Description of the Related Art Vancomycin is a glycopeptide antibiotic having an excellent antibacterial action against Gram-positive bacterial infection, and has conventionally been used as a water-soluble injection in order to maximize its efficacy. On the other hand, liposome is a package consisting of a lipid bilayer composed of lipids of biological origin, and has been used since the 1970s as a drug carrier capable of sustained-release of drug and maintaining high blood drug concentration. .

できる。[Means for Solving the Problems] The present inventors have completed the present invention by discovering that vancomycin can be used as a liposome preparation to simplify the complicated administration method and sustain the antibacterial action. That is, the present invention is a liposome preparation containing vancomycin. Examples of lipids that form liposomes include phospholipids and glycolipids. As the phospholipid, any phospholipid, natural or non-natural, can be used in the present invention as long as it is a phospholipid that can be metabolized in vivo. Examples of the natural phospholipid include egg yolk, soybean, and other natural phospholipids derived from plants and animals. Non-natural phospholipids include compounds obtained by hydrogenating natural phospholipids, and synthetic compounds such as phosphatidylcholine, phosphatidylserine, phosphatidylglycerol, phosphatidylethanolamine, phosphatidic acid, sphingomyelin, and dicetylphosphate. Phospholipids of. Any glycolipid can be used in the present invention as long as the glycolipid can be metabolized in vivo. Examples include phosphatidylinositol, cerebroside, sulfatide, ganglioside and the like. When preparing liposomes,
Usually, stabilizers such as cholesterol and tocopherol,
Alternatively, a fatty acid such as oleic acid that controls the release of vancomycin can be added. In the case of a stabilizer such as cholesterol or tocopherol, the blending amount thereof is 1.0 or less, preferably 0.3 to 1.0, relative to the lipid, and in the case of fatty acid such as oleic acid, the molar ratio is 1.0 relative to the lipid.
Below, preferably 0.1 to 0.5. Furthermore, the present invention is used for additives such as isotonicity agents, preservatives, pH regulators and the like, which are usually used for injections, or for oral agents such as antioxidants, preservatives, flavoring agents, and colorants. Additives can be added.

よい。The liposome preparation of the present invention can be obtained by an ordinary method for producing a liposome preparation. For example, vancomycin may be incorporated into the liposome by preparing the liposome after homogenizing vancomycin with phospholipid or glycolipid. Alternatively, vancomycin may be contained after preparing liposomes with phospholipids or glycolipids in advance. Specific examples of methods for preparing liposomes include known methods such as a thin film method, a surfactant removal method, an ultrasonic method, an ether injection method, a high-pressure injection emulsification method, and an extrusion method. To contain vancomycin in the liposomes prepared in advance, solid vancomycin and the liposome solution may be mixed and freeze-thawed or sonicated.

At this time, the freeze-drying method is performed according to a conventionally used method. The thus obtained vancomycin-containing liposome preparation is reconstituted as it is or in the case of a freeze-dried form, and then administered as an oral preparation or an injection, intravenously, or intramuscularly, intradermally, subcutaneously, or locally. be able to.

Weigh 0.9 g and put chloroform 20 in an eggplant-shaped flask.
It was dissolved in ml. Then, chloroform was removed by a rotary evaporator, and a completely dried film component mixture was obtained by a vacuum dryer. To this mixture, 20 ml of a 5% glucose solution and 0.4 g of vancomycin were added, and the mixture was shaken to disperse the mixture and further freeze-thawed to obtain vancomycin-containing multilamellar vesicles. Further, vancomycin which was not encapsulated was removed using a Sephadex (registered trademark) G-50 column, and a liposome fraction was obtained to obtain the preparation of the present invention.
