Claims (1)
- 【特許請求の範囲】 1.式I: ▲数式、化学式、表等があります▼I [式中、R1は水素またはメチルを表し、R2,R3及びR4は独立して水素、 メチル、メトキシ、ヒドロキシ、カルバモイル、スファノイルまたはハロゲンを 表し、Xは式II:▲数式、化学式、表等があります▼ (ここで、R5及びR6は非芳香族ヒドロカルピル基を表し、これらは同じでも 異なっていてもよく、両方合わせて少なくとも3個の炭素原子を含有し、そして 、R5とR6はアミン窒素と共に環を形成してもよい)の第三アミノ基を表す] の3,3−ジフェニルプロピルアミン、及び該化合物の生理学的に許容される酸 との塩、及び該化合物が光学異性体の形態をとるときにはそのラセミ混合物及び 各光学対掌体。 2.各R5及びR6が独立して飽和ヒドロカルピル基、特に飽和脂肪族ヒドロカ ルピル基例えばC1−8−アルキル特にC1−6−アルキル、またはアダマンチ ルであり、R5とR6が合わせて少なくとも3個好ましくは少なくとも4個の炭 素原子を含んでいる請求項1の3,3−ジフェニルプロピルアミン。 3.R5及びR6が一緒になってアミン窒素と共に環を形成する請求項1または 2の3,3−ジフェニルプロピルアミン。 4.R5及び/またはR6が少なくとも1つのヒドロキシ置換基を有しているる 請求項1、2または3の3,3−ジフェニルプロピルアミン。 5.R5及びR6の少なくとも1つが分枝炭素鎖を有している請求項1から4の いずれかの3,3−ジフェニルプロピルアミン。 6.Xが下記の基a)−f)のいずれかを表し、その各々は少なくとも1つヒド ロキシ置換基を有しうる請求項1から5のいずれかの3,3−ジフェニルプロピ ルアミン:a)▲数式、化学式、表等があります▼,b)▲数式、化学式、表等 があります▼,c)▲数式、化学式、表等があります▼,d)▲数式、化学式、 表等があります▼,e)▲数式、化学式、表等があります▼,f)▲数式、化学 式、表等があります▼7.下記の化合物群から選択した3,3−ジフェニルプロ ピルアミン、該化合物の生理学的に許容される酸との塩、並びに可能であればそ のうセミ体及び各光学対掌体である請求項1の3,3−ジフェニルプロピルアミ ン: N,N−ジイソプロピル−3−(2−ヒドロキシ−5−メチルフェニル)−3− フェニルプロピルアミン,N−メチル−N−第三ブチル−3−(2−ヒドロキシ フェニル)−3−フェニルプロピルアミン, N−メチル−N−第三ブチル−3−(2,4−ジヒドロキシフェニル)−3−フ ェニルプロピルアミン,N−メチル−N−第三ブチル−3,3−ピス−(2−ヒ ドロキシフェニル)プロピルアミン, N,N−ジイソプロピル−3,3−ピス−(2−ヒドロキシフェニル)プロピル アミン, N,N−ジイソプロピル−3−(2,5−ジヒドロキシフェニル)−3−フェニ ルプロピルアミン, N−メチル−N−第三ブチル−3−(2,5−ジヒドロキシフェニル)−3−フ ェニルプロピルアミン,N,N−ジイソプロピル−3−(2−メトキシフェニル )−3−フェニルプロピルアミン, N−[3−(2−メトキシフェニル)−3−フェニルプロピル〕−2,2,6, 6−テトラメチルピペリジン,(+)−N,N−ジイソプロピル−13−(2− ヒドロキシ−5−メチルフェニル)−3−フェニルプロピルアミン。 8.薬剤活性物質特に抗コリン剤として使用するための請求項1−7のいずれか の3,3−ジフェニルプロピルアミン。 9.請求項1−7のいずれかの3,3−ジフェニルプロピルアミンと相容性の薬 剤担体とからなる薬剤組成物。 10.抗コリン剤の製造への請求項1−7のいずれかの3,3−ジフェニルプロ ピルアミンの使用。 11.a)式III: ▲数式、化学式、表等があります▼III[式中、R1−R4は上記と同義であ り、いずれのヒドロキシ基も保護されていてよく、Yは脱離基である〕の反応的 にエステル化した3,3−ジフェニルプロパノールと式IV:H−X IV [式中、Xは上記と同義である〕のアミンとを反応させ、または、 b)式V: ▲数式、化学式、表等があります▼V [式中、R1−R4及びXは上記と同義であり、いずれのヒドロキシ基も保護さ れていてよい]の3,3−ジフェニルプロピオンアミドを還元し、または、 c)式VI: ▲数式、化学式、表等があります▼VI[式中、R1−R4は上記と同義であり 、いずれのヒドロキシ基も保護されていてよく、Zはメチル以外のR5及びR6 と同義である]の第二3,3−ジフェニルプロピルアミンVIをN−メチル化し 、または、 d)式VIIaまたはVIIb: ▲数式、化学式、表等があります▼VIIa▲数式、化学式、表等があります▼ VIIb[式中、R1−R4及びXは上記と同義であり、いずれのヒドロキシ基 も保護されていてよく、Wはヒドロキシ基またはハロゲン原子を表している]の 3,3−ジフェニルプロピルアミンを還元し、そして、 i)所望であればフェニル環の一方または両方をモノ−またはジ−ハロゲン化し た後、得られた化合物のヒドロキシ保護基を必要に応じて分離し、及び/または 、 ii)所望であれば、生理学的に許容される酸により、得られた式Iの塩基をそ の塩に変換し、またはその反対を行い、及び/または、 iii)所望であれば、得られた光学異性体混合物を各光学対掌体に分離し、及 び/または、 iv)所望であれば、R1が水素であり及び/またはR4がヒドロキシである得 られた式Iの化合物のオルト−ヒドロキシ基をメチル化することからなる請求項 1−7のいずれかの3,3−ジフェニルプロピルアミンの製法。
