In esters obtained by esterifying terpene acids such as farnesylic acid, homofarnesylic acid, and 7-arnesyl acetic acid with reactive derivatives such as acid halides and acid anhydrides, side effects of the pregnane derivative as the starting material may be avoided. It is known that the activity is generally improved compared to the pregnane derivatives when applied externally (see US Pat. No. 3,488,421). Several people are interested in prednisolone 21-farnesylate [11β, 17α, which is not specifically mentioned in the above US patent specification.

Although we found that the expression was low,
It would be desirable to provide a terpene-modified compound of prednisolone that has even better anti-inflammatory effects in treating inflammation.

Reactive derivatives of prednisolone include halides, alkanesulfonates, arenesulfonates, carboxylates, etc., and reactive derivatives of 2E, 6-farnesylic acid include lower alkyl esters, acid halides, mixed acid anhydrides, and alkali metals. Examples include salts, silver salts, and salts of organic tertiary or quaternary bases. The reaction between prednisolone or its reactive derivative and 2E,6E-farnesylic acid or its reactive derivative can be carried out under conventionally known general ester synthesis reaction conditions. A representative example is shown.

Arthritis was induced by injecting Q and lme into the tail base of 30 male Lewis rats (body weight 195-245F). The grinding volume was measured on the 15th day after inoculation, and the rats were divided into three groups (10 rats per group) so that the volumes of the right hind paws were comparable. 100 mg of a gel ointment containing the test compound prepared according to the method in Example 4 below was applied to the right hind paw of the rat once a day from day 15 to day 22 after inoculation. What is the volume of the rat's right hind paw in l? , [18th day after, 20th day after,
Measurements were taken on the 22nd, 25th, 27th and 29th days. In addition, as a control, a similar test was conducted except that no gel ointment was applied. The results of these tests are shown in FIG.

farnesylate) and the control compounds prednisolone 2l-(2Z, 6Z-farnesylate) and prednisolone 2l-(2Z, 6E-farnesylate), human lymphocytes blasted with phytohemagglutinin absorbed thymidine. The effect of suppressing the uptake effect was evaluated. That is, lymphocytes separated from human peripheral blood were placed in a RPMI-1640 culture medium containing fetal bovine serum at a concentration of 3.3 x 10 volumes.
They were suspended at a concentration of 5 pieces/d. Add 20 µ of an ethanol solution of the test compound at the specified i degree to the obtained mixture and incubate for 30 minutes at a temperature of 37°C in air containing carbon dioxide at a concentration of 5 volumes. I did it.

, 6E-farnelate), there is a clear exception from the results of the pharmacological tests described above, and the geometric isomers of prednisolone 21-farnelate other than the control compound prednisolone and prednisolone 2l-(2E,6E-farnesileto) It has been shown to have a significantly superior anti-inflammatory effect compared to prednisolone, and the side effects seen with prednisolone have been reduced. Also, prednisolone 2l-(2E
, 6g-7 alnesylate) effectively exhibits the excellent anti-inflammatory action of prednisolone 2l-(2E, 6F2-7 alnesylate).
