Description
本発明はω−インドール−3−イルアルカノー
ル化合物の改良された製造方法に関するものであ
る。
ω−インドール−3−イルアルカノール化合物
は、広く天然に存在し、多くのインドール誘導体
やインドールアルカロイド類および医薬品の合成
原料として有用な化合物である。
ω−インドール−3−イルアルカノール化合物
の合成については古くからいくつかの方法が提供
されているが、そのうちフエニルヒドラジンを出
発物質とするFischer合成法を利用した方法とし
て下記の方法が知られている。
置換フエニルヒドラジンの塩酸塩とジヒドロ
フランとを含水ジオキサン中で長時間加熱する
方法〔Khim、Geterotsikl.Soedin.、1972、
1366−1367〕
置換フエニルヒドラジンと2−ヒドロキシテ
トラヒドロフランとをベンゼン中で還流し、共
沸により水を留出させ、得られる縮合物を更に
高温減圧下に加熱分解する方法〔同誌;1972、
No.8、1083〜1084〕
置換フエニルヒドラジンとα−ホルミル−γ
−ブチロラクトンとを塩化水素の存在下に含水
アルコール中で長時間加熱反応させる方法〔同
誌;1974、No.1、90〜91〕
上記の既知方法はいずれも反応に長時間を要し
たり、操作が煩雑であつたり、原料の入手が難し
いなどの点により工業的製造方法としては不満足
なものである。このような見地から本発明者らは
ω−インドール−3−イルアルカノール化合物の
経済的合成方法について鋭意研究し、本発明を完
成した。
すなわち本発明は、下記一般式()の化合物
〔但し上式中R1は水素原子、低級アルキル基、
アリール基又はアルアルキル基を表わし、R2は
水素原子、低級アルキル基、アリール基、アルア
ルキル基、低級アルコキシル基、アリーロキシル
基、アルアルコキシル基、又はハロゲン原子を表
わし、R3は水素原子、又は、低級アルキル基を
表わし、nは0又は1を表わす。〕
を製造するために、
下記一般式()のフエニールヒドラジン化合
物:
〔但し上式中R1、R2はそれぞれ前記定義に同じ〕
の強酸塩と、下記一般式()のオキサシクロア
ルカン−2−オール化合物:
〔但し、式中R4は低級アルキル基又はアシル基
を示し、R3及びnはそれぞれ前記定義に同じ〕
とをアルコール系溶媒中で反応させることを特徴
とするものである。
本発明方法において、一般式()のフエニル
ヒドラジン化合物と、一般式()のオキサシク
ロアルカン−2−オール化合物との反応に際し、
フエニルヒドラジン化合物が強酸塩として存在す
ることが重要である。
前記()式において、R1で表わされる低級
アルキル基は1個から4個迄の炭素原子を有する
ものであることが好ましく、メチル、エチル、プ
ロピルおよびブチル基から選ぶことができる。ま
たR1で表わされるアリール基はフエニル基又は
トリル基であることが好ましく、更にR1で表わ
されるアルアルキル基は、ベンジル又はフエニル
エチル基であることが好ましい。
前記()式において、R2で表わされる低級
アルキル基は1個から4個までの炭素原子を有す
ることが好ましく、メチル、エチル、プロピル、
およびブチル基から選ぶことができる。また、
R2で表わされるアリール基は、フエニル基又は
トリル基であることが好ましく、R2で表わされ
るアルアルキル基はベンジル又はフエニルエチル
基であることが好ましく、R2によつて表わされ
るアルコキシル基は、メトキシ、エトキシ、又は
プロポキシ基であることが好ましく、R2によつ
て表わされるアリーロキシル基は、フエノキシ、
又はトリルオキシ基であることが好ましく、更
に、R2によつて表わされるアルアルコキシル基
は、ベンジルオキシ、又はフエニルエチルオキシ
基であることが好ましい。
一般式()において、R3によつて表わされ
る低級アルキル基は、1個から3個までの炭素原
子を有するものが好ましく、メチル、エチル、お
よびプロピル基から選ぶことができる。
更に、一般式()において、R4で表わされ
る低級アルキル基は、1個から6個までの炭素原
子を有するものが好ましく、また、R4で表わさ
れるアシール基はアセチルおよびプロピオニル基
から選ぶことができる。
本発明方法において使用される、一般式()
のフエニルヒドラジン化合物としては、フエニル
ヒドラジン、トリルヒドラジン、p−ベンジルフ
エニルヒドラジン、メトキシフエニルヒドラジ
ン、ベンジルオキシフエニルヒドラジン、クロル
フエニルヒドラジン、1,1−ジフエニルヒドラ
ジン、1−ベンジル−1−フエニルヒドラジン、
および1−メチル−1−フエニルヒドラジンなど
がある。
本発明方法に用いられる一般式()のオキサ
シクロアルカン−2−−オール化合物としてはテ
トラヒドロフラン、テトラヒドロピラン、4−メ
チルテトラヒドロピランなどの2の位置がメトキ
シ、エトキシ、プロポキシ、などのアルコキシ
基、又は、アセトキシなどのアシール基で置換さ
れた化合物がある。
すなわち一般式()のオキサシクロアルカン
−2−オール化合物中のオキサシクロアルカン環
は、テトラヒドロフラン環およびテトラヒドロピ
ラン環から選ばれることが好ましい。
本発明方法において、フエニルヒドラジン化合
物とオキサシクロアルカン−2−オール化合物と
の反応は、少くとも1種のアルコール系化合物を
含む溶剤中で行われる。このアルコール系化合物
は、1価の脂肪族アルコール類、例えば、メチル
アルコール、エチルアルコール、プロピルアルコ
ール、およびブチルアルコールなど、2価の脂肪
族アルコール類、例えば、エチレングリコールお
よびプロピレングリコールなど、脂肪族エーテル
アルコール類、例えばエチレングリコールメチル
エーテル、および、エチレングリコールモノエチ
ルエーテルなど、並びに、前記アルコール化合物
の少くとも1種と水との混合物とから選ぶことが
できる。溶剤のタイプは、製造されるべきω−イ
ンドール−3−イル−アルカノール化合物の性質
を考慮して選択される。この溶剤は、1種のアル
コール化合物からなるものであつてもよいし、2
種以上のアルコール化合物の混合物であつてもよ
いし、或は少くとも1種のアルコール化合物と水
との混合物であつてもよい。後者の場合、アルコ
ール化合物が水に対して10%以上含有されている
ことが好ましい。
本発明方法において、フエニルヒドラジン化合
物は、オキサシクロアルカン−2−オール化合物
1モル当り1モル以上で用いられることが好まし
く1.3モルから3.0モルまでの量で用いられること
が更に好ましい。この場合、フエニルヒドラジン
化合物と、オキサシクロアルカン−2−オール化
合物との反応を強酸の存在で行い、この強酸の量
が、使用されたフエニルヒドラジン化合物の少く

1モルのフエニルヒドラジン化合物に相当する量
を、その強酸塩に変成するのに必要な量であるこ
とが重要である。
前記強酸は、一般式()のフエニルヒドラジ
ン化合物と塩を形成することのできる、塩酸、硫
酸およびp−トルエンスルホン酸からなる群から
選ばれる少くとも1種からなるものであることが
好ましい。
若し、フエニルヒドラジン化合物を、オキサシ
クロアルカン−2−オール化合物1モル当り1モ
ル上の量で用いられるならば、強酸塩の状態にあ
るフエニルヒドラジン化合物の量の、強酸塩の状
態にないフエニルヒドラジン化合物の残余量に対
するモル比が2以上であることが好ましい。強酸
塩の状態にないフエニルヒドラジン化合物は、遊
離塩基の形をとつていてもよいし、或は、弱酸、
例えば、酢酸との塩の形をとつているものであつ
てもよい。
前記強酸の存在により、反応混合物は酸性状態
に維持され、この酸性状態は、フエニルヒドラジ
ン化合物と、オキサシクロアルカン−2−オール
化合物との間の反応を行わせるためには必須の条
件である。
しかしながら、反応混合物中に添加されるべき
強酸の量は、反応混合物中のフエニルヒドラジン
化合物の全量を、その強酸塩に変成するために必
要な量を越えて過剰にならないことが好ましい。
フエニルヒドラジン化合物と、オキサシクロア
ルカン−2−オール化合物との間の反応が、特定
量の強酸の存在によつて促進される理由は、未だ
完全には明らかではない。しかしながら、反応混
合物中の強酸の量が、反応混合物中のフエニルヒ
ドラジン化合物の全量を、その強酸塩に変成する
のに必要な量よりも過剰であるときは、1種の分
子内アセタール化合物であるオキサシクロアルカ
ン−2−オール化合物が分解し、この分解生成物
が重合、又は縮重合して、所望の生成物すなわ
ち、ω−インドール−3−イル−アルカノール化
合物(トリプトホール化合物)、の収量を低下さ
せることが見出されている。若し、反応混合物が
塩基性の状態にあるならば、フエニルヒドラジン
化合物とオキサシクロアルカン−2−オール化合
物との間の反応は生起しない。強酸が反応混合物
中のオキサシクロアルカン−2−オール化合物1
モル当り少くとも1モルのフエニルヒドラジン化
合物を、その強酸塩に変成するのに必要な特定の
量で、しかも、フエニルヒドラジン化合物の全量
を完全に変更するに必要な量をこえる過剰量では
ない量で使用れる場合においてのみ、フエニルヒ
ドラジン化合物はオキサシクロアルカン−2−オ
ール化合物と直接に縮合し、望ましくない副反応
を防止しつつインドール環を形成することができ
る。
フエニルヒドラジン化合物とオキサシクロアル
カン−2−オール化合物との間の反応は好ましく
は70℃から170℃までの温度で、反応を完結する
のに必要な時間だけ、一般には30分以上、例えば
30分から数時間行われる。反応が70℃より低い温
度で行われると、反応速度が低くなるので好まし
くない。反応速度は170℃の温度でピークに達す
る。従つて170℃より高い反応温度は反応速度を
高めるためには効果がない。
フエニルヒドラジン化合物が、そのベンゼン環
に付加され、高い反応性を有するエーテルタイプ
の置換基、例えばメトキシ又はベンジルオキシ基
を有する場合、このフエニルヒドラジン化合物が
用いられるときは、その反応温度は、その他のフ
エニルヒドラジン化合物が用いられるときよりも
低いこと、例えば70℃から90℃であることが好ま
しい。
本発明方法において、上記反応を行わせるに
は、フエニルヒドラジン化合物と、オキサシクロ
アルカン−2−オール化合物とを別々に所定量の
アルコール系溶剤に溶解し、得られた溶液のいず
れか一方を他方に滴下し、このとき、反応混合物
の温度を所望のレベルに保持する方法を用いても
よい。
また、他の方法としては、フエニルヒドラジン
化合物とオキサシクロアルカン−2−オール化合
物とを一緒に溶剤中に溶解し、得られた溶液を所
望温度に加熱する方法を用いてもよい。
得られる生成物、すなわち、ω−インドール−
3−イル−アルカノール化合物は、反応混合物か
ら通常の分離方法より分離される。例えば反応混
合物を水と混合し、次に水不溶性有機溶剤、例え
ばクロロホルム、と混合して反応生成物を有機溶
剤に抽出してもよい。得られた抽出液を蒸留に供
し、有機溶剤を除去する。この蒸留残留物を減圧
蒸留を供するが、或は再結晶に供して精製された
反応生成物を得る。
本発明のω−インドール−3−イル−アルカノ
ール化合物の種類は、反応したフエニルヒドラジ
ン化合物の種類とオキサシクロアルカン−2−オ
ール化合物の種類とに依存して定まる。本発明の
ω−インドール−3−イル−アルカノール化合物
は、たとえば2−(インドール−3−イル)エタ
ノール(通称トリプトフオール、以下通称名を用
いる)、4−メチルトリプトフオール、5−メチ
ルトリプトフオール、6−メチルトリプトフオー
ル、7−メチルトリプトフオール、5−クロルト
リプトフオール、7−クロルトリプトフオール、
5−ブロムトリプトフオール、N−メチルトリプ
トフオール、N−フエニルトリプトフオール、N
−ベンジルトリプトフオール、5−メトキシトリ
プトフオール、5−ベンジルオキシトリプトフオ
ール、3−(インドール−3−イル)プロパノー
ル、3−(インドール−3−イル)−ブタノールな
どを包含する。
本発明方法を更に実施例により説明する。下記
実施例中、得られた生成物の収率は、この生成物
の現実に得られた収量の、使用したオキサシクロ
アルカン−2−オール化合物の使用量から計算さ
れた生成物の理論収量に対する比(%)で表わさ
れている。
実施例 1
エチレングリコールモノメチルエーテル50mlに
フエニルヒドラジン塩酸塩2.17g(15ミリモル)を
溶解して、この溶液を加熱撹拌して液褐が123℃
に達したときに、2−メトキシテトラヒドロフラ
ン1.02g(10ミリモル)をエチレングリコールモノ
メチルエーテル20mlに溶解した溶液を30分間で滴
下した。反応温度を122〜124℃に保つて1時間撹
拌を行つた。反応終了後、反応液を室温まで放冷
し、水100mlを加え、クロロホルム100mlで2回抽
出し、クロロホルム層を合一して、洗液が中性と
なるまで水洗した。クロロホルムを減圧下に留去
し、得られた褐色油状物を減圧蒸留して167〜173
℃/2mmHgの留分をトリプトフオールとして
1.32g(収率82%)を得た。このものの融点は56℃
であつた。
実施例 2〜8
第1表に示すオキサシクロアルカン−2−オー
ル化合物、フエニルヒドラジンの強酸塩及び溶媒
を用い実施例1に準じて反応させトリプトフオー
ルを第1表の通りに得た。

コールモノメチルエーテル25mlに溶解したものを
30分間で滴下した。この後反応液を沸騰させなが
ら1時間撹拌した。反応終了後溶媒の半量を留出
させて冷却した後水50mlを加え、クロロホルム
100mlで2回抽出し、クロロホルム層を合一して
洗液が中性となるまで水洗した。クロロホルムを
減圧下に留去し、得られた褐色油状物を減圧蒸留
し、182〜185℃/0.5mmHgの留分を3−(インド
ール−3−イル)−ブタノールとして5.19gを得
た。このものの収率は70%であつた。

トラヒドロピランに代え2−メトキシテトラヒド
ロピラン4.55g(39.2ミリモル)を用いて実施例11

実施例 21
エタノール40mlにp−ベンジルオキシフエニル
ヒドラジン塩酸塩2.26g(9ミリモル)を溶解し、
この溶液を撹拌しながら加温し、76℃をなつたと
き2−メトキシテトラヒドロフラン0.61g(6ミリ
モル)をエタノール20mlに溶解したものを30分間
で滴下した。反応液を76〜78℃に保ちながら1時
間撹拌して反応させた。反応終了後水50mlを加え
たものをクロロホルム100mlで2回抽出し、クロ
ロホルム層を合一(洗液が中性となるまで水洗し
た。クロロホルムを減圧下に留去し、得られた褐
色油状物をカラムクロマトグラフイーにより分別
し、5−ベンジルオキシトリプフオール0.71g(収
率44%)を得た。このものの融点は93−93.5℃で
あつた。
実施例 22
フエニルヒドラジン塩酸塩21.6g(0.15モル)と
フエニルヒドラジン3.24g(0.03モル)を水90mlと
n−プロパノール90mlの混合液に溶解した溶液を
加熱撹拌し、液温が90℃に達したとき、2−メト
キシテトラヒドロフラン12.3g(0.12モル)を水20
mlとn−プロパノール20mlの混合液に溶解したも
のを2時間で滴下した。この後、反応液を沸騰さ
せながら3時間撹拌した。反応終了後溶媒の約半
量を留出させた後、残液をクロロホルムで70mlず
つ2回抽出を行い、クロロホルム層を合一して水
洗した。クロロホルムを減圧下に留去し、得られ
た褐色油状物を減圧蒸留して175〜178℃/2mm
Hgの留分をトリプトフオールとして14.3g(収率
74%)を得た。
実施例 23
フエニルヒドラジン塩酸塩24.5g(0.17モル)と
フエニルヒドラジン1.09g(0.01モル)を水40mlと
エチレングリコール120mlの混合液に溶解した溶
液を加熱撹拌し、液温が108℃に達したとき、2
−メトキシテトラヒドロフラン12.3g(0.12モル)
をエチレングリコール25mlに溶解した溶液を1.5
時間で滴下した。この後反応液を沸騰させながら
3時間撹拌して反応を行つた。反応終了後反応液
を冷却したのち、水300mlを加え、クロロホルム
で200mlずつ3回抽出し、クロロホルム層を合一
して水洗した後、減圧下にクロロホルムを留去し
た。得られた褐色抽状物を減圧蒸留して173〜178
℃/1.5〜2.5mmHgの留分をトリプトフオールとし
て11.2g(収率58%)を得た。
比較例
フエニルヒドラジン塩酸塩10.1g(0.07モル)と
フエニルヒドラジン12.0g(0.11モル)を水90mlと
n−プロパノール90mlの混合液に溶解した溶液を
加熱撹拌し、液温が90℃に達したとき、2−メト
キシテトラヒドロフラン12.3g(0.12モル)を水20
mlとn−プロパノール20mlの混合液に溶解したも
のを2時間で滴下した。この後、反応液を沸騰さ
せながら3時間撹拌して反応させた。反応終了後
実施例22に準じて処理したが目的のトリプトフオ
ールは得られず、反応液のTLCよりヒドラゾン
