はトキンンをその
受容体を担持する細胞に向けるのに使用できる。ハイブリッド蛋白分子をコード
する融合遺伝子によってコードされたトキシンおよび抗体によって、あるいは別
々に産生されたリガンドおよびトキシン分子を連結するのに使用される非ペプチ
ド共有結合によって、これらの抗体または抗体断片を細胞毒に融合させることが
できる。いくつかの有用なトキシンは後述する。
抗体/トキシンハイブリッドは、IL、−2R担持細胞につき前述したのに同様
の毒性アッセイを用い、受容体担持細胞を殺すその能力につきテストできる。
本発明の方法で有用なモノクローナル抗体は、ヒト・IL−2Rまたは培養した
T−リンパ球でマウスを免疫し、マウス肺臓細胞を適当な骨髄腫細胞と融合し、
エライザアソセイによって必要なT L + −2R結合特性について、得られ
たハイブリトーマ系によって産生された抗体をスクリーニングすることによって
作製できる。抗体の産生およびスクリーニングはウチャマ(Uchiyama)
ら(ジャーナル・オブ・イミュノロジー0.Ii+5uno1.) 126 +
1393.1981)に従って達成できる。
本発明では、無傷のモノクローナルまたはポリクローナル抗体のみならず、免疫
学的に活性な抗体断片、例えば、Fabまたは(Fab)z断片、抗体重鎮、抗
体軽鎖:遺伝子工学により作製した一本鎖FV分子(ラドナー(Ladner)
ら、米国特許第4946778号):またはキメラ抗体、例えば、マウス抗体の
結合特異性を含有するが、残りの部分のほとんどまたはすべてがヒト起源である
「ヒューマナイズド(humanized)J抗体を使用できる(ライヒマン(
Reict++5an)ら、ネイチャー (Nature)332 : 323

トキシン
好ましくは、本発明の方法で有用なトキシン分子は、細胞内に存在する場合のみ
有意に細胞毒であるペプチドトキシンのごときトキシンである。勿論、これらの
状況下で、該分子は標的化受容体を担持する細胞に侵入できなければならない。
この能力は、分子の性質および細胞受容体の性質に依存する。例えば、天然にて
リガンドの摂取を可能とする細胞受容体は、その受容体を担持する細胞に侵入す
るトキシンを包含する分子につき手段を提供するようである。好ましくは、ハイ
ブリッド蛋白分子をコードする組換えDNA分子を産生ずることによって、ペプ
チドトキシンをIL−2R結合ドメインに融合させる。かかるアプローチは組成
物のコンンステンシーを保証する。
多くのペプチドトキシンは一般化された真核細胞受容体結合ドメインを有し:こ
れらの例において、該トキシンは受容体非担持細胞の中毒化を防ぐように修飾さ
れなければならない。いずれのかかる修飾も、分子の細胞毒機能を保持する方法
でなされなければならない(ヘルス・アンド・ヒユーマン・サービンズ(Hea
lth and Human 5ervice)の米国部会、米国特許出願40
1412号参照)。
可能性として有用なトキシンは、コレラトキシン、リシン、O−シカ様トキ、/
ン(SLT−T、5LT−n、SLT rIV)、LTトキンン、C3トキンン
、シガ(Shiga) トキシン、百日咳トキシン、破傷風トキシン、シュード
モナスのエンドトキシン、アロリン(alorin)、ザポリン(sapori
n)、モデンン(modeccin)、およびゲラニン(gela口in)を包
含する。
ジフテリアトキシンをヘースとした分子ジフテリアトキシンは本発明の方法で有
用な分子を生産するのに使用できる。
その配列が公知のジフテリアトキシンは、ここに参照して一体化させるマーフィ
ー (klurphy)の米国特許第4675382号に詳細に記載されている
。コリネバクチリウム・ジフテリア(Corynebacteriui dip
htheriae)によって分泌される天然ンフー)リアトキシン分子は、当該
分子のアミ5ノ末端で出発し、酵素的に活性なフラグメントA(アミノ酸Gly
+ Argue3)およびフラグメントB(アミノ酸S e r 194 S
e r 535)として特徴付けられるいくつかの機能性ドメインよりなり、こ
れは、トランスローケーンマンドメインおよび一般化された細胞結合ドメイン(
アミノ酸残基475ないし535)を包含する。
477子す7トキンンが感受性真核細胞を中毒化する方法は、少な(とも以下の

(tv)飲食作用・j1胞のpHが6未鳩まで低下するにつれ、トキシンは核内
体膜を横切り、フラグメントAのサイドシルへの送達を容易とする; (V)フ
ラグメントAの触媒活性(すなわち、「伸長因子2」なる用語の真核細胞蛋白合
成のニコチンアミドアデニンジヌクレオチド−依存性アデニンニリン酸(ADP
)リボンル化)は中毒化細胞の死滅を引き起こす。サイドシルに導入されたフラ
グメント2・〜の単一分子は細胞の蛋白合成装置を阻害し、細胞を殺すのに十分
にようである。
ノユードモナスエキソトキンンAによる、および恐らくはある種の他の天然トキ

3に融合した成熟ジフテリアトキシンのアミノ酸残基1ないし485よりなる。
かくして、DAB41+61L 2は、当該分子の酵素的活性部分をコードする
シフテリアトキノンフラグメントAのすべて、およびフラグメントBの一部を包
含する。
DA B4s61 L 2に存在するフラグメントBの一部は一般化された受容
体結合ドメインは包含しないが、サイドシルへの酵素的活性部分の送達を容易と
するトランスローケーンマンドメインを包含する。
DAB4861L 2およびDABsgelL 2の調製DABu+5lL−2

DA83s、lL 4の細胞毒仕
種々の細胞タイプの活性を減少させるDAB、。IL−4の能力は蛋白合成アッ
セイの阻害を用いて測定した:このアッセイの結果は表3に示す。IC,。(M
)は蛋白合成を50%減少させるのに必要なりAB、。IL〜4の濃度である。
ラットのConA活性化した正常稗臓リンパ球は、いずれの他の細胞または細胞
系よりも、DA83sslL−4に対して感受性がかなり低かった。ラット・イ
ンターロイキン−4受容体はヒト・インターロイキン−4に結合しないので、こ
の結果はDAB3solL−4の特異性を示す。これらのラット細胞はジフテリ
アトキシン/ラット・インターロイキン−2ハイブリツド分子に感受性である。

ることによってDABsmeIL 2につき前述したごと(に産生できる。
混合トキシン
本発明で有用ないくつかの分子の細胞毒性部分は、混合トキシン分子によって提
供できる。混合トキシン分子は、2つの異なるポリペプチドトキシンから誘導さ
れた分子である。一般に、ジフテリアトキシンに関連して前記したごと(、ポリ
ペプチドトキシンは、−膜化された真核細胞結合の原因となるドメインに加えて
、酵素的に活性な部分およびトランスローケーンタンドメインを有する。結合お
よびトランスローケーンタンドメインは、各々、細胞の認識およびトキシンの侵
入に必要である。酵素的に活性なドメインは、当該分子が細胞内に入ると、細胞
毒性活性の原因となるドメインである。
トランスローケーンタンドメインを有することが知られている天然に存在する蛋
白はジフテリアトキシン、シュードモナスエキソトキシンA、および恐らくは他
のペプチドトキシンを包含する。ジフテリアトキシンおよびシュードモナスエキ
ソトキシンAのトランスローケーンタンドメインはよ(特徴付けられており(例

この3部分ハイブリッド分子5LT−A/DTB’ /IL−2は、前記したD
A84as I L−2と同様に、本発明の方法で有用である。該3部分ハイブ
リッドのrL−2部分は、当該分子のIL−2R担持細胞への特異的付着を引き
起こし、ジフテリアトキシンのトランスローケーンタンドメイン部分はシカ様ト
キシンの酵素的活性なAサブユニットに作用して標的化細胞へ挿入させる。ジフ
テリアトキシンと同様に、シカ様トキシンの酵素的に活性な部分は、当該細胞の
蛋白合成装置に作用して蛋白合成を妨げ、か<シ、て該細胞を殺す。これらの2
つのタイプのハイブリッドトキシン間の差異は、その酵素的活性の性質である+
DAB<5alL 2の酵素的部分は、伸長因子2のニコチンアミドアデニンノ
ヌクレオチドによってADP−リボンル化を触媒し、それにより、蛋白合成に必
要なこの因子を不活化し、その一方で、5LT−A/DTB’ /IL−2の酵
素的部分は臨界的部位においてリポソームRNAを切断でき、それにより、当該
リポソームを不活化できるリボヌクレアーゼである。5LT−A/DTB’/I
L−2ハイブリツドは、従って、DAB<5srL−2と同一の症状の治療とし
て有用であり、それで置き換えたり、それと−緒に使用できるであろう。
トキシンの結合リガンドへの連結
有用なハイブリッド分子の結合リガンドおよび細胞毒はいくつかの方法で連結で
きる。ハイブリッド分子を融合遺伝子の発現によって産生ずるならば、ペプチド
結合は細胞毒と結合リガンドの間の連結として働く。別法として、該トキシンお
よび結合リガントを別々に産生じ、非ペプチド共有結合によって後にカップリン
グさせることができる。
例えば、共有結合連結は、ジスルフィド結合の形態を採り得る。この場合、IL
−2R結合リガンドが蛋白、例えば、IL−2である場合、IL−2をコードす
るDNAは、ここに参照して一体化させるマーフィ(llurpt+y)らの米
国特許第313599号に記載されているごとくに、余分なシスティンコドンを
含有するように設計できる。該システィンは当該分子の1l−2R結合活性に干
渉しないように位置させなければならない。例えば、システィンコドンはIt−
2の成熟形態のPro”をコードするDNAの丁度上流に挿入することができる
。トキノン分子は、システィンと反応性のスルフヒドリル基で誘導体化されてい
なければならない。ペプチドトキシンの場合、これは、当該トキシンをコードす
るDNA配列に7ステインコドンを挿入することによって達成できる。別法とし
て、スルフヒドリル基は、それ自体あるいはシスティン残基の一部として、固相
ポリペプチド法を用いて導入することができる。例えば、スルフヒドリル基のペ

る。次いで、細胞毒およびIL−2R結合リガンドを生産し、精製し、両硫黄基
を還元し、約1:5ないし】:20の比でトキシンとリガンドを混合し、室温に
てジスルフィド結合の形成を完了まで進行させ(一般に20ないし30分)るこ
とによって、精製した分子間にジスルフィド結合を形成させた。次いで、混合物
をリン酸緩衝化生理食塩水に透析して未反応リガンドおよびトキンン分子を除去

−2H結合能を測定することができる。種々の分子のIL−4H結合活性はパー
ク(Park)らによって記載されているアッセイ(ジャーナル・イブ・エクス
ベリメンタル・メデイシン(J、 Explled、)166 : 176.1
.984)またはフtツクスウェル(Foxwell)らによって記載さねてい
るアッセイ(ニーロビーアン・ジャーナル・イブ・イミュノロジ−(Eur、J
、fmunol、)19 :1637.1989)を用いて測定できる。
毒性についての了、セイ
本発明の分子(抗体および)\イブリッド分子双方)は、後述するごときア・ソ
セイじより、標的化受容体を担持する細胞の活性を減、少させる能力につきスク
リーニングできる。
IL−2担持細胞に対する毒性は以下のごとにテストできる。培養したHUT1
02/6TG (ラド(Tsudo)ら、プロンーデイングズ・イブ・ナショナ
ル・アカデミ−・イブ・サイエンシズ(Proc、 Natl、^cad、Sc
i、)USA83 :9694.1986)またはYT2C2(テノギワリ(T
eshigiwarj )ら、ジャーナル・イブ・1クスベリメンタル・メデイ
シン0. Exp、 Med、)165 :223.1987)細胞を、25m
M HEPES (pH7,4) 、2mM 1−グルタミン、100U/ml
ベニンジン、10(b1g/mlステレブトマインン、および10%胎児ウン血
清(ハセルトン(Hazel ton)、レネクセア(Lenexa)、カンザ
ス州)を補足したRPMI 1640培地(ジブコ(Gibco)、グランド・
アイランド(Grand l5land)、ニューヨーク州))に維持する。完
全培地中のウェル当たり1×105の濃度にて、細胞を、96−ウェルV−底プ
レート(リンブローフロラ・ラボラトリーズ(Linbro−Flow Lab
oratories)、マクリーン(McLean)、ノクーンニア州)に接種
する。トキンン候捕物を添加して種々の濃度(10−12Mないし10−’M)
とし、該培養を5%CO0雰囲気中、37℃にて18時間インキュベートする。
インキュベーンランに続き、170xgにて5分間プレートを遠心し、培地を除
去し、8μCi/m+ (3H−ロイシン、ニュー・イングランド・ヌクレアー
<New England Nuclear)、ボストン、マサチュセ・ソツ州
)を含有する100μm無ロイシン培地(MEM、ジブコ(Gibco))で置
き換えた。37℃でさらに90分後、170Xgにて5分間プレートを遠心し、
セル・ノ\−ベスター(cell harvester) (ヌ、カドロン(S
katron)、スターリング(Sterling)、ノ(−ジニア州)を用い
、ガラス繊維フィルター上に収集する。フィルターを洗浄し、乾燥し、標準的ブ
;方法により計数する。培地単独で培養した細胞を対照として供した。
IL−4Rを担持する細胞に対する毒性は、MLA 144細胞(ラビン(Ra
bin)ら、ジャーナル・イブ・イミュノロジ−(J、 Immunol、 )
127 : 1852.1981)または)ltJT 102/6TG細胞を
利用することを除き、IL−2R担持細胞につき前記したのと同様のア・ソセイ
によってテストすることができ、ウェル当たりI X 105細胞にて接種し、
40時間インキュベートする。
血!
一般に、本発明の分子は静注によって投与する。まt二、皮下投与することもで
きる。本発明の方法で有用な分子の投与量は、基質が細胞毒、溶解性抗体、また
はl L−2Rブロッキング分子であるか否かのごとき因子に依存して変動する
。
細胞内にて作用する毒性分子の場合、細胞摂取の程度は重要な因子であり、透過
性の低い分子は高用量で投与しなければならない。
60人を超える患者に相1/’II臨床プロトコルにてDA84sglL 2を
摂取させた。最大許容用量(MTD)にて治療した患者の約3096における一
過性無症候肝トランスアミナーセ上昇によってMTDを確立した。抗−腫瘍効果
がほぼ40%の憶者で観察され、応答はB−細胞白血病およびリンパ腫、皮膚T
−細胞リンパ腫およびホンキシ病(レメストル(Leklaistre)ら、ブ
ラッド(B1.ood) 360a アブストラクト1429.1990 ;ウ
ッドワース(Woodvorth)ら、ヒト・レトロウィルス学についての第4
回国際会議、1991)において見られた。
10”M DAB4,61L−2の血清濃度が悪性発現IL−2受容体をもつ患
者で達成された。抗−腫瘍効果が高治療抵抗性白血病/リンパ腫患者で観察され
、これらの効果はすべての患者にお1ブる可溶性IL−2Rレベルの上昇の存在
にも拘わらず起こった。この観察は、可溶性TL−2Rが高親和性インターロイ
キン−2受容体へのIL−2の結合に干渉しないことを示唆するデータに合致す
る。
動物およびヒトの研究は、DAB4ss I L 2が一般的な免疫抑制効果を
有しないことを示した(レメストル(LeMajstre)、前掲;ウッドワー
ス(foodworth)ら、前掲)。
実験は、高親和性IL−2受容体を担持する細胞によるD A B 4sa l
l 2の結合および内在化は、暴露30分以内に起こることを示し、その結果
数時間内に蛋白合成の最大阻害が起こる。従って、該分子は血清半減期がむしろ
短くとも効果的である。
一般に、IL−2受容体に標的化された薬物は血管負傷後直ちに(例えば、数分
またはそれ以内)投与する。好ましくは、治療は、血小板および白血球の蓄積前
に始める。裸出バルーンカテーテル負傷の動物モデルを用いて、血小板の凝集お
よび血栓の形成が負傷直後に起こること、および白血球付着は数時間内に始まる
ことが示された。血管形成術の5日後に死亡した患者の検死により、増殖する平
滑筋細胞が拡張領域を侵しつつあることが明らかにされた(オースチン(^us
tjn)ら、ツヤ−ナル・イブ・アメリカン・カレッジ・イブ・カルジオロブ−
(J、 Amer、 0a11. Cardiol。)6°369.1985)
。
一旦開始されたら、内皮細胞の再生は1ないし2週間内に完了する。血管壁の再
内皮化は平滑筋細胞の増殖を阻害するようなので(イブ(Ip)ら、ジャーナル
・イブ・アメリカン・カレッン・イブ・カルシオロジー0.^mer、 Co1
1. Cardiol、)15:1667.1990)、はんの数週間治療を継
続すればよい。従って、内皮細胞の再生を可能とするのに適当な期間にわたって
、本発明の化合物を周期的に投与するのが望ましい。
ハイブリッド分子は、非修飾分子として、あるいは治療上許容される塩、例えば
、生理食塩水と混合した、医薬上許容される塩の形態で投与することができる。
好ましい塩の例は治療上許容される有機塩、例えば、酢酸、乳酸、マLツイン酸
、クエン酸、またはサリチル酸である。例えば、ハイブリッド分子は精製し、2
ミクロンフィルターを用いて滅菌濾過し、滅菌リン酸緩衝生理食塩水(0,15
MNaC] ; 00.02Mリン酸緩衝液pH7,2)に懸濁することができ
る。
他の具体例
内因性IL−2の利用をブロックするIL−2の誘導体は、IL−2R担持細胞
の増殖を妨げるのに有用である。11−2を欠く活性化細胞は増殖せず、IL−
2によって供される必須の同化刺激の不存在下では、結局は死滅するであろう。
所与のIL−2誘導体のIL−2機能への干渉能はシュ(Ju)らによって記載
されているごときIL−2生物活性アツセイでテストできる(ジャーナル・イブ
・バイオロジカル・ケミストリー(J、 Biol、 Chew、 )262
: 5723.1987)。トキ、/ンが不活化されてしまっているIL−2R
/)キシンノ\イブリットもまたIL−2受容体をブロックするのに使用できる
。非毒性突然変異体ンフテリアトキシン分子は記載されており(ウチダ(Uch
ida)ら、ジャーナル・イブ・バイオロジカル・ケミストリー(J、 Bio
l、 Chem、)248 : 3838.1973)、この分子は非毒性IL
−2/ジー2リアトキシンハイブリッドを生産するのに使用できる。かかるハイ
ブリッド分子の例については、ここに参照して一体化させるスブルルガ(Svr
luga)らの米国特許第590113号を参照されたい。
IL−2の結合および/または摂取に干渉するモノクローナル抗体は、l L−
2を奪われたIL−2R担持細胞は増殖しないゆえ、本発明の方法で有用である
。ブロックするモノクローナル抗体は、シュOu)ら(前掲)の方法を用いて、
IL−2生物活性へのその干渉能につきテストすることができる。
補体を誘導するモノクローナル抗体はIL−2R担持細胞を破壌するのに使用で
きる。補体誘導抗体は一般にIgG1.1gG2.1gG3、およびTgMイソ
タイプのものである。モノクローナル抗−TL−2R抗体は、補体依存性細胞毒
性テストを用いて、以下の如く、補体誘導能につきスクリーニングできる。
ヒト・T−リンパ球およびEBS形賞転換B−リンパ球を6IC,クロム酸ナト
リウムで標識し、標的細胞として使用する。これらの細胞はハイブリドーマ培養
上澄および補体とともにインキュベートし、次いで、上澄を収集し、ガンマカウ
ンターて計数する。活性化T−リンパ球に対して毒性を示すが、休止T−または
B−リンパ球に対しては示さない1澄みを選択し、次いで、さらにスクリーニン
グに付して、(レオナルト(Leonard)ら(ブロノーディングズ・イブ・
ナショナル・アカデミ−・イブ・サイエン・/ズ(Proc、NatlAcad
、Sci、)USA80 : 6957.1983)に詳細に記載されている)
50kd糖蛋白IL−2受容体を沈殿させる(すなわち、それと特異的に反応性
の)抗体を含有する上澄を選択する。
所望の抗−IL−2受容体抗体を常法を用いて上澄から精製する。
Claims (29)
- 1.医薬上許容される担体と、非リンパ球および非単球起源の腫瘍性細胞に発現 されたサイトカイン受容体に特異的に結合できる分子とを混合することよりなり 、該サイトカイン受容体はリンパ球および単球起源の細胞に通常発現されるもの であり、該分子は該腫瘍性細胞の活性を減少できることを特徴とする非リンパ球 および非単球起源の該腫瘍性細胞の増殖によって特徴付けられる肉腫を治療する ための医薬品の製法。
- 2.該腫瘍性細胞が造血幹細胞起源でない請求の範囲第1項記載の方法。
- 3.該肉腫が骨肉腫である請求の範囲第1項記載の方法。
- 4.該肉腫が繊維肉腫である請求の範囲第1項記載の方法。
- 5.該肉腫が平滑筋肉腫である請求の範囲第1項記載の方法。
- 6.該肉腫が横紋筋肉腫である請求の範囲第1項記載の方法。
- 7.医薬上許容される担体と、非リンパ球および非単球起源の腫瘍性細胞に発現 されたサイトカイン受容体に特異的に結合できる分子とを混合することよりなり 、該サイトカイン受容体はリンパ球および単球起源の細胞で通常に発現されるも のであり、該分子は該腫瘍性細胞の活性を減少できることを特徴とする該腫瘍性 細胞の増殖によって特徴付けられる肺カルシノーマを治療するための医薬品の製 法。
- 8.該サイトカイン受容体がインターロイキン受容体である請求の範囲第1項記 載の方法。
- 9.該インターロイキン受容体がインターロイキン−2受容体である請求の範囲 第8項記載の方法。
- 10.該インターロイキン−2受容体が高親和性インターロイキン−2受容体で ある請求の範囲第9項記載の方法。
- 11.該インターロイキン受容体かインターロイキン−4受容体である請求の範 囲第8項記載の方法。
- 12.該インターロイキン受容体がインターロイキン−6受容体である請求の範 囲第8項記載の方法。
- 13.該分子が該サイトカイン受容体を担持する細胞を殺す請求の範囲第1項記 載の方法。
- 14.該分子が、一緒に共有結合により連結した第1および第2の部分よりなる ハイブリッド分子であり、該第1の部分が細胞活性を減じることができる分子よ りなるものであって該第2の部分が該サイトカイン受容体に特異的に結合できる 分子よりなる請求の範囲第1項記載の方法。
- 15.該第2の部分が該サイトカイン受容体に対して特異的な抗体のすべてまた はその結合部分よりなる請求の範囲第14項記載の方法。
- 16.該第2の部分が該サイトカイン受容体についてのリガンドのすべてまたは その結合部分よりなる請求の範囲第14項記載の方法。
- 17.該サイトカイン受容体がインターロイキン受容体である請求の範囲第16 項記載の方法。
- 18.該リガンドがインターロイキンである請求の範囲第16項記載の方法。
- 19.該第1の部分が細胞毒よりなる請求の範囲第14項記載の方法。
- 20.該細胞毒が酵素的に活性であるが一般化された真核細胞受容体結合活性を 有しないペプチドトキシンの断片である請求の範囲第19項記載の方法。
- 21.ペプチドトキシンの該断片がジフテリアトキシンのフラグメントAおよび サイトゾルヘのトランスローケーションを容易とするための充分なジフテリアト キシンのフラグメントBよりなる請求の範囲第20項記載の方法。
